A Technique to Enhance Insecticidal Efficacy Using Bt Cry Toxin Mixture and Eicosanoid Biosynthesis Inhibitor (혼합 비티 독소단백질과 아이코사노이드 생합성 억제자를 이용한 약효 증진 기술)

  • To enhance Bacillus thuringiensis (Bt) efficacy, four Cry toxins were purified from four different Bt strains and assessed in their combined efficacy. The Cry mixtures significantly expanded their target insect spectra. Bacterial culture broth of Xenorhabdus nematophila (Xn) significantly suppressed insect cellular immune response and increased Cry toxicity. The addition of Xn culture broth to Cry mixture significantly enhanced Bt efficacy in target insect spectrum and insecticidal activity.

  • Calcium carbonate $(CaCO_3)$ bead immobilized with alginate were developed as buffer system to enhance the cultivation efficiency of bifidobacteria. When Bifidobacteriuim longum KCTC 3128 and HLC 3742 were independently cultivated in 2.5-liter fermenter buffered the $CaCO_3$ bead, NaOH, $Na_2CO_3$, and $NH_4OH$. The proliferation of bifidobacteria and their storage stability were higher in culture broth buffered $CaCO_3$ beads than in culture broth buffered with NaOH, $Na_2CO_3$, and $NH_4OH$. Therefore, $CaCO_3$ bead may be useful as a buffer to enhance of the cultivation efficiency and viability of bifidobacteria.

  • A bacterium(Y-5) which has potent antibacterial activity against swine atrophic rhinitis bacteria (Bordetella bronchiseptica, Pasteurella multocida) was isolated from soil and identified as a strain of Bacillus sp. Y-5 upon investigation of the morphological and physiological characteristics. The culture broth obtained from incubation of the Y-5 strain at $30^{\circ}C$ for 21 h in tryptose-bouillon agar medium (pH 6.0) showed active antibacterial activity against Bordetella bronchiseptica and the culture broth that of $30^{\circ}C$, pH 6.5, 18 h showed active antibacterial activity against Pasteurella multocida.

  • Several bacterial strains producing biosurfactants were isolated from polluted marine and soil by oil. One of the strains named LSC11 showed strong production activity of biosurfactants. This strain was identified as a Bacillus sp. LSC11 based on the morphological, biochemical, and physiological characteristics. The biosurfactant, produced by the strain, emulsified crude oil, vegetable oil, and hydrocarbons. The surface tension of the culture broth of Bacillus sp. LSC11 decreased to 32 mN/m. The crude biosurfactant was obtained from the culture broth by acid precipitation, freeze drying, solvent extraction, and evaporation. The emulsifying activity of the biosurfactant showed better than the chemically synthesized surfactant (SDS, Span40, Span 85). The biosurfactants had strong properties as an emulsifying agent and as an emulsion-stabilizing agent.

  • The fruiting bodies of Paecilomyces japonica been used for an anticancer and immuno-stimulating agent as an oriental medicine, Antimutagenecity test with SOS chromotest and antioxidant test with NBT method were carried out using the concentrated culture broth, the extract of mycelia, and that of fruiting bodies. Among the sample extracts tested, the extract of fruiting bodies was most effective to antimutagenecity against the mutagens tested such as MNNG, ethidium bromide(EtBr), 2-aminofluorene(AF) and nitrofluorene(NF). Antimutagenic activity of all the extracts were very effective against mutagen, MNNG. When the extracts were added to certain concentration, antimutagenic activity was enhanced against mutagen, MNNG and NF. Antioxidant activity of the extract from fruiting bodies was highest. However, its activity was very low, compared to ascorbic acid.

Clitocybin D, a Novel Human Neutrophil Elastase Inhibitor from the Culture Broth of Clitocybe aurantiaca

  • Clitocybin D, a novel human neutrophil elastase inhibitor, was isolated from the culture broth of Clitocybe aurantiaca. This compound was purified by solvent extraction, silica gel column chromatography, Sephadex LH-20 column chromatography, and preparative HPLC. The compound was determined to be 4-(4,6-dihydroxy-3-methoxy-3H-isoindol-1-yl)-benzoic acid on the basis of 1D and 2D NMRs and MS spectroscopic analysis. Analysis of the human neutrophil elastase (HNE) inhibitory activity of the isolated compound revealed that it showed significant HNE inhibitory activity with an $IC_{50}$ value of $17.8{\mu}M$.

