Prevalence and Characterization of Plasmid-Mediated Quinolone Resistance Determinants qnr and aac(6')-Ib-cr in Ciprofloxacin-Resistant Escherichia coli Isolates from Commercial Layer in Korea

  • The prevalence and characterization of plasmid-mediated quinolone resistance (PMQR) determinants in ciprofloxacin-resistant Escherichia coli isolated from a Korean commercial layer farm were studied. A total of 45 ciprofloxacin-resistant E. coli isolates were recovered and all isolates were multidrug-resistant. Eight isolates have the PMQR genes aac(6')-Ib-cr, qnrS1, and qnrB4, and seven isolates exhibited double amino acid exchange at both gyrA and parC, and have high fluoroquinolone minimum inhibitory concentrations. Five transconjugants demonstrated transferability of PMQR and β-lactamase genes and similar antimicrobial resistance. Because PMQR genes in isolates from commercial layer chickens could enter the food supply and directly affect humans, control of ciprofloxacin resistance is needed.

A Study on the Mudfish Raising using of Raw Hens Excreta (생(生)계분을 이용한 미꾸리 양식에 관한 연구)

  • for 50% of Rash water every week during experimental period. Eight of 100L of aquarium, eight kg of mudfish and four of female korean native chicken were used this study. This study are divided according to the mudfish fed on commercial mudfish diet in the four of control group and fed on raw hens excreta in the four treatment group. The chemical composition was compared with commercial mudfish diet and hens excreta. In the both sample, crude protein contents was almost same. The crude fat and crude ash were higher in hens excreta than commercial mudfish diet. The growth performance of mudfish tend to high when fed hens excreta. There were no differences in contents of E. Coli and Salmonella and pH of water in aquariums between the groups. Mortality of mudfish tend to decrease when fed hens excreta. No problem with mudfish health was observed during the experimental period of 35 days. These results indicated that it is possible to raising mudfish fed on hens excreta only.

  • This study was conducted to investigate the high-value added new products from spent hens. Spent hens were randomly designed to one of the four dietary treatments: 1) Control (commercial feed),2) T1 (commercial feed supplemented with 100 lU ${\alpha}$-tocopherol, 3) T2 (commercial feed with 10% squid liver oil and 4) T3 (commercial feed with 100 IU ${\alpha}$-tocopherol plus 10 % squid liver oil). They were fed one of the experimental diets for 15 days and slaughtered. And, also, somking of spent hens as follows : digestion in sources, boiling and somking.

Effect of Dietary Mugwort on the Physico-Chemical Properties of Chicken Meat (쑥의 급여가 계육의 이화학적 특성에 미치는 영향)

  • This study was conducted to investigate the influence of dietary mugwort on physico-chemical characteristics and processing characteristics in meat sample of chicken meat. Broilers were randomly assigned to one of four dietary treatments: 1) Control(commercial feed) 2) T1(commercial feed supplemented with 1% mugwort) 3) T2(commercial feed with 3% mugwort) and 4)T3(commercial feed with 5% mugwort). They were fed one of the experimental diets for five weeks and slaughtered. After that, the meat samples were vacuum packaged and stored at 4${\pm}$1$^{\circ}C$. The physico-chemical characteristics and processing characteristics were analyzed for meat samples stored over a period of 0, 1, 2, 3 and 4 weeks. The pH of all treatments significantly increased during the storage periods(U<0.05). The pH of the thigh was rather higher than that of the breast. The drip loss and heating loss tend to decrease in dietary mugwort group(p<0.05). The WHC(water holding capacity) of all treatment was significantly increased during storage(p<0.05). The meat color was increased during storage.

However, the VBN was not significantly different between control and treatment groups. The meat color (b*) significantly increased during storage periods.

  • Indigenous (native) breeds of livestock have higher disease resistance and adaptation to the environment due to high genetic diversity. Even though their extinction rate is accelerated due to the increase of commercial breeds, natural disaster, and civil war, there is a lack of well-established databases for the native breeds. Thus, we constructed the native pig and chicken breed database (NPCDB) which integrates available information on the breeds from around the world. It is a nonprofit public database aimed to provide information on the genetic resources of indigenous pig and chicken breeds for their conservation. The NPCDB (http://npcdb.snu.ac.kr/) provides the phenotypic information and population size of each breed as well as its specific habitat. In addition, it provides information on the distribution of genetic resources across the country. The database will contribute to understanding of the breed's characteristics such as disease resistance and adaptation to environmental changes as well as the conservation of indigenous genetic resources.

