Nematodes Associated with Rice in Korea - II. A Test of Rice Resistance and Control Effect of Hot Water Treatment to White Tip Nematode (한국(韓國)에 있어서 벼기생선충(寄生線蟲)에 관(關)한 연구(硏究) - II. 벼잎선충(線蟲)(Aphelenchoides besseyi)에 대(對)한 저항성(抵抗性) 품종(品種) 검정(檢定) 및 온탕침법(溫湯侵法)에 의(依)한 방제효과(防除效果)에 대하여)

  • This study was carried out to investigate the effect of hot water immersion treatment on the control of white tip nematode (Aphelenchoides besseyi) in rice seeds. Disease incidence and yield losses induced by white tip nematode were alos investigated in association with resistance of various rice cultivars. The hot water immersion treatment of rice seeds at $61^{\circ}C$ for 10-15 minutes was effective for the control of the white tip nematode. The stem of most Japonica-type cultivars was damaged by white tip nematode. The Tongil-type cultivars, however, were not infected by the nematode at all. Diseas incidence of Japonica-type cultivars, Odaebyeo and Bogkwangbyeo were 95.6% and 81.8%, respectively. The number of nematodes in infected rice seeds of both Japonica-type and Togil-type cultivars was over 100 per 10gr rice seeds. Although Tongil-type cultivars didn't show white tip symptoms, the yield loss was found in many cultivars. The yield was reduced by 40% Japonica-type cultivars, Odaebyeo and Bogkwangbyeo that were infected by white tip nematode. The yield loss in Japonica-type cultivars was more severe than that in Tongil-type cultiveas. The resistance of Youngdege 5 was highest than any other cultivars showing no disease symptoms and yield loss.

Resistance of Cultivars of Chinese Cabbage to Plasmodiophora brassicae Isolates of Several Races Collected in Korea (국내 재배포장에서 수집한 뿌리혹병균(Plasmodiophora brassicae) 균주들에 대한 배추 품종들의 저항성 반응)

  • Clubroot disease caused by Plasmodiophora brassicae, induces damage to cruciferous vegetables worldwide. For control of the disease, many CR (clubroot resistant) $F_1$ hybrid cultivars of Chinese cabbage have been bred and released in Korea. In this study, we determined the race of 10 field isolates of P. brassicae collected from ten regions in Korea using Williams' differential varieties and investigated the degree of resistance of 25 commercial CR cultivars of Chinese cabbage to the isolates. The clubroot pathogens were assigned into two (HS and YC), two (HN1 and HN2), two (DJ and SS) and four (GS, GN, JS, and PC) isolates for race 2, race 4, race 5, race 9, respectively. All CR cultivars showed similar response, resistant or susceptible, to each isolate and the P. brassicae isolates were divided into two groups. Among them, the DJ, GS, GN, HS, and JS isolates could not infect the CR cultivars. In contrast, the SS, HN1, HN2, PC, and YC isolates caused severe clubroot disease on the CR cultivars like susceptible cultivars. Even though they belong to the same race, the CR cultivars showed a different response to the pathogens. The results suggest that the breakdown of CR in Chinese cabbage has already occurred in cultivation areas of Korea and resistance source introduced in CR cultivars may be very limited. In addition, it is likely that resistance genes of Williams' differential varieties to P. brassicae are different from the gene of CR cultivars of Chinese cabbage used in the study.

Impact of low temperature during ripening stage, amylose content and activities of starch biosynthesis in rice endosperm

