Effects of Sonication and Vacuum Infiltration on Agrobacterium-Mediated Transformation in Immature Embryos of Korean Wheat Genotypes

  • The effects of sonication and vacuum infiltration on transformation efficiency was investigated by using immature embryos of Korean wheat as explants. Two Agrobacterium tumefaciens strains, KYRT1 and EHA105, carrying pCAMBIA 1305.1 were used. Transformation efficiency was demonstrated by the detection of $\beta-glucu-ronidase$ (GUS) activity. GUS expression showed clear difference among Korean wheat cultivars. Geurumil showed higher GUS expression efficiency $79.1\%$ compared with other cultivars. The effects of the duration of vacuum infiltration and sonication treatment showed a tendency high GUS expression efficiency by their combination. In comparison with other Agrobacterium strains, KYRT1 showed high efficiency in most Korean cultivars.

Amino Acid Composition and Antioxidative Activities of Mutant Pyropia yezoensis (방사선 돌연변이 방사무늬김(Pyropia yezoensis)의 성분 분석과 항산화 활성)

  • Recently, several mutant laver Pyropia yezoensis strains were developed. In this study, control laver Pyropia yezoensis and its mutant Py500G were compared for their amino acid contents and antioxidative activities. There was no significant difference of total amino acid contents between wild-type and mutant Py500G, but mutant had higher free amino acid contents than wild-type. Also, total phenolic content was higher in mutant Py500G. Phycoerythrin and phycocyanin contents were higher in wild-type. But, antixoidative activities were shown to be higher in mutant Py500G. These results will be useful for the development of new cultivar of Pyropia.

Plant Regeneration from Seed-Derived Callus in Perennial Ryegrass (Perennial ryegrass의 종자유래 캘러스로부터 식물체 재분화)

  • This study was camed out in order to establish plant regeneration via seed-derived callus of perennialryegrass.Varietal difference in callus growth and plant regeneration was obvious between two cultivars of perennialryegrass. "Reveille" showed a relatively high capacity for plant regeneration. The MS medium was superiorto SH or Bj in callus formation and plant regeneration. The highest regeneration frequency (60%) from calluswas obtained in presence of 5 mg/ l 2,4-D and 1 mg/ t kinetin. Regeneration response varied among calluscultures initiated from the same cultivar. Regeneration frequency was the most effective in 6 weeks-old calliafter initiation and lost their regeneration capacity gradually over a period of 12 weeks.(Key words : Perennial ryegrass (Lolium perenne L.), Plant regeneration)e L.), Plant regeneration)

Breeding of Korean Raisin Tree (Hovenia dulcis var. koreana Nakai) for High Productivity in Korea

  • For the purpose of breeding a high-yield superior variety of Korean raisin tree (Hovenia dulcis var. koreana Nakai), whose value as an edible and medicinal resource is increasing, tree candidates for superior individuals were selected from its 11 habitats in Korea from 1996 to 1998. A clone bank preserve was created in 1998 with 70 clones proliferated by grafting; the fruition traits (e.g., the number of fructified laterals, the average number of bunches per fructified lateral, the average number of bunches per fruiting lateral, the fruitpetiole weight per individual, and the yield per individual) of 47 clones that had bloomed and borne fruit were investigated and analyzed in 2002; five upper-ranking clones whose yield per individual exhibited a 261% improvement against the total average were picked in 2005; and three clones, including 'Poong-Sung 1', that showed a difference in their fruit petiole ripening stage, were finally selected in 2007 as high-yield new cultivars of Korean raisin tree.

Response of Kneeing Ability of Rice Varieties and Echinochloa Species (벼 품종(品種)과 피의 굴기력(屈起力) 반응(反應))

  • The experiment was carried out at the Yeongnam Crop Experiment Station in 1984 and 1990 to obtain the basic information on kneeing responses of rice cultivars and Echinochloa species. Kneeing response of rice cultivars was not directly related with cultivar group(Tongil-type vs Japonica-type), maturity, culm length, or other agronomic traits and thus concluded that kneeing ability was soly peculiar cultivar character based on its genetic background. Cultivars having relatively great kneeing ability were Yeongdeog 1(Japonica), Seogwangbyeo(Tongil-type). Somjinbyeo(Japonica), Pungsanbyeo(Tongil -type), Hangangchalbyeo(Tongil-type), etc. while these for low kneeing ability cultivars were Chilseoungbyeo, Gayabyeo, and Samgangbyeo that were all belonged to Tongil-types, respectively. Two Echinochloa species were not much differed from each other in kneeing ability. However, kneeing ability of Echinochloa species was much greater than rice cultivars. Kneeing ability of Echinochloa species was not differed by culm node order : all the visual nodes(5~6 nodes) exhibited similar kneeing ability having more or less 30${\circ}$ in each node.

