Lactic Acid Bacterial Fermentation Increases the Antiallergic Effects of Ixeris dentata

  • Ixeris dentata (ID, family Asteraceae), called Seumbakuy in Korea, was fermented with lactic acid bacteria (LAB) and their antiallergic activities were investigated. Fermentation of ID with Bifidobacterium breve or Lactobacillus acidophilus increased its inhibition of degranulation in RBL-2H3 cells induced by the IgE-antigen complex. Oral administration of these extracts to mice inhibited the passive cutaneous anaphylaxis (PCA) reaction induced by the IgE-antigen complex and scratching behaviors induced by compound 48/80. The fermented ID more potently inhibited the PCA reaction and scratching behaviors than the non-fermented one. These extracts also inhibited mRNA expression of TNF-${\alpha}$ and IL-4, as well as NF-${\kappa}B$ activation in RBL-2H3 cells induced by the IgE-antigen complex. These findings suggest that LAB fermentation improves ID-mediated inhibition of IgE-induced allergic diseases such as rhinitis and asthma, and that ID works by inhibiting degranulation and NF-${\kappa}B$ activation in mast cells and basophils.

Exploring a zero food waste system for sustainable residential buildings in urban areas

  • This study explores the environmentally innovative and low-impact technology, a zero food waste system (ZFWS) that utilizes food waste and converts it into composts or biofuels and curtails carbon emissions. The ZFWS not just achieves food waste reductions but recycles food waste into fertilizer. Based on a fermentation-extinction technique using bio wood chips, the ZFWS was employed in a field experiment of the system installed in a large-scale apartment complex, and the performance of the system was examined. The on-site ZFWS consisted of three primary parts: 1) a food waste slot into which food waste was injected; 2) a fermentation-extinction reactor where food waste was mixed with bio wood chips made up of complex enzyme and aseptic wood chips; and 3) deodorization equipment in which an ultraviolet and ozone photolysis method was employed. The field experiment showed that food waste injected into the ZFWS was reduced by 94%. Overall microbial activity of the food waste in the fermentation-extinction reactor was measured using adenosine tri-phosphate (ATP), and the degradation rate of organic compounds, referred to as volatile solids, increased with ATP concentration. The by-products generated from ZFWS comply with the national standard for organic fertilizer.

A Fermentation Strategy for Anti-MUC1 C595 Diabody Expression in Recombinant Escherichia Coli

  • The development of fermentation conditions for the production of C595 diabody fragment (dbFv) in E. coli HB2151 clone has been explored. Investigations were carried out to study the effect of carbon supplements over the expression period, the comparison of C595 dbfv production in synthetic and complex media, the influence of acetic acid upon antibody production, and comparison of one-stage and two-stage processes operated at batch or fed-batch modes in bioreactor. Yeast extract supplied during expression yielded more antibody fragment than any other carbon supply. The synthetic medium presented higher specific productivity (0.066 mg dbFv $g^{-1}$ dry cell weight) when compared to the complex medium (0.044 mg dbFv $g^{-1}$ DCW). The comparison of fermentation strategies demonstrated that (1) one-stage fed-batch fermentation performed higher C595 dbFv production than that operated in batch mode which was significantly affected by acetate concentration; (2) a two-stage batch operation could enhance C595 dbFv production. It was found that a concentration of 12.3 mg $L^{-1}$ broth of C595 dbFv and a cell concentration of 10.8g $L^{-1}$ broth were achieved at the end of two-stage operation in 5-L fermentation.

Electron Flow Shift in Clostridium acetobutylicum Fermentation by Lactate

  • Clostridium acetobutylicum produced more butanol in the medium containing corn steep liquor (CSL) than in a complex medium without CSL Addition of CSL to CAB medium increased sugar consumption by the bacterium. Similar results were obtained in the fermentation using CAB medium containing lactate. The ratio for the butanol produced to acetone of the control culture was 1.8, whilst that of the culture containing 44 mM lactate was 5.2. From these results it is hypothesized that lactate functions as an electron flow modulator in the fermentation. This finding has been utilized for the successful butanol fermentation of a non-corn based agricultural byproduct, palm oil waste.

Production of Yeast Diet for Aquaculture in Batch Fermenters

  • Production of yeast diet has been carried out in batch fermenters under optimum culture conditions. The fermentation of Candida utilis on a $2\%$ complex medium resulted in 1.22 g/L/h productivity and $65\times10^8$ viable cells/ml, and the addition of $15{\mu}M$ zinc to the medium increased both the productivity and the number of viable cells just a little more. In the case of the fermentation of Kluyveromyces fragilis, the highest value of the biomass productivity, 1.94 g/L/h, was obtained on a $2.5\%$ fructose medium with $70\times10^8$ viable cells/ml, and $1\%$ peptone was found to be a growth factor in this fermentation. When $3.5\%$ NaCl was added to the given medium, both the biomass productivity and the number of viable cells decreased significantly in each fermentation, but this may be considered to preserve yeast diet long without osmotic lysis.