  • This research study was conducted to analyze the characteristics of different rice cultivars in abnormal temperature conditions (low temperature) for ripening period abnormalities, and to investigate the physiological causes behind the abnormalities. Four Korean high quality japonica-type rice cultivars, Jinbu (JB), Junamjosaeng (JJ), Geumyoung (GY), Hwawang (HW) were used in the experiment. The following day after flowering, they were then moved into two phytotrons under natural daylight with 65% RH but controlled at different temperatures - one at $19/29^{\circ}C$ (night/day) and the other at $13/23^{\circ}C$ as the low - temperature study on ripening. For the cultivars at $13/23^{\circ}C$ (low temperature study), JB and JJ had a ripening rate of 93% which is similar to the ripening rates of cultivars at $19/29^{\circ}C$ at 45 days after heading (DAH). In contrast, GY and HW recorded lower ripening rates of 86% and 57% respectively. However, when the cultivars at $13/23^{\circ}C$ were harvested at 61 DAH (when the accumulated temperature reached $1100^{\circ}C$), the difference in ripening rates compared to the 4 cultivars of $19/29^{\circ}C$ harvested at 45 DAH was not obvious (JB 94%, JJ 97%, GY 97%, HW 88%). Starch content showed little difference among the 4 cultivars at different temperature conditions while amylose content was higher for cultivars at $13/23^{\circ}C$ compared to those at $19/29^{\circ}C$. In addition, the enzyme activities of starch biosynthesis were about 5~10 days slower in cultivars at $13/23^{\circ}C$ compared to cultivars at $19/29^{\circ}C$. The grain-filling rate showed highly significant correlations with the enzyme activities of Sucrose synthase ($R^2=0.70^{***}$), ADP glucose pyrophosphorylase ($R^2=0.63^{***}$), UDP glucose pyrophosphorylase ($R^2=0.36^{***}$), Starch synthase ($R^2=0.51^{***}$), and Starch branching enzyme ($R^2=0.59^{***}$). Among the enzymes, Sucrose synthase activity had the highest correlation coefficient with grain-filling rate. In conclusion, the activity of enzymes such as Sucrose synthase, UDP glucose pyrophosphorylase, ADP glucose pyrophosphorylase, Starch synthase, Starch branching enzyme in starch biosynthesis is proven to be highly related to the grain filling process. Notably, the decrease in the activity of Sucrose synthase and Starch branching enzyme and the late increase in ADP glucose pyrophosphorylase activity at low temperature in the ripening stage are considered to be disadvantageous as they delay ripening and increased amylose content.

Quality and Physicochemical Characteristics of the Korean Cowpea Cultivars Grown in Different Seeding Periods (품종 및 파종시기별 동부의 품질 및 이화학 특성)

  • To begin with, this study identified and reviewed the proximate compositions, quality and physicochemical characteristics of three Korean cowpea (Vigna unguiculata L. Walp.) cultivars grown in two different seedling periods. The proximate compositions of the cowpea showed significant differences according to three cultivars (Seowon, Yeonboon and Okdang) and two different seeding periods (5 June and 5 July). Through this lens, the lightness, redness and yellowness of the cowpea cultivars increased slightly during the 5 July cultivation compared to the 5 June cultivation. This brings us to understand that the water binding capacity, water solubility index, and swelling power were significantly different among the cultivars. In this case, the water binding capacity of the cowpea was 108.87~143.19%, and decreased during the 5 July cultivation compared to the 5 June cultivation. As shown, the total polyphenol and flavonoid contents of cowpea showed significant difference according to cultivars. In this case, the total polyphenol content of 5 June and 5 July cultivation on the Seowon 4.17 and 4.12 mg GAE/g, respectively. In these terms, the DPPH radical scavenging activity of cowpea showed a significant difference according to the cultivars, and the Yeonboon and Okdang increased slightly during 5 July cultivation. The ABTS radical scavenging activity of the cowpea showed significant difference according to the cultivars, and all cultivars decreased slightly during the 5 July cultivation. In other words, the phenol contents and radical scavenging activity differ measurable depending on the cultivars and seeding periods. Therefore, it is necessary to study the proper cultivar and seeding time considering the environment of the applicable cultivation area.

