Antifungal Activities of Metarhizium anisopliae against Fusarium oxysporum, Botrytis cinerea, and Alternaria solani (Metarhizium anisopliae의 Fusarium oxysporum, Botrytis cinerea, Alternaria solani 등에 대한 항진균활성)

  • This study was conducted to find out antifungal activities of entomopathogenic fungus, Metarhizium anisopliae, against phytopathogenic fungi, Fusarium oxysporum, Botrytis cinerea and Alternaria solani. M. anisopliae was confirmed its antagonistic effect through mycelial inhibition zone of phytopathogenic fungi by culture filtrate of the antagonist. The filtrate (30%: v/v) inhibited the conidial germination of B. cinerea and F. oxysporum to 21.5% (control: 88.2%) and 53.0% (control: 78.6%), respectively and delayed the start of spore germination about 8hours. Microscopic observations proved that the addition of 10% culture filtrate of M. anisopliae restricted the growth of phytopathogenic fungus, F. oxysporum, to the formation of chlamydospore. From these results, we concluded that an addition effect of the filtrate from M. anisopliae on culturing F. oxysporum was fungistatic.

  • One fungal isolate CM2 parasitic to Sclerotinia sclerotiorum and S. minor causing Sclerotinia rot of lettuce was identified as Paraconiothyrium minitans based on its morphological and molecular characteristics. P. minitans CM2 grew best on PDA with pH 6.5 at $22^{\circ}C$ under alternating cycles of 12 hr near ultraviolet light and 12 hr darkness. Scleroria of S. sclerotiorum and S. minor treated with conidial suspension of P. minitans CM2 did not directly germinate and produced no apothecia.

  • This study was conducted to evaluate the antifungal activity of 19 agro-materials that have been registered for organic cultivation in Korea, after inoculation of pear leaves with Venturia nashicola and Gymnosporangium asiaticum. In V. nashicola, most of the nine agro- materials containing sulfur and copper completely inhibited spore germination, and some of the spores that germinated did not form appressoria. However, in only lime sulfur, Neobordeaux (cupric sulfate), and Wheengaris (sulfur)showed antifungal activity against G. asiaticum. Among the agro-materials containing plant extracts, Wheengarujaba (wood vinegar+spirits+rhubarb) inhibited conidial germination in V. nashicola and G. asiaticum by 100% and 71.6%, respectively. Among the agro-materials containing antifungal microorganisms, Cheongotan (Streptomyces griseus) reduced spore germination rate of V. nashicola to 88.8%; moreover, formation of appressoria or intracellular accumulation was not observed. Application of Topsid (Paenibacillus polymyxa) reduced spore germination rates in V. nashicola and G. asiaticum to 71.0% and 90.6%, respectively, and the formation of appressoria was not observed. Studying the antifungal activity of agro-materials because of cumulative applications under the field conditions is necessary, owing to their contact fungicidal effect and the induced-resistance by microbial metabolites and natural compounds.

Effects of Dryness, Moisture Interruption, and Temperature on Germination of Diaporthe citri Pycnidiospores on Yuzu (건조, 습윤중단 및 온도가 유자 검은점무늬병원균 Diaporthe citri 포자 발아에 미치는 영향)

  • Melanose caused by Diaporthe citri is a serious disease on yuzu fruits. Moisture is the most important to infect after the pycnidiospores are released. In order to understand how dryness and moisture interruption affect on germination, the conidial suspensions on slide glass were dried for 0-48 h. For the moisture interruption experiment, moisture was supplied for 10 h then interrupted for 0-6 h depending on the treatments. Germinations on the treatments with longer than 15 h of dryness were less than 10%. And those with longer than 3 h of moisture interruption were less than 30%. Compared with other fungal phytopathogens, D. citri was xero-sensitive. Germination on yuzu feel medium was measured under 15, 20, 25, 30, $35^{\circ}C$ with 3, 6, 9, 12, 15, 18, and 24 h of wetness. The minimum wetness periods achieving 10% and 50% germinations were 4.5 and 13 h, and the optimum temperatures were $29.1^{\circ}C$ and $29.2^{\circ}C$, respectively. From the yuzu medium study, the regression equations to be required wetness period achieving 10% and 50% germinations on various temperatures were $Wh_{50%}=0.1093{\times}T^2-6.3762{\times}+106.08$ and $Wh_{10%}=0.082{\times}T^2-4.8025{\times}T+74.861$.

