Efficiency and Midgut Histopathological Effect of the Newly Isolated Bacillus thuringiensis KS ${\delta}$-Endotoxins on the Emergent Pest Tuta absoluta

  • Tuta absoluta (Povolny, 1994) is a devastating moth to the Solanaceae plants. It is a challenging pest to control, especially on tomatoes. In this work, we studied the entomopathogenic activity of the Cry-forming ${\delta}$-endotoxins produced by Bacillus thuringiensis strain KS and B. thuringiensis kurstaki reference strain HD1 against T. absoluta. These strains carried the cry2, cry1Ab, cry1Aa/cry1Ac, and cry1I genes, and KS also carried a cry1C gene. The ${\delta}$-endotoxins of KS were approximately twofold more toxic against the third instar larvae than those of HD1, as they showed lower 50% and 90% lethal concentrations (0.80 and 2.70 ${\mu}g/cm^2$ (${\delta}$-endotoxins/tomato leaf)) compared with those of HD1 (1.70 and 4.50 ${\mu}g/cm^2$) (p < 0.05). Additionally, the larvae protease extract showed at least six caseinolytic activities, which activated the KS and HD1 ${\delta}$-endotoxins, yielding the active toxins of about 65 kDa and the protease-resistant core of about 58 kDa. Moreover, the histopathological effects of KS and HD1 ${\delta}$-endotoxins on the larvae midgut consisted of an apical columnar cell vacuolization, microvillus damage, and epithelial cell disruption. These results showed that the KS strain could be a candidate for T. absoluta control.

Insecticidal Activity and Molecular Characteristics of Bacillus thuringiensis CAB530 Isolated from Anomala albopilosa (Rutelidae: Coleoptera) (청동풍뎅이에서 분리한 Bacillus thuringiensis CAB530 균주의 살충활성 및 분자학적 특성)

  • Bacillus thuringiensis CAB530 was isolated from dead Anomata albopilosa (Rutelidae: Coleoptera) and soil of green tea field, and confirmed its insecticidal activities. CAB530 isolate showed a high insecticidal activity against the beet armyworm among the many lepidopteran insects that are difficult to control. $LC_{50}$ value of CAB530 isolate against the second larva of Spodoptera exigua was $1.49{times}10^4$ spore concentration (cfu/$m{\ell}$). SDS-PAGE result of insecticidal toxin protein of CAB530 isolate showed a band at 130 kDa that is similar pattern with B. thuringiensis subsp. kurstaki that took insecticidal activity against S. exigua. Otherwise, the crystal protein of the CAB530 isolate was conformed at 65 kDa level after 30 minute of incubation in S. exigua midgut juice. Six crystal genes (cry1Aa, cry1Ab, cry1C, cry1D, cry1F and cry1I) were identified by PCR. It different from genes of B. thuringiensis subsp. kurstaki. Crystal shape and pattern of toxin protein was similar with B. thuringiensis subsp. kurstaki, however, insecticidal activity and PCR result of CAB530 isolate was similar with B. thuringiensis subsp. aizawai.

Spacing Effect of the Intervening Sequences between Ribosome Binding Site and the Initiation Codon on Expression of Bacillus thuringiensis $\delta$-Endotoxin

  • To verify importance of the intervening sequence between the ribosome binding site (RBS) and the initiation codon for expression of Bacillus thuringiensis $\delta$-endotoxin, the pProMu, containing SphI and NcoIsites between RBS and the initiation codon of the cry1Ac gene, and the deletion derivatives of pProMu were constructed and transformed into the B. thuringiensis subsp. kurstaki $Cry^{-B}$ strain. The pProMu-ΔSphIhad identical six bases of intervening sequence to pProAc though the arrangement of sequence was different. Other mutants containing pProMu had 1 or 10 or 14 bases between RBS and the initiation codon. Among deletion mutants, only ProMu-ΔSphI/CB only produced 130 kDa typical bipyramidal crystals like those seen for ProAc/CB. However, ProMu/CB, $ProMu-{\Delta}NcoI$, and ProMu-ΔSphI+NcoIdid not produce Cry1Ac crystals. In conclusion, the results suggest that 6-base intervening sequence was important for expression of cry1-type class gene. Furthermore, spacing effect of the intervening sequences may play an important role in expression of individual crystal proteins in B. thuringiensis without doubt.

