Classification and Review of Diseases that Traditional Chinese Medicine is Better at Treating (중의우세병종의 분류 및 고찰)

  • Objective : This study was aimed to classify diseases that Traditional Chinese Medicine(TCM) is better at treating. Method : Literature was searched on China National Knowledge Infrastructure(CNKI) and categorized according to literature type, published date and research method. Studied six types of research papers and four types of published books. Results : Experts were surveyed and interviewed, medical records were studied retrospectively, and doubleblind method was used in selecting diseases that TCM was better at treating. There were a total of 372 diseases that TCM was better at treating. By the KCD classification, 45 were in gastrointestinal (12.1%), 39 in urogenital (10.5%), 36 in circulatory (9.7%), 35 in musculoskeletal or connective tissues (9.4%). Conclusion : Total of 372 diseases were classified as diseases that TCM was better at treating, and if the results are used adequately, the values of western and TCM can be maximized and benefit the government, patients and the medical practitioners.

Controlled Release Dosage Form of Narcotic Antagonist(II) : Biocompatibility and Pharmacokinetics of Naloxone Implant (마약길항제의 방출 제어형 제제 (제2보): 나록손 이식제제의 생체적합성 및 약물속도론적 평가)

  • For the effective administration of narcotic antagonist, the application of sustained release implantable systems with biodegradable polyphosphazene was examined. Using poly[(diethyl glutamate)-co-(ethyl glycinate) phosphazene], the implantable devices containing naloxone hydrochloride were prepared and in vivo implantation studies were carried out subcutaneously in rat and rabbit with this preparation for the biocompatibility and pharmacokinetics. The histological finding in rats at initial time period was the inflammation that occurred focally around the implants, but they were showed subsequent mild and limited chronic inflammations and the irreversible changes such as necrosis and degeneration of the muscle or connective tissues were not observed. Therefore the placebo and naloxone implants are considered to be biocompatible formulations histologically. In pharmacokinetic studies, the release of naloxone from the naloxone implants into blood plasma was maintained in 192 hours, but the initial burst effect was observed. If this problem was solved, the application for the narcotic antagonist sustained release systems can be expected.

Protective actions of Rubus coreanus ethanol extract on collagenous extracellular matrix in ultraviolet-B irradiation-induced human dermal fibroblasts

  • Solar ultraviolet (UV) irradiation leads to distinct changes in the skin connective tissues by degradation of collagen, which is a major structural component in the extracellular matrix. UV irradiation induces the production of matrix metalloproteinases (MMP) capable of attacking native fibrillar collagen and responsible for inhibiting the construction of collagenous extracellular matrix. In this study, we attempted to investigate the protective actions of Rubus coreanus ethanol extract (RCE) on the MMP production and the consequent procollagen/collagen degradation in UV-B-irradiated human dermal fibroblasts. The analytical data showed that Rubus coreanus ethanol extract was mostly comprised of cyanidin 3-rutinoside. Pre-treatment of fibroblasts with this extract inhibited UV-B-induced production of MMP-1, MMP-8 and MMP-13 in dose-dependent manners. In addition, Western blot analysis and immunocytochemical staining assay revealed that RCE markedly augmented the cellular levels of procollagen/collagen declined in UV-B-exposed dermal fibroblasts. These results demonstrate that RCE blocks UV-B-induced increase of the collagen degradation by inhibiting MMP production. Thus, RCE may act as an agent inhibiting excessive dermal collagen degradation leading to the skin photoaging.

Reproductive Cycle of the Ark Shell, Scapharca subcrenata, on the West Coast of Korea

  • Monthly changes in the gonad index (GI), egg-diameter composition, gonadal development, reproductive cycle of the ark shell, Scapharca subcrenata, were investigated by histological method and morphometric data. This species is dioecious and oviparous. The gonad is located among the subregion of mid-intestinal gland, digestive diverticula and the outer fibromuscular layers compacted by the fibrous connective tissues and muscle fibers. The gonad index sharply increased in May, reached the maximum value in June, and then gradually decreased from July to December. The reproductive cycle of this species can be divided into six successive stages: early active stage (January to May), late active stage (June to July), ripe stage (June to August), partially spawned stage (July to September), degenerative stage (August to December), and resting stage (January to April). S. subcrenata spawns once a year between July and early September, and the main spawning occurred between July and August when the water temperatures were above $20^{\circ}C$. This evidence suggest that timings of maturation and spawning are closely related to water temperatures. Even though the spawning period was once a year, it is assumed that the number of spawning frequencies (broods) may occur more than twice during the spawning season.

The Effect of L-Ascorbic Acid on the Formation of Immature Crosslink in Bone Collagen in vitro (비타민 C가 in vitro 계에서 콜라겐 미숙가교 생성에 미치는 영향)

  • Intermolecular collagen cross links stabilize collagen fibrils and are necessary for normal tensile strength in collagen fibrils. Once the fibrils are aligned, hydroxyllysine, hydroxylysine derived aldehyde modified enzymatically, reacts with hydroxylysine to form the dehydrodihydroxylysinonorleucine (DHLNL), an immature crosslink. Pyridinoline, one of matured cross links is presumably formed nonenzymatically through condensation of DHLNL and hydroxylysine residue. It is widely distributed in hard connective tissues such as cartilage, bone and tendon. L ascorbic acid(AsA) is well known to be required for the enzymatic hydroxylation of proline and lysine in collagen fibrils. The purpose of this study is to clarify the role of AsA on the biosynthesis of DHLNL in vitro. We examined the effect of AsA on the formation of hydroxylysine and DHLNL in collagen. Pyridinoline and DHLNL were measured as a function of time. The contents of DHLNL was increased, reached maximum within 2 hr and was held until 24 hr, then it decreased slowly. On the contrary, pyridinoline increased gradually after 24 hr and continued to increase for 2 weeks. Moreover, the contents of DHLNL remarkably decreased at 60 min after incubation, the contents of DHLNL was decreased by addition of AsA or dehydroascorbic acid(DHA). These results suggest that the supplementation of AsA causes decrease in DHLNL formation and pyridinoline formed by nonenzymatic reaction of DHLNL.

