Autocrine or paracrine mediators released by the early embryo are implicated in the support of embryonic development. Their mechanisms and optimal embryo density in the medium, however, are uncertain. This study was conducted to establish the optimal embryo density and culture medium volume in mouse parthenogenetic embryo culture. In experiment 1, culture of parthenogenetirally activated oocytes at a concentration of $2{\~}4$ embryos/${\mu}L$ significantly improved development to the blastoryst stage ($72{\%}{\leq}$) compared with culture at the lower ($0.2{\~}1$e mbryos/${\mu}L,\;0\~37.5\%$) and the higher ($5{\~}6$ embryos/${\mu}L,\;30\~53\%$) concentration for 120 h when the oocytes were cultured in a 5 ${\mu}L$ drop under mineral oil In experiment 2, the embryos cultured at a concentration of $2{\~}4$ embryos/${\mu}L$ in a 10 ${\mu}L$ drop ($81.1{\%}$) showed significantly higher blastocyst rates than those in a 5 ${\mu}L$ drop ($68.5{\%}$). This study optimizes in vitro culture condition by modifying embryo density and the volume of culture medium It may give appropriate level of autocrine and/or paracrine factors to enhance viability and subsequent normal development of mouse parthenogenetic embryos in vitro.
The possibility of application of membrane type immobilized adsorbent to the fed-batch or perfusion culture system with anchorage-independent cells as well as batch system was investigated. The improvement in cell density and cell viability due to the combination of immobilized adsorbent with each culture system was evaluated for the investigation, and the optimum culture system employing immobilized adsorbent system was suggested based on the results. It was observed that the system with immobilized adsorbent showed better cell growth and cell viability than that without immobilized adsorbent in every operation system of batch, fed-batch, and perfusion. In case of batch system, 200% improvement of maximum cell density was observed in the system where ammonium chloride was added on purpose. And 50% improvement of maximum cell density was observed in the fed-batch system where ammonium ion accumulates significantly, while small increase in maximum cell density was observed in the perfusion system where dilution of waste byproducts exists. Especially, the fed-batch system showed the most significant improvement on cell growth because both compensation of nutrient and removal of ammonium ion occurred simultaneously in the system. Therefore a combined system of immobilized adsorbent and fed-batch operation could be suggested as an optimum system with in situ removal of ammonium ion.

Previous research indicated that free-living sporangial filament keep hollow morph under high-culture density and form bipartite cells under low-culture density, while the following conchospore release was inhibited by high light. Here, we further explored the molecular bases of these affects caused by light and culture density using a transcriptome analysis. Many differentially expressed genes (DEGs) related to carbon dioxide concentration and fixation, photosynthesis, chlorophyll synthesis and nitrogen absorption were upregulated under high-light conditions compared with low-light conditions, indicating the molecular basis of rapid vegetative growth under the former. The stress response- and ion transport-related DEGs, as well as the gene encoding the vacuole formation-brefeldin A-inhibited guanine nucleotide exchange protein (BIG, py05721), were highly expressed under high-density conditions, indicating the molecular basis of the hollow morph of free-living sporangial filaments under high-culture density conditions. Additionally, the brefeldin A treatment indicated that the hollow morph was directly influenced by vacuole formation-related vesicle traffic. Others DEGs related to cell wall components, zinc-finger proteins, ASPO1527, cell cycle and cytoskeleton were highly expressed in the low density with low-light group, which might be related to the formation and release of conchospores. These results provide a deeper understanding of sporangial filaments in Neopyropia yezoensis and related species.
Copepod is useful live food organism in the larval rearing. However of fin fish, culcures of copepod species (Specially calanoida and cyclopoida) were very difficult under artificial management conditions. In this study, we report successful culture of a free swimming cyclopodia copepod species (Apocyclops sp.) by easily baker's yeast as food. The maximum culture density of Apocyclops sp. was observed 11,200±2,660 ind/ℓ during the 30 days culture periods in the 1.5ℓ culture tanks. The nauplii were growth up maximum density 6,400±432 ind/ℓ on day 16th. The number of egg carring female was observed maximum density (666.7 ind./ℓ) of two times after 6days and 28 days of initial culture.