The Layer of Emotion that Makes up the Poem "Falling Flowers(落花) " by Cho Ji-Hoon

  • This study of Cho Ji - Hoon's Poem "Falling Flowers" was attempted to find the mechanism of poetic healing and utilize it for literary therapy. In this study, I examined how Cho Ji-Hoon's poem "Falling Flowers" encoded crying. Especially, we focused on the organic relationship of each layer represented by poem and put emotional codes on the layer of functor and argument. The results are as follow. It represents the Separation Layer of 1-3strophes, 4-6strophes constitute the Time Layer, and 7-9strophes the Sadness Layer. This poem proceeds the encoding of the sentence in which the crying of cuckoo in the 1-3strophes transforms into the crying of the poetic narrator in the last 9strophe. The relation of emotional layers in this poem is in the same function relations as "(1-3strophes) ${\subset}$ (4-6strophes) ${\subset}$ (7-9strophes)". Since these functional relations consist of the encoding of sadness, encrypts emotion signals of sadness as "U+U+U" becomes "UUU". 1-3strophes' U is the cry of the cuckoo, and U of the 4-6strophes is blood cry. Therefore, "UUU" is the blood cry of poetic narrator. This Cho Ji-Hoon's poem has a Han(恨) at its base. So, as Cho Ji-Hoon's poem "Falling Flowers" is uttered, the poetic mechanism of U, the code of sadness, is amplified. Then we get caught up in the emotions we want to cry. The poetic catharsis of "crying" is providing the effect of literary therapy. In the future, it will be possible to develop a more effective literary therapy technique by developing a literary therapy program like this poetic structure.

Directed Mutagenesis of the Bacillus thuringiensis Cry11A Toxin Reveals a Crucial Role in Larvicidal Activity of Arginine-136 in Helix 4

  • Based on the currently proposed toxicity model for the different Bacillus thuringiensis Cry $\delta$-endotoxins, their pore-forming activity involves the insertion of the ${\alpha}4-{\alpha}5$ helical hairpin into the membrane of the target midgut epithelial cell. In this study, a number of polar or charged residues in helix 4 within domain I of the 65-kDa dipteranactive Cry11A toxin, Lys-123, Tyr-125, Asn-128, Ser-130, Gln-135, Arg-136, Gln-139 and Glu-141, were initially substituted with alanine by using PCR-based directed mutagenesis. All mutant toxins were expressed as cytoplasmic inclusions in Escherichia coli upon induction with IPTG. Similar to the wild-type protoxin inclusion, the solubility of each mutant inclusion in the carbonate buffer, pH 9.0, was relatively low When E. coli cells, expressing each of the mutant proteins, were tested for toxicity against Aedes aegypti mosquito-larvae, toxicity was completely abolished for the alanine substitution of arginine at position 136. However, mutations at the other positions still retained a high level of larvicidal activity Interestingly, further analysis of this critical arginine residue by specific mutagenesis showed that conversions of arginine-136 to aspartate, glutamine, or even to the most conserved residue lysine, also abolished the wild-type activity The results of this study revealed an important determinant in toxin function for the positively charged side chain of arginine-136 in helix 4 of the Cry11A toxin.

  • Porous silica ceramics were prepared using DCCA(Drying Control Chemical Additives) Such as uncharged polymer(Polyethylene glycol) and protein (Lipase) under H2O/Low-grade TEOS=10 C2H5OH/Low-grade TEOS=1 HC1/Low=grade TEOS=0.01 After Plain which doesn't added DCCA and samples of 11 sorts which varied molecular weight of PEG(Mw=600, 1000, 2000) quantity of Lipase and concentration of wat-er were synthesized gellation time and thermal analysis were investigated. After heat-treated at 600, cry-stal structures analyses of SiO2 polymer and characteristics of pores were investigated. Gellation time was retarded about 2-6 times as compared with plain resulting in addition of DCCA and crystal structures ex-hibited amorphous state. Moreover as increase of water a short gellation time was obtained. The samples added PEG showed increase of specific surface areas up to 20-40% and had micropores while those of Lipase were decreased about 90% and showed broad pore size distribution.

