Assessment of Allelopathic Potential and Antioxidant Activity of Leaf Extracts from Three Compositae Plants

  • Some Compositae plants are known to contain biologically active substances that are allelopathic to weeds species as well as antioxidant to foods. Aqueous extracts from leaves of 3 plant species, Cirsium japonica, Kalimeris yomena, and Lactuca saliva, were bioassayed against alfalfa (Medicago saliva) to determine their allelopathic effects. The extracts applied on filter paper in bioassay significantly inhibited root growth of alfalfa. Extracts of 20 g dry tissue $\textrm{L}^{-1}$ from Lactuca sativa showed the most inhibitory effect on alfalfa seedling growth and followed by Cirsium japonica and Kalimeris yomena. Oxidative stability by Rancimat method and antioxidant activity by TBA method for the ground samples were the greatest in Lactuca sativa although were less than that of a commonly used antioxidant, 1% ascorbic acid. Antioxidant activity of methanol extracts on storing meat was stably kept for 28days and was excellent compared to control. These results suggest that three Compositae plants have potent allelopathic and antioxidant effects, and that their activities differ, depending on plant species.

  • Compositional differences in flavonoids are varied in the big family of Compositae. By summarizing our previous analytical studies and other scientific evidences, new strategy will be possible to further analyze flavonoids and utilize them for human health. The HPLC analytical method has been established in terms of linearity, sensitivity, accuracy, and precision. Herbs of the family of Compositae have considerable amounts of peroxynitrite ($ONOO^-$)-scavenging effects and their phenolic substances. These effects may contribute to the prevention of disease associated with excess production of $ONOO^-$, depending on the high content of flavonoid substances.

Isolation of Angiotensin Converting Enzyme Inhibitors from Compositae Plants

  • Thirty plants of the Compositae family were screened for inhibitory activity of angiotensin converting enzyme (ACE). Among them, Chrysanthemum boreale Makino and Ixeris dentate Nakai were selected for further investigation since they had the highest inhibitory activity. Crude water extracts of the flowers of Chrysanthemum boreale Makino and the roots of Ixeris dentate Nakai were prepared by heating at 95$^{\circ}C$ and 6$0^{\circ}C$ for 2 hr, respectively, followed by centrifugation at 8000$\times$ g far 30 min. Crude extracts were then filtered using YM-3 and YM-1 membranes. The ACE inhibitors were isolated using consecutive chromatographic methods including: Sephadex G-15, ion exchange, FPLC, and reverse phase HPLC. The inhibitors were identified to have molecular masses of 204 and 283 daltons, respectively, by mass spectrometry. These results demonstrate that crude extracts of Compositae plants may be useful as functional food ingredients with anti-hypertensive properties.

Effect of Compositae Plants on Protein Levels in Streptozotocin-induced Diabetic Rats

  • The investigation assessed the influence of Compositae plants consumption on the protein profile in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats by injection of STZ (45 mg/kg body weight) into tail vein. The rats were randomly assigned to five groups: normal and STZ-control fed an AIN-93 diet, and groups whose diets were supplemented with 10% Compositae powder containing Artemisia iwayomogi (A. iwayomogi), Atractylodes lancea (A. lancea) or Taraxacum mongolicum (T. mongolicum). To observe the effects of Compositae plants in the animal model, the levels of protein in liver, kidney, lung, pancreas, and brain were determined after 4 weeks. The level of protein in kidney increased significantly in rats receiving the A. iwayomogi- and T. mongolicum-supplemented diet compared to the STZ-control group. The level of protein in lung was increased significantly in the A. iwayomogi-supplemented group. Blood glucose level correlated well with brain protein level but did not correlate with other protein levels. Also, blood glucose correlated inversely with kidney, lung and brain protein levels. It is suggested that supplementation with A. iwayomogi in diabetic rats leads elevates protein in kidney and lung.

  • 대구광역시 팔공산에서 국화과 참취속의 1종인 Aster hispidus var. lleptocladus를 발견하여 긴쑥부쟁이로 명명하였다. 일본의 사문암지대에 유존적으로 분포하는 일본 고유종으로 알려져 온 긴쑥부쟁이는 모종인 갯쑥부쟁이와는 하부 경생엽이 좁고 선형이며 예두로써 털이 없으며, 두화와 총포의 지름이 좁고, 통상화의 관모 길이가 1-2 mm인 점이 다르다.

삽주의 18S rRNA 유전자의 염기서열 결정, 계통분류학적 분석 및 atractylon 분석 (DNA Sequencing and Phylogenetic Analysis of the 18S rRNA Gene of Atractylodes japonica Koidz and Analysis of Atractylon)

  • The region containing 18S rRNA gene, ITS 1 and part of the 5.8S rRNA gene of the Atractylodes japonica Koidz was amplified by PCR and the product cloned in a pBluescript SK II plasmid. DNA sequence of the cloned DNA was determined and submitted to the GenBank (accession number EU678363). Phylogenetic analysis of the ITS 1 DNA showed close similarity with the other plant species of the family Compositae. The extract of the plant materials of five different members of the family Compositae was analyzed by HPLC to detect atractylon. Extract of the A. japonica Koidz showed presence of significant amount of atractylon. However, noticeable amount of atractylon was not detected by the same analyses from the extracts of the other plants belonging to the family Compositae including Artemisia capillaris, Chrysantemum zawadskii, Eclipta prostrata or Taraxacum platycarpum.

  • Cirsium xanthocanthum (Compositae) is a perennial herb. From the roots of this plant which were used for arthritis, neuralgia and carbuncular abscess as folkloric remedies, linarin, mp $255{\sim}257^{\circ}$, was isolated and identified by chemical reactions and spectral data with authentic samples.

Antioxidative Components from the Aerial Parts of Lactuca scariola L.

  • The antioxidant activity of Lactuca scariola (Compositae) was investigated by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl) radical. The methanolic extract of the aerial parts of Lactuca scariola showed strong radical scavenging activity. The EtOAc soluble fraction exhibited a stronger activity than the others, and was purified by silica gel and Sephadex LH-20 column chromatography. Quercetin-3-O-$\beta$-D-glucopyranoside, luteolin-7-O-$\beta$-D-glucopyranoside, luteolin, quercetin and kaempferol, together with 11$\beta$,13-dihydrolactucin were isolated from the EtOAc soluble fraction as active ingredients.

천연물 연구에서의 Peroxide 성분 (Peroxide Constituents in the Natural Product Research)

  • Peroxides in natural products have been recently received a considerable attention due to their various biological and pharmacological properties. Nearly 300 peroxides have been isolated and structually characterized from natural sources, mainly as constituents of Compositae and marine sponge, and occur randomly in about 10 other plant families. Among peroxides studied, sesquiterpene endoperoxide, quinghaosu, has been already clinically applied as a new antimalarial drug. Based on the peroxides reported, structural classification, natural distribution and biological and pharmacological activities are reviewed. Color reagent and spectroscopic identification of peroxide are also described.

A Polyacetylene and Flavonoids from Cirsium rhinoceros

  • Cirsium rhinoceros Nakai (Compositae) is a herbaceous perennial native to Korea, whole plant of which has been used in folklore medicine. C. rhinoceros was extracted by a standard extraction procedure. Its n-hexane, $CHCl_3$ and n-BuOH extracts were fractionated by column chromatography to provide a polyacetylene, a coumarin and five flavonoids. Ciryneol C, scopoletin, acacetin, cirsimarin were isolated for the first time from this plant.