Mongolian Gerbil as a Novel Animal Model for Ligature-induced Periodontitis

  • Inflammation from chronic and acute infections of distal organs and tissues such as periodontitis is a risk factor for atherosclerotic vascular processes. Recently, a new model of atherosclerosis with vascular pathologies was developed in the Mongolian gerbil. In this study, we attempted to develop a model of ligature-induced periodontitis in gerbils and compared the characteristics of that periodontitis model with that in rats and mice. Each gerbil, rat, and mouse was randomly assigned to groups of control and periodontitis. A thread was placed around the cervix of the right and left first molars in the mandible with knots placed on the mesial side of each molar. At day 14 after the ligation, the animals were sacrificed and their mandibles were dissected. To measure alveolar bone loss along with inflammation, histopathological and micro-CT analyses were carried out. Gerbils showed tooth characteristics of deeper gingival crevice, longer cusp, longer root trunk and shorter root than those of rats and mice. The increased CEJ-ABC distance in distal and PDL area in furcation was also observed in ligated gerbils. An inflammatory response in the connective tissue under the junctional epithelium was also shown in all the animals. As a result, we confirmed the induction of periodontitis by ligature in the gerbils. We therefore consider the gerbil to be a useful model for investigating relationship between periodontitis and vascular disease in the same animal.

Space-maintaining and osteopromotive effect of freezedried bone graft in the procedure of GBR (동결건조골이 재생공간 유지 및 골재생에 미치는 영향)

  • This study was performed to evaluate the effect of freeze-dried bone graft on space-making capacity and bone formation in the procedure of guided bone regeneration with titanium reinforced ePTFE membrane. After decortication in the calvaria, GBR procedure was performed on 8 rabbits with titanium reinforced ePTFE membrane filled with human FDBA(Rocky Mountain Tissue Bank,Aurora Co., USA). Decortication was performed to induce the effect of bone forming factor from bone marrow. The animals were sacrificed at 2 weeks, 4 weeks, 8 weeks and 12 weeks after the surgery. Non-decalcified specimens were processed for histologic analysis. πle results of this study were as follows: 1. Titanium reinforced-ePTFE membrane was biocompatable and capable of maintaining the space-making. 2. FDBA particle was surrounded with connective tissues but there was no evidence on new bone formation. 3. FDBA particle resorbed continuously but it remained until 12weeks after the surgery. Within the above results, TR-ePTFE membrane could be used effectively for Guided bone regeneration but It was assumed that FDBA does not appear to contribute to bone formation.

Tensile stress regulation of NGF and NT3 in human dermal fibroblast

  • Fibroblast is constantly subjected to mechanical loads in connective tissues where mechanical signals are converted to intercellular biochemical events. The aim of this study is to understand the effects of tensile stress on the neurotrophin (NT) and transforming growth factor (TGF) expression of fibroblast in vitro. Nerve growth factor (NGF) stimulates fibroblast migration, and TGF is related to tissue repair. In this study, at the uniaxial stretch of 10% strain and frequency of 0.5 Hz, different resting times of 0, 20, and 60 min are placed in between 10 min stimulations periods. Results show increase in NGF mRNA levels and a substantial decrease in NT3 mRNA after 1 hr of stimulation, indicating that the tensile stress may regulate NGF and NT3, key factors for the neurocosmetic applications. The mRNA level for TGF-${\alpha}$ and TGF-${\beta}2$ had increased up to two-folds after 1 hr of stimulation, showing that the tensile stress may control TGF, an important part of wound healing.

  • The present study was conducted to evaluate inhibition effect of oxidation and discoloration in yellow sea bream, treated with some antioxidants, dried and stored for one month. The effect was observed microscopically from the aspects of distribution and movement of fat, and changes of muscular system. On the initial stage of drying, a part of hypodermic fat was penetrated into intermuscular fissile passing through connective tissue and myocommata, but the rest part was exuded to the out side of surface. The tissues from which the hypodermic fat was removed seemed to be hardned with the gathering of adjacent muscle fibers. The inhibition effect of antioxidants was better in order of Tenon-11, BHA, NDGA from the point of retention of fat and preservation of the muscular tissue.

  • Background: The vascularity of the peri-implant tissue is a very important parameter in establishment and maintenance of a healthy tissue after dental implant insertion. Objective: The purpose of this study was to compare the vascularity of the peri-implant mucosa between flap and flapless implant surgeries by using a canine mandible model. Study Design: In six mongrel dogs, bilateral, edentulated, flat alveolar ridges were created in the mandible. After 3 months of healing, two implants were placed in each side by either the flap or flapless procedures. After another healing period of 3 months, biopsies were obtained, prepared for light microscopy and exposed to morphometric measurements. Results: The supracrestal connective tissue lateral to the implant was found to be more richly vascularized in the flapless group than in the flap group. Conclusion: These results suggest that the flapless procedure may have an effect on increasing the vascularity of the peri-implant mucosa.
