갈항미생물이 콩나물 부패균에 미치는 향균능력과 콩나물 생육 특성 (Study on the Antagonism of Useful Microbes against Soybean Sprout Rotting Pathogens and Their Effect on the Growth of Soybean Sprouts)

  • 유용미생물을 이용한 콩나물 부패 원인균에 대한 길항작용과 콩나물 생육에 미치는 영향에 대한 연구의 일환으로 Pseudomonas areofacience 14H-3, Pseudomouas fluorescens Rl-12, Bacillus cereus Yell균을 이용하여 콩나물 부패 원인균에 대하여 길항능력과 콩나물 생육에 미치는 영향에 대하여 조사한 결과는 다음과 같다. P. areofacience 14H-3균주와 B. cereus Yell 균주는 Rizoctonia solani에 높은 길항력을 보였으며, P. areofacience 14H-3과 B. cereus Yell은 세균병에 특히 높은 항균력을 나타내었다. 길항세균의 배양여액을 PDA에 첨가하여 콩나물 부패균의 증식 억제 효과를 조사한 결과 P. areofacience 14H-3과 P. fluorescens Rl-12은 78.8% 억제율을 보였으며 B. cereus. Yell은 52.9%의 억제율을 보였다. 길항미생물과 배양여액을 처리하여 콩나물을 재배한 결과 B. cereus Yell 처리구가 대조구에 비해 콩나물의 총무게가 증가하는 것으로 나타났다. 길항세균 배양여액에 콩을 침지하여 재배한 결과 200배액 처리구가 대조구에 비해 생육이 양호하였다. 콩나물의 일반성분을 분석한 결과 수분, 회분, 총당에서는 유의적인 차이가 나타나지 않았으나, 조단백의 경우 B. cereus Yell(8.9%)가 control(3.6%)보다 약 2배 정도 증가하였으며, 조지방과 조섬유의 경우는 P. areofacience 14H-3, P. fluorescens Rl-12가 control보다 각각 2배 정도 함량이 증가하였다. 비타민의 경우도 대조구(9.9mg%)에 비하여 길항미생물 처리구(9.4∼10.8mg%)가 높게 나타났다.

돼지 난포난자의 체외성숙에 있어서 Catalase와 Xanthine의 영향 (Effect of Catalase and/or Xanthine on In Vitro Maturation of Porcine Follicular Oocytes)

  • 본 연구는 미성숙 돼지 난포 난자의 체외성숙에 있어서 catalase (0.1 mg/$m\ell$)와 xanthine (5 mM)의 역할에 대하여 검토하였다. 그 결과, 체외에서 성숙배양 48시간 후 metaphase-II 단계로 발육한 난자의 비율은 xanthine (54%) 첨가 보다는 대조구 (72%), catalase (73%) 및 catalase+xanthine (70%) 첨가구에서 유의적으로 높은 성숙율을 나타냈다 (P<0.05). 한편, 체외에서 30시간 동안 성숙배양한 경우 모든 실험구에서 성숙율의 유의적인 차이는 인정되지 않았으나, 성숙배양 36, 42 및 48시간 후 xanthine의 첨가 여부에 관계없이 catalase 무첨가 (29~50%) 보다는 첨가시 (49~70%)에 유의적으로 높은 성숙율을 나타냈다 (P<0.05). 체외에서 xanthine의 첨가 또는 무첨가시 배양시간의 연장이 난자의 성숙에 미치는 영향을 검토한 결과 배양 72시간에서 높은 성숙율을 나타냈으며, 퇴행난자의 비율은 배양 120시간에서 catalase무첨가(47%)에 비하여 첨가시 (28%) 유의적으로 낮게 나타났으나 xanthine이 첨가된 배양액내에서 catalase첨가유무에 의한 차이는 인정되지 않았다. 또한 xanthine을 첨가하여 72시간 배양한 경우 단위발생란이 처음으로 관찰되었지만 catalase의 첨가 유무에 의한 차이는 인정되지 않았지만 배양시간이 길어짐에 따라 발생비율이 증가하였다. 이와 같은 결과에서 돼지 난포난자는 catalase와 xanthine을 첨가한 배양액내에서 배양 72시간까지 성숙율이 증가할 수 있으며, 배양기간의 연장시 catalase에 의하여 난자의 퇴행을 억제하며 단위발생란의 증가를 가져오는 것으로 생각된다.

