Operational Strategy for Increasing Ethanol Production in Repeated Fed-batch Ethanol Fermentation Using Saccharomyces cerevisiae (Saccharomyces cerevisiae 를 이용한 반복 유가식 ethanol 발효에서 ethanol 생산량을 증가를 위한 운전 전략)

  • We designed the optimal operational strategy in repeated fed-batch ethanol fermentation using Sacchromyces cerevisiae ATCC 24858 in views of ethanol yield, specific ethanol production rate, and ethanol productivity, when the aeration rate were controlled at 0.0 and 0.33 vvm. Coincidentally, the time intervals of withdrawal-fill of culture medium (24 and 36 h) were investigated. Ethanol yield and ethanol productivity when the aeration was carried out at 0.33 vvm were superior to those when the aeration was not carried out. Additionally, those parameters when the time interval of withdrawal-fill of culture medium was 24 h were superior to those when time interval of withdrawal-fill of culture medium was 36 h. The total ethanol production reached at the greatest value, 703.8 g-ethanol, when the aeration was carried out at 0.33 vvm and the time interval of withdrawal-fill of culture medium was 24 h. In this study, we verified experimentally the necessity of designing the operational strategy for increasing ethanol production in terms of aeration rate and time interval of withdrawal-fill of culture medium in the repeated fed-batch ethanol fermentation.

  • Methods of lichen-derived culture were spore- and thallus-derived cultures. Spore-derived culture was modified and thallus-derived culture was established for our two decades. We maintained about 500 lichen-derived cultures by using both methods. In recent several years, we have investigated to screen various pharmacological activities in lichen-derived cultures for the first time and found some activities in our cultures.

  • Relationship of soil properties and seasonal variation on microbilogical population to-continuous culture and first-time culture of ginseng was investigated by bimonthly from May 1976 to January 1977. pH and P contents of 2 years continuous culture of soil were higher than other culture plot of soil, and contraty to the above, 2 years first-time culture of ginseng soil was conplot of soil, and contraty to the above, 2 years first-time culture of ginseng soil was contained more potassium contents than other culture plot of soil. In microbiological fluctuation with seasonr in various soil conditions, the population, trends of Fusarium spp., Erwiniaspp., and flourescent Psedudomonas spp. were increased in May and July in general, but decreased in the other month. It was observed that in all type of soil, Fusarium spp. was distributed in abundance in and on rihizosphere, and decreased the propagules numbers as soil depth increase. The numbers of Erwinia spp. and fluorescent Pseudo-monas spp. were distributed greater in numbers on the surface zone of soil depth decreasing the numbers along the soil layer increase, and also in 2years continuous culture of soil especially, a great numbers of Erwinia spp. and fluorescent Pseudomonas were evenly distributed in surface zone and rhizosphere. Ginseng disease with a high incidence of bacterial disease in continuous culture of 2 and 4 years was seemed to be associated with soil bacteria that was high in numbers of Erwinia spp. and fluorescent Pseudomonas spp. in May and July.

  • Consumer behavior is driven by culture; and culture is contextual. Therefore, human behaviors such as those exhibited in consumption behavior should not be measured and compared cross-culturally by using cultural specific mea-sures or paradigms which assume a universal reality, time and context free. Since it is known that consumption behavior is influenced by culture, and cultures in the United States differ from those in Korea, the assumption of universal ‘truths’ which can be known is inappropriate. To employ a paradigm with invalid assumptions automatically leads to the lack of validity, a must for truth claims in the positivist paradigm. Thus, 'truths' in the research reported must be suspect.

  • Information culture is a concept with multiple and sometimes conflicting definitions. Nevertheless at a time when social and cognitive aspects of information and communication systems are recognized as being of similar significance as the technological functionality, the concept of information culture is demonstrating considerable value. This paper explains the concept of information culture and considers its value from two main perspectives: firstly as providing an analytical tool, and secondly as a means of providing practitioners with insight and sensitivity to the organisational contexts of their workplaces.

Yeast Cell Cultivation of Produce Active Dry Yeast with Improved Viability (생존능이 증진된 활성 건조효모 생산을 위한 효모세포배양)

  • Optimum conditions for vacuum-drying ad cultivation of yeast cells for the production of active dry yeast were examined. At lower temperature, more drying time was required to dry the yeast pellet to reach the desirable water content(8%). Optimum temperature of vaccum oven and time for drying was 63$^{\circ}C$ and 90 min, respectively. Optimum medium composition for flask culture using cane molasses as the substrate were 0.25% sugar, 0.013% $K_2$HPO$_4$, 0.1% $K_2$HPO$_4$. and 0.125% (NH$_4$)$_2$SO$_4$. Culture temperature $25^{\circ}C$ gave the highest survival rate of dired yeast. After finishing fed-batch culture and the culture was left in the fermentor without adding any sugar or nutrient, survival of the dried yeast harvested from the fermentor increased to 86.0% after 36 hr. It was also observed that the yeast cells with higher budding rates showed lower survival rate.