The Development of Container-type Plant Factory and Growth of Leafy Vegetables as Affected by Different Light Sources (컨테이너 식물공장의 개발과 이를 활용한 광원별 엽채류의 생장특성)

  • For the energy-saving production of fresh vegetables in poor environment such as the Antarctic, a container-type plant factory was designed and developed. To maximize space usage of the 20 feet container ($L5.9m{\times}W2.4m{\times}H2.4m$), a three-level hydroponic cultivation system was installed and the nutrient solution was supplied by bottom watering. Using this system, 3 lettuce cultivars were grown under different the light source (light intensity). After 2 weeks from the transplanting, fluorescent lamp ($145\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) showed the best fresh weight of top part and leaf area. However, After 4 weeks, fluorescent lamp plus metal halide lamp ($150\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) led to the optimum growth of the each lettuce cultivar. The cultivar, 'Cheongchima', showed the best fresh weight of top part and leaf area, followed by 'Jeokchukmyeon' and 'Lollo rosa'. The chlorophyll concentrations (SPAD) showed no significant difference among the sources of lights. However, 'Cheongchima' showed relatively high chlorophyll concentration. With the above results, we found that the growth of lettuce is depend on light intensity and even at same intensity, the growth is different among the cultivars. Therefore, the selection of optimum cultivar should be considered in the plant factory system that has only weak light density.

Effect of seed priming on germination and sprouting vigor of colored rice

  • The study was conducted to select optimal materials for promoting germination rate, high sprouting vigor by priming treatment using PEG 6000 (water potential -0.5 to 2.0 Mpa) and Azolla extracts (0.1, 1.0, and 10.0%) extracted by ethanol, distilled, and/or hot water in colored rice cultivars. Each rice seed (three black rice cultivars and two red rice cultivars) was soaked 24 and 48 hr including untreated control. In black rice, Joseongheugchal rice cultivar, azolla ethanol extract (0.1%) induced highest germination rate, germination speed was taken to 5 days in distilled water and to 3days in Azolla extracts extracted hot water. Otherwise, degree of bacterial inhibition (number of colony, $10^3cfu$) in dry seed, water soaking for 24hr, soaking with fungicide for 24hr and 48hr, soaking with fungicide and aeration for 24hr and 48 hr was 22, 500, 95, and 0.46, respectively. In order to minimize fungal inhibition, a method can be chosen to combination of soaking fungicide and aeration for 48 hr. In seed priming treatments using growth pouch, seed soaking with fungicide did not affect change of germination percentage and germination speed, it delayed only 2 or 4day in the Joseongheugchal rice cultivar. It differs from rice cultivars and priming materials, Azolla extract(0.1 to 1%) promotes seed germination percentage in the Ilpum, Hongjinju, and Joseonghuegchal, in addition, germination in Jeogjinju cultivar was only promoted by PEG solution(10 to 20%), otherwise, it showed much lower or inhibited on the germination in Heugjinju and Sinmyungheugchal rice cultivars. In a paddy field trial, seedling establishment rate by applying PEG6000 and azolla extract did not show significantly statistical difference. When it compared with untreated control, seedling establishment rate was increased over 50% in priming treatments. Interestingly, seedling establishment rate under azolla extract (0.1%) extracted with ethanol was promoted over 2.5 times compared to the control in a black rice, Joseongheugchal and red rice, Jeogjinju.