  • ${\gamma}-linolenic$ acid를 함유한 유지를 생산하기 위해 포도당을 주기적으로 첨가하면서 Mucor sp. KCTC 8405P를 유가배양하였다. 배양 시작 후 2일까지 효모 모양으로 배양한 후 배지의 pH를 높여 균사체 모양으로 유도하여 배양하면, 지방질은 포도당이 고갈되는 배양 7일째까지 축적되었다. 본 배양조건에 의하여 건조 균체량 99.3 g/l, 총지방질 38.0 g/l와 3.5 g/l의 ${\gamma}-linolenic$ acid를 얻을 수가 있었다.

  • The color difference value of fabrics dyed with browning extracts from mushroom were as follows. 1. Two hours of dyeing depending on pH at 95℃ exhibited colors of YR and Y. The color of wool and nylon was darkest brown at pH 4, but light brown or yellow as pH increased. The color of silk was darkest brown at pH 5 and pH changes resulted in the same tendency as in wool. 2. In wool the color was changed to darker brown as time prolonged, however, there was no further color change after 9hr and value and chroma of wool declined. 3. At pH 4, the increase in temperature turned colors of sil, wool and nylon into dark brown from light yellow. 4. As the result of repetitive dyeing, colors of silk, wool and nylon gradually turned into darker brown. The repetitive dyeing was more effective in dark color and high colorfastness compared to a prolonged dyeing. 5. The amount of mordant resulted in color changes between light yellow and dark brown without various color changed. 6. In color change according to methods of mordant, post-mordant with CuSO₄·5H₂O and with SnCl·2H₂O showed colors of green and light yellow, respectively.

토마토 주스의 미생물 발효 산도 특성에 관한 연구 (A Study on the pH Characterization for Microbial Fermentation in Tomato Juice)

  • This study was conducted to know the behavior of pH behavior in the tomato juices to find out an effective medium for microbial cultivation. Bacterial culture media is a material consist a mixture of nutrients used to grow microorganisms on or in it. In addition, microbial culture media can also be used for isolation, propagation, testing the nature physiological, and calculation of the number of microorganisms. Fresh tomato juice is used for basic ingredient, therein added salt, sugar and EM (Effective Microbial). The fermented solution placed in a room with a temperature of 40oC. Data retrieval before the pH value reached a constant value is done every 12 hours, after constant rate data collection was done every 24 hours. The pH value has been steady after 372 hours of fermentation process (15.5 days). From the results obtained that the amount of additional ingredient which added into tomato juice does not affect final pH value of solution. Thereby the most effective treatment for microbial cultivation media is treatment number four.

평판형 막 생물 반응기를 이용한 구연산의 생산 (Production of Citric Acid in a Flat-type Membrane Bioreactor)

  • 호기성 미생물 고정화를 위해 새로 개발한 평판형막 생물 반응기(FMBR)을 개발하여Aspergillus niger (KCTC 1232)로부터의 구연산 생산을 연구하였다. 반응기는 삼단으로 구성되어 있으며 상층부에는 산소 공급을 위한 공기흐름이 있고 주층부에는 미생물이 고정화되어 있으며 하층부에는 배지가 연속적으로 공급된다. 배지의 초기 pH는 구연산 생성에 있어서 중요한 영향을 미치며 pH가 낮을수록 구연산 생산수율이 높았다. 산소원으로 공기와 순수산소를 이용하여 생산성은 각각0.20, 0.40h/Lh에 달하였다. EH한 배지의 공급속도가 높을수록 생산성이 어느 정도 증가함을 알 수 있었다.

Improvement of Unsaturated Fatty Acid Production from Porphyridium cruentum Using a Two-Phase Culture System in a Photobioreactor with Light-Emitting Diodes (LEDs)

  • In this study, the culture conditions for Porphyridium cruentum were optimized to obtain the maximum biomass and lipid productions. The eicosapentaenoic acid content was increased by pH optimization. P. cruentum was cultured with modified F/2 medium in 14-L photobioreactors using a two-phase culture system, in which the green (520 nm) and red (625 nm) light-emitting diodes (LEDs) were used during the first and second phases for biomass production and lipid production, respectively. Various parameters, including aeration rate, light intensity, photoperiod, and pH were optimized. The maximum biomass concentration of 0.91 g dcw/l was obtained with an aeration rate of 0.75 vvm, a light intensity of 300 μmol m-2s-1, and a photoperiod of 24:0 h. The maximum lipid production of 51.8% (w/w) was obtained with a light intensity of 400 μmol m-2s-1 and a photoperiod of 18:6 h. Additionally, the eicosapentaenoic acid and unsaturated fatty acid contents reached 30.6% to 56.2% at pH 6.0.

