Physiological and Ecological Characteristics of Indigenous Soybean Rhizobia Distributed in Korea -I.   • In order to improve effectiveness of rhizobia-legume symbiotic nitrogen fixation, ecological and physiological characteristics of indigenous rhizobia distributed in Korea soil, that is, the effects of soybean cultivation, physico-chemical properties and climate on the population of indigenous rhizobia and other soil microbes were investigated. The results were summarized as follows: The population of indigenous rhizobia were ranged from $5.1{\times}10^4$ cells to $196.8{\times}10^4$ cells per gram of soil in soybean cultivated soils but from $1.6{\times}10^4$ cells to $78.6{\times}10^4$ cells per gram of soil in soybean un-cultivated soils sampled from 9 different agro-climate zone. The highest population was observed in a soybean cultivated loamy soil from southern part of Korea. The content of available phosphate, exchangeable Ca, Mg, Cu, and B in soil were positively correlated but active Fe, exchangeable Al, Na, and $SO_4$ were inversely correlated to the population of indigenous rhizobia. The inverse relationship was observed between the number of indigenous rhizobia and actinomycetes.

Effects of Paddy-Upland Rotation on Soil Characteristics and Crop Productivity in Rice Fields on Reclaimed Tidal land (신간척지 벼 재배 농지의 답전윤환에 따른 토양 특성 및 작물 생산성 변화)

  • Paddy-upland rotation system is one of the important cropping system for improving soil quality and crop productivity. we conducted to investigate the effect of paddy-upland rotation system on soil properties and crop productivity in reclaimed tidal land. The paddy-upland rotation could be effective to conserve soil water contents and prevent from salt damage when cultivating upland crops. The first two years of maize cultivation after rice cultivation could be effective to secure stable production. However, in case of soybean crop, the rotation effect might be lower than that of maize. In the first year, the yield of soybean was 214 kg/10a. In the second and third year, the yields of soybean decreased consecutively to 152, 123 kg/10a respectively. In this paper, it would be suggested that maize be cultivated for up to two years and soybean be cultivated for one year after rice crop grown in reclaimed tidal land. This study could be provide basic data of the physico-chemical properties applicable to paddy-upland rotation system at reclaimed tidal lands.

Study of Soybean Culture and Analysis on the Chemical Composition of Soybean for Making Use of the Mountainous Uncultivated Land (산지유휴지활용을 위한 대두재배 및 성분연구)

  • This study was undertaken to learn the over-all influence of fertilizer on soybean culture and the chemical composition of soybean in the reclaimed land. Treatments included five rates of N.P.K: 4.10.8, compost: 1, 000. N.P.K.: 4.10.8 + compost: 1, 000, N.P.K: 2.5.5 kg/10 are as well as unfertilized plot in the reclaimed land and N.P.K: 4.10.8 kg/10 are as a check plot of the cultivated area. The yield of green plant above-ground, grain yield and protein content were higher in the cultivated plot than in the reclaimed land. Maximum yield of green plant or the greatest grain yield occurred in the plot of N.P.K: 4.10.8+compost: 1, 000 kg application and they diminished as the fertilizer application decreased. Yet there was no statistical significance in the chemical seed composition among treatments.

Quality Evaluation for Vegetable Use in Local Soybean Cultivars with Various Seed Coat Color

  • From the evaluation of physical properties such as springiness, gumminess, adhesiveness, chewiness and hardness by the texture analyzer, vegetable soybean lines with green seed-coat were best as compared with those with black, brown, mixed, and yellow seed-coats. A panel test evaluated on the basis of taste, sweetness, chewiness, and total scores also indicated that soybean lines with green seed-coat were the best. The total scores of panel test was decreased in the order of green > yellow> black> brown seed-coat colored soybean. The mean value of sucrose content obtained by HPLC analysis was highest in black seed-coat colored soybean, and followed by green, yellow, and brown soybeans. The highest sucrose content (8.22%) was observed in 180362, a soybean line with green seed-coat. The full-season type soybeans showed much higher sucrose content than summer types which are mainly cultivated on farmer's fields for vegetable purposes. The final 13 lines selected from 300 colored soybeans showed nearly the same panel scores as Miwongreen. However, these lines had a great deal of variation in sucrose content, and much higher readings in texture analysis than Miwongreen, especially in chewiness and hardness which were the most important properties in vegetable soybeans.

