Background: Inhaled indacaterol (Onbrez Breezhaler), a long-acting ${\beta}_2$-agonist, is approved in over 100 countries, including South Korea, as a once-daily bronchodilator for maintenance and treatment of chronic obstructive pulmonary disease (COPD). Here, we present an interim analysis of a post-marketing surveillance study conducted to evaluate the real-world safety and effectiveness of indacaterol in the Korean population. Methods: This was an open-label, observational, prospective study in which COPD patients, who were newly prescribed with indacaterol (150 or $300{\mu}g$), were evaluated for 12 or 24 weeks. Safety was assessed based on the incidence rates of adverse events (AEs) and serious adverse events (SAEs). Effectiveness was evaluated based on physician's assessment by considering changes in symptoms and lung function, if the values of forced expiratory volume in 1 second were available. Results: Safety data were analyzed in 1,016 patients of the 1,043 enrolled COPD patients receiving indacaterol, and 784 patients were included for the effectiveness analysis. AEs were reported in 228 (22.44%) patients, while 98 (9.65%) patients reported SAEs. The COPD condition improved in 348 patients (44.4%), while the condition was maintained in 396 patients (50.5%), and only 40 patients (5.1%) exhibited worsening of ailment as compared with baseline. During the treatment period, 90 patients were hospitalized while nine patients died. All deaths were assessed to be not related to the study drug by the investigator. Conclusion: In real-life clinical practice in South Korea, indacaterol was well tolerated in COPD patients, and can be regarded as an effective option for their maintenance treatment.
The purpose of this study was to examine the residents' opinions on the degree of importance, and satisfaction with, the managerial service of multi-family housing. Residents' participation in managerial activities was also examined, because residents' opinions should be taken into serious consideration in order to improve the efficiency of housing management. Research methods used in this study were survey, field study, and content analysis of professionals' opinions. Survey data of 520 residents who live in multi-family housing were analyzed by means, frequencies, correlation, chi-square tests, t-tests, one-way ANOVA, post-hoc estimation (Scheffee test), and regression. The major findings of this study were as follows: First, many residents were dissatisfied with the quality of managerial service, but they thought that managerial services were very important. For example, they thought that safety inspection of the playground was very important, yet the current level of service was not satisfactory. Second, the characteristics of residents that determined the degree of residential satisfaction were income the location of the residence, and degree of participation in the community events. Third, the residents generally did not want to participate in the managerial services and community programs through such activities as participation in resident committee. In order to improve the housing management efficiency, residents' participation is critical. Some participants in this study pointed out that the details of management service and future plans need to be made public. New management models need to be developed to promote residents' participation and increase their satisfaction in the management.
Environmental factors such as light and low $CO_2$ concentrations trigger events which may result in stomatal opening. Stomatal aperature is largely controlled by the solute contents of guard cells, but not exclusively, by through changes in their content of potassium salts, with $K^+$ balanced either by $Cl^-$ or malate, depending on the species and conditions. However, how these signals are sensed and how they are transduced into driving the ion fluxes that control stomatal movements is not still fully understood. The basic role of stomata is regulating transpiration and photosynthesis. Photosynthesis plays a central role in the physiology of plants and an understanding of its response to light is, therefore, critical to any discussion of how plants sense and respond to light. It had been proposed that the evidences pointed three possible mechanisms for the light response. Firstly, there is a direct response of stomata to light. Secondly. there is an indirect response of stomata to light through the effect of $CO_2$. Lastly, there are some evidences for a third effect of light on stomata. However, attempts to investigate how these three possible mechanisms explained in detail in response to light have not been made. Therefore, this study is examined the differences among these three possible mechanisms.
