Study for comparison with male & female rat locus coeruleus using confocal laser scanning microscopy (공초점현미경을 이용한 암수 흰쥐 청색반점의 비교연구)

  • The locus coeruleus(LG) is known to be observed a sexual dimorphism in rat CNS. LC is the largest collections of norepinephrine(NE)-containing neurons in the mammalian brain. Especially in rat, all LC neurons contained NE unlike other mammalians, so that specific reactions were found in the tyrosine-hydroxylase(TH) immunoreactive neurons. Sexual dimorphism of rat LC has affected by genes before sex hormone appeared, thereafter affected by sex hormones. In these day, many scientists founded morphological differences between male and female LC morphology, but differences of entire structure was not founded. Thus we investigated sex differences of the LC neuron's morphology in rat by three-dimensional(3-D) reconstruction using Confocal laser scanning microscopy(CLSM). We reported that neuron's shape was relatively-large multipolar neurons and neuron's processes in dorsal LC proceeded to ventral direction in the male and female rat. Male had a longer anterior-posterior length than female had in dorsal LC. In addition to middle-LC, male rat's LC had a more thicker posterior region but had not viewed in a previous study. In reverse, female rat's LC had a thicker anterior region like a previous study. This results using 3-D reconstruction by CLSM showed that the male's LC was more wide-ranging than female's relatively.

Autofocus of Infinity-Corrected Optical Microscopes by Confocal Principle and Fiber Source Modulation Technique (공초점 원리와 광섬유 광원 변조를 이용한 무한보정 현미경 자동초점)

  • The autofocus is one of the important processes in the automated vision inspection or measurements using optical microscopes, because it influences the measuring accuracy. In this paper, we used the confocal microscope configuration based on not a pinhole but a single-mode optical fiber. A single mode fiber has the functions of source and detector by applying the reciprocal scheme. As a result, we acquired a simple system configuration and easy alignment of the optical axis. Also, we embodied a fast autofocus system by acquiring the focus error signal through a source modulation technique. The source modulation technique can effectively reduce physical disturbances compared with objective lens modulation, and it is easily applicable to general optical microscopes. The focus error signal was measured with respect to the modulation amplitude, reflectance of the specimen and inclination angle of the measuring surface. The performance of the proposed autofocus system was verified through autofocusing flat mirror surface. In addition, we confirmed that source modulation rarely degrades the depth resolution by the comparison between the FWHMs of axial response curves.

  • A laser ultrasonic inspection system is a non-contact inspection device which generates and measures ultrasonics by using laser beam. A laser ultrasonic inspection system provides a high measurement resolution because the ultrasonic signal generated by a pulse laser beam has a wide-band spectrum and the ultrasonic signal is measured from a small focused spot of a measuring laser beam. In this study, galvanic corrosion phenomenon was measured by non-destructive and non-contact method using the laser. The case of mixed foreign material on the part of corrosion was assumed and laser ultrasonic experiment was conducted. Ultrasonic was generated by pulse laser from the back side of the specimen and ultrasonic signal was acquired from the same location of the front side using continuous wave laser and Confocal Fabry-Perot Interferometer(CFPI). The characteristic of the ultrasonic signal of exist foreign material part was analyzed and the location and size of foreign material was measured.

A CONFOCAL MICROSCOPIC STUDY ON DENTINAL INFILTRATION OF ONE-BOTTLE ADHESIVE SYSTEMS AND SELF-ETCHING PRIMING SYSTEM BONDED TO CLASS V CAVITIES (제 5급 와동에서의 단일용기 상아질 접착제와 자가 산부식 접착제의 상아질에 대한 침투도 평가)

  • Objective : The purpose of this study was to evaluate the resin infiltration into dentin of one-bottle adhesive systems and self-etching primer bonded to Class V cavities using confocal laser scanning microscope(CLSM). Material and Methods : Forty Class V cavities were prepared from freshly extracted caries-free Human teeth. These teeth were divided into two groups based on the presence of cervical abrasion: Group I, cervical abrasion : Group II, wedge-shaped cavity preparation. Resin-dentin interfaces were produced with two one-bottle dentin bonding systems-ONE COAT BOND(OCB; Coltene$^R$) and Syntac$^R$SPrint$^{TM}$(SS; VIVADENT)-, one self-etching priming system-CLEARFIL$^{TM}$ SE BOND (SB : KURARAY)- and one multi-step dentin bonding system-Scotchbond$^{TM}$Multi-Purpose (SBMP, 3M Dental Products)-as control according to manufacturers' instructions. Cavities were restored with Spectrum$^{R}$(Dentsply). Specimens were immersed in saline for 24 hours and sectioned longitudinally with a low-speed diamond disc. The resin-dentin interfaces were microscopically observed using CLSM. The quality of resin-infiltrated dentin layers were evaluated by five dentists using 0~4 scale. Results : Confocal laser scanning microscopal investigations using primer labeled with rhodamine B showed that the penetration of the primer occurred along the cavity margins. Statistical analysis using one-way ANOVA followed by Duncan's Multiple Range test revealed that the primer penetration of the group 2(wedge-shaped cavity preparation) was more effective than group 1(cervical abrasion) and that of the gingival interfaces was more effective than the occlusal interfaces. In the one-bottle dentin bonding systems, the resin penetration score of OCB was compatible to SBMP, but those of SS and self-etching priming system, SB were lower than SBMP.

A CONFOCAL LASER SCANNING MICROSCOPIC STUDY ON THE INTERFACE BETWEEN TOOTH COLORED RESTORATIVE MATERIALS AND DENTIN (공초점레이저주사현미경을 이용한 심미수복재와 상아질의 접착계면에 관한 연구)

  • The purpose of this study was to evaluate on the interfacial morphology between dentin and restorative materials. In this in vitro study, the cavity wall restorated with 3 different kinds of tooth colored restorative materials [resin-modified Glass Ionomer cement (Fuji II LC), composite resin (Z-100), compomer (Dyract)]. The thirty extracted human molar teeth without caries and/or restorations are used. The experimental teeth were randomly divided into three groups of ten teeth each. In each group, Wedge shaped cavities (width: 3mm, length: 2mm, depth: 1.5mm) were prepared at the cementoenamel junction on buccal and lingual surfaces. The adhesive of composite resin were mixed with rhodamine B. Primer of composite resin, Prime & Bond 2.1 of Dyract and liquid of Fuji II LC were mixed with fluorescein. In group 1, the cavity wall was treatment with dentin conditioner, and then restorated with Fuji II LC. In group 2, the cavity wall was treatment with Prime & Bond 2.1 and then restorated with Dyract. In group 3, the cavity wall was etching with 10% maleic acid, applied with primer and bonding agent and then restorated with Z-100. The interface between dentin and restorative materials was observed by fluoresence imaging with a confocal laser scanning microscope. The results were as follows : 1. In Glass ionomer group, adaptation of resin modified Glass-ionomer restoration against cavity wall is tight, but the crack formed inside of restoration were observed. 2. In Dyract group, the penetration of resin tag is shorter and the width of hybrid layer is narrower than composite resin group. 3. In Z-100 group, primer penetrated deeply through dentinal tubule. Also bonding agent was penetrated along the primer, but the penetration length is shorter than primer part, and in 3-D image, the resin tag is conical shape and lateral branch is observed.