연속 발효조를 이용한 soluble glucan 생산 공정 개발 (Development of Soluble Glucan Production Process with Continuous Stirred Tank Fermentor)

  • Continuous fermentation process for the production of soluble glucan using mutant Agrobacterium sp. ATCC31750 has been developed in this research. When the concentration of soluble glucan was higher than 6 g/l, the viscosity of the fermented broth was too high and it needs complex separation process to separate from culture broth. Mathematical models which describe the cell growth and glucan production was developed and they kinetic parameters were estimated with experimental data. They are used for the optimization of continuous fermentation process and calculate optimal dilution rate for easy separation of glucan 4 g/l. With continuous fermentation, glucan production rate was increased 1.8 times more than that with batch fermentation.

  • A highly productive fed-batch fermentation process was developed for the production of L-ornithine by using a new stabilized strain, Breviabcterium ketoglutamicum BK52. Fed-batch cultures with a continuous feeding of the complex medium were conducted on various operating conditions. The optimal concentration of phosphate in the complex medium was 2.1g/L. The optimal feeding rate of L-arginine was 0.028g/L/hr. The optimal feeding point of the complex medium was determined to be at 40 OD of the cell mass. The final L-ornithine concentrations within 64hrs of cultivation in 5 and 50 liter fermenters were 73g/L and 71g/L, respectively. The maximum overall L-ornithine productivity was 1.14g/L/hr which was about 2 times higher than that of the conventional fed-batch culture with intermittent feeding. The overall productivity of the fermentation system is remarkably improved by employing the optimized conditions, and it offers a significant potential for industrial application.

김치 독자성의 근거와 형성 과정에 대한 고찰 (A Study on the Basis and Formation Process of Kimchi's Uniqueness)

  • The Chinese Sigyeong records the foods of the Primitive Pickling Period, pickling being a universal vegetable storage method, but does not indicate the origin of the pickled vegetables or the location of the source of transmission. Kimchi mainly used salt and sauce-based soaking materials at the beginning of the Fermented Pickling Period (beginning in the 1st to 3rd centuries A.D.), and it differed from the Chinese method, which used alcohol and vinegar. In the Umami-Flavored Pickling Period (beginning in the 14th and 15th centuries A.D.), jeotgal, fermented seafoods, were added, and pickles with a completely new identity were created, one different from any other pickles in the world. Lastly, entering the Complex Fermentation and Pickling Period (beginning in the 17th and 18th centuries), the technical process evolved using a separate special seasoning containing red pepper as the secondary immersion source after pickling in brine, the primary immersion source. As a result of this, kimchi was transformed into a food with a unique form and taste not found anywhere else. The unique characteristic of kimchi is that the composition of original materials, a combination of salted marine life and vegetable ingredients, is its core identity, and there is a methodological difference in that it is completed through a second process called saesaengchae (生菜)-chimchae (沈菜).

홍삼함유 과채류 복합 추출물의 증숙열처리 및 유산균 발효에 의한 장관면역 활성 (Evaluation of Intestinal Immunity Activity by Steam-Heat Treatment and Fermentation of Lactic Acid Bacteria of Fruit and Vegetable Complex Extracts containing Red Ginseng)

  • 본 연구의 목적은 과채류 복합추출물에 소량의 홍삼을 첨가하여 증숙가열 및 유산균 발효에 의한 진세노사이드 대사산물의 활성 증가와 장내면역 활성이 현저히 개선되는지를 알아보고자 하였다. 홍삼 단독 추출물 또는 과채류복합추출물에 비해 홍삼을 함유한 과채류 복합추출물에서 시너지 효과에 의한 장내면역 활성의 증가가 관찰됨을 확인하였다. 다음으로 홍삼을 함량별로 첨가하여 유산균 발효에 따른 시너지 효과를 얻을 수 있는 홍삼의 농도를 결정하였다. 그 결과, 홍삼을 과채류 혼합물 질량에 대하여 3~10%의 질량비로 혼합하여 증숙 및 유산균 발효 추출하였을 때 시너지 효과가 나타나는 것을 확인하였다. 또한 홍삼을 함유한 과채류 복합추출물은 200㎍/ml의 농도로 처리하였을 때 홍삼이 함유되지 않은 과채류 복합추출물에 비해 NO 생성을 약 60% 억제하였으며, IL-1β의 발현은 63%, IL-6의 발현 69%, TNF-α의 발현 76% 수준으로 억제하여 장내면역 활성이 크게 향상되었음을 확인하였다.

A Strategy of modeling for fermentation process by using genetic-fuzzy system

  • An algorithm for modeling of yeast fermentation process using genetic-fuzzy algorithm is presented in this work. The algorithm involves developing the fuzzy modeling of the process and model update capability against the system change. The membership functions of state variables and specific rates and the decision table were generated using genetic algorithm. This algorithm could replace the complex mathematical model to simple fuzzy model and cope with the change of process characteristics well.
