In this study, the genetic diversity and the population structure of 77 wild strains and 23 cultivars of Lentinula edodes from Korea were analyzed using 20 genomic SSRs, and their genetic relationship was investigated. The tested strains of L. edodes were divided into three sub-groups consisting of only wild strains, mainly wild strains and several cultivars, and mainly cultivars and several wild strains by distance-based analysis. Using model-based analysis, L. edodes strains were divided into two subpopulations; the first one consisting of only wild strains and the second one with mainly cultivars and several wild strains. Moreover, AMOVA analysis revealed that the genetic variation in the cultivars was higher than that in the wild strains. The expected and observed heterozygosity and values indicating the polymorphic information content of L. edodes cultivars from Korea were also higher than that of the wild strains. Based on these results, we presume that the cultivars in Korea have developed by using numerous strains from other countries. In conclusion, the usage of wild strains for the development of new cultivars could improve the adaptability of L. edodes to biotic and abiotic stress.
Pyogo (Shiitake, Lentinula edodes) is one of the most important edible mushrooms because of its outstanding nutritive and medicinal value. In the registration and protection procedure for newly developed mushroom cultivars, the application of molecular markers that can supplement the morphological characteristic-based distinction has been strongly requested. Sanjo 701 and Chamaram, newly developed at the Federation Forest Mushroom Research Center of Korea, have been characterized as innovative cultivars suitable for customer demands because of their high yields and cultivation rates. However, no technical tools can protect the rights to these important cultivars. In this study, using comparative genomic information from 23 commercially available pyogo cultivars, we identified single nucleotide polymorphisms (SNPs) that accurately differentiated Sanjo701 and Chamaram from the other cultivars. We also developed high-resolution melting analysis (HRM)-based SNP markers that discriminate among the tested 23 pyogo cultivars. The developed SNP markers can be utilized for rapid, accurate identification of pyogo cultivars with low genetic diversity and to prevent cultivar contamination caused by illegally distributed inocula. In addition, these markers can serve as a crucial scientific basis for securing the right to conserve new cultivars in international markets.
Rose crown gall caused by Agrobacterium tumefaciens is a major disease that damages the production of cutroses in Korea. The effective prevention methods for this disease include the use of resistant varieties. This study was conducted to evaluate the resistance of 58 Korean cultivars and six foreign cultivars to crown gall disease with nodal explants in vitro. Among 180 A. tumefaciens strains, pathogenic strain RC12 was selected as an inoculant strain. The strain RC12 was identified based on characteristics of some selective media, pathogenicity test, and polymerase chain reaction analysis. Forty rose cultivars formed tumors on explants inoculated with A. tumefaciens RC12. However, 24 cultivars, including 22 Korean cultivars and 2 foreign cultivars, showed resistance to A. tumefaciens RC12 without forming any tumors. Six cultivars with tumor formation rates of over 30% formed initial tumors within 23 days after inoculation. Six cultivars with low tumor formation rates of around 5% formed initial tumors after 28 days of inoculation. It was found that gall formation rate was highly correlated with the initial gall formation period. Thus, the relationship between the period of gall formation and the rate of gall formation could be useful for assessing resistance to crown gall disease. In vitro inoculation methods could be used to evaluate resistance of cut-rose cultivars to crown gall diseases.

Recently NERICA (New Rice for Africa) was developed by a crossing of African rice (Oryza glaberrima Steud.) and Asian rice (Oryza sativa L.) in West Africa, and is considered to be drought resistant, but drought resistance of NERICA and differences between Asian rice are not clarified enough. In this research, NERICA (four cultivars and two lines), Asian rice (three cultivars and sativa parent of NERICA) and African rice (glaberrima parent of NERICA) were cultivated in the field in Shizuoka University under drought and traits of each cultivar and line relevant to drought resistance, stomatal conductance by porometer, soil water content of individual depths by TDR method, SPAD values by SPAD meter and leaf thickness by micrometer, were measured and compared with dry matter production and yield. Effects of compost were also compared among sativa parent, one NERICA cultivar and two NERICA lines. Glaberrima parent showed highest top dry weight. One NERICA line, one drought resistant Asian rice cultivar and sativa parent, showed higher top dry weight and yield (ear weight) than other Asian rice cultivars and NERICA cultivars and line tested. Compost tended to increase top dry weight and yield in one of NERICA line and sativa parent. But in one NERICA cultivar and line, top dry weight and yield were not increased. In one of Asian rice, one of NERICA line and sativa parent that showed high top dry weight and yield, stomatal conductance was high. On the contrary the glaberrima parent and in other NERICA cultivars and line it was low. In sativa parent compost increased stomatal conductance but in NERICA cultivar and lines it was not. Among cultivars and lines that showed high top dry weight and yield sativa parent and one of NERICA line SPAD value and leaf thickness were high but in one of Asian rice and glaberrima parent they were low. Cultivar and line differences in yield and top dry weight among Asian rice and NERICA were significantly correlated with those in stomatal conductance ($r=0.778^{**}$ and $r=0.654^*$, respectively) and those in leaf thickness ($r=0.600^*$ and $r=0.640^*$, respectively). In Asian rice cultivars average soil water content was significantly correlated with yield ($r=0.886^*$) but in NERICA cultivars and lines it was not significant correlated (r= -0.256). Cultivar and line differences in leaf thickness were significantly correlated with SPAD value ($r=0.773^{**}$). In Asian rice cultivars it was significantly correlated ($r=0.962^{**}$), but in NERICA cultivars and lines it was not significantly correlated (r=0.559). Asian rice cultivars tended to consume soil water to increase yield but in NERICA cultivars and lines the tendency was not clear. Correlation between SPAD value and leaf thickness was different between Asian rice and NERICA cultivars and lines, and in Asian rice cultivars it was significantly correlated but in NERICA cultivars and lines it was not significant. Importance of maintaining high stomatal conductance by high leaf thickness was clarified.