Effectiveness of various nonpharmacological analgesic methods in newborns

  • Background: Pain during the developmental period may adversely affect developing neuronal pathways and result in adverse neurodevelopmental, cognitive, and behavioral effects in later life. Immunizations, e.g., hepatitis B vaccine (HBV), administered at birth are painful experiences to which neonates are universally subjected. Purpose: Here we aimed to study and compare the effectiveness of various nonpharmacological pain management methods in newborns to enable the development of safe and effective analgesic methods for newborns. Methods: This prospective study was conducted at a tertiary care hospital in the Himalayan region. Three hundred term healthy neonates were divided into 6 groups of 50 each. Groups 1-5 were intervention groups, patients of which received a nonpharmacological intervention (breastfeeding, nonnutritive sucking, rocking, 25% sucrose, or distilled water) before the intramuscular HBV, while patients in group 6 received no intervention. The pain response in each group after the HBV injection was assessed and compared using cry duration and Douleur Aigue Nveau-ne (DAN) score, a behavioral acute pain rating scale for newborns. Results: Cry duration was decreased in all intervention groups, significantly so in the sucrose (19.90 seconds), breastfeeding (31.57 seconds), and nonnutritive sucking (36.93 seconds) groups compared with controls (52.86 seconds). DAN scores decreased significantly (P<0.05) at one or more points i.e. 30, 60, or 120 seconds in the breastfeeding and 25% sucrose intervention groups compared with controls. Conclusion: Oral sucrose and nonnutritive sucking are simple yet underutilized nonpharmacological interventions that effectively reduce pain in newborns.

  • If baby disease not treats at early, case that cause problem in health hereafter much exists. Specially, because baby expresses own inexpedience by crying, parents or practitioner's is not apt to grasp. Therefore, In this paper, I wish to develop system for analyze if disease is in some region to analyze face and baby cry that organism signal appears intensive for baby disease diagnosis. In this paper, I wish to propose methodology that analyze organism signal that appear in face of whole baby organism signal analysis system. For this, I necessary region binary input baby face picture and to draw necessary five sensory organ and Myungdang region in baby disease diagnosis. Finally, the usefulness of this paper is verified by several experiments.

  • The application of reflex media in education has been widely used in the aspects of teaching & learning method, humanitarian education and the culture of school lives. It has been resulted from the generality of the applicable scope within conveying the information, propagation velocity, and the efficiency of the amount of conveyable information through the reflex. To utilize this kind of efficiency of reflex media in producing new graduation culture, I intend to show students the graduate reflex including their 3years' school lives and try to find out its effect from them. And then with this result I have studied the changes of the students behavior in the graduation ceremony. As a result I can notice that the intent of graduation reflex which aims to look back into their past and keep in mind it has been reflected to students. In addition I can ascertain their friendship and love for their school have been lifted while watching the reflex with laugh and cry.

Comparison of Postoperative Analgesic Efficacy of Caudal Block versus Dorsal Penile Nerve Block with Levobupivacaine for Circumcision in Children

  • Background: Circumcision is a painful intervention frequently performed in pediatric surgery. We aim to compare the efficacy of caudal block versus dorsal penile block (DPNB) under general anesthesia for children undergoing circumcision. Methods: This study was performed between July 1, 2009 and October 16, 2009. Fifty male children American Society of Anesthesiolgists physical status classification I, aged between 3 and 12 were included in this randomized, prospective, comparative study. Anesthetic techniques were standardized for all children. Patients were randomized into 2 groups. Using 0.25% 0.5 ml/kg levobupivacain, we performed DPNB for Group 1 and caudal block for Group 2. Postoperative analgesia was evaluated for six hours with the Flacc Pain Scale for five categories; (F) Face, (L) Legs, (A) Activity, (C) Cry, and (C) Consolability. For every child, supplemental analgesic amounts, times, and probable local or systemic complications were recorded. Results: No significant difference between the groups (P > 0.05) was found in mean age, body weight, anesthesia duration, FLACC pain, and sedation scores (P > 0.05). However, on subsequent measurements, a significant decrease of pain and sedation scores was noted in both the DPNB group and the caudal block group (P < 0.001). No major complication was found when using either technique. Conclusions: DPNB and caudal block provided similar postoperative analgesic effects without major complications for children under general anesthesia.