  • Reflecting a particular place and time, from the agrarian society, through the Industrial Age, furniture has been one of the closest factors to our housing life and developed continuously. The information age representing digital, information and telecommunication generally gave a wide range of individuals and societies various phases of culture shock that had never been experienced so far, among which the change of the attitude toward work and marriage influenced on housing life style, they have required unexplored style and function of furniture additionally. People so called the singles has recently become a culture code reflecting the present time, they already have built their own territories in society, economy and culture. The ideas about a variety of furniture for the singles, which consider the housing culture of the singles, has been floated, therefore furniture for the singles are main streamed into home furniture category, required characteristics of furniture for the singles are drawn, furthermore some design examples are suggested on the basis of those characteristics in this study.

Optimization of submerged culture conditions for roridin E production from the poisonous mushroom Podostroma cornu-damae

  • Roridin E, produced by Podostroma cornu-damae, is a mycotoxin with anticancer activity. To increase the content of roridin E, submerged culture conditions were optimized using response surface methodology. Three factors, namely, medium initial pH, incubation time and agitation speed were optimized using a Box-Behnken design. The optimum submerged culture conditions to increase the content of roridin E included a medium with an initial pH of 4.0, an incubation time of 12.90 days, and an agitation speed of 63.03 rpm. The roridin E content in the submerged culture, under the aforementioned conditions, was 40.26 mg/L. The findings of this study can help lower the current price of roridin E and promote its related research.

Background to the Formation of the Term Hyangjang (香粧) and Change in Cosmetic (化粧) Culture -Focusing on Change from Visual Make-up to Olfactory Make-up- (향장(香粧) 용어의 성립배경과 화장(化粧)문화의 변화 -시각적 화장에서 후각적 화장으로의 변화를 중심으로-)

  • Modernization drastically changed the cosmetic culture of Korea and Japan. A classic case that shows this is the appearance of the term 'Hyangjang (香粧)'. This paper investigated the background to the formation of the term Hyangjang (香粧), and reviewed the aspects of cosmetic culture that changed with the emergence of Hyangjangpum (香粧品), or cosmetics containing perfume. The investigation revealed that the term Hyangjang (香粧) appeared for the first time in Hirano (1899), a literature published in the Meiji period in Japan, and that the new term Hyangjang (香粧) had been formed against the background of advanced Western synthetic perfume and played an important role in contemporary techniques for the manufacture of cosmetics. The term Hyangjang (香粧) and cosmetics containing perfume, or Hyangjangpum (香粧品) were then introduced from Japan to Korea. In Korea, the term 'Hyangjang (香粧)' appeared for the first time in an advertising copy written by Hyeon Hui-un, a pioneer of the Korean modern theater movement during the period of Japanese colonial rule. At that time, cosmetic companies in Korea and Japan were releasing cosmetic products that contained perfume that stimulated a women's desire to purchase them by emphasizing 'fragrance' in their advertising. Existing public tendencies to regard a fragrant smell from a made-up face as vulgar were changing and the public perception of fragrance were also changing. The appearance of Hyangjangpum (香粧品) indicates that the existing cosmetic culture revolving around visual sense changed into a complex cosmetic culture involving olfaction. This change in culture is significant in that it heralded the direction of future development towards cosmetic culture that uses increase combinations of different senses including touch, taste, and hearing.

Relationship between the time to positivity of blood culture and mortality according to the site of infection in sepsis

  • Objective: The time to positivity (TTP) of blood culture reflects bacterial load and has been reported to be associated with outcome in bloodstream infections. This study was performed to evaluate the relationship between the TTP of blood culture and the mortality rates associated with sepsis and septic shock according to the site of infection. Methods: We performed a retrospective cohort study on patients with sepsis and septic shock. The rates of blood culture positivity and mortality as well as the relationship between the TTP and 28-day mortality rate were compared among patients with different sites of infection, such as the lungs, abdomen, urogenital tract, and other sites. Results: A total of 2,668 patients were included, and the overall mortality rate was 21.6%. The rates of blood culture positivity and mortality were different among the different infection sites. There was no relationship between the TTP and mortality rates of total, lung, and urogenital infections. Patients with abdominal infections showed a negative correlation between the TTP and 28-day mortality rate. In patients with abdominal infections, a TTP<20 hours was independently associated with 28-day mortality compared with patients with negative blood culture (hazard ratio, 1.73; 95% confidence interval, 1.16-2.58). However, there was no difference in mortality rates of patients with a $TTP{\geq}20$ hours and a negative blood culture. Conclusion: The shorter TTP in patients with abdominal infections in sepsis and septic shock was associated with a higher 28-day mortality rate.
