Phylogenetic Analysis Using Cytochrome c Oxidase Subunit I of Silver Croaker(Pennahia argentata) Mitochondria DNA (미토콘드리아 DNA의 cytochrome c oxidase subunit I을 이용한 보구치(Pennahia argentata) 계통 분석)

  • Silver croaker (Pennahia argentata) is a turbulent species that is widely distributed worldwide and is mainly found in the bottom of the ocean. In the study, we characterized the cytochrome c oxidase subunit I (COI) gene of the mitochondrial DNA (mtDNA) on P. argentata inhabiting Gwangyang Bay and analyzed the phylogenetic location of marine fish species. As a result of multiple arrangement of 605 bp COI sequences, high homology of mtDNA nucleotide sequences was confirmed in the silver croakers from Gwangyang Bay (98~100%). However, the nucleotide variation was different according to the catching points of the inland and the open seas of Gwangyang Bay. The nucleotide sequence variation in COI was high in P. argentata from the open seas of Gwangyang Bay (43.2~70.3%). Furthermore, the phylogenetic analysis of 13 fish showed that P. argentata from Gwangyang Bay were grouped into one clade with P. argentata reported in Taiwan, and the evolutionary distance was 0.036. In addition, it was identified that the evolutionary distance was close to that of fish belonging to the Mi-iuy croaker (Miichthys miiuy) and the Big-head pennah croaker (Pennahia Macrocephalus) (0.041~0.048). The result of these studies will be used as the key genetic information for fisheries resources monitoring and species diversity management according to the coastal environment.

  • The tendency of rancidity in roasted and/or reheated yellowfin sole and yellow croaker was investigated as typical white muscled fish. In fatty acid composition of the total lipid, saturated fatty acid was $27.4\%,\;33.4\%$; monoenoic acid, $36.5\%,\;38.7\%$ and polyenoic acid, $34.5\%,\;26.5\%$ in yellowfin sole and yellow croaker, respectively. The ratio of unsaturated fatty acid (UFA)/saturated fatty acid (SFA) were 2,6, 2.0 and content of total lipid was $1.4\%,\;0.8\%$, respectively. Peroxide content decreased after heating in yellowfin sole while decreased after heating and tended to increase after reheating in yellow croaker. Thiobarbituric acid value increased during roasting and heating in all the samples but decreased after reheating. Acid value increased after roasting, heating and reheating in all samples, particularly higher in yellowfin sole which have a high content of UFA. Conjugated dienes continuously increased during the repeats of heating, showing more increase in the fillet with Skin than the skinless.

Annual Reproductive Cycle of Wild Female Yellow Croaker, Larimichthys polyactis (자연산 암컷 참조기, Larimichthys polyactis의 생식년주기)

  • Annual reproductive cycle of small yellow croaker, Larimichthys polyactis were histologically investigated based on samples captured nearby the coast around Chujado, Youngkwang and Kanghwa, South Korea from February $2004{\sim}February$,2005. By histological survey, the annual reproductive cycle was classified into the following successive stages: The primary growth stage from September to October, the second growth stage from October to December, the mature stage from January to April, the final mature, ovulation and spawning stage from May to June, the degeneration stage from June to July and the rest stage from August to September. Throughout anatomical observation of the ovary, it is found that the small yellow croaker, L. polyactis belongs to an asynchronous and multiple spawner from late spring to early summer.

Age and Growth of Small Yellow Croaker, Larimichthys polyactis in the South Sea of Korea (한국 남해 참조기의 연령과 성장)

  • Age and growth of the small yellow croaker, Larimichthys polyactis were estimated using right sagittal otoliths of 506 fish specimens from March to December, 2002 and from January to February, 2005 in the South Sea, part of the East China Sea of Korea. Examination of outer margins of the otolith showed that the opaque zone was formed once a year. Marginal increment of the otolith formed annual rings from May and June at the beginning of spawning season. In the relationship between total length and body weight, a multiplicative error structure was assumed because variability in growth increased as a function of the length, and the estimated equation was $BW=0.0044TL^{3.2502}$ ($R^2=0.97$). The relative growth as body weight at total length has significant difference between females and males (P<0.05). For describing growth of the small yellow croaker, Larimichthys polyactis a von Bertalanffy growth model was adopted. The von Bertalanffy growth curve had an additive error structure and the growth parameters estimated from non-linear regression were $L_{\infty}=33.88cm$, K=0.20/year and $t_0=-2.39year$. Growth at age of males and females shows no significant difference (P>0.05). Most examined fish were 1, 2 and 3 years old, although the oldest fish were 7 old for males and 8 for females.

  • As a series of studies about improvement of yellow croaker drift gill net fishing gear and development of the labor saving fishing system, this study analyzed the behavior characteristics of fishing gear, which does use three types of different sinker materials in the field tests. The result of maritime performance test indicated that the fishing gear which used the lead reached the maximum depth and bio ceramics hauled to the surface of water the fastest. The correlation between the sinking time (St) and depth (Dsl, Dsc, Dsb) of maritime performance test can be shown such experimental equations as Dsl=2.70St - 0.75, Dsc = 2.38St - 1.15, Dsb = 1.77St - 4.00. The correlation between the hauling time (Ht) and depth (Dhl, Dhc, Dhb) of maritime performance test can be shown such experimental equations as and Dhl = 7.88Ht + 35.48, Dhc = 7.80Ht + 40.01, Dhb = 7.95Ht + 36.44.