Evaluation of Agronomic Characteristics and Forage Production of Domestic and Foreign Italian Ryegrass Cultivar in Korea (국내육성 및 외국도입 이탈리안 라이그라스 품종의 생육특성 및 사초 생산성 평가)

  • This experiment was conducted to evaluate the agronomic characteristics and forage production of Italian ryegrass cultivars at three locations (Gurae, Sungju and Cheonan) from 2013 to 2014. The experiment employed a randomized complete block design with three replications. The seven cultivars used in this experiment were 'Korwinearly', 'Florida80', 'YounbongI', 'YounbongII', 'YounbongIII', 'KeimyungI' and 'KeimyungII' varieties. Italian ryegrass varieties were seeded on October 4 at Gurae, October 8 at Seongju and October 3 at Cheonan in 2013. The Italian ryegrass varieties were harvested on April 26 at Gurae and Seongju, and on April 24 at Cheonan in 2014. Although the seven cultivars had resistance to foliar diseases and insects and high winter survival, they has little lodging resistance. The heading dates for 'Younbong I', 'Younbong II' and 'Keimyung I' were earlier than for other varieties at the three locations. Dry matter (DM) content and plant height in the early maturing cultivars were higher than in other cultivars at the three locations. The fresh and DM yields of the early maturing cultivars were also higher than others at the three locations. The results of this experiment indicate that the heading date for early maturing cultivars is earlier, and the plant height is higher than for other cultivars. Early maturing cultivars had higher DM content as well as fresh and DM yields of Italian ryegrass. Therefore, early maturing cultivars should be recommended as high-yield forage in double-cropping systems because of their superior production of forage and early heading date.

Physicochemical Characteristics and Functional Components of Mudeungsan Watermelon and the other Cultivars from Korea (일반 수박과 무등산 수박의 이화학적 특성)

  • Physicochemical characteristics of Citrullus lanatus cv. Mudeungsan and 5 other cultivars of Citrullus lanatus were investigated. The color of watermelon (redness) was not significantly different among the cultivars. Hardness ranged from 38.76 to 52.12 force (g) among cultivars. Soluble solid content in Taeyangggul and Mudeungsan were higher than that in other cultivars. pH ranged from 5.39 to 6.02 among cultivars, and total acidity was the highest in Mudeungsan (0.147 mg/100 g). No significant difference in total carotenoid content was observed among the cultivars. Lycopene content in Mudeungsan cultivar was higher than that in other cultivars. Citrullin contents of watermelon flesh and rind in Mudeungsan and GangryeokSambok were higher than those in other cultivars. The major free sugars of watermelon were sucrose and fructose, and the predominant organic acids were succinic acid and citric acid. In conclusion, Mudeungsan cultivar had better properties as outlined above and contained soluble solids and functional components, compared to the other watermelon cultivars.

Discrimination of Korean Soybean Cultivars by SSR Markers (SSR 마커에 의한 한국 콩 품종의 판별)

  • The objective of this study was to develop a technique for the cultivar discrimination using SSR markers in soybean. A total of 91 soybean cultivars developed from 1913 to 2002 in Korea were evaluated by five polymorphic SSR markers (Sat_043, Sat_036, Sat_022, Sat_088 and Satt045). Five SSR markers generated a total of 64 alleles and the number of alleles for each SSR marker ranged from 10 to 15 with average of 12.8. Polymorphic information contents (PIC) by five markers of 91 cultivars were ranged from 0.790 to 0.905 with average of 0.857. A total of 82 cultivars (90%) among 91 soybean cultivars could be individually discriminated by combination of five SSR markers through five step analysis. A cultivar, Buseok, by Sat_043 at the first step, 34 cultivars including Hojangkong by Sat_036 at the second step, 29 cultivars including Dankyeongkong by Sat_022 at the third step, 12 cultivars including Sinpaldalkong 2 by Sat_088 at the fourth step, and 6 cultivars including Saebyeolkong by Satt045 at the fifth step were discriminated. Soybean cultivars which were not discriminated by SSR markers could be discriminated by morphological characteristics.

