Biological Activities of Various Solvent Extracts from Propolis (프로폴리스 용매별 추출물의 생리활성)

  • The scavenging effects of four extracts on the DPPH radical were increased with increasing amount of extract. However, these effects were not statistically significant. The reducing power of the extracts is increased as the amount of extract is increased. To compare reducing powers of various solvent extracts, 50% ethanol extract showed the highest reducing power. The various solvent extracts were also capable of scavenging nitrite in a manner dependent on concentration. They exhibited scavenging effects 90% on nitrite at the dose 500 p.g of water extract from propolis. The propolis extract significantly inhibited all the microorganisms tested, showing the largest inhibitory zone for Bacillus subtilis and Bacillus cereus. The 70% ethanol extract from propolis exhibited the strongest inhibitory effect on A549. The inhibitory effect of four extracts showed in that order 70% ethanol extract>95% ethanol extract>water extract>50% ethanol extract.

Physiological Activities of Ethanol Extracts from Different Parts of Allium hookeri (삼채부위별 에탄올 추출물의 생리활성탐색)

  • Biological compounds such as crude saponin and total polyphenol were determined in Allium hookeri roots and leaves. In addition, activities of DPPH radical scavenging activity, hydroxyl radical scavenging activity, ${\alpha}$-amylase inhibition activity and nitrite scavenging were detected in concentrates of 70% ethanol extracts of Allium hookeri roots and leaves. The crude saponin content of the roots and leaves were 4.28 mg/g, and 4.17 mg/g, respectively. Total polyphenol and total flavonoid content was higher in leaves than roots. Furthermore the DPPH radical scavenging activity and hydroxyl radical scavenging activities of leaves was higher than roots. For ${\alpha}$-glucosidase inhibitory activity, 70% ethanol extracts of Allium hookeri roots and leaves showed 60% activity inhibition at a 10 mg/mL concentration. Nitrite scavenging abilities under acidic conditions were most effective for Allium hookeri leaves extracts. These results suggested that Allium hookeri leaves might be used as the components of health functional food.

Molecular Mechanism of NO-induced Cell Death of PC12 Cells by $IFN{\gamma}\;and\;TNF{\alpha}$

  • Nitric oxide (NO) is a small, diffusible, and highly reactive molecule, which plays dichotomous regulatory roles under physiological and pathological conditions. NO promotes apoptosis in some cells, and inhibits apoptosis in other cells. In the present study, we attempted to characterize the NO signaling pathway and cellular response in PC12 cells treated with cytokines. $IFN{\gamma}\;and\;TNF{\alpha}$ treatment resulted in a synergistic increase of nitrite accumulation, with the induction of inducible nitric oxide synthase (iNOS) in the PC12 cells. Moreover, as nitrite concentration increased, cell viability decreased. In order to explore MAP kinase involvement in nitric oxide production resultant from $IFN{\gamma}\;and\;TNF{\alpha}$ stimulation, we measured the activation of MAP kinase using specific MAP kinase inhibitors. PC12 cells pretreated with SB203580, a p38 MAP kinase-specific inhibitor, resulted in the inhibition of iNOS expression and NO production. However, PD98059, an ERK/MAP kinase-specific inhibitor, was not observed to exert such an effect. In addition, Stat1 activated by $IFN{\gamma}\;and\;TNF{\alpha}$ was interacted with p38 MAPK. These data suggest that p38 MAP kinase mediates cytokine-mediated iNOS expression in the PC12 cells, and Jak/Stat pathway interferes with p38 MAPK signaling pathway.

DPPH Radical Scavenging Effect and Antimicrobial Activities of Plantain (Plantago asiatica L.) Extracts (질경이 추출물의 DPPH 라디칼 소거효과 및 항균활성)

  • To develop the functional food, methanol extracts of dried plantain (Plantago asiatica L.) was fractionated in hexane, chloroform, ethyl acetate, butanol and aqueous fraction. DPPH radical scavenging effect, reducing power, nitrite scavenging effect and antimicrobial activities of these fractions were investigated. Ethyl acetate fraction among these fractions showed the highest effect of all experiments. Ethyl acetate fraction showed about 60% scavenging effect on the DPPH radicals at 400 $\mu$g/5 mL. Nitrite scavenging effect of ethyl acetate and butanol fraction was more than 70~80% at concentration above of 500 $\mu$g. Among various solvent fractions from methanol extracts of plantain, ethyl acetate fraction showed the highest inhibitory effect on the microorganism such as Staphylococcus aureus, Bacillus cereus and Vibrio parahaemolyticus at 5 mg/disc.