MoJMJD6, a Nuclear Protein, Regulates Conidial Germination and Appressorium Formation at the Early Stage of Pathogenesis in Magnaporthe oryzae

  • In plant-pathogen interactions, Magnaporthe oryzae causes blast disease on more than 50 species of 14 monocot plants, including important crops such as rice, millet, and most 15 recently wheat. M. oryzae is a model fungus for studying plant-microbe interaction, and the main source for fungal pathogenesis in the field. Here we report that MoJMJD6 is required for conidium germination and appressorium formation in M. oryzae. We obtained MoJMJD6 mutants (ΔMojmjd6) using a target gene replacement strategy. The MoJMD6 deletion mutants were delayed for conidium germination, glycogen, and lipid droplets utilization and consequently had decreased virulence. In the ΔMojmjd6 null mutants, global histone methyltransferase modifications (H3K4me3, H3K9me3, H3K27me3, and H3K36me2/3) of the genome were unaffected. Taken together, our results indicated that MoJMJD6 function as a nuclear protein which plays an important role in conidium germination and appressorium formation in the M. oryzae. Our work provides insights into MoJMJD6-mediated regulation in the early stage of pathogenesis in plant fungi.

Effect of Water Activity and Temperature on Growth, Germination, Sporulation, and Utilization of Carbon Source of Penicillium oxalicum (PENOX) as a Biocontrol Agent(BCA) for control of Clover(Trifolium repens L.) (토끼풀(Trifolium repens L.) 방제용 생물제제 Penicillium oxalicum (PENOX)의 발아, 생장, 포자생성 및 탄소원이용에 미치는 수분활성 및 온도의 영향)

  • Penicillium oxalicum (PENOX) has shown the potential as a biocontrol agent(5CA) for control of a weed, clover(Trifolium repens L.) in grass plots. The bioherbicidal activity may be due to germinative and growth capacities and substrate availability of the agent over a range of environmental factors. The influences of different water activities($0.94{\sim}0.995\;a_w$) and temperatures($18{\sim}30^{\circ}C$) on mycelial growth, conidial germination, sporulation oil 2% MEA(malt extract agar) adjusted to different water activities with glycerol, and carbon source utilization using BIOLOG GN MicroPlate were determined in vitro. Decreases in $a_w$ on MEA caused a reduction in mycelial growth and conidial germination depending on temperature. The mycelial growth of PENOX was greatest at $30^{\circ}C/0.995\;a_w$. At some lowered water activity($0.97\;a_w$), the growth was similar between 25 and $30^{\circ}C$, and considerably decreased at lowered temperature($20^{\circ}C$). The germination rate was also greatest at $30^{\circ}C/0.995\;a_w$. Lag phase times for PENOX at $18^{\circ}C$ on MEA were >6hrs at tile whole $a_w$ level tested, and at 18 and $25^{\circ}C$ they were >18hrs and >12hrs at $0.94\;a_w$, respectively. However, its sporulation was some better at $0.97\;a_w$ than $0.995\;a_w$ or $0.94\;a_w$, and better at $20^{\circ}C$ than $30^{\circ}C$. In contrast, the number of carbon sources(niche size) utilized by PENOX varied with $a_w$ and temperature. Under some water stress condition($0.95\;a_w$), the agent utilized smaller number of carbon sources than $0.995\;a_w$ depending on temperature. The niche size at 0.995 and $0.95\;a_w$ were highest at $25^{\circ}C$, and showed 86 and 65, respectively. At $30^{\circ}C$, the niche size at 0.995 and $0.95\;a_w$ showed 84 and 50, respectively. There was no carbon source utilized by PENOX at $0.90\;a_w$ regardless of temperature. These information of tile fungal ecophysiology will be useful for the effective development of BCA.