Pharmacokinetic Study of Florfenicol in Healthy and Vibriosis-infected Pseudosciaena crocea after Oral Administration

  • The pharmacokinetics of florfenicol were studied in healthy and vibriosis-infected large yellow croaker (Pseudosciaena crocea) following administration of a single oral dose of $20mg{\cdot}kg^{-1}$ at $25{\pm}2^{\circ}C$. After oral administration, florfenicol levels in tissues (liver, kidney, muscle, serum, and skin) were analyzed using high-performance liquid chromatography. A two-compartment open model was used to describe the pharmacokinetics of florfenicol following oral administration. Compared to the healthy group, the absorption rate of vibriosis-infected fish significantly decreased, peak-time ($T_{max}$) delayed, maximum concentration ($C_{max}$) declined, total body clearance decreased, the elimination half-life ($T_{1/2{\beta}}$) was extended, and the area under the curve increased. These results indicate that a $20mg{\cdot}kg^{-1}$ oral dose of florfenicol administered once daily continuously for 4 or 5 days can be used for the treatment of Vibrio alginolyticus infection in large yellow croaker (Pseudosciaena crocea).

Predicting Changes in Fishing Conditions for the Small Yellow Croaker Larimichthys polyactis based on Expansions of the Yellow Sea Bottom Cold Water (황해저층냉수에 따른 참조기(Larimichthys polyactis) 어황의 변화와 예측 가능성)

  • We consider changes in the fishing ground of the small yellow croaker Larimichthys polyactis and discuss their utility in predicting fishing conditions for this species. The fishing ground, which having been formed around Jeju Island since the 1970s, is dominated by the Yellow Sea Bottom Cold Water (YSBCW), and variation in its southward expansion from the Yellow Sea is the single most key environmental factor affecting the L. polyactis catch. When the YSBCW showed strong expansion and the fishing ground shifted to the west and southwest of Jeju Island, as occurred in the late 1980s, late 1990s, and early 2000s, the L. polyactis catch was low; conversely, when expansion was weak, as in the early 1990s and late 2000s, the L. polyactis catch was high. This relationship was statistically significant and should be useful in predicting fishing conditions for L. polyactis.

A Study on the Combined Equipment for the Pre-cooling and the Thawing using the Low Temperature Vacuum System (저온진공기술을 이용한 예냉 및 해동 겸용장치에 관한 연구)

  • We need refrigeration system which can maintain the freshness of agricultural products, because of being distance from a tiller to a consumer. Vacuum Pre-cooling system has an advantage in quality maintenance through vapid cooling down by using latent heat of evaporation of stored products. A number or thawing methods in current use have also several disadvantages in thawing time. discoloration mass loss caused by drying, capital costs and running cost. These damages are, it is claimed, either eliminated or improved by the vacuum thawing system. An experimental study on the pre-cooling for the bean sprouts and cabbage, and thawing for hairtail and croaker by the low temperature vacuum system were carried out. The cabbage cooling time with this Pre-cooling vacuum system took about 60 minutes to reach from $23.2^{circ}C to 4.5^{\circ}C$ at 5 mmHg abs. ($6.66\times10^{-4}$ MPa). The croaker thawing time with this low temperature vacuum thawing system took about 170 minutes to reach from $-10.3^{circ}C to -0.8^{\circ}C$ at 20 mmHg abs ($2.67\tiems10^{-3}$MPa). The vacuum Pre-cooling and thawing system have merits compared with present systems in their short intervals to cool down and to thaw without any quality losses.

Acoustic Identification of Six Fish Species using an Artificial Neural Network (인공 신경망에 의한 6개 어종의 음향학적 식별)

  • The objective of this study was to develop an artificial neural network (ANN) model for the acoustic identification of commercially important fish species in Korea. A broadband echo acquisition and processing system operating over the frequency range of 85-225 kHz was used to collect and process species-specific, time-frequency feature images from six fish species: black rockfish Sebastes schlegeli, black scraper Thamnaconus modesutus [K], chub mackerel Scomber japonicus, goldeye rockfish Sebastes thompsoni, konoshiro gizzard shad Konosirus punctatus and large yellow croaker Larimichthys crocea. An ANN classifier was developed to identify fish species acoustically on the basis of only 100 dimension time-frequency features extracted by the principal components analysis (PCA). The overall mean identification rate for the six fish species was 88.5%, with individual identification rates of 76.6% for black rockfish, 82.8% for black scraper, 93.8% for chub mackerel, 90.6% for goldeye rockfish, 96.9% for konoshiro gizzard shad and 90.6% for large yellow croaker, respectively. These results demonstrate that individual live fish in well-controlled environments can be identified accurately by the proposed ANN model.

  • The feasibility of passive detection of fishes which had been caught in the fishing grounds near the Korean peninsula was theoretically investigated. Considering the commercial importance and the acoustical informations readily identified, although many species of fish make noise, Croaker is clarified to be a representative fish for passive fish detection. Assuming a source level of the sound produced by croakers is given as 150-18OdB (re 1l'pa, 500 Hz bandwidth), The range detected by a passive line array sonar is estimated to be about 3-20km. In addition, the tonal noise (700~800Hz) made by croaker that is easily separated from underwater noise is expected to increase the ability to discriminate from the other species of fish.
