Fate and Characteristics of Dissolved Organic Matters in a Water Reclamation Facility, Korea (하수처리수 재이용시설의 공정별 용존유기물질 거동 및 특성)

  • This study investigated the fate of dissolved organic matter (DOM) in a water reclamation facility (WRF) in Korea. The WRF consists of coagulation, sedimentation, microfiltration, and reverse osmosis (RO) components. The production capacity of WRF is 90,000 m3/day. The reclaimed water is reused as industrial water. We also characterized DOM in raw, processed, and finished waters based on analysis of dissolved organic carbon (DOC), ultraviolet absorbance at 254 nm (UVA254), fluorescence excitation emission matrix (FEEM), and DOC fractions via liquid chromatography-organic carbon detection (LC-OCD). Based on the results of DOC, UVA254, and FEEM analyses, neither the coagulation/sedimentation nor the microfiltration at the WRF effectively removed DOM. The RO process removed more than 94% of DOM. The raw water (i.e., secondary treated effluent obtained from a wastewater treatment plant) exhibited tryptophan-like peaks, which are a promising marker of wastewater, in the FEEM analysis. Coagulation and microfiltration failed to eliminate the wastewater marker, whereas RO completely removed it. The raw water also carried high levels (89.4%) of hydrophilic and low-molecular weight substances, which are difficult to remove via coagulation-sedimentation or microfiltration. Humic substance was a major component of the hydrophilic fractions. Based on the LC-OCD analysis, RO effectively removed the humic and polymeric materials from DOM.

  • The International Agency for Research on Cancer (IARC, 1989) has classified whole diesel exhaust as probably carcinogenic to humans. Diesel exhaust particulate matter (DPM) adsorbs different chemical substances including PAHs and nitroarenes. DPM is emphasized because it is a major component of diesel exhaust, it is suspected of contributing to a health hazard. Diesel exhaust is a complex mixture of carbon particles and associated organics and inorganics, and it is not known what fraction or combination of fractions cause the health effects [cancer effects, noncancer effects (respiratory tract irritation/inflammation and changes in lung function)] that have been observed with exposure to diesel exhaust. In order to identify which chemical classes are responsible for the majority of the observed biological activities, we performed a particular biological/chemical analysis. Respirable particulate matter (PM2.5: <2.5mm) was collected from diesel engine exhaust using a high-volume sampler equipped with a cascade impactor. Particulate oganic matter was extracted by the dichloromethane/sonication method and the crude extract was fractionated according to EPA recommended procedure into seven fractions by acid-base partitioning and silica gel column chromatography. We examined genotoxic potentials of diesel exhaust particulate matter using novel genotoxicity tests, which are rapid, simple and sensitive methods for assessing DNA-damage at the DNA and chromosomal level (comet assay, in vitro MN test and Ames test). Higher genotoxic potency was observed in non polar fractions and several PAHs were detected by GC-MS, such as 1,2,5,6 dibenzanthracene, chrysene, 1,2-benzanthracene, phenanthrene and fluoranthene.

Antioxidant and Antiallergic Activity of Compounds from the Aerial Parts of Scutellaria baicalensis Georgi (황금 지상부의 항산화 및 항 알러지 활성 성분)

  • Roots of Scutellaria baicalensis have been used for fever remedy; diuresis, antiphlogistic. For the investigation of the active component from aerial parts of Scutellaria baicalensis, MeOH extracts from aerial parts of Scutellaria baicalensis were suspended with $H_2O$, and partitioned by $CHCl_3$. In order to investigate the efficacy of antioxidative activity the activity guided fraction and isolation of physiologically active substance were peformed. Its $H_2O,\;30\%,\;60\%$ MeOH and MeOH fractions were examined on antioxidative activity using DPPH method and TBARS assay; It was revealed that $30\%\;and\;60\%$ MeOH fractions have significant anti-oxidative activity. its fractions testing type I allergy, compound 48/80 induced systemic anaphylaxis was applied. As a result, compared with reference (cromolygate), these fraction significantly inhibited systemic anaphylaxis by $71\%\;and\;57\%$, respectively. From $30\%,\;60\%$ MeOH fraction, five compounds were isolated and elucidated apigenin 6-C-${\alpha}$-L-arabinopyranosyl-8-C-${\beta}$-D-glucopyranoside (isoschaftside, I), scutellarein 7-O-${\beta}$-D-glucuronopyranoside (scutellarin, II), apigenin 7-O- ${\beta}$-D-glucuronopyranoside (III), isoscutellarein 8-O-${\beta}$-D-glucuronopyranoside (IV), kaempferol 3-O-${\beta}$-D-glucopyranoside (V) through their physicochemical data and spectroscopic methods. We measured radical scavenging activity with DPPH method and anti-lipid peroxidative efficacy on human LDL with TBARS assay. [$I] showed antioxidant activities in order. Type I allergy compound 48/80 induced systemic anaphylaxis was applied. $[V inhibited systemic anaphylaxis in order.