It have been investigated how algal and bacterial symbiotic reaction influences on removal of organic carbon in river ecosystem. And artificial experimentation apparatus was made for algae'and bacteia'culture as lab scale. Investigating and researching minutely the change of concentration of organic carbon substrate and the change of population density of algae'and of bacteria'with this artificial experimentation apparatus, the next results could be obtained. 1. Successful decrease of DOC(dissolved organic carbon) could not be expected unless algal and bacterial biomass floe was nut formed effectively and unless biosorption was not proceeded effectively in the very culture system in which artificial synthetic wastewater was supplied continuously at constant rate. 2. In conditions of culture liquid of 1335 glucnse mg/L(type 1) and of 267 glucose mg:L(type 2), the algal dominant species was always Chlorella vulgaris in both types in which artificial synthetic wastewater were supplied continuously at constant rate and algae population density was around maximum 107 cells/mL. 3. It was around 108 ~ 107 cells/mL that the population density of heterotrophic bacterium. In culture medium systems type 1 and type 2 in which artificial wastewater were supplied continuously at constant rate, the same density appeared initially when using the population density of Escherichia coli w 3110 as indirect indicator. And this density decreased rapidly till the culturing date 35 days were passed away, while this density increased with gentle slope after same date and then the trend of change at type 2 was more severe than one at type 1. 4. When seeing such a change of population density of Escherichia coli w 3110, the growth of heterotrophic bacterium appeared as survival instinct pattern of broader requirement of nutrient at condition of low concentration of organic carbon substrate than condition of high concentration of same substrate.
This paper introduces to a simple culture method and growth of axenic (bacteria-free) rotifer Brachionus rotundiformis for seed stock of rotifer mass culture. This rotifer axenic culture method is based on the washing and transferring with sterilized sea water and modified antibiotic mixture AM9 solution. Population growth (final density on day 16) of axenic cultured rotifer were maintained with a high density and stable growth compared with the control of non-axenic culture (general culture style) through the 3 times-rerunning experiments (trial 1, 2 and 3). But the egg carrying rates of amictic females were not different between the axenic-and non-axenic culture condition. Although, rotifer density was higher in axenic culture, the food (Nannochloropsis oculata) was still remained unutilized than that of the non-axenic culture in third trial culture. These results suggest the possible existence of harmful bacteria for the rotifer population growth in the trial 1 and 3 of non-axenic culture compared to the trial 2. This axenic rotifer culture method is valuable for seed stock of the stable rotifer mass cultures.
Low efficiency in microalgal biomass production was largely attributed to the low density of algal cell cultures. Though mutations that reduced the level of chlorophyll or pigment content increased efficiency of photon usage and thus the cell-culture density under high-illumination growth conditions (e.g., >$500{\mu}mol\;photon\;m^{-2}\;s^{-1}$), it was unclear whether algae could increase cell-culture density under low-illumination conditions (e.g., ${\sim}50{\mu}mol\;photon\;m^{-2}\;s^{-1}$). To address this question, we performed forward genetic screening in Chlamydomonas reinhardtii. A pool of >1,000 insertional mutants was constructed and subjected to continuous subcultures in shaking flasks under low-illumination conditions. Complexity of restriction fragment length polymorphism (RFLP) pattern in cultures indicated the degree of heterogeneity of mutant populations. We showed that the levels of RFLP complexity decreased when cycles of subculture increased, suggesting that cultures were gradually populated by high cell-culture density (HCD) strains. Analysis of the 3 isolated HCD mutants after 30 cycles of subcultures confirmed that their maximal biomass production was 50-100% higher than that of wild type under low-illumination. Furthermore, levels of chlorophyll content in HCD mutant strains were similar to that of wild type. Inverse polymerase chain reaction analysis identified the locus of insertion in two of three HCD strains. Molecular and transcriptomic analyses suggested that two HCD mutants were a result of the gain-of-function phenotype, both linking to the abnormality of mitochondrial functions. Taken together, our results demonstrate that HCD strains can be obtained through continuous subcultures under low illumination conditions.
