Inheritance of Anthracnose Resistance in a New Genetic Resource, Capsicum baccatum PI594137

  • Pepper fruit anthracnose caused by Colletotrichum species is an economically important disease that causes serious yield loss and quality deterioration in many Asian countries including South Korea and Taiwan. Recently, Capsicum baccatum PI594137 was found to exhibit broad-spectrum resistance to Colletotrichum acutatum. The inheritance of anthracnose resistance to C. acutatum was analyzed in an intraspecific population derived from a cross between C. baccatum Golden-aji and PI594137. Detached mature green fruits were inoculated using the microinjection method. The disease response was evaluated as the disease incidence and the overall lesion diameter at 7 days after inoculation(DAI). The segregation ratios of resistance and susceptibility to C. acutatum in the $F_2$ and $BC_s$ populations significantly fit a 3:1 Mendelian model. This result indicates that the resistance of PI594137 to C. acutatum is controlled by a single dominant gene.

  • Zoysiagrass has been used in Korea as a native grass. Informations on distributions, classifications, breeding efforts and outputs, current production and uses of zoysiagrasses in Korea are summarized. During past a few decades, collection of naturally growing zoysiagrass ecotypes has been continued, followed by evaluation, selection and cross-pollination. Classification efforts using important morphological, biochemical traits and DNA polymorphisms resulted in the several suggestions of grouping zoysiagrass species and varieties. Zoysia japonica consistently form separate group, while Zoysia sinica and Zoysia macrostackya have been separately or inter-mixed. Z. matrella with some variations frequently grouped separately but with some similarity with Z. tenuifolia, which was recently renamed as Z. pacifica. Hybrid types of zoysiagrasses were frequently found in Korea. New varieties such as 'Kunhee' and 'Green Ever' etc. were bred by ecotype selections, while 'Senock', 'Semil', 'Millock' were bred through pollination. Molecular approaches were also used to introduce beneficial genes into zoysiagrasses with no practical outputs by legal regulations. Currently popular varieties are inter-specific hybrid types such as 'Anyang' and other local zoysiagrasses as well as common zoysiagrass (Zoysia japonica) Production area in Korea was estimated as 2,947 ha with increasing problems of mixed or contaminated species and varieties.

Alstroemeria plants and its biotechnological applications

  • Alstroemeria plants are widely cultivated in many countries especially in Western Europe and North America and popularity has increased in recently due to its long-base life, large variety of colors and low energy requirement during cultivation period. So far, more than 60 species have been released on the commercial market in the world. To meet the demand of consumer and develop the elite Alstroemeria cultivars, conventional breeding including cross-hybridization and selection as well as mutation breeding were used. However, as other important ornamental plants such as lily, rose, carnation and orchids accepted the biotechnological methods, this newly-born approach should be applied and developed an optimized the genetic transformation system. Then, this biotechnological approach can be fused with the conventional breeding methods and thus can be contributed to the production of elite Alstroemeria plants containing agriculturally good genetic traits which are useful for the both farmers and consumers in the future. In this paper, we reviewed the botanical and genetical features of Alstroemeria plants and its biotechnological approaches in the last decades.

Inheritance of Resistance to Nuclear Polyhedrosis Virus in Silkworm, Bombyx mori

  • Inheritance pattern of resistance to Bombyx mori nuclear polyhedrosis virus (BmNPV) was studied in an Indian silkworm stock TX by single back-cross test method. The resistant parent [TX], susceptible parent [HM], their Fl, F2, and Fl progeny back-crossed to TX [BC(R)] and HM [BC(S)] were inoculated per os with a fixed concentration of BmNPV($0.5{\times}10^{th} PIB/ml$) on the first day of second stadium. The cumulative mortality was recorded until day $10^{\times}$ post-inoculation. The results show that the resistance to BmNPV in TX fellow mono Mendelian inheritance pattern. The resistance dominated over the susceptibility at Fl. At F2, the resistant and susceptible offspring segregated in 3:1 ratio whereas at BC(S), the resistant and susceptible offspring segregated in 1:1 ratio. The response of BC(R) was more or less like the resistant parent TX which confirms the involvement of a major dominant gene conferring resistance to BmNPV in TX. The possible mechanism of inheritance of resistance in TX is discussed.

Analysis and mapping of the re-1 gene for reduced embryo size in rice

  • The objective of this study was to map the gene for reduced embryo size in rice using DNA markers. The reduced embryo size mutant was induced from N-methyl-N-nitrosourea (MNU) treated Taichung 65. Genetic analysis revealed that the phenotype of the reduced embryo was controlled by a single recessive gene, designated as re-1. For mapping the gene controlling embryo size, an $F_2$ population was developed from a cross between the Korean Tongil-type, Milyang 23 (Oryza sativa ssp. indica) and the mutant. The ratio of $F_2$ seeds nearly fitted to 3:1 ratio, indicating that this phenotype was controlled by a single recessive gene. Bulked sergeant analysis was performed with SSR markers. The gene for the reduced embryo size was detected on chromosome 1. The gene was further mapped between two SSR markers, RM315 and RM265 on chromosome 1 (approximately 1.5 Mb interval). The linked markers will facilitate selection of this grain character in a breeding program and provide the foundation for positional cloning of this gene.