Effect of Flavonoid Fractions Extracted from Rhus verniciflua STOKES on the Reproductive Parameters in SD Male Rats (옻나무(Rhus Verniciflua STOKES) Flavonoid 분획 투여가 정자생성 및 생식관련 장기에 미치는 영향)

  • Rhus verniciflua Stokes (RVS) has been used as a food supplement and a traditional herbal medicine. In this study, we prepared various flavonoid fractions (RS, RW1, RW2 and RWE) from a hot water extract of RVS and their influence on male reproductive organs and spermatogenesis were studied in rats which were orally administered 200 mg/kg of them for 8 weeks. All experimental groups did not show any significant changes in body weight and blood clinical chemistry for liver function. Plasma testosterone level was elevated about 3.7, 5.2 and 6.3 folds in RW1, RW2 and RWE groups, respectively. The weights of testes and epididymides tended to increase slightly without the statistical significance in RW2 and RWE. The spermatozoon motility and epididymal sperm concentration were significantly increased (P<0.05) in RWE and RW1, respectively, when compared to the control group. There was no significant difference in histology and apparent shape of testes and epididymides among the control and the experimental groups. Collectively, RWE showed effectively the elevation of plasma testosterone level, spermatozoon motility and the epididymal sperm concentration without the significant increase of testis and epidiymides weights. When the component HPLC profile among the flavonoids fractions of RVS was compared, the ratio of components were only different. These findings suggest that the Rhus flavonoid fraction, particularly RWE, can stimulate the androgen-dependent male sexual function and it can be applied to the material of functional food for enhancing the sexual function.

  • The influence of addition of arrow(Pueraria hirsuta Matsum) root flour and its fractions by ultrafiltration on the flow properties of the rice flour-water systems were investigated. The flow properties of rice flour pastes during cold storage$(5^{\circ}C)$ were measured and the components from arrow root flour responsible for changes of flow properties were screened. Addition of arrow root flour significantly changed the flow properties of rice flour pastes. Permeate fractions by ultrafiltration, representing low molecular weight component fraction, significantly affected the flow property of rice flour paste stored at $5^{\circ}C$. Addition of permeate fraction to rice flour pastes rapidly decreased the consistency index, yield stress and pseudoplasticity, and showed a stability of flow property during storage suggesting the retardation of rice starch retrogradation. Permeate fractions of ultrafiltration were identified as puerarin, daidzein and daidzin known to representive isoflavonoid from arrow root.

  • The antimutagenic activity of a methanol extract of Ecklonia stolonifera (Laminariaceae) against aflatoxin $B_1\;(AFB_1)$ was demonstrated with the Salmonella typhimurium assay. The numbers of revertants per plate decreased significantly when this extract was added to the assay system using S. Salmonella typhimurium TA100. The methanol extract also exhibited significant inhibitory effects on the micronuclei formation in mouse peripheral blood reticulocytes and the DNA damage in mouse spleen lymphocytes induced by N-methyl-N-nitrosourea (MMU) and benzo(a)pyrene (B(a)P). The MeOH extract was then sequentially partitioned with $CH_2Cl_2,\;CH_2Cl_2$ insoluble intermediate, EtOAc, n-BuOH, and $H_2O$. All fractions possessed antimutagenic activity but the $H_2O$ fraction was inactive. Among active fractions, the EtOAc and $CH_2Cl_2$ insoluble intermediate fractions showed the highest activity. Column chromatography using $SiO_2$ and Sephadex LH-20 yielded phloroglucinol from the EtOAc fraction. Phloroglucinol also demonstrated significant antimutagenic activity, and inhibitory effects on the micronuclei formation in mouse peripheral blood reticulocytes and DNA damage in mouse spleen lymphocytes induced by MMU and B(a)P.