This study was conducted to establish the optimal culture conditions for in vitro production of bovine embryos derived from slaughter house ovaries. Cumulus-oocyte- complexes (COCs) collected by aspiration from follicles of 2~7 mm in diameter were matured in Ham's F-10 medium supplemented with 0.01 $\mu\textrm{g}$/m1 epidermal growth factor (EGF) at 39$^{\circ}C$ in a humidified atmosphere of 5% $CO_2$in air. After 24 hrs of culture, the oocytes were co-cultured with epididymal sperm selected off by Percoll-density gradient in TALP medium for 24 hrs. The presumptive zygotes were cultured in HECM-6 medium for 3 d post-insemination, and followed by cultured in TCM199 medium until 7 to 10d post-insemination. The cultures were compared of their cleavage and development into later stage in culture medium by additions of different protein sources (PVA, BSA and BCS) and by different embryo density. The rates of cleavage and development rates into blastocyst were not significantly (P<0.05) different among the culture media containing with BSA (75.0% and 40.5%), BCS (76.7% and 38.0%) and PVA (72.5% and 42.2%), respectively. Significantly (P<0.05) higher blastocysts rates were obtained in culturing of 30 and 40 embryos in each 50$\mu$l droplets of culture medium than in 5, 10 and 20 embryos. These results indicate that the optimal density of embryos is 30~40 embryos in a 50$\mu$l droplet of culture medium. Furthermore there is no effect of different protein sources on early embryonic development.

This study was carried out to investigate the effect of rotifer fed the different diets in high density culture on larval flounder Paralichthys olivaceus. Potifer was enriched with enrichment supplements, Marine ${\alpha}\;and\;{\omega}-yeast$ for 6 hours after being cultured with freshwater Chlorella for 18 hours during high density culture before it was fed to larval flounder. And rotifer was culutured with marine Chlorella and freshwater Chlorella for 24 hours during semi-continuous high density culture before it was fed to larval flounder. Culture tanks(21 working volum) set for rotifer culture in a water bath($28{\circ}C$) were continuously supplied with oxygen gas. The content of n-3 HUFA to fatty acids in rotifer(dry weight ${\%}$) enriched with Marine ${\alpha}$ for 6 hours and cultures with marine Chlorella for 24 hours were higher than that in rotifer enriched with ${\omega}-yeast$ for 6 hours or cultured with freshwater Chlorella for 24 hours. The growth and survival rates of larval flounder fed on rotifer enriched with Marine ${\alpha}$ for 6 hours and cultured with marine Chlorella for 24 hours were higher than those of larval flounder fed on rotifer enriched with ${\omega}-yeast$ for 6 hours or cultured with freshwater Chlorella for 24 hours. And the content of n-3 HUFA of larval flounder fed on rotifer enriched with Marine ${\alpha}$ for 6 hours was higher than that of larval flounder fed on other rotifers, The results from this study indicated that rotifer culture with marine Chiorelia would be suitable for the high density culture and effective diet for the growth of larval flounder.
Optimization of the preimplantation mammalian embryo culture condition was widely focused on refining medium composition under the name of chemically defined media. However, recent research revealed that the alteration of physical environment can be a crucial factor to a successful embryo development. In this study, under the same embryo density, a novel culture device named oil-free micro tube culture (MTC) system was evaluated using porcine parthenogenetic embryos. The activated oocytes were placed into the 0.2 ml thin-wall flat cap PCR tube and cultured to the blastocyst stage. As a preliminary step, embryo density and culture medium volume were optimized under a standard drop culture system. The optimal embryo density range for in vitro culture was 0.5 embryos per ${\mu}l$ in $20\;{\mu}l$ drop (20.5%) and 1.0 embryos per ${\mu}l$ in $10\;{\mu}l$ drop (20.6%). Based on these results, we compared drop culture system and 'MTC' system in terms of the developmental rate to the blastocyst stage. In $20\;{\mu}l$ medium volume, the 'MTC' system showed similar blastocyst formation rate when compared with drop culture system (20.2% versus 20.5%, respectively) while the 'MTC' system showed lower blastocyst formation rate than drop culture system in $10\;{\mu}l$ one (12.7% versus 20.0%, respectively). Therefore the $20\;{\mu}l$ MTC system may be an alternative incubation system for short-distance embryo transport without carrying the $CO_2$ incubator and this provides novel embryo culture device to clinical veterinary embryologists.
