Simple Assay Method for Determination of Capsaicinoid Synthetase Activity

  • A new method to assay the capsaicinoid synthetase (CS) activity was developed by utilizing NADHcoupled enzyme systems involving pyruvate kinase and lactate dehydrogenase. CS activities in Capsicum placenta, depending upon the kinetics of the NADH oxidation, revealed almost the same profile as compared with those shown using an HPLC-based method. When the substrates, 8-methyl nonanoic acid and vanillylamine, for the CS enzyme were employed separately or simultaneously, it appeared that the two-step reaction, acyl-CoA formation and condensation with vanillyla~ne, of the CS enzyme was a coupled reaction. Thus, this assay method of the CS enzyme can be considered as an alternative to the HPLC-based method, since it has the advantages of rapidity and simplicity as well as reliability when compared with the existing method.

Synthesis and Surface Active Properties of Long Chain N-Acyl Collagen Derivatives (고급 지방산 N-아실 콜라겐 유도체의 합성 및 계면활성)

  • The Surfactants composed of acylated aterocollagen which is produced by the acylation of the side chain amino radicals of aterocollagen with an aliphatic acid having 12 to 18 carbon atoms will be discussed in this study. This condensation is done at moderate reaction temperature (<$25^{\circ}C$) in aqueous alkaline solution. The products of this reaction were identified by UV/VIS spectroscopy and infrared spectroscopy. For these compounds, surface active properties and physical properties including isoelectric point, Krafft point, surface tension, critical micelle concentration(cmc), foaming power, viscosity behaviour, water holding capacity, skin irritation and emulsifying power were measured respectively. The experimental results received that the products have a good emulsifying power, excellent water holding capacity while having low skin irritation. Thus, these derivatives will be expected to be used as an emulsifying agent for O/W type cosmetic emulsion.

  • In recent years the interest in organic/inorganic hybrid materials has increased at a fast rate. Nano organic-inorganic hybrid composites have shown advantages for preparing hard coating layers. Especially, nano hybrid composite has low environmental pollution. It has high transparency, hardness, toughness, thermal dissociation temperature, hydrophobicity by using nano sized inorganic material. There are many ways in which these materials may be synthesized, a typical one being the use of silica and silanes using the sol-gel process. The structure of sol-gel silica evolves as a result of these successive hydrolysis and condensation reactions and the subsequent drying and curing. The sol-gel reactions are catalyzed by acids and produce silica sol solutions. The silica sol grows until they reach a size where a gel transition occurs and a solid-like gel is formed. Colloidal silica(CS)/silane sol solutions were synthesized in variation with parameters such as different acidity and reaction time. In order to understand their physical and chemical properties, sol-gel coating films were fabricated on glass. From all sol-gel solutions, seasoning effect of sol-gel coating layer on glass was observed.

  • To develop a recycling process of magnesium alloy scrap, a fundamental study on the distillation of magnesium alloy melt was carried out. Melt temperature, vacuum degree and reaction time were considered as experimental variables. The amount of vaporized magnesium melt per unit surface area of melt increases with the increase of melt temperature, reaction time and vacuum degree. The vapor condensed at the tip of water cooling Cu-condenser as a form of pine cone. Magnesium and zinc were vaporized easily from the melt. However, It's difficult to separate magnesium and zinc by vacuum distillation because vapor pressure of zinc is similar to one of magnesium. The contents of aluminum, manganese and iron, etc. in residual melt increase due to the decrease of magnesium and zinc content after the distillation of magnesium alloy.

  • The condensation reaction of 9,9'-bis(4-hydroxyphenyl)fluorene with epichlorohydrin to prepare 9,9'-bis[4(glycidyloxy) phenyl]fluorene (2), an important building block for fluorene-containing epoxy polymers, has been studied. The reaction is found to be quite sensitive to several experimental conditions such as reaction temperature and time, added amount of epichlorohydrin, the presence of catalysts and the use of co-solvent. Several conditions for obtaining the best yield in the reaction are: the reaction temperature is below 373 K and the reaction time is shorter than 1.5 h, and the ammonium salts act as a catalyst. Also, the use of ternary solvent (toluene, DMSO, water) has been proved to be crucial to maintain the reaction temperature and for an easy purification. Thus, the reaction proceeds in an environment-friendly manner where the use of reactants and the production of chemical wastes is minimized.

  • Recently, synthetic polymers containing units of carbohydrate derivatives with pendatnt functional groups have been much studied. Epoxidation of Styrene using Nanosized γ-Al2O3/NiO Heterogeneous Catalyst Derived from the P123 Surfactant

  • $Al_2O_3$/NiO powder was obtained through hydrolysis-condensation reactions and thermal treatments. An organic additive, triblock copolymer surfactant P123, was added to the starting materials to control the surface area and morphology. The synthesized powder was characterized by X-ray diffractometry (XRD), field-emission scanning electron microscopy (FE-SEM) and a Brunner-Emmett-Teller surface analysis (BET). The heterogeneous catalytic activity of this powder was applied to an epoxidation reaction of styrene and was monitored using a gas chromatograph with mass spectrophotometry (GC/MS).

  • Phenol-free resin was synthesized and its printing ink properties were investigated. The phenol-free resin was produced by esterification of poly phthalate and Diels-Alder adduct of rosin anhydride. Compared to rosin modified phenolic resin, eco-friendly phenol-free resin showed better vehicle properties in terms of gloss, yellowing, runability, and storage stability. The results suggest the utility of phenol-free resin instead of conventional rosin modified phenolic resin.

  • Optically pure(-)-and (+)-$\alpha$-hydroxy-$\alpha$-(p-chlorobiphenyl)acetic acids were prepared. The racemate was synthesized through three steps. By condensation of p-cnorobiphenyl with diethyl ketomalonate in the presence of SnCl$_{4}$, diethyl $\alpha$-hydroxy-$\alpha$-(p-chlorobiphenyl)malonate (1) was formed and subsequently ($\pm$)-$\alpha$-hydroxy-$\alpha$-(p-chlorobiphenyl)acetic acid (3) was obtained through hydrolysis and decarboxylation. For the separation of the racemate the classical resolution method, derivatization of a racemate by reaction with an optically pure compound was employed. In this case the optically pure compound were [R]-(+)-$\alpha$-methylbenzylamine and [S]-(-)-$\alpha$-methylbenzylamine. Diastereomeric salts between acids and bases could be easily separated by crystallization in absolute ethanol.
