The Magas1 Gene is Involved in Pathogenesis by Affecting Penetration in Metarhizium acridum

  • Appressorium is a specialized infection structure of filamentous pathogenic fungi and plays an important role in establishing a pathogenic relationship with the host. The Egh16/Egh16H family members are involved in appressorium formation and pathogenesis in pathogenic filamentous fungi. In this study, a homolog of Egh16H, Magas1, was identified from an entomopathogenic fungus, Metarhizium acridum. The Magas1 protein shared a number of conserved motifs with other Egh16/Egh16H family members and specifically expressed during the appressorium development period. Magas1-EGFP fusion expression showed that Magas1 protein was not localized inside the cell. Deletion of the Magas1 gene had no impact on vegetative growth, conidiation and appressorium formation, but resulted in a decreased mortality of host insect when topically inoculated. However, the mortality was not significant between the Magas1 deletion mutant and wild-type treatment when the cuticle was bypassed by injecting conidia directly into the hemocoel. Our results suggested that Magas1 may influence virulence by affecting the penetration of the insects' cuticle.

Analysis of roughness of wave hair formed by thermal perm (열 펌으로 형성된 웨이브 모발의 거칠기 분석)

  • Appearance management through hair beauty forms the basis of the beauty industry, while permanent waves using heat are often used in hair salons, but hair damage due to thermal permanent wave treatment is an inevitable reality. Therefore, this study was conducted for the purpose of presenting an efficient method for thermal permanent wave that can further increase hair wave formation ability and minimize customer's hair damage. After collecting virgin hair from the occipital region, thermal rod pretreatment and thermal permanent wave treatment were performed, and hair roughness analysis and 3D-image were studied using an Atomic Force Microscope. As a result of the study, both the average roughness (Ra) and the ten point average roughness (Rz) were calculated as 223 nm and 853 nm for 4 sections, respectively, showing the highest values. Although the number of samples of the experimental data is limited, the wave forming power can be further increased through this study, and it is expected that it will be practically possible to propose an objective method for thermal permanent wave that can minimize hair damage as well as protect the cuticle of the customer's hair.Judge.

Ultrastructure of the Integument of Capillaria hepatica (syn. Calodium hepatica) (간모세선충(Capillaria hepatica) 표피의 미세구조)

  • Capillaria hepatica is a parasitic nematode which causes hepatic capillariasis in rodents and other mammals, including man. Rat species of the genus Rattus are main primary host and rates of genus Rattus of up to 100% have been reported. Infection to reservoir and other mammalian hosts occur incidentally due to ingestion of water or food contaminated with C. hepatica embryonated eggs. The worms mature exclusively inside the liver, but they die and disassemble soon after egg spawning in rats. Dead worms and their eggs cause immune response of focal necrosis and inflammation within the liver. C. hepatica adult with a thin and long body is similar to capillary. The members of Order Trichurida are characterized by having a stichosome and the bacillary bands in front of the body. As already mentioned, the adult C. hepatica residesin the liver, where it deposits groups of eggs, and finally die in the encapsulated tissue of the liver. They produce eggs that elicit a marked granulomatous reaction that eventually destroy the worms. And the adult worms were mixed with eggs. So the complete isolation of the worm and observation of intact ultrastructure is very difficult. In this study, integument structure of C. hepatica isolated from the liver of mouse at 7 weeks after inoculation of embryonated eggs were observed with scanning and transmission electron microscopy. As a results, body length of isolated C. hepatica was about 99 mm. Cuticle, bacillary band and bacillary pore were obtained in the integument of worm. Bacillary pore across cuticular surface of the worm were observed. According to the existence of cap material, external forms of bacillary pore can be divided into three types such as flat, ingression, and ingression with the cap material type. The complete isolation of the worm and observation of ultrastructure of integument will provide the fundamental data which is important in the nematode research including C. hepatica.