Confined and mechanically ventilated broiler house has been recently come into wide use to increase productivity and control rearing conditions. However, high dust concentration inside facility can threaten respiratory welfare of farmers and broilers. In Korea, there is no regulation in terms of air quality control inside agricultural facilities and sufficient data is not available. To cope with these, periodic monitoring of inhalable and respirable dust concentration were conducted according to season, broiler's activity and specific events such as shipment work for broilers in mechanically ventilated broiler house. Chamber experiment was also designed to analyze mechanism of dust generation from ground beds according to water contents and surrounding wind environment. Results showed that significant increase of inhalable (p=0.002) and respirable (p=0.03) dust were observed when activity of broilers was high according to entrance of the workers. Even shipment work for matured broilers, high level of dust were observed; inhalable dust was exceeded the threshold limit over maximum 303 % and respirable dust was over maximum 1,550 %, implying that acute respiratory symptoms could be manifested for the workers. From the chamber experiment, critical water contents for interrupting of dust generation were measured; about 45 % for inhalable dust and about 50 % for respirable dust. These results can be a trigger for designing plan of dust control however it still needs consideration of various environmental conditions, hygiene problems, etc.
Mitogen-activated protein kinase (MAPK) signaling cascades play critical roles in various cellular events in plants, including stress responses, innate immunity, hormone signaling, and cell specificity. MAPK-mediated stress signaling is also known to negatively regulate nitrogen-fixing symbiotic interactions, but the molecular mechanism of the MAPK signaling cascades underlying the symbiotic nodule development remains largely unknown. We show that the MtMKK5-MtMPK3/6 signaling module negatively regulates the early symbiotic nodule formation, probably upstream of ERN1 (ERF Required for Nodulation 1) and NSP1 (Nod factor Signaling Pathway 1) in Medicago truncatula. The overexpression of MtMKK5 stimulated stress and defense signaling pathways but also reduced nodule formation in M. truncatula roots. Conversely, a MAPK specific inhibitor, U0126, enhanced nodule formation and the expression of an early nodulation marker gene, MtNIN. We found that MtMKK5 directly activates MtMPK3/6 by phosphorylating the TEY motif within the activation loop and that the MtMPK3/6 proteins physically interact with the early nodulation-related transcription factors ERN1 and NSP1. These data suggest that the stress signaling-mediated MtMKK5/MtMPK3/6 module suppresses symbiotic nodule development via the action of early nodulation transcription factors.

In marine ecosystems, the biosynthesis and catabolism of dimethylsulfoniopropionate (DMSP) by marine bacteria is critical to microbial survival and the ocean food chain. Furthermore, these processes also influence sulfur recycling and climate change. Recent studies using emerging genome sequencing data and extensive bioinformatics analysis have enabled us to identify new DMSP-related genes. Currently, seven bacterial DMSP lyases (DddD, DddP, DddY, DddK, DddL, DddQ and DddW), two acrylate degrading enzymes (DddA and DddC), and four demethylases (DmdA, DmdB, DmdC, and DmdD) have been identified and characterized in diverse marine bacteria. In this review, we focus on the biochemical properties of DMSP cleavage enzymes with special attention to DddD, DddA, and DddC pathways. These three enzymes function in the production of acetyl coenzyme A (CoA) and CO2 from DMSP. DddD is a DMSP lyase that converts DMSP to 3-hydroxypropionate with the release of dimethylsulfide. 3-Hydroxypropionate is then converted to malonate semialdehyde by DddA, an alcohol dehydrogenase. Then, DddC transforms malonate semialdehyde to acetyl-CoA and CO2 gas. DddC is a putative methylmalonate semialdehyde dehydrogenase that requires nicotinamide adenine dinucleotide and CoA cofactors. Here we review recent insights into the structural characteristics of these enzymes and the molecular events of DMSP degradation.
본 연구에서는 몇 가지 암세포를 대상으로 마늘 열수추출물(WEAS)의 항암활성을 조사하였으며, 비교적 높은 감수성을 보인 백혈병세포를 대상으로 세포증식억제가 apoptosis 유도와 연관성이 있음을 제시하였다. WEAS에 의한 U937 세포의 apoptosis 유도는 세포주기 G2/M arrest와 연관성이 있었으며, 다양한 apoptosis조절 유전자들의 발현 변화를 동반하였음을 확인하였다. 이러한 apoptosis조절 인자들의 발현변화는 미토콘드리아 막 전위의 소실과 연관성이 있었으며, WEAS에 의한 암세포의 G2/M arrest에 의한 apoptosis 유발에 ROS가 중요한 조절자로 작용함을 알 수 있었다.