Quality Characteristics of Korean Native Chicken Meat (한국산 토종 닭고기의 품질 특성)

  • A study was conducted to compare the quality characteristics among commercial broiler, Wangchoo (imported dual purpose breed) and Korean native chicken(KNC). Thigh and breast meat of the broiler(7-wk old), Wangchoo(15-wk old), and Korean native chicken(15-wk old) stored for 24 h at 5t were used to analyze chemical composition, physico-chemical characteristics, textural traits and sensory evaluation test. Crude fat and moisture contents in broiler meat and crude protein content in KNC were significantly(P<.05) higher than those in the other breeds regardless of parts of the body. Total collagen content in broiler meat was significantly higher than those of the other breeds, however, the heat-soluble and the acid-soluble collagen content in Wangchoo were significantly lower than those of the other breeds. Water-holding capacities of KNC in breast meat, and of broiler in leg meat were significantly higher than that of the other breeds, while the results of the water-holding capacity and the cooking loss were reversed. Myofibrillar fragmentation index in broiler meat was significantly higher than that in the other breeds regardless of body parts. Hardness, elasticity and cohesiveness in Wangchoo were significantly higher than those in the other breeds. The prominent fatty acids were oleic, palmitic and linoleic acids and run up to 79.03~83.82 %, regardless of breeds and parts. The sensory evaluation score of tenderness, taste and preference in Wangchoo were lower compared to the broiler and KNC, however, they were not significantly different between broiler and KNC. In conclusion, the quality characteristics of KNC were excellent compared to Wangchoo.

Polymorphism of Ghrelin Gene in Twelve Chinese Indigenous Chicken Breeds and Its Relationship with Chicken Growth Traits

  • A 2,656 bp fragment of chicken ghrelin gene was cloned and SNPs were detected by PCR-RFLP and Allele Specific PCR (ASP) in 12 Chinese indigenous chicken breeds and a commercial chicken population. The results showed that there were 23 base variations and an amino acid change ($Gln{\rightarrow}Arg$) in cloned chicken ghrelin gene. Three SNPs were confirmed in 13 populations and associations between this gene and growth traits of Tibetan chicken (TC) and Recessive White chicken (RW) were investigated. The results of haplotype analysis revealed that 26 haplotype genotypes were composed of eight haplotypes. The results of $x^2$ tests indicated that there were significant differences between genotypes or haplotype genotype frequencies in some of the breeds or sexes at 0.05 or 0.01 levels. The results of ANOVA revealed that there were significant differences between genotypes or haplotype genotypes on some growth traits of TC and RW chicken breeds at 0.05 or 0.01 levels. Multiple comparisons showed that there were significant associations between genotype CT at site 71 and some growth traits of two chicken breeds and between genotype AG at site 1,215 and body weight at 16 wk of two chicken breeds, and there was a significant association between haplotype genotype CAA/CAG and body weight and shank girth at 16 wk of two chicken breeds.

Study of six different commercial Koran-native chicken crossbreeds from hatch to twelve weeks of age

  • A study was conducted to compare growth performance of six female commercial Korean native chicken (KNC) crossbreeds from hatching to twelve weeks of age. Three hundred and twelve, 1-day-old female commercial KNC were used within 1 paternal line and 6 maternal lines. The chickens were allocated to 24 battery cages to give 4 replicates per strain with 13 chickens per cage. The chickens were reared under continuous lighting (24 h) and water was available at all times. Ad-libitum feeding was practiced throughout the experimental period. Among the six different strains, 2A had the greatest bodyweight (BW) at 42 days after hatching (p < 0.05). No BW difference between six crossbreed strains (p > 0.05) was found thereafter. Crossbreed 1A had the higher average daily gain (ADG) than crossbreed 2A and 3A chickens (p < 0.05), whereas crossbreed 4A, 5A, and 6A had similar ADGs to that of crossbreed 1A (p > 0.05) at 84 days after hatching. Furthermore, crossbreed 4A had a great average daily feed intake (ADFI) from hatching to 84 days (p < 0.05). Nonetheless, there was no difference in the feed conversion ratio (FCR) and uniformity between six crossbreed strains for the experimental period (p > 0.05). Despite that 1A, 4A, and 6A had the higher viability (p < 0.05) than crossbreed 2A and 5A, they had a similar viability than crossbreed 3A (p > 0.05). With this in mind, crossbreed 2A had greater BW, ADG, and FCR than other chicken crossbreeds from hatching to 84 days, although they had a lower viability than others.