Nitrate Reduction and Pigment Formation of Chinese-Style Sausage Mixes Caused by Micrococcaceae

  • This study investigated the nitrate reduction ability of Micrococcaceae on pigment formation in Chinese- style sausage. One hundred ppm sodium nitrite and 150 ppm sodium nitrate was added asepticly to ground pork which was then inoculated with $10^7CFU/g$ of either Micrococcus varians, Staphylococcus carnosus or Staphylococcus xylosus. All samples were cured at $20{^{\circ}C}$ or $30{^{\circ}C}$ for 3 days and then color, residue nitrite, nitrosyl pigment and residue nitrate were determined. The results indicated that samples inoculated with S. xylosus had higher a- and b- values due to nitrate reduction and pigment production after 3 days curing and these values were higher at the higher curing temperature. The nitrosyl pigment of the samples with S. xylosus had highest values after 3 days curing at both $20{^{\circ}C}$ and $30{^{\circ}C}$. However, sample inoculated with S. carnosus and S. xylosus had lower nitrate contents than the sample inoculated with M. varians. At $30{^{\circ}C}$ as well as S. carnosus and M. varians had a stronger decreasing in nitrate concentration during curing at $20{^{\circ}C}$. Moreover, samples inoculated with S. xylosus and S. carnosus had a higher residual nitrite content during curing at $20{^{\circ}C}$ or $30{^{\circ}C}$. In conclusion, two Staphylococci strains tested were most optimum starter cultures for improving pigment formation during Chinese-style sausage curing.

Antioxidative Activities and Nitrite Scavenging Abilities of Extracts from Ulmus devidiana (느릅나무 추출물의 항산화 효과 및 아질산염 소거능)

  • To measure antioxidative activities, the various extracts from Ulmus devidiana were examined in oil emulsion. Water, ethanol, methanol and butanol were used as extract solutions. The activity oxygen species ($H_2O_2,-OH,\;KO_2$) bound the extracts for antioxidative activities were excellent. The extracts bound $Fe^{2+}$ ion and $Cu^{2+}$ ion showed effective antioxidative activities and strong chelating effects. The concentration of $Fe^{2+}$ ion and total ion in ethanol and methanol extracts from Ulmus devidiana root parts (Chinese) were higher than those of the other products. The highest superoxide dismutase-like activities showed butanol extracts from Ulmus devidiana root parts (Chinese) and water extracts from Ulmus devidiana bark parts (Korean). Electron donating abilities and nitric scavenging abilities of ethanol, methanol and butanol extracts were higher than those of water extracts. The nitrite scavenging abilities also reached the maximum at pH 1.2 and the minium at pH 6.0.

Estimation of Nutrients Released from Sediments of Deukryang Bay (득량만 퇴적물로부터 영양염 용출 평가)

  • The purpose of the present study is to estimate the release of dissolved Inorganic nitrogen and photophorous from sediments of Deukryang Bay. One method used In this study is to calculate nutrients released from a concentration gradient between sediment porewaters and the overlying water based on periments. The calculated and measured ammonium released from the sediments were 8.93mg-atN/$m^2$ hr and 60.4mg/atN/$m^2$ hr, respectively In July. 8.57mg-atN/$m^2$. hr and 32.9mg/atN/$m^2$ hr, respectively in October. The ammonium was released more highly in truly than in October, and the measured ammonium flux was higher than the calculated one. The calculated nitrate plus nitrite released from the sediments were 0.31mg/atN/$m^2$. hr in July and 0.84mg/atN/$m^2$. hr in October. The measured nitrate plus nitrite released from the sediments was 282mg/atN/$m^2$. hr in October. The calcuated was lower than the measured because the content of the nutrients in the sediments was always much more than in the overlying waters, and it has shown a differently seasonal pattern compared to the ammonium flux. The calculated phosphorous released from the sediments were 0.97mP/atN/$m^2$ hr and measured negative fluxed -6.50mP/atN/$m^3$ hr In truly, and alculated 0.18mg/atP/$m^2$. hr and measured 24.6mg/atP/$m^2$. hr, respectively in October.

