산양산삼약침이 고지방식이에 의한 당뇨병 유발 흰쥐에 미치는 영향 (Effects of distilled Cultivated Wild Ginseng Herbal Acupuncture in Rats with Diabetes Induced by High Fat Diet)

  • Objectives : This study was to verify the effects of distilled cultivated wild ginseng herbal acupuncture(CWGHA) on diabetes by hematological analysis. Methods : Rats were fed with high fat diet for 8 weeks and the rats with hyperglycemia were selected for the experiment. Various treatments of distilled cultivated wild ginseng herbal acupuncture were administered intravenously and glucose, ${\beta}-lipoprotein,$ triglyceride, total-cholesterol, HDL-cholesterol, LDL-cholesterol, Free Fatty acid(FFA), TBARS, superoxide dismutase(SOD), catalase and glutathione peroxidase activities in the liver were analyzed. Results : 1. Experiment group 3(0.1 ml of CWGHA was injected intravenously 10 times) showed significant decrease in serum glucose, ${\beta}-lipoprotein,$ triglyceride, LDL-cholesterol levels and liver TBARS compared to the control group, whileas showed significant increase in liver glutathione peroxidase activity. 2. Experiment group 2 and 3 (treated with 0.5 ml, 1 ml, respectively), showed significant decrease in serum FFA, total cholesterol and TBARS levels compared to the control group, and showed significant increase in liver superoxide dismutase and catalase activities. 3. Serum HDL-cholesterol didn't show significant changes in both experiment and control groups. Conclusions : Above results indicate that distilled cultivated wild ginseng herbal acupuncture plays significant role as a hypoglycemic agent and in lipid metabolism. Increase in the number of administrations yielded more significant results.

Genetic Diversity of Wild and Cultivated Populations of American Ginseng (Panax Quinquefolium) from Eastern North America Analyzed by RAPD Markers

  • The objective of this study was to assess genetic diversity among 6 different wild ginseng populations from New York, Kentucky, North Carolina, Pennsylvania, Tennessee and Virginia, and to compare these wild populations to one cultivated population. RAPD markers were used to estimate the genetic difference among samples from the 7 populations. The 64 random primers were screened, and 15 primers were selected which exhibited the 124 highly reproducible polymorphic markers. The ratio of discordant bands to total bands scored was used to estimate the genetic distance within and among populations. Multidimensional scaling (MDS) of the relation matrix showed distinctive separation between wild and cultivated populations. The MDS result was confirmed using pooled chi-square tests for fragment homogeneity. This study suggests that RAPD markers can be used as population-specific markers for American ginseng.

고려인삼의 재배 역사와 본초학적 고증을 통한 인삼 기미 고찰 (Discussion of Ginseng Properties through a Historical Research of Korean Ginseng)

  • Objectives : The purpose of this study is to discuss ginseng properties by historical research. Methods : Historical documentary records of ginseng were used to develop this review. Results : According to the historical research, the first Korean ginseng cultivation started from the Koryo Dynasty around A.D. 1000. Later, during the Chosun Dynasty around A.D. 1500$\sim$1600, the ginseng cultivators established a method of cultivation. Gasam (cultivated ginseng) was cultivated ginseng on the skirts of their domicile. It was reported that Gasam cultivation was broadened throughout the country around A.D. 1790 on Cheongjosilrok. On the other hand, ginseng properties were reported slightly cool in the oriental herbal medicine books from A.D. 250 to A.D. 1600. But, they were reported slightly warm or warm after A.D. 1600. Korean ginseng cultivation started in the Koryo Dynasty around A.D. 1000. Later, during the Chosun Dynasty, the ginseng cultivators established a standard method of cultivation. Gasam, cultivated ginseng, was grown on the outskirts of their domicile. It was reported that Gasam cultivation was broadened throughout the country around 1790 A.D. in Cheongjosilrok. On the other hand, ginseng properties were reported slightly cool in the oriental herbal medicine books from 250 to A.D. 1600, but they were reported slightly warm or warm after A.D. 1600. Conclusions : It suggests that ginseng properties (slightly cool) before A.D. 1600 are wild ginseng. Also, wood-grown ginseng and ginseng properties (slightly warm, warm) after A.D. 1600 are Gasams.

