  • This study aimed to investigate the single nucleotide polymorphisms (SNPs) of the porcine MC4R gene and validate the effect of the MC4R genotype for marker assisted selection (MAS). Six amplicons were produced to analyze the entire base sequences of the porcine MC4R gene and six SNPs were detected (c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, c.892A>G-Asp298Asn, and c.*430A>T). Linkage disequilibrium (LD) of the six SNPs was analyzed by performing haploid analysis. There was a perfect linkage disequilibrium in c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, and c.*430A>T. Only the c.892A>G (Asp298Asn) SNP showed a very low LD with an $r^2$ value of 0.028 and the D' value of 0.348. As a result, the two SNPs-c.707A>G (Arg236His) and c.892A>G (Asp298Asn)-were selected to extract the genotype frequencies from the 5 pig breeds by using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) genotype analysis method. The SNP frequency of c.707A>G (Arg236His) indicated the presence of the A (His) allele only in Yorkshire, while the G allele was fixed in the KNP, Landrace, Berkshire, and Duroc. Association analysis was carried out in 484 pigs with the c.707A>G (Arg236His) SNP and the meat quality traits of four different pig cross populations: a significant association was noted in crude fat, sirloin moisture, meat color, and the degree of red and yellow coloration. The frequency of the c.892A>G(Asp298Asn) SNP genotype varied among the breeds; while Duroc showed the highest frequency of the A (Asn) allele, KNP showed the highest frequency of the G (Asp) allele. Association analysis was carried out in 1126 pigs with the c.892A>G (Asp298Asn) SNP and the meat quality traits of four pig populations: a highly significant linkage was noted in the back-fat thickness (P<0.002). It was found that the back-fat thickness was higher in individuals with the AA genotype than in those with the AG or GG genotype. Thus, in this study, we verified that the c.892A>G (Asp298Asn) SNP in the pig MC4R gene has a sufficient effect as a gene marker for MAS in Korean pork industry.

  • Dry matter productivity at silage stage of two corn hybrids varying resistance to rice blackstreaked dwarf virus(RBSDV) and two sorghum hybrids varying growth duration was evaluated at six planting dates (Apr. 1, Apr. 15, Apr. 29, May 13, May 27, and June 10) in Kyongsan where RBSDV is prevalent.Corn was harvested 38-40 days after silking. For sorghum 1st-cut was made 19-22 days after heading and some regrowth (2nd-cut) on October 13. 1. Percent stand of both corn hybrids was over 91% at all planting dates, but that of sorghum ranged 37.2-80.3% depending on hybrids and planting dates. 2. The number of days from planting to emergence decreased as planting date delayed in both crops. The number of days from emergence to silking of both corn hybrids and a sorghum hybrid(P 956) decreased as planting date delayed, but that of the other sorghum hybrid(P 931) increased significantly after May 13 planting. 3. RBSDV infection rate of corn hybrids was highest at April 1 planting (Kwanganok 24.896, Jinjuok 63.0%) and decreased up to April 29 planting and then increased as planting date delayed. Sorghum was not infected by RBSDV at all. 4. EarfTotal dry matter ratio of corn ranged 30.6-47.9% and that of sorghum 3.1-30.7% depending on hybrids and planting dates. All the hybrids developed grain normally at all planting dates except a sorghum hybrid (P 931) which developed few grains after May 13 planting. 5. Crude (C) protein and C. fat contents were similar among all treatments. C. fiber content of P 931 was higher than that of both corn hybrids and P 956, but nitrogen free extract (NFE) was lower. C. fiber and NFE of both corn hybrids and 1st-cut sorghum were similar among the planting dates, but C. fiber and NFE of 2nd-cut of sorghum decreased and C. protein increased as planting date delayed due to plants were younger. 6. Dry matter (DM) yield of all hybrids decreased as planting date delayed except Jinjuok of which yield was highest at April 29 planting. DM yield of corn was similar to 1st-cut sorghum, but lower than total DM yield of sorghum at April 1 and 15 plantings where regrowth yield was high.

  • This study was to investigate the chemical content and antioxidant activity by the part of Salicornia herbacea as part of using Salicornia herbacea as materials of functional foods. On a basis of the materials, this study was to clarify the nutritional excellence, chemical composition, and antioxidant activity of Salicornia herbacea. From the result of this research above, it may be summed up as follows. The Salicornia herbacea used in the experiment contained the moisture and crude fiber in the stalks a lot, and the protein, fat, and ash in the branches more. As to the content of free amino acid, the content of arginine took in the branches and stems most. Then, it contained threonine, glycine, tryptophan, valine, isoleucine, and lysine, etc. in the branches a lot. Also, it contained threonine, glutamic acid, tyrosine tryptophan alanine, and isoleucine in the stalks a lot. It was shown to be contained of essential amino acids like isoleucine, leucine, threonine, valine, methionine, lysine, phenylalanine, and tryptophan in the branches and steams more. As to the content of fatty acid in Salicornia herbacea, it found out that it contained the unsaturated fatty acid more than the saturated fatty acid. It took the content of eicosenoic acid(20:1) in the branches and stalks most. And then, it contained linoleic acid(18:2), pehtadecenoic acid(15:1), palmitic acid(16:0), and oleic acid(18:1), etc. a lot. As to the antioxidant activity in Salicornia herbacea using the DPPH radical, it was shown to be existed in the largest antioxidant activity when the concentration of methanolextract from Salicornia herbacea was 1 mM. There was higher antioxidant activity than 100 ppm BHT used as control plot when the concentration of methanol extract from the stalks was $100{\mu}M$. From the result of experiment above, Salicornia herbacea contained the essential amino acid a lot. It will be possible to be used as natural antioxidants because it has excellent antioxidant effect. Therefore, this researcher concludes that it will be available in using it as materials of functional foods.

  • This study aimed to provide basic evidence regarding the development of materials by analyzing the physicochemical properties and antioxidant activities of Streptococcus thermophilus KCCM 3782 strain-fermented Cordyceps militaris grown on Tenebrio molitor. Among the solvents tested, DPPH radical scavenging activity was the highest in the hot water-extracted sample after 30 min of extraction. Moisture content decreased, whereas crude protein and fat content increased, after lactic acid bacteria-mediated fermentation. Sodium, magnesium, calcium, and zinc contents increased, with potassium levels attaining the maximum value, whereas free amino acid content decreased after the fermentation. Among Hunter's color values, a value increased to $66.7{\rightarrow}149.92$ after fermentation, whereas the L and b values decreased to $15.79{\rightarrow}-15.75$ and $54.45{\rightarrow}0.01$, respectively. Cordycepin content assay increased from 7.02 mg% to 8.66 mg%. The DPPH free radical scavenging activity of the fermented product was 91.92 EDA%, which was higher than that of the extract (84.69 EDA%). The ABTS free radical scavenging and superoxide dismutase (SOD) activities were also higher in the fermented products.