Effect of Temperature, Relative Humidity on Germination and Development of Powdery Mildew (Leveillula taurica) on Pepper and Its Inoculation Method (고추 흰가루병균(Leveillula taurica)의 발아특성 및 접종법)

  • Pepper powdery mildew, Leveillula taurica is one of the most important pathogens of pepper in the greenhouses and fields in Korea and is becoming a worldwide disease. These experiments were carried out to investigate the optimal germination, disease development conditions, disease incidence and effective inoculation methods of pepper powdery mildew. The incidence of powdery mildew was investigated throughout the country based on the major pepper growing areas in 2009. The average rate of infected leaves ranged from 15.3% to 81.8% in greenhouses and fields. Powdery mildew incidences were more severe in greenhouses than those of fields. The optimal temperature for conidial germination was $25^{\circ}C$ and showed the highest germination at 6 hs after treatment. The range of temperature for germination was $10^{\circ}C$ to $35^{\circ}C$. Temperature of greater than $30^{\circ}C$ and below $20^{\circ}C$ affected the abnormal germination and germ tube elongation. The optimal relative humidity for germination and germ tube elongation was 85% and germination increased as relative humidity increased. Disease development started 8 days after inoculation and showed the highest disease severity at 15 days in greenhouse and field regardless of varieties. Among three different inoculation methods tested the spore dropping method was better than touching with infected leaves and spraying with spore suspension. However, the method has limitation in mass inoculation due to the amount of time consumed.

Studies on Sheath Rot of Rice Caused by Acrocylindrium oryzae Sawada -Testing Varietal Reactions and Culture Filtrates of the Causal Fungus - (벼 잎집썩음병에 관한 연구 -품종저항성검정, 병균의 배양여액-)

  • Occurrence of sheath rot rice caused by Acrocylindrium oryzae on indica varieties, IR lines and IR varieties was light to moderate in the field, whereas generally none of the recommended japonica varieties showed the disease. In general, percentage infection in the field agreed with that obtained from seed inoculation tests made in the laboratory. Culture filtrates of the fungus inhibited the sprouting of seeds of rice, barley, wheat, rye and rape. A new variety, Tongil obtained from a hybrid between japonica and indica was inhibited significantly more than was Jinheung. Conidial germination of Pyricularia oryzae was inhibited by the filtrate while no inhibitory effects were found on conidia of Cochliobolus miyabeanus.

In Vitro and Greenhouse Evaluation of Cucumber Growth Enhanced by Rhizosphere Microorganisms (실험실내와 비닐하우스에서 근권 미생물에 의한 오이 생육증진의 검정)

  • We developed an in vitro assay method for evaluating plant growth promotion and providing an evidence that the growth promotion is rendered by growth enhancing factors. The amendment of culture filtrates of Trichoderma harzianum T95 and Gliocladium virens G872 and G872B in Murashige and Skoog (MS) agar medium enhanced the cotyledon growth of cucumber in terms of fresh weight and primary leaf area of cucumber cotyledon cuttings, of which the treatment of G. virens G872B was the most effective. The mycelial culture filtrate of G872B was more effective in the growth promotion than its conidial germling filtrate. The addition of 1% sucrose in MS mineral medium with 0.1% culture filtrates of the antagonists (T95 and G872B) was optimum for enhancing the effect of the filtrates on the growth of cotyledon cuttings in vitro. When cucumber seeds treated with G872B, Pseudomonas putida Pf3 or the G872B-Pf3 mixture were planted in a greenhouse, the rate of seed germination, biomass of shoot and root, and yield of cucumber fruits were increased, especially by G872B or the G872B-Pf3 mixture. Correspondingly, cucumber fruit yields in early to middle-cycles of harvest were significantly greater in the plots of G872B than the control and Pf3-treated plots, and the final yield was highest in the plots of the G872B-Pf3 mixture applications.

Pre-Infection Behavior of the Pitch Canker Fungus Fusarium circinatum on Pine Stems

  • Pre-infection behavior of Fusarium circinatum on stems of pine species was investigated with scanning electron microscopy. Two-year-old stems of Pinus densiflora and p. rigida were inoculated with the fungal conidial suspension and subjected to $25^{\circ}C$ for up to 16 hr. Most microconidia germinated 12 hr after inoculation on pine stems. Conidia produced germ tubes from either one or both ends of microconidia. Germ tubes grew over the stem surface and appeared to enter host tissues through natural openings on pine stems. Surface cracks in the cork were entrance sites of germ tubes of F. circinatum. In addition, host cell wall cracks were often found at the tip of germ tubes. The cuticle appeared to be eroded either at the tip of germ tubes or around germlings. Germ tubes also produced appressoria-like structures, exhibiting swollen tips of germ tubes on the stem surface. There seems to be no significant differences in the pre-infection behavior of F. circinatum on stems between the two pine species.
