The characteristic Ramularia type conidia of Mycosphaerella nawae were formed on naturally infected leaves. Artificial inoculation with the conidia induced typical symptom on leaves, which was not distinguishable from those of ascospore infection, which has been considered as a primary inoculum source in nature. Also the morphology of the conidia produced on PDA was not different from those formed on artificially inoculated leaves or on naturally infected leaves at later stage of symptom development. Accordingly, we report the role of the conidia as a secondary inoculum of the circular leaf spot pathogen of persimmon for the first time.
The effects of media, incubation time, temperature and pH on production of conidia and chlamydospore of Cylindrocarpon destructans (Zinssm.) Scholen causing root rot of Panax ginseng were studied. Microconidia of the pathogen were abundantly produced on V-8 juice agar as a solid substrate with 5.73(log conidia/mm2) and in V-8 broth as a liquid substrate with 6.65 (log conidia/ml) among media tested. No difference was observed on the length of microconidia produced from the media with a range of 9.50∼11.38 $\mu\textrm{m}$. However, tryptic soy agar produced the broadest microconidia (average 5.00 $\mu\textrm{m}$) among the media tested. All the media produced chlamydospores In a range of 1.06∼4.37 (log chlamydospores/mm2) without a significant difference in number, while V-8 juice agar produced the bigger one (18.39 $\mu\textrm{m}$ in diameter) as compared to the tested media. The fungus began to sporulate conidia after three days of incubation and reached maximum at the 8th day. It seemed to be in a stationary phase until 30 days of incubation but was decreased thereafter. Chlamydospore was produced at 4th day after incubation. Maximum production was observed at 8th day and the number seemed to be maintained during the observation period. Both conidia and chlamydospore of the pathogen were able to be spoluated at 10∼25$^{\circ}C$. However, optimum temperatures of conidia and chlamydospore formation were 15∼25$^{\circ}C$ and 10∼20$^{\circ}C$, respectively. C. destrmtans produced conida with an wide range of pH from 3.3 to 8.0 and chlamydospore from 2.8 to 8.0. Number of conidia was increased with an increase of pH up to 4.0. There was no significant difference in the number between 4.0 to 8.0. It seemed to have two optimum pH ranges, 3.3∼4.0 and 7.1∼8.0 for the chlamydospore formation.
Conidial germination of Septoria glycines Hemmi, brown spot fungus of soybean, was studied by slide germination test. Poor conidial germination of S. glycines was observed on sterile distilled water, but potato dextrose agar(PDA) and distilled water floated with soybean leaf disc furnished a satisfactory medium for conidial germination. Exogenous supply of carbon source was essential for conidial germination, while phosphorous and potassium were not evident as that for carbon. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $5\times10^{-2}mol$. concentration of glucose. Germination was decreased with increment of conidial concentration and was almost completely suppressed in the density of 10,000 conidia per $mm^2$. It suggested existing a self-inhibitor(s). Non-washed conidia germinated more than washed conidia and this was obvious when the conidia density was over $2\times10^3$ conidia per $mm^2$ on the dry agar block.
Aspergillosis is a life-threatening disease in individuals with compromised immune systems. Fungal invasion is a highly critical process during host cellular infection. Several papers have reported that transcription factors are responsible for the infection process. To investigate what transcription factors are involved in the process in an effort to inhibit fungal infection into cells, I checked the surfactant protein family and PU.1 transcription factor levels in A549 cells infected with A. fumigatus conidia. PU.1 and surfactant protein-D levels were reduced in cells infected with fungal conidia. I then observed an increase in surfactant protein-D on PU.1-overexpressed cells. Infection of A. fumigatus conidia was decreased in PU.1-overexpressed cells, whereas the suppression of PU.1 did not lead to any changes in cases of A. fumigatus conidia infection. These results indicate that PU.1 inhibits the infection of A. fumigatus conidia via the expression of surfactant protein-D, suggesting that PU.1 is a key transcription factor for protection against A. fumigatus invasion.
Direct effects of atmospheric ozone on conidia of the rice blast pathogen, Magnaporthe grisea, were investigated to evaluate ozone-induced effects on infection potential of the rice blast fungus. Acute ozone exposure (200 nl $1^{-1}$, 8 h $day^{-1}$3 days) during sporulation significantly affected conidial morphology, appressorium formation, and disease development on rice loaves. Ozone caused reduction in conidial size and change in conidial shape. Relative cytoplasmic volume of lipids and vacuoles were increased in ozone-exposed conidia. Inhibition of appressorium formation and simultaneous increase in endogenous levee of polyamines were found in ozone-exposed conidia. The inverse relationship between appressorium formation and level of polyamines implies that ozone-mediated increase in intracellular level of polyamines may inhibit appressorium formation in rice blast fungus. Furthermore, rice plants inoculated with ozone-fumigated conidia exhibited less severe disease development than those with unfumigated conidia. This result suggests that the anti-conidial consequence of acute ozone will eventually weaken the rice blasts potential for multiple infection cycle. This further suggests that consequently, rice blast can be transformed from an explosive disease to one that has limited epidemiological potential in the field.