Isolation and Characterization of Antimicrobial Substance Macrolactin A Produced from Bacillus amyloliquefaciens CHO104 Isolated from Soil

  • A strain antagonistic to Fusarium solani, CHO104, was selected from approximately 100 microorganisms isolated from soil. Strain CHO104 was identified as Bacillus amyloliquefaciens and found to be Gram-positive based on the Biolog system and 16S rRNA methods. A culture broth of B. amyloliquefaciens CHO104 also exhibited antimicrobial activity against various microorganisms. As such, the EtOAc extract of the culture broth was isolated by various column chromatographic procedures and HPLC. The antimicrobial and antifungal substance was then characterized as macrolactin A $(C_{24}H_{34}O_5)$ using high-resolution EI-MS and NMR analyses, and found to be very effective in inhibiting the growth of Staphylococcus aureus, E. coli, and Botrytis cinerea, even when using a concentration of one-twentieth of the benzoic acid as the control compound.

  • The bacteria which secrete surface-active agent and decrease the surface tension of culture broth were isolated from soil samples. Among them, biosurfactant producing strain KK-7 was selected and emulsification was also detected. The KK-7 produced biosurfactant not only lipid but also glucose by using carbon source. Taxonomical characterization tests have demostrated the strain KK-7 to be Pseudomonas aeruginosa. The media composition of the P. aeruginosa KK-7 for the biosurfactant production was 1% glucose, 0.5% tryptone, 0.2% yeast extract, 0.15% potas sium phosphate mono-dibasic, 0.05% MgSO$_{4}$, initial pH 8.5, at 30$\circ $C for 2 days. In this condition, the concentration of biosurfactant was reached CMC 5 in the culture broth. Surface active material was produced maximum at stationary8 phase, but emulsification power was higher at log phase than stationary phase. It was considered that P. aeruginosa KK-7 produced biosurfactant more than one type having defferent properties and each maximum production time was different. The minimun surface tension of biosurfactant in 50 mM Tris buffer (pH8.0) was 28 dyn/cm, and CMC was 1 g/L.

Isolation and Characterization of the Auxin producing Plant Growth Promoting Rhizobacterium from Soil in a Ginseng Field (인삼토양으로부터 옥신 생성 식물생장촉진세균의 분리 및 특성)

  • An auxin-producing bacterium (AMT-54) was isolated from ginseng cultivating soil of Geumsan area. The isolate AMT-54 was confirmed to produce indole-3-acetic acid (IAA) which is one of auxin hormone by TLC analysis. When the concentration of IAA was assessed by performing HPLC quantitative analysis, the maximal 457ppm of IAA was detected from the culture filtrate after culturing in R2A broth containing 0.1% tryptophan for 24h at $35^{\circ}C$. The molecular weight of the main peak obtained by LC-mass analysis was correspondent well to 175, that of IAA. The strain AMT-54 was identified as a novel species belongs to Klebsiella mobilis by a chemotaxanomic and phylogenetic analysis. To investigate the growth promoting effect of crop, when the culture broth of K. mobilis AMT-54 was infected onto seed pot of mung bean, the adventitious root induction and root growth of mung bean were 3.3times higher than control.

Biosurfactant Production from Phenanthrene Degrading Bacteria (Phenanthrene 분해균주로부터 미생물 계면활성제의 생산)

  • Phenanthrene degrading bacteria were isolated from the petroleum contaminated soil near an oil tank. Four of 15 strains decreased surface tension of culture broth of phenanthrene-containing minimal media. H6, one of the isolated bacteria decreased surface tension of culture broth below 33 dyne/cm during growth on glucose. H6 was identified as Bacillus subtilis and biosurfactant produced by H6 was lipopeptide. The biosurfactant was produced at 0.13 g/L in the mineral medium containing 2% glucose. Critical micelle concentration(CMC) of the biosurfactant was 52 mg/L. Foaming power was similar to Tween 80 and dispersing power was superior to Tween 80m SDS and Brij30. High thermal stability and emulsion index were also observed.