A New Spray Chrysanthemum Cultivar, "Cherry Blossom" with Resistant to White Rust, Single Flower Type and Bright Pink Petals for Cut Flower

  • A new spray chrysanthemum cultivar, 'Cherry Blossom' was released by National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), Korea in 2007. A cross was made in 2002 between 'Relance', a spray cultivar with red petals and resistant to white rust and 'Yeonja', a spray cultivar with pink petals. Trials were conducted from 2005 to 2007 for the evaluation and selection of this cultivar, including shading culture in summer and retarding culture in autumn. The natural flowering time of "Cherry Blossom" is late October, but year-round flowering is possible by shading and lighting treatment. This cultivar is single type flowers with dark pink petals and green flower center and resistant to white rust. It is very stable color of petals when the variety is cultivated under high temperature conditions in summer season. The diameter of flower is 55.0 mm. The number of flowers per stem is 10.5 and the number of petals per flower is 24.0. The days to flowering under the short day treatment is about 45 in spring season.

  • A new spray chrysanthemum cultivar 'Prima Donna' was released by National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA), in 2008. The cross was made between 'Plano' and 'Yeonja' in 2005. Trials were conducted from 2006 to 2008 for the evaluation and selection of this cultivar, including shading cultures in summer and retarding cultures in spring. The natural flowering time of 'Prima Donna' is late October, but year-round flowering is possible by photo-periodic control. The cultivar has single type flowers with pink petals, green center and good inflorescence. The growth of plant is very vigorous. The diameter of flower is 7.0cm. The number of flowers per stem and petals per flower are 13 and 42, respectively. Days to flowering under the short day treatment is about 59 and its vase life is 26.1 days in autumn season.

A cross was made in 2004 between 'Lineker Salmon', a spray cultivar with salmon petals, and 'Baeksokuk', a spray cultivar with white petals. Trials were conducted from 2005 to 2007 for evaluation and selection of this cultivar, including shade cultivation in summer and retarding cultivation in autumn. The natural flowering time of 'Pink Berry' is late October, but year-round flowering is possible by photo-periodic control. The cultivar has single type flowers with pink petals and a green flower center. It shows long vase life of 22 days and resistance to white rust. The diameter of flower is 52.0 mm. The numbers of flowers per stem and petals per flower are 15.5 and 29.5, respectively. Days to flowering under the short day treatment is about 56 in autumn seasons.

  • A New Cymbidium variety "Yellow Evening" was developed from a cross between "Oji" and "Valley Flower Cherry Life" followed by seedling and line selections at the National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) in 2004. After investigation of the characteristics and selection for 9 years (1996-2004), it has been selected specifically pot orchid flower. "Yellow Evening" has medium plant height and semi-erect peduncle. It has medium flowering habit and brownish yellow flowers (RHS, Y11B: petal and sepal) with grayed purple GP183A) spot on lip, when fully opened. Each peduncle has 15.3 flowers with diameter of about 7.4 cm. It has adequate peduncle height, with some curved and spreading petal and sepals. Under optical culture conditions, it started to bloom early-winter (November).

QTL Mapping of Cold Tolerance at the Seedling Stage using Introgression Lines Derived from an Intersubspecific Cross in Rice

  • Low-temperature stress is an important factor controlling the growth and development of rice (Oryza sativa L.) in temperate region. In this study, a molecular linkage map consisting of 136 SSR markers was employed to identify QTL associated with cold tolerance at the seedling stage. 80 recombinant inbred lines (RILs) from an intersubspecific cross between Milyang23 (O. sativa ssp. Indica) and Hapcheonaengmi3, a japonica weedy rice and the parents were evaluated for leaf discoloration and SAPD value of seedlings. Rice plants were grown for 15 days in the low-temperature condition (13/20℃ day/night) and the control condition (25/20℃ day/night) in the growth chamber. The degree of leaf discoloration showed a highly significant correlation with the SPAD value in the low-temperature plot (r = -0.708, P < 0.0001). A total of four QTLs for SPAD were identified and the phenotypic variance explained by each QTL ranged from 5.4 to 16.0%. Two QTLs detected in the control condition were located on chromosomes 2 and 5, respectively. Two QTL on chromosomes 1 and 4 were detected at the low-temperature condition and Hapcheonaengmi3 alleles increased the SPAD values at these loci. Substitution mapping was conducted to delimit the position of qSPA-4 using introgression lines derived from the same cross. Results indicated that qSPA-4 was located in a 810-Kb region flanked by RM16333 and RM16368. The results indicated that Hapcheonaengmi3 contains QTL alleles that are likely to improve cold tolerance of Indica rice.