Optimal Conditions for Chitinase Production by Serratia marcescens

  • A chitinase-producing bacterium was isolated from seashore mud around Beobseongpo in Chunmam province through the use of a selective enrichment culture. The best chitinase producing strain was isolated and identified as Serratia marcescens KY from its characteristics. For effective production of chitinase, optimum pH, temperature, and agitation speed were investigated in flask cultures. The optimum pH using Serratia marcescens KY was between pH 6 and 7 and the chitinase produced was 37.9 unit/mL. On the other hand, the optimal pH of the Serratia marcescens ATCC 27117 was 7.5, and the produced amount of chitinase was 35.2 unit/mL. The optimal temperature for chitinase production for Serratia marcescens KY and Serratia marcescens ATCC 27117 was $30^{\circ}$. The cell growth pattern at different temperature was almost identical to the chitinase production. To investigate the optimal shaking speed under optimal culture, speeds were varied in the range of 0∼300 rpm. The maximum production of chitinase was carried at 200 rpm although the cell growth was the highest at 150 rpm. It indicates that oxygen adjustment is required for the maximum chitinase production. Using optimal conditions, batch cultures for comparing Serratia marcescens KY and Serratia marcescens ATCC 27117 were carried out in a 5 L fermentor. The oxygen consumption was increased with the increase of culture. Especially, at 120 h of culture Serratia marcescens KY and Serratia marcescens ATCC 27117 produced 38.3 unit/mL, and 33.5 unit/mL, respectively.

Investigation of the Microbiological and Biochemical Properties of Kimchi in the Submerged Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of lactic acid bacteria (LAB) from kimchi as a starter culture in the production of fermented sausages. To achieve this, a submerged model medium that contained LAB as part of a complex system of kimchi (0.5, 1.0, 1.0, 3.0, and 5.0%) and lyophilized kimchi powder (0.2 and 0.5%) was fermented for 120 h. During the fermentation period, the growth of total viable organisms and LAB, and the changes in the pH and the titratable acidity, were investigated. The initial LAB counts ranged from 6.4 to 7.7 Log CFU/mL for the kimchi media, and from 6.9 to 6.9 Log CFU/mL for the kimchi powder media. In all the kimchi batches, the LAB increased logarithmically, and the highest LAB counts (around 9 Log CFU/mL) were reached in 24 h. An evident lag phase of the LAB was observed in the kimchi powder samples and reached 8.8 Log CFU/mL in 8 h. The decrease in the pH and the formation of lactic acid were rapid in the kimchi batches, and reached pH values of 3.4-3.5 in 12 h. With these results, the LAB that was integrated with the addition of kimchi or kimchi powder demonstrated its potential utility as a substitute for starter culture.

Paenibacillus woosongensis의 만난분해 효소활성 (Mannanolytic Enzyme Activity of Paenibacillus woosongensis)

  • 만난의 분해활성을 갖는 Paenibacillus woosongensis의 배양 상등액으로부터 mannanase, ${\beta}$-mannosidase와 ${\alpha}$-galactosidase 활성이 관찰되었다. 배양상등액 내의 locust bean gum를 분해하는 mannanase는 pH 5.5와 $60^{\circ}C$, para-nitrophenyl-${\beta}$-D-mannopyranoside를 분해하는 ${\beta}$-mannosidase는 pH 6.5, $50^{\circ}C$, para-nitrophenyl-${\alpha}$-D-galactopyranoside를 분해하는 ${\alpha}$-galactosidase는 pH 6.0-6.5와 $50^{\circ}C$에서 각각 최대활성을 보였다. 배양상등액의 만난 분해효소는 mannotriose, mannotetraose, mannopentaose, mannohexaose와 같은 만노올리고당을 분해할 뿐 아니라 mannobiose도 분해하였다. 또한 melibiose, raffinose, stachyose 등의 ${\alpha}$-1,6 결합형 galacto-oligosaccharides를 분해하여 galactose를 생성하였다. 이러한 결과로 보아 P.woosongensis는 galactomannan을 완전히 분해하는데 필요한 3종류 효소를 모두 생산하는 것으로 판단된다.