선택실험법을 이용한 산양삼의 경제적 가치평가 (Estimation on Economic Value for Cultivated Wild Ginseng using Choice Experiment)

  • 이 연구는 산양삼의 상품 속성에 대하여 소비자들이 얼마만큼의 가치를 부여하고자 하는지를 파악하고 그것에 대한 경제적 가치를 평가하고 한다. 이를 위해 선택실험기법을 이용하였으며, 선택실험기법에 사용된 속성은 재배연령, 식재방법, 산양삼의 몸통 크기, 산양삼의 수염뿌리 길이, 색채 등과 같이 외관 특징에 기초하였다. 분석을 위해 모집단은 서울을 포함한 수도권에 거주하는 일반시민을 대상으로 하였으며, 조사방법은 설문면접조사로 총 173부의 설문지를 획득하였다. 분석결과, 모형은 안정적인 범위에 속하였으며, 응답자는 직파삼에 대하여 가장 높은 가 치를 부여한 반면 몸통크기에 대해서는 상대적으로 그 가치가 낮은 것으로 나타났다.

Rapid characterization of ginsenosides in the roots and rhizomes of Panax ginseng by UPLC-DAD-QTOF-MS/MS and simultaneous determination of 19 ginsenosides by HPLC-ESI-MS

  • Background: Ginsenosides are the characteristic and principal components which manifest a variety of the biological and pharmacological activities of the roots and rhizomes of Panax ginseng (GRR). This study was carried out to qualitatively and quantitatively determine the ginsenosides in the cultivated and forest GRR. Methods: A rapid and sensitive ultra-high-performance liquid chromatography coupled with diode-array detector and quadrupole/time of flight tandem mass spectrometry (UPLC-DAD-QTOF-MS/MS) was applied to the qualitative analysis of ginsenosides and a 4000 QTRAP triple quadrupole tandem mass spectrometer (HPLC-ESI-MS) was applied to quantitative analysis of 19 ginsenosides. Results: In the qualitative analysis, all ingredients were separated in 10 min. A total of 131 ginsenosides were detected in cultivated and forest GRR. The method for the quantitative determination was validated for linearity, precision, and limits of detection and quantification. 19 representative ginsenosides were quantitated. The total content of all 19 ginsenosides in the forest GRR were much higher than those in the cultivated GRR, and were increased with the growing ages. Conclusion: This newly developed analysis method could be applied to the quality assessment of GRR as well as the distinction between cultivated and forest GRR.

General Introduction of American Ginseng Indigenous in USA and Canada

  • American ginseng (Panax quinquefolium) is herbaceous perennial plants indigenous to North American forests. This is highly valued as medicinal herbs with a long history of collection from wild populations since 1716. Wild American ginseng distributed from Quebec in Canada to northern Florida in USA. A heavy concentration is found in the Appalachian mountains, although wild American ginseng is considered endangered. The price paid for field cultivated ginseng has dropped dramatically in the past 10 years, while the price for wild or woods cultivated ginseng has rised significantly. The price curve for ginseng resembles a roller coaster, reflecting not only supply and demand but many other factors. This information will be useful to understand American ginseng compared to Korean ginseng.

HPLC-CAD에 의한 산양삼의 극성 및 비극성 ginsenoside 동시 분석 (Simultaneous Analysis Method for Polar and Non-polar Ginsenosides in Cultivated Wild Ginseng by Reversed-phase HPLC-CAD)