Effect of Cultivar and Processing on the Hemagglutinin Activity of Soybean

  • Effects of cultivars, cooking, and processing on hemagglutinin activity were evaluated by observing macroscopic hemagglutination using serial twofold dilution of trypsinized human blood type-O or rabbit blood. Hemagglutinin activity was expressed as maximal geometric dilution fold. Agglutination of rabbit blood was more sensitive compared to human blood. Hemagglutinin activities of glyphosate-tolerant soybean, HS2906, and imported conventional soybeans were not statistically different, although significant differences were observed among conventional soybean cultivars cultivated in Korea (286 to 1535 HU/mg protein). Time required to reach fifty percent inhibition of hemagglutinin activity ($IT_{50}$) value decreased with increasing cooking temperature and pressure. Most effective conventional cooking method to inhibit hemagglutinin activity was pressure-cooking ($IT_{50}$: 1.36 min). Calculated activation energy based on reaction rate constant was 4.88 kcal. No hemagglutinin activities were detected in processed soybean products such as tofu, soybean paste, and soysauce.

Chemical properties of soybean-cultivated field soils (대두재배 농가포장 토양의 화학적 특성)

  • In order to obtain the basic informations on the reasonable fertilization and soil management systems for a high soybean yield, both soybean leaves and soils were collected from 24 soybean-cultivated fields in central area of Korea, and analyzed. For this study, soybean leaves and soil were sampled together in July of 1988(growing season), and soil alone in October of 1988 (harvesting season) and April of 1989(sowing season) at each sampling site. The results might be summarized as follows: 1. The soil pH ranged between 4.8 and 6.8. and the pH after harvest decreased to 5.4 which compared with the pH 5.7 of growing season. 2. Both the 1N KCl extractable Al and the 1N $NH_4OAc$ extractable Al in soils were inversely correlated with the soil pH. the former concentration was negligible above pH 5.8. 3. The soil total-N and Bray 1-P after harvest were lower than those of growing season. The Bray 1-P's of sample soils were very high irrespective of sampling time. 4. The exchangeable Ca and Mg in soils even after harvest varied little whereas the exchangeable K decreased with time. 5. The N content in soybean leaves was not correlated with soil total-N, but highly correlated with the K content in soybean leaves. The Mg content in soybean leaves was also highly correlated with Ca content in suybean leaves and exchangeable Mg in soils.

Impact of vitamin-A-enhanced transgenic soybeans on above-ground non-target arthropods in Korea

  • In order to confirm the safety of a genetically modified organism (GMO), we assess its potential toxicity on non-target insects and spiders. In this study, the effects of GM soybean, a type of vitamin-A-enhanced transgenic soybean with tolerance to the herbicide glufosinate, were assessed under a field condition. The study compared this vitamin-A-enhanced transgenic soybean and a non-GM soybean (Gwangan) in a living modified organism (LMO) isolated field of Kyungpook National University (Gunwi) and the National Institute Agricultural Sciences (Jeonju) in the Republic of Korea in 2019 - 2020. In total, 207,760 individual insects and arachnids, representing 81 families and 13 orders, were collected during the study. From the two types of soybean fields, corresponding totals of 105,765 and 101,995 individuals from the vitamin-A-enhanced transgenic soybean and Gwangan samples areas were collected. An analysis of variance indicated no significant differences (p < 0.05). A multivariate analysis showed that the dominance and richness outcomes of plant-dwelling insects were similar. The data on insect species population densities were subjected to a principal component analysis (PCA) and an orthogonal partial least squares-discriminant analysis (OPLS-DA), which did not distinguish between the two varieties, i.e., the vitamin-A-enhanced transgenic soybean and the non-GM soybean in any cultivated field. However, the results of the PCA analysis could be divided overall into four groups based on the yearly survey areas. Therefore, there was no evidence for the different impact of vitamin A-enhanced transgenic soybean on the above-ground insects and spiders compared to non-GM soybean.