This experiment was carried out to assess the growth and development of five rye(Secale cereale L.) cultivars based on the Feekes scale in relation to sowing date at the Experimental Livestock Farm, Seoul National University(SNU), Suwon from 30 September 2001 to 4 May 2002. The experiment was conducted in split plot design with three replications. The main plots consisted of two sowing date such as early(30 September) and late(15 October). The subplots consisted of rye cultivars of five different maturity groups such as 'Kodiak', 'Koolgrazer', 'Danko', 'Homil22' and 'Olhomil'. The plant height of early sowing rye cultivars was higher than that of late one. Among the rye cultivars tested, plant heights of early maturing cultivars were higher than those of the other cultivars. The tiller number at early sowing(48.0) was higher than that of late(24.3), however, late maturing cultivars were higher than those of the other cultivars among the rye cultivars tested. Growth and development in the early sowing were generally three days earlier than those of late sowing cultivars. Growth and development in 'Olhomil' rye cultivars was earlier than 'Danko' rye cultivars when sown in early, but 'Olhomil' and 'Koolgrazer' rye cultivars showed an early maturity than 'Danko' rye when sown in late. Therefore, the developmental stages of winter rye cultivars could be categorised the following maturity; 'Olhomil' and 'Koolgrazer' are early in maturity, 'Homil22' is medium, and 'Kodiak' and 'Danko' are late in maturity.

Powdery mildew and root-knot nematode are very important diseases occurred in cucurbits. This study was conducted to evaluate the resistance of commercial cucurbit cultivars (21 cultivars of cucumber, 9 cultivars of watermelon, 7 cultivars of oriental melon, and 2 cultivars of melon) to powdery mildew and root-knot nematode. At 60 days after transplanting, disease severity of powdery mildew of commercial cucurbit cultivars was investigated. Two cucumber cultivars, 'Gangryeogsamcheok' and 'Sunhobaegchimdadagi' were moderately resistant but the rest of cucumber cultivars were susceptible to powdery mildew. All examined watermelon and oriental melon cultivars were susceptible to powdery mildew, while two melon cultivars ('PMR Turbo' and 'PMR Victory') were resistant. At 45 days after inoculation of seedlings, disease severity of root-knot nematode of commercial cucurbit cultivars were investigated. One cucumber cultivars, 'Baegbongdadagi' was moderately resistant and the others were susceptible to root-knot nematode. In case of watermelon, 'Dalgonakkul' was resistant and the others were moderately resistant or susceptible to root-knot nematode. All examined oriental melon and melon cultivars were susceptible to root-knot nematode. On these results, we recommend that resistant cultivars to powdery mildew and/or root-knot nematode will be more suitable in cultivation of cucurbits for organic farming.