  • 산양삼은 식품 또는 약초로써 많이 사용되고 있다. 이번 연구의 목표는 high performance liquid chromatography (HPLC) - charged aerosol detection (CAD)에 의해 산양삼의 추출물에 사포닌들의 분석을 최적화 하는 것이다. CAD는 물리적인 특성은 물론이고 입자크기, 스펙트럼 특성과는 무관하게 비휘발성 물질도 분석할 수 있다. 산양삼 추출물 중 Gensenoside Rb1, Rd, Rg1, Rf, Re 및 Rh1을 분석하였다. HPLC-CAD분석을 위한 사포닌 추출물은 건조 산양삼 10 g에 95% 에탄올 100 ml을 넣어 80℃에서 24시간 동안 추출하여 사포닌을 얻었다. 6개의 주요한 사포닌을 정량화하였을 경우 값은 다음과 같다. 사포닌들의 함량은 Rb1 (5.48±0.12 mg/g), Rd (5.33±0.14 mg/g), Rg1 (12.80±0.05 mg/g), Rf (19.08±0.68 mg/g), Re (19.87± 0.05 mg/g), 및 Rh1 (16.47±0.16 mg/g)로 확인하였다. 대체적으로 산양삼 추출물의 사포닌은 protopanaxatriol (Rg1, Rf, Re, Rh1)의 값이 protopanaxadiol (Rb1, Rd)보다 높게 확인 되었다. 이번 연구에서는 산양삼 사포닌의 분석조건을 HPLC-CAD를 이용하여 최적화하였고 제품화를 위해 약리활성에 관한 연구도 매우 중요할 것이다.

Genetic diversity among cultivated and wild Panax ginseng populations revealed by high-resolution microsatellite markers

  • Background: Ginseng (Panax ginseng Meyer) is one of the world's most valuable medicinal plants with numerous pharmacological effects. Ginseng has been cultivated from wild mountain ginseng collections for a few hundred years. However, the genetic diversity of cultivated and wild ginseng populations is not fully understood. Methods: We developed 92 polymorphic microsatellite markers based on whole-genome sequence data. We selected five markers that represent clear allele diversity for each of their corresponding loci to elucidate genetic diversity. These markers were applied to 147 individual plants, including cultivars, breeding lines, and wild populations in Korea and neighboring countries. Results: Most of the 92 markers displayed multiple-band patterns, resulting from genome duplication, which causes confusion in interpretation of their target locus. The five high-resolution markers revealed 3 to 8 alleles from each single locus. The proportion of heterozygosity (He) ranged from 0.027 to 0.190, with an average of 0.132, which is notably lower than that of previous studies. Polymorphism information content of the markers ranged from 0.199 to 0.701, with an average of 0.454. There was no statistically significant difference in genetic diversity between cultivated and wild ginseng groups, and they showed intermingled positioning in the phylogenetic relationship. Conclusion: Ginseng has a relatively high level of genetic diversity, and cultivated and wild groups have similar levels of genetic diversity. Collectively, our data demonstrate that current breeding populations have abundant genetic diversity for breeding of elite ginseng cultivars.

산양삼 재배지의 입지, 토양 및 재배특성 분석 (An Analysis on Site, Soil and Cultivation Characteristics of Korean Mountain Cultivated Ginseng (Panax ginseng) Field)

  • 산양삼 재배를 위한 기초자료를 제공할 목적으로 9개도 18 재배지를 대상으로 입지환경, 토양환경, 생육과정, 재배방법과 시기를 조사 분석하였다. 산양삼 재배 입지환경은 활엽수림, IV영급 기준 ha당 잔존본수 500~1,000본, 해발 500 m 미만, 경사 $25^{\circ}$ 이하, 재배사면은 북, 북동, 동, 북서방향에서 주로 재배되는 것으로 나타났다. 토양은 유기물함량 3.43~24.07%, pH 3.8~5.7, 토성은 사토~사양 토로 재배지 간에 많은 차이를 나타내었다. 생육과정은 발순, 개엽, 개화, 결실, 홍숙, 낙과, 낙엽까지 조사되어 평균 98일의 생육기간이 소요되었다. 재배방법은 파종과 이식 형태로 증식되고 있었으며, 파종은 개갑처리(발아촉진처리)와 직파형태로 파종하고 있으며 각각 70%, 30%로 처리되는 것으로 나타났다.

Variation in the number of nucleoli and incomplete homogenization of 18S ribosomal DNA sequences in leaf cells of the cultivated Oriental ginseng (Panax ginseng Meyer)

  • Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.