  • As a result of examining the specimens collected from various freshwater bodies since 1995, four eutardigrades are added to Korean fauna: lsohypsibius brevitubulatus n. sp., I. baldii (Ramazzotti), I. marcellinoi (Binda and Pilato), and Diphascon (Diphascon) higginsi Binda. I. brevitubulatus n. sp. differs from its congeners by having a combination of the following characteristics: smooth cuticle, rather big body size, short and broad buccal tube, presence of lunules, and pharynx bearing 3 rod-like macroplacoids without microplacoid. All the species are illustrated and briefly commented on their habitats with the associated fauna.

Pre-Penetration Behavior of Botryosphaeria dothidea on Apple Fruits

  • Pre-penetration behavior of Boytryosphaeria dothidea on apple fruits was investigated with scanning electron microscopy. Once conidia were deposited on the fruit surface, they germainted and produced germ tubes from one or both ends of the conidia. Germ tubes grew over the fruit surface and entered the fruits through lenticels or surface cracks formed naturally. Germ tubes of the fungus also appeared to penetrated the fruits directly with or without forming appressoria. Globose appressoria were frequently formed at the tip of germ tubes on the fruit surface, where no lenticels or surface cracks were found. The conidia collapsed and became flattened to the fruit surface after appressorial formation. Cuticles of fruit surface underneath appressoria and tips of some germ tubes were evidently altered, indicating possibility of direct penetration of the fungus by enzymatic degradation of the cuticle layers. This the first report on the formation of appressoria by B. dothidea.

Effects of the Insect Growth Regulator Dimilin on Larval Development of Hemigrapsus sanguineus (Crustacea, Brachyura) Reared in the Laboratory (무늬발게 幼生에 對한 Dimilin 의 致死 影響)

  • The effects of insect growth regulator Dimilin which interfere with the synthesis of chitin in the cuticle of insect larvae were investigated at various concentrations using the crab larvae of Hemigrapsus sanguineus. The larvae were cultured at control, 0.5, 1, 5 and 10 $\mu g$ Dimilin solutions and three replicate experiments were carried out to give correct analysis. Significant differences in percent mortality have occurred between control and 10$\mu g$ when the larvae were exposed to Dimilin whereas no differences were found between 5 and 10$\mu g$ Dimilin concentrations. If lethal concentration is defined as concentration at which less than 10 percent of crab larvae reach to the last zoeal stage from hatching it can be concluded that insect growth regulator Dimilin is lethal to the larvae of Hemigrapsus sanguineus at 5 and 10$\mu g$ Dimilin.

  • Lipophilic glandular trichomes form secretory vesicles that accumulate in a distended noncellular secretory cavity . Imidazole-buffered osmium tetroxide solution was used to visualize unsaturated lipids in glands of Cannabis sativa. This method of staining revealed two kinds of secretory vesicles in the cavity of glands. Some smaller and rounded vesicles in the secretory cavity and secretory cells were positively stained with the imidazole, and they appeared electron-dense. The other type of vesicles which have bigger sizes more or less were not reacted, however, they appeared transparent. A high contrast of the cuticles which cover the gland was also strongly reacted with that processing. Those result suggest that the dark vesicles in the cavity may contribute to enlarge the subcuticular wall and cuticle when contents of these vesicles are dispersed into wall.

Redescription of Free-Living Marine Nematode, Dracograllus filipjevi Allen and Noffsinger, 1978 (Nematoda: Draconematidae) from Korea

  • A draconematid nematode species, Dracograllus filipjevi Allen and Noffsinger, 1978, is described from the washings of shallow littoral coralline algae at Guryongpo in the eastern coast of Korea. This species is recognized by the following characteristics: larger body size, number of posterior sublateral adhesion tubes (9 in male and 12-13 in female) and subventral adhesion tubes (8-9 in male and 9-10 in female), the absence of cephalic acanthiform setae on rostrum, the presence of longitudinally areolated body cuticle with dot-like punctations, the presence of some cuticular collar in swollen pharyngeal region, the shape of amphidial fovea (large, elongate, and loop-shaped in both sexes), the absence of preanal corniform setae, and the absence of lateral differentiation on narrow body region. Morphological features of the species using differential interference contrast photomicrographs are described in detail in the present work. The is the first report of D. filipjevi in Korea.