Evaluation of Agronomic Characteristics and Dry Matter Yield of Forage Rye for High-Yielding (우량품종 선발을 위한 사초용 호밀의 생육특성 및 건물수량 평가)

  • This experiment was performed to evaluate the agronomic characteristics and dry matter yields of the early maturing cultivars(c.v.'Koolgrazer', '9909,' 'Ol-homil' and 'Jlee select') the medium maturing cultivar(c.v. 'Homil 22') and late maturing cultivars(c.v. 'Danko', 'Kior', 'Amilo' and 'Spooner'.) Seeds were sown on October 3, 2002 and October 8, 2003, respectively at the Livestock Experiment Farm of Keimyung College, Seongju, Kyeongbuk Province. Forage rye was harvested on April 22, 2003 and April 23, 2004, respectively. The height of early maturing cultivars were higher than that of late maturing cultivars at the harvest. Fifty percent heading date of the early maturing cultivar was on April 14 and that of late maturing cultivars were on April 24. In the first year, dry matter(DM) yield of 'Jlee select' was significantly the highest in among all cultivars(p < 0.05). DM yield of early maturing cultivars were higher than that of medium and later maturing cultivars. In the second year, DM yield of '9909' was significantly the highest in among all cultivars(p < 0.05). The early maturing cultivar of 'Koolgrazer' and 'Ol-homil' were similar and 'Jlee select' was low, late maturing cultivar of 'Danko', 'Kior', 'Amilo' and 'Spooner' were lower than early maturing cultivar. According to the results obtained from the study, it is suggested that the early maturing cultivar would be recommendable for fall sown rye for double cropping forage production.

Starch characteristics of foxtail millet and sorghum cultivars grown in Korea (국내 육성 조 및 수수 전분의 이화학적 특성)

  • In this study, we investigated the starch characteristics of foxtail millet (FM) and sorghum (SG) cultivars grown in Korea. The amylose contents were 29.42 and 6.42% in the Daname and Samdachal FM cultivars, and 9.09 and 11.11% in the Nampungchal and Sodamchal SG cultivars, respectively. The amylopectin polymerization analysis showed that the highest degree of polymerization (DP) was in the range of DP13-24 for all samples, at approximately 60%. The resistant starch content was very low (<0.10%) among the FM and SG cultivars. The starch gelatinization analysis showed low setback values in the Samdachal and Nampungchal cultivars. The gelatinization enthalpy, calculated based on an endothermic reaction, did not differ significantly between the SG cultivars but was significantly higher in the Samdachal FM cultivars than in the Daname FM cultivars. The results of this study might be used as basic data for the development of FM and SG products.

Morphological Characteristics and Classification of Zizyphus Cultivars in Korea by Multivariative Analysis (다변량 분석에 의한 국내산 대추나무 품종의 형태적 특성과 유연관계)

  • The objectives of this study, an analysis of fruit and leaf morphological characteristics among the five Zizyphus cultivars could be used for the investigation of cultivars classification and could provide information to make out the UPOV TG(Test Guidelines). ANOVA tests showed that there were statistically significant differences in all fruit and leaf morphological characteristics among the five Zizyphus cultivars at 1% level. But, for kernel characteristics, differences were statistically non-significant among the cultivars. Approximately, the Wolchul and Boeun cultivars showed larger and smaller values in overall characteristics and cultivars, respectively. The results of principal component analysis(PCA) for the fruit and leaf morphological characteristics showed that the first for principal components(PC's) explained about 65.3% of the total variation. The first PC was correlated with those characteristics that were mainly related to the terminal leaf length(TLL), leaf length(LL), fruit length(FL), terminal leaf width(TLW), and leaf petiole length(LPL). The second and third PC was mainly correlated with the terminal leaf morphological index(TLMI). Therefore, these characteristics were important to analysis of the fruit and leaf morphological characteristics and classification among the five Zizyphus cultivars. Cluster analysis using UPGMA method based on principal components showed that five Zizyphus cultivars could be clustered into two groups. Group I comprises Mudung, Wolchul, and Bokjo and Geumsung cultivars, Group II is Boeun cultivar. These results well similar to that of principal component analysis.