An entomopathogenic fungus, Paecilomyces fumosoroceus SFP-198 was isolated in Kyungbuk province, Korea and screened out for the control of Trialeurodes vaporariorum nymphs. It showed 72.5% efficacy against second instars of T. vaporariorum nymphs at 6 days after treatment in the laboratory. To select an active ingredient for the mass production and the formulation, SFP-198 culture products were compared based on their insecticidal activities against T. vaporariorum nymphs. Among them, conidia and blastospores showed much higher insecticidal activity than supernatants. Furthermore, SFP-198 conidia were more heat-resistant than blastospores, and also powder form of conidia was more stable than their suspension form. SFP-198 conidia showed high pathogenicity on not only T. vaporariorum but also Tetranychus urticae in the glasshouse. This result suggested that SFP-198 conidia can be used as a multi-targeting biological control agent against sucking agri- cultural pests, such as whiteflies and mites.
Conidial development of Cordyceps militaris was observed from germinating ascospores and vegetative hyphae through light and scanning electron microscopy (SEM). Ascospores were discharged from fresh specimens of C. militaris in sterile water as well as Sabouraud Dextrose agar plus Yeast Extract (SDAY) plates. We observed ascospore germination and conidial formation periodically. Under submerged condition in sterile water, most part-spores germinated unidirectionally and conidia were developed directly from the tips of germinating hyphae of part-spores within 36 h after ascospore discharge, showing microcyclic conidiation. First-formed conidia were cylindrical or clavate followed by globose and ellipsoidal ones. Germination of ascospores and conidial development were observed on SDAY agar by SEM. Slimy heads of conidia on variously arranged phialides, from solitary to whorl, developed 5 days after ascospore discharge. Besides, two distinct types of conidia, elongated pyriform or cylindrical and globose, were observed in the same slimy heads by SEM. Conidia were shown to be uninucleate with 4,6-diamidino-2-phenylindole staining. Conidiogenous cells were more slender than vegetative hyphae, having attenuated tips. Microcyclic conidiation, undifferentiated conidiogenous hyphae (phialides), polymorphic conidia and solitary, opposite to whorled type of phialidic arrangement are reported here as the characteristic features of asexual stage of C. militaris, which can be distinguished from other Cordyceps species.
Sporulation patterns of rice blast fungus were studied at relatively later stages of leaf blast and neck blast seasons in Icheon, Korea. This experiment was done by detaching lesion-bearing leaves and panicle bases. The number of conidia remaining on the leaf blast lesions of different cultivars from Jul 20 to Jul 23 ranged from 3,640 to 82,740 spores. More conidia were observed on the adaxial surface because they were released from abaxial surface. After heading, sporulation was observed from the lesions on the flag leaves but the number of spores was less than in the late July. Detached panicle bases or uppermost internodes infected by Pyricularia grisea produced abundant amount of conidia. Among these panicle bases, 30.1 mm size lesion recorded the highest count of 244,560 spores. When we compared the sporulation amount using the KY-type spore trap, more conidia were recorded from intact lesions than from the lesions which removed conidia and conidiophore The ratio of conidia release against total sporulation ranged from 20.5%-25.0% for leaf blast and 8.2%-25.3% in the neck blast. Effective inoculum potential was also discussed.
Biological control of forest insect pests, Agelastica coeruleci, Meganola n~elancholia,a nd Glyphodes perspectalis, turfgrass insect pest, Blitoperfhu orientr~lis,v egetable insect pests, Plutella xylostella and Agrotis segetun~ with entomopathogenic fungus, Beciuveria hassinna GY 1 - 17 isolated from rice paddy of Yangsan in the southern part of Korea were investigated. Mortalities of A. coeruleu and P. ~ylo.~rellluar vae were 100% at the rate of 7.0 and 2.0 x 107conidia/ml after 7 and 5 days and those of M. melancholia were 66.7 - 100% at the rate of 0.03875-3.1 X 107conidia/ml. However, G. perspectulis was not affected at the rate of 2.0 x lo7 to X I04conidia/ml. And mortalites of B. orientcilis and A. segetum larvae were 46.7% at 3.7 x 107conidia/ml and 63.3% at 2.5 X 107conidia/ml.
To find optimum application methods of Epicoccosorus nematosporus for control of water chestnut, five different concentration of conidial suspensions ($10^[3}$ conidia/ml to $10^[7}$ conidia/ml) fo the fungus were applied 1 to 4 times on 10~40 days old seedlings of water chestnut in greenhouse. Inoculum levels equal to or greater than $10^[5}$ conidia/ml killed significantly more shoots (82.6%~92.1%) and suppressed significantly underground tuber formation compared to inoculum concentration less than $10^[4}$ conidia/ml. When the conidial suspension of ($6.3 {\times} 10^{5}$conidia/ml of E. nematosporus was sprayed 2 times in 7 days interval, percentage of the killed plants was up to 98.7%. Numbers of reshoots and tubers were alsosuppressed significantly compared to one time application. The percents of killed shoots were similar between 10-day-old and 20-day-old seedlings, and significantly higher than those treated on 30- or 40-day-old seedlings. The fungus treated on 20-day-old seedlings was the most effective because there was high number of reshoots from 10-day-old seedlings. Therefore, optimal application conditions for E. nematosporus is 2~3 times of application in 7 days interval with $10^[5}$ conidia/ml on 20-day-old seedling age.