The Nitrite-scavenging Effects by the Component of Traditional Tea Materials (기호음료 성분의 아질산염 소거작용)

  • The present study was conducted to investigate nitrite-scavenging ability of the components of traditional tea materials in Korea. The nitrite-scavenging ability was appeared in all the samples examined. Ethanol soluble fractions obtained from water soluble fractions of Cassiae torae semen, Perilla semen, Acantopanacis cortex and Zingiberis rhizoma exhibited higher the nitrite-scavenging effect than ethanol precipitate frecipitate fractions. While opposite results were obtained from those of Zizyphi fructus, Chaenomelis fructus and Schizandrae fructus in the nitrite-scavenging effect.

Identifying antibacterial activity components of cosmos flower extracts (코스모스 추출물의 항균활성 성분 탐색)

  • This study investigated whether the extracts from cosmos flowers exhibit antibacterial activities and identified which components were ascribed to the antibacterial effects. The antibacterial effects of extracts from white, pink, and violet cosmos flowers were observed for 24 h after inoculation with four kinds of bacteria, including Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Escherichia coli. Among the three fractions of cosmos flower extracts, the best antibacterial activity against the four bacteria was observed in the extracts isolated from the EtOAc layer. However, the extracts from the CHCl3 layer were also effective against S. aureus. Moreover, the first of white, second of pink, and first of violet silica gel fractions (Fr.) isolated from the EtOAc layer exhibited minimal inhibition at a concentration of 0.1 mg/mL. Comparison of NMR and High-Pressure Liquid Chromatography results between silica gel Fr. and apigenin suggested that the effective fractions can contain a component including apigenin moiety.

Antioxidant Activity of Solvent Extract from Onion Skin (양파껍질에서 분리된 용매 추출물의 항산화효과)

  • The antioxidant activity of the solvent fractions extracted from onion skin was examined. The antioxidant activity of methanol extract at the cencentration of 0.02% and 0.03% was stronger than that of mixed tocopherol. The antioxidant activity of the fractions of methanol extract increased in the order of butanol> ethyl acetate> ethyl ether> water fraction. The antioxidant activity of each fractions was strongly related with total phenol content and HDA. Further separation of butanol fraction by TLC yielded 6 fluorescent bands with Rf values of 0.20, 0.33, 0.49, 0.60 and 0.94. The total phenol content and HDA of fluorescent band, Rf 0.96, were remarkable higher than those of the other band and exhibited a strong UV absorption at 255 nm and 317 nm, which would be specifically produced by flavonol. Spectral analyses indicated that the major antioxidant component was quercetin aglycone (3,3',4',5,7-pentahydroxyflavone).

The Nitrite-Scavenging Effects by Component of Green Tea Extracts (녹차 추출물의 아질산염 분해작용)

  • This paper was investigated to elucidate the nitrite-scavenging action as one of biological actions of green tea extracts. Green tea extracts exhibited remarkable nitrite-scavenging action. Among the green tea extracts, steamed green tea showed higher nitrite-scavenging action than roasted green tea. Nitrite-savenging actions of gren tea extracts were hardly influenced until three times extraction with hot water. Furthermore , the nitirte-scavenging action of green tea extracts showed ph dependent , highest at pH 1.2 and lowest at pH 6.0. The nitrite -scavenging rate in sample extracts /amine /nitrite systems exhibited faster than that in amine/nitrite systems. In order to screen the nitirte-scavenging factors, samples extracts were fractionated with methanol. in both samples, methanol-soluble fractions possessed greater nitrite-scavenging action than methanol-insoluble fractions .The nitrite-scavenging action of methanol-soluble fractions increased with proportion to absorbance at 420nm and at 280nm, while that of green tea extracts increased with the contents of total phenols and absorbance at 280nm.