To investigate the genetic variation of high-and low-molecular-weight glutenin subunits (BMW-GS and LMW-GS), granule-bound starch synthase I (GBSSI) and puroindoline in 24 Korean wheat cultivars. At the BMW-GS compositions, three Glu-A1 alleles, five Glu-B1 alleles and three Glu-D1 alleles were identified. The high frequency of alleles at each locus was Glu-A1c allele (15 cultivars), Glu-B1b allele (16 cultivars) and Glu-D1f allele (16 cultivars). Four alleles were identified at the Glu-A3 and Glu-B3 loci and three at Glu-D3 locus and Glu-A3d, Glu-B3d and Glu-D3a were mainly found at each Glu-3 locus. Glu-A3d, Glu-B3d, Glu-D3b or c (4 cultivars, respectively) and Glu-A3d, Glu-B3d, Glu-D3a and Glu-A3c, Glu-B3d or h, Glu-D3a (3 cultivar, respectively) were predominantly found in Korean wheats. At the GBSS compositions, 2 waxy wheat cultivars, Shinmichal and Shinmichal1, showed null alleles on the Wx loci and other cultivars were wild type in GBSS compositions. At the puroindoline gene compositions, Korean wheat cultivars carried 3 genotypes, which 10 cultivars (41.7%) were Pina-D1a and Pinb-D1a, 11 cultivars (45.8%) had Pina-D1a and Pinb-D1b and 3 cultivars (12.5%) carried Pina-D1b and Pinb-D1a. These genetic variations could present the information to improve flour and end-use quality in Korean wheat breeding programs.
Greenhouse studies were conducted to evaluate the tolerance of 42 pepper cultivars to postemergence applications of bentazon [3-(1-methylethyl)-(1H)-2,1,3-benzothiadiazin-4(3H)-one 2,2-dioxide]. Cultivars, Jopoong, Singsing House, Sweet Green, Kwangbok, and Ilcheon showed relatively tolerant response to bentazon, while cultivars, Dahhong, Early Glory, Korea, Cheongyang, Nostalgia, and Daejanggyeong were susceptible ones to it. At rates over 2.40 kg ai/ha, the tolerant cultivars appeared to be clearly or more tolerant than the susceptible cultivars. For the determination of growth inhibition by bentazon, the concentration required to reduce growth by 50% (GR_(50)) was 2.00 to 2.40 kg ai/ha for susceptible cultivars, and 10.00 to 12.00 kg ai/ha for tolerant cultivars. Moreover, the herbicide rate required to inhibit growth by 50% $(I_{50})$ was 2.40 kg ai/ha for susceptible cultivars and 9.60 kg ai/ha for tolerant cultivars, respectively. On the $I_{50}$ and $GR_{50}$ estimates of growth, the tolerant cultivars were 5- to 6-fold more tolerant to bentazon than susceptible ones.
To understand the heading habit of recently developed 20 Korean rice cultivars, rice plants in a phytotron were exposed to different temperature: $22.5^{\circ}C(day\;27^{\circ}C/night\;18^{\circ}C),\;27.5^{\circ}C(day\;32^{\circ}C/night\;23^{\circ}C)$, and day-length conditions: 10, 12, 13, 14, 15 hours. Four rice cultivars (Sobibyeo, Juanbyeo, Ilpumbyeo and Shindongjinbyeo) showed relatively short Basic Vegetative Phase (BVP) of 17 to 18 days, while Dasanbyeo showed the longest (35 days) BVP, compared to other remaining 15 tested cultivars which exhibited 24 to 31 days of BVP. In this experiment, it was tried out to separate the eliminable vegetative phase into photosensitivity and thermo-sensitivity with two different pathways. Many tested cultivars, however, exhibited quite different responses under low temperature and / or long day-length conditions. Especially, Surabyeo and Juanbyeo were the most difficult cultivars to separate into photo- or thermo-sensitivity in that the eliminable vegetative phase of these two cultivars increased greatly only under low temperature $(22.5^{\circ}C)$ and long day-length (15 hr.) conditions. Regarding the heading response to temperatures, tested cultivars could be categorized into 2 groups. In 1st group of rice cultivars, the eliminable vegetative phase decreased almost equally as the temperature changed from $20.0^{\circ}C\rightarrow22.5^{\circ}C\rightarrow25.0^{\circ}C\rightarrow27.5^{\circ}C$. In contrast, the 2nd group of rice cultivars exhibited eliminable vegetative phase slowly decreasing when the temperature changed from $22.5^{\circ}C\rightarrow25.0^{\circ}C\rightarrow27.5^{\circ}C$, but rapidly decreasing when the mean temperature changed from $20.0^{\circ}C\;to\;22.5^{\circ}C$. All the cultivars belonged to 2nd group, the heading date would be very delayed if cool summer comes.

Stomatal traits such as stomata density (SD), aperture length (APL) and width (APW), guard cell length (GCL) and width (GCW), and distance between stomata (DIS) were investigated to identify correlation with agronomic traits for 35 Korean wheat cultivars. Flag leaf width (FLW) of Korean wheat cultivars was the widest in Ol-mil, and the narrowest in Keumkang. SD tended to be higher on the adaxial side than on the abaxial side in Korean wheat cultivars. SD of adaxial and abaxial sides was classified into a cultivar with a significantly different or not. In APL, 18 wheat cultivars showed significant differences according to leaf side, and APL of adaxial was longer than APL of abaxial in 13 wheat cultivars. In APW, 15 wheat cultivars showed a significant difference, and APW of abaxial was wider than APW of adaxial among them. In GCL, 14 wheat cultivars showed a significant difference, and the GCL of abaxial was longer than the GCL of adaxial in 10 wheat cultivars. In GCW, 10 wheat cultivars showed a significant difference, GCW of adaxial was wider than GCW of abaxial and in 6 wheat cultivars. FLW of adaxial and abaxial showed a negative correlation with GCL and a positive correlation with grain number per panicle. FLW of only abaxial showed a positive correlation with DIS. The SD of the adaxial showed a negative correlation with GCL, while the SD of the abaxial showed a negative correlation with APL. APL of both sides of the leaf showed a positive correlation with GCL, and APW of only abaxial showed a negative correlation with GLC. DIS of adaxial showed a negative correlation with tiller number (TN), while DIS of abaxial showed a positive correlation with GNP.