Occupational hazards of firefighting and rescue works include frequent exposure to emergencies and life-threatening situations. These stressful work conditions of being constantly under pressure and exposed to potentially traumatic events put them at higher risk of developing posttraumatic stress disorder (PTSD), compared to the general population. PTSD is a potentially debilitating mental disorder, due to persistent intrusive thoughts, negative alterations of mood and cognition, hypervigilance, avoidance of similar situations and reminders, and re-experiences of the traumatic event. Previous studies have shown a relatively high prevalence of PTSD among firefighters, indicating the need for a systematic approach of early detection and prevention. Therefore, a critical review of the current literature on PTSD in firefighters would provide valuable insights into developing effective prevention and intervention programs. Literature indicated that there are risk factors of PTSD in firefighters, such as pre-existing depression, anxiety, sleep disorders, occupational stress, physical symptoms, and binge drinking, whereas social support and adequate rewards are protective factors. Although there are differences in the prevalence of PTSD across studies, partly due to various assessment tools utilized, different sample sizes, and sample characteristics, over one tenth of the firefighters were estimated to have PTSD. The current review warrants further investigations to precisely assess PTSD and co-morbid mental disorders, functional outcomes, and associated factors, and to develop evidence-based preventive and interventional programs to help firefighters with PTSD.
The actin cytoskeleton plays an important role in macrophage-mediated inflammatory responses by modulating the activation of Src and subsequently inducing nuclear factor (NF)-${\kappa}B$ translocation. In spite of its critical functions, few papers have examined how the actin cytoskeleton can be regulated by the activation of toll-like receptor (TLR). Therefore, in this study, we further characterized the biological value of the actin cytoskeleton in the functional activation of macrophages using an actin cytoskeleton disruptor, cytochalasin B (Cyto B), and explored the actin cytoskeleton's involvement in morphological changes, cellular attachment, and signaling events. Cyto B strongly suppressed the TLR4-mediated mRNA expression of inflammatory genes such as cyclooxygenase (COX)-2, tumor necrosis factor (TNF)-${\alpha}$, and inducible nitric oxide (iNOS), without altering cell viability. This compound also strongly suppressed the morphological changes induced by lipopolysaccharide (LPS), a TLR4 ligand. Cyto B also remarkably suppressed NO production under non-adherent conditions but not in an adherent environment. Cyto B did not block the co-localization between surface glycoprotein myeloid differentiation protein-2 (MD2), a LPS signaling glycoprotein, and the actin cytoskeleton under LPS conditions. Interestingly, Cyto B and PP2, a Src inhibitor, enhanced the phagocytic uptake of fluorescein isothiocyanate (FITC)-dextran. Finally, it was found that Cyto B blocked the phosphorylation of vasodilator-stimulated phosphoprotein (VASP) at 1 min and the phosphorylation of heat shock protein 27 (HSP27) at 5 min. Therefore, our data suggest that the actin cytoskeleton may be one of the key components involved in the control of TLR4-mediated inflammatory responses in macrophages.
PURPOSES : The occurrence of unexpected disasters, including fire events, increases as the road network becomes complicated and traffic volume increases. When a fire event occurs on and under bridges, the damage extensively influences direct damage to structures, vehicles, and human life and secondary socioeconomic issues owing to traffic blockage. This study investigated potential fire-hazard risks on bridges of the Korean national route road. METHODS : The investigation was conducted using field investigation and analysis with satellite pictures and road views from commercial websites and the Bridge Management System (BMS). From the filed investigation, various potential fire resources were identified. The satellite pictures and road views were helpful in measuring and recognizing conditions underneath bridges, stowage areas, etc. RESULTS : There are various potential fire resources underneath bridges such as piled agricultural products, parked petroleum tanks, construction equipment, and attached high-voltage cables. A total of 94.6% of bridges have underneath clearances of less than 15 m. A bridge underneath volume that can stow a potential fire hazard resource was $7,332m^3$ on average, and most bridges have about $4,000m^3$ of space. Based on the BMS data, the amounts of PSC and steel girders were 29% and 25%, respectively. CONCLUSIONS : It was found that the amount of stowed potential fire hazard resources was proportional to the underneath space of bridges. Most bridges have less than 15 m of vertical clearance that can be considered as a critical value for a bridge fire. The fire risk investigation results should be helpful for developing bridge fire-protection tools.