Removal Characteristic of Ammonia Nitrogen and Behavior of Nitrogen in Synthetic Wastewater Using Leclercia Adecarboxylata (Leclercia Adecarboxylata를 이용한 합성폐수의 암모니아성질소 제거특성 및 질소거동)

  • In this study, the removal characteristic of ammonia nitrogen and behavior of nitrogen was investigated using Leclercia adecarboxylata, which was derived from the culture contaminated by ammonia nitrogen of high concentration. The method of ammonia nitrogen removal was not biological nitrification and denitrification but elimination of nutrient salt with internal synthesis of microorganisms which use ammonia nitrogen as substrate. L. adecarboxylata(one of ammonia synthesis microorganisms) was highly activated and showed the most high removal efficiency in free salt condition but the removal efficiency decreased badly in salt concentration of more than 4%. About 80 mg/L of $NH_3-N$ was mostly removed within 20 hours and 500 mg/L of $NH_3-N$ showed less then removal efficiency of 50% because carbon source was not enough. However, ammonium nitrogen concentration was decreased again when the carbon source was inserted additionally thus, ammonium nitrogen removal efficiency by L. adecarboxylata, was related to amount of carbon source. pH decreased from 8.0 to 6.36 according to growth of L. adecarboxylata. Concentration of nitrite nitrogen and nitrate nitrogen did not increase and TKN concentration showed no variation while ammonia nitrogen was removed by L. adecarboxylata. In addition to, when content of protein in organic nitrogen was measured, protein was not detected at the beginning of microorganism synthesis but protein of 193.1 mg/L was detected after 48 hours. Hence, ammonium nitrogen was not decomposed as nitrate nitrogen and nitrite nitrogen but synthesized by L. adecarboxylata, which has excellent ability of nitrogen synthesis and can threat ammonia nitrogen of high concentration in wastewater.

  • In order to study on the variational characteristics of water quality and chlorolphyll a concentration the water samples were collected daily or three times a week during the period from April 1990 to November 1991 at Kukdong port located in the northern Kamak bay of Southern Korea I made an analysis on biological factor as chlorophyll a concentration as well as physico-chemical factors such as water temperature salinity sigma-t dissolved oxygen, nutrients (ammonia, nitrite, nitrate, phosphate, and silicate) N/P ratio and chemical oxygen demand. In Northern Kamak bay seasonal variations in physical factors such as water temperature salinity and sigma-t were very marked. On the other hand chemical factors such as nutrients concentration and COD were not so. Chemical factors, in particular silicate were influenced by input of freshwater. And the roles of silicate on the seasonal succession of phytoplankton species composition was very low. Phytoplankton biomass as measured by chlorophyll a concentration was very high all the year round and it was controlled by the combination of several factors especially of N/P ratio determined by dissolved inorganic nitrogen.

Macrophage Activation of Chitosan Hydrolysates with Different Molecular Weights in vitro (분자량에 따른 Chitosan의 in vitro 대식세포 활성화)

  • This study was performed to investigate the average molecular weight range of chitosan hydrolysates showing maximum effect in macrophage activation. Nitrite production by continuous macrophage cell line J774A.1 was the highest at $10\;{\mu}g/ml$ concentration of intact chitosan. Hydrogen peroxide production by J774A.1 showed the high value of $894\;{\mu}M/mg$ macrophage protein at $1,000\;{\mu}g/ml$ concentration of chitosan hydrolysate fraction 5 and $1,044\;{\mu}M/mg$ macrophage protein at $100\;{\mu}g/ml$ concentration of the fraction 6. Chitsan hydrolysate fraction 4, fraction 6 and intact chitosan enhanced $IL-1{\alpha}$ production, while the others did not. The production of tumor necrosis factor showed the high value at $1,000\;{\mu}g/ml$ concentration of chitosan hydrolysate fraction 4, $100\;{\mu}g/ml$ concentration of the fraction 5 and fraction 6, and $10\;{\mu}g/ml$ concentration of intact chitosan. In conclusion, fractions 4, 5 and 6 of the chitosan hydrolysatets with average molecular weight of $24,000{\sim}64,000$ calculated by HPLC analysis are the most effective in macrophage activation tested in this study.