Differences of Growth Characteristics and Colorant Level in Two Breeding Lines of Persicaria tinctoria H. Gross (쪽 선발계통의 생육특성 및 색소함량 차이)

  • Indigo crop, Persicaria tinctoria H. Gross is an annual plant containing natural colorant, the blue dye indigo, and local cultivars had been cultivated to produce natural indigo for textile dyeing in Korea since ancient times. Naju No. 2 is a new mass-selected line from the mother population, Naju Local cultivar. In this study, two breeding lines of Naju Local and Naju No. 2, have been cultivated in four different locations, the South regions of Korea, to compare plant growth and yield characteristics between two lines. Naju No. 2 was higher in plant height, and Naju Local has more 1st branches. Naju No. 2 has larger leaf area and higher width/length ratio of leaf, showing the round leaf type as morphological stable character without regional differences. Though there was considerable regional variation in fresh and dry top weight of harvested plant, the significant difference of plant weight between two lines were not shown. The ratio of leaf to total shoot of dry weight of Naju No. 2 was higher than one of Naju Local, indicating that Naju No. 2 has better yielding of colorant which is synthesized mostly in leaf. Naju No. 2 contained more Niram (crude indigo extract) and indigo, and showed much blueness at dyeing of silk using fresh leaves than Naju Local. We concluded that a new line, Naju No. 2 could be a superior cultivar due to having higher leaf yield and better quality of natural colorant than Naju local cultivar.

Effect of Cut on Forage Production and Grain Yield of Naked Barley Cultivars (예취가 쌀보리 품종의 청초생산과 종실수량에 미치는 영향)

  • Eight naked barley cultivars were grown in three production systems to select proper cultivars for dual production of forage and grain and to determine production system x cultivar interactions. In the forage systems. barley was seeded on September 27 and October 17, harvested for forage on November 1. December 10 and February 12 from the September 27 planting (forage system I) and on December 10 and February 12 from the October 17 seeding(forage system II). In the grain-only system, barley was planted on November 5. In forage system I, oven-dried forage yields of eight cultivars ranged 195 to 296kg/10a and Saessalbori and Naehanssalbori yielded significantly higher than the other cultivars recording 280 and 296kg/ l0a, respectively. In forage system II, oven-dried forage yields of eight cultivars ranged 106 to 143kg/10a showing no significant difference among cultivars. Production system x cultivar interactions were significant for lodging at maturity, powdery mildew rating, 1000 kernel weight and grain yield. Leaf area index and biomass at heading, no. of spikes per $m^2$, no. of kernels per spike and test weight were not affected by the production system. Forage utilization delayed heading by 3 days and reduced culm length, spike length, lodging due to heavy rain on May 5 and lodging at maturity except for Songhagbori and Naehanssalbori. Forage utilization did not significantly affect grain yield from the September 27 planting but reduced 9% from the October 17 planting. while Saessalbori and Hyangcheonggwa 1 yielded significantly less than in the grain-only system. Songhagbori appears to be a proper cultivar for dual production of forage and grain in Cheju, considering forage and grain yields. and lodging and powdery mildew resistance.

Growth, Yield and Photosynthesis of Introduced Kenaf Cultivars in Korea (신도입 케나프 품종의 파종시기에 빠른 생육 및 수량 변동과 광합성 특성)

  • Kenaf (Hibiscus cannabinus L.), an annual plant of the family Malvaceae, is considered to be the most promising for alternative plants with potential use as a non-wood fiber source. The objectives of this study were to select the optimum seeding date and adaptable cultivar among newly introduced kenaf cultivars (Everglades-41, Tainung-2 and Chingpi-3) in Jeju island, and to clarify the photosynthetic characteristics of those cultivars. Among the three cultivars, the fresh shoot weight per unit area of Chingpi-3 at harvest season showed highest through all seeding dates, that of while Everglades-41 was the lowest. The difference of shoot yield at harvest mainly due to seedling stand rate and plant number per unit area among the cultivars. The Chingpi-3 showed the highest shoot fresh weight in the seeding date of May 11 and decreased trend in the late seeding dates. Net photosynthesis rate was higher on Everglades-41 with entire type leaf than other two cultivars with palmate type leaf. The activity of ribulose-1,5-bisphosphate carboxylase/foxygenase (rubisco) estimated from the A-Ci curve showed highest in Chingpi-3 among three cultivars. These results suggest that Chingpi-3 might be adaptable cultivar with seeding date of around May 10 for kenaf production in Jeiu island.
