Preparation of Amorphous Adefovir dipivoxil using Reverse Phase Column Chromatography and Solid Dispersion Method (역상컬럼 및 고체분산체를 이용한 무정형 아데포비어디피복실의 제조)

  • Adefovir dipivoxil which was originally developed by Gilead Sciences has been used as treatments of HIV and HBV, especially a therapeutics for HBeAg positive and negative chronic patients. We developed highly efficient purification method using reverse phase column chromatography for mass production and a stable amorphous Adefovir dipivoxil using solid dispersion method. Reverse phase column chromatography led to highly pure product, more than 99.7% by HPLC and can be used for mass production compared with normal column chromatography. Solid dispersion method containing watersoluble polymer and Isomalt showed improved stability of amorphous Adefovir dipivoxil against heat and moisture.

The Study of Improvement Effect of Ground Settlements and Bearing Capacity by Stone Columns (스톤컬럼 공법의 지내력 증진 효과에 대한 연구)

  • Aseries of in-situ tests and ground water level measurements with stone and slug materials had been conducted to find out effect of ground settlement reduction and bearing capacity improvement by the Stone Column method. As the result of the tests, it was proved that the Stone Column method is effective for reduction of ground settlement and improvement of bearing capacity. In addition the ground water level went down without overburden load. These results show that the Stone Column method is effective for an increase in density and resistance to liquefaction. The results of estimation of ground settlement and bearing capacity by general theoretical equation, it show that the Stone Column method increases bearing capacity by 2.7~5.7 times and decreases ground settlement by 2~3.5 times.

Minimum stiffness of bracing for multi-column framed structures

  • A method that determines the minimum stiffness of baracing to achieve non-sway buckling conditions at a given story level of a multi-column elastic frame is proposed. Condensed equations that evaluate the required minimum stiffness of the lateral and torsional bracing are derived using the classical stability functions. The proposed method is applicable to elastic framed structures with rigid, semirigid, and simple connections. It is shown that the minimum stiffness of the bracing required by a multi-column system depends on: 1) the plan layout of the columns; 2) the variation in height and cross sectional properties among the columns; 3) the applied axial load pattern on the columns; 4) the lack of symmetry in the loading pattern, column layout, column sizes and heights that cause torsion-sway and its effects on the flexural bucking capacity; and 5) the flexural and torsional end restrains of the columns. The proposed method is limited to elastic framed structures with columns of doubly symmetrical cross section with their principal axes parallel to the global axes. However, it can be applied to inelastic structures when the nonlinear behavior is concentrated at the end connections. The effects of axial deformations in beams and columns are neglected. Three examples are presented in detail to show the effectiveness of the proposed method.

  • The virbration characteristics of a passenger car steering column are studied by using a modal test and a finite element (FE) analysis. To verify the FE model and the results, an experiment using the impact exciting method is performed. Two types of the steering column in this study are considered as follows; (ⅰ) the non-tilt type steering column and (ⅱ) the upper-tilt type steering column. The experimental results are compared with those o the FE analysis, and it ti shown that the results agree with each other. The effects of various design parameters such as the bracket thickness, the column diameter on the natural frequencies are also investigated by FE analysis.

Theoretical study of UHPCC composite column behaviors under axial compression

  • To improve the durability and service life of reinforced concrete column such as bridge piers, an advanced composite column made of Ultra High Performance Cementitious Composites (UHPCC) permanent form is proposed. Based on elasticity plasticity theory, axial compression behavior of the composite column was studied theoretically. The first circumferential cracking load and ultimate limit loading capacity are derived for the composite column. Short composite column compression tests and numerical simulations using FEM method were carried out to justify the theoretical formula. The effects of UHPCC tube thickness on the axial compression behavior were studied. Using the established theoretical model and numerical simulation, the large dimension composite columns are calculated and analyzed with different UHPCC tube thickness. These studies may provide a reference for advanced composite column design and application.

  • The coner high column method that is used in the multi-roof building Sungryemoon seems to have been used for the first time during the Choseon Dynasty, and it's characteristics and trend of usage are studied in this paper. In the results, it was confirmed that the coner high column method is economic and structurally safe, and selectively combines the structural factors used in the Chinese seats. It was found that this method was a newly adopted construction method to effectively express the authority of the country with the establishment of a new dynasty. Also the meaning of the method could be guessed since it was continuously used in important structures from the Choseon Dynasty to the Daehan Empire, in spite of the new multi-roof building technologies.

  • In this study, experimental research was carried out to investigate the seismic and structural performance of precast concrete exterior beam-column joints using bolt type connection and prestressing method. A total of five full-scale exterior beam-column joints were constructed and tested under reversed cyclic loading, controlled by displacement. Results of the test are as follows: Energy dissipation capacity and pinching phenomenon of PC beam-column joints showed disadvantageous behavior compared to RC beam-column joints. However, drift capacity of the PC joint was excellent. Also, yield mechanism concentrated on embedded nuts was suitable as an exterior beam-column joint of lateral load resistance frame. Additional application of prestressing method was also very effective to control excessive pinching and cracking in the joint region, and thus improved an overall seismic performance of the PC joint.

  • Recently, a great progress has been made in bridge construction technology through the development of high performance materials and new structural types. However, most of attention has been paid to the cast-in-place technologies and material cost saving. The cast-in-place method is always subject to some environmental damages in construction sites, which frequently causes conflicts with residents. To overcome the disadvantages, a lot of fabrication construction method was developed. Most fabrication construction methods developed up to now have been applied for superstructure of bridges. In contrast, such fabricable methods developed for substructures are extremely rare. A fabricated column using ICH CFT(Internally Confined Hollow CFT) column was developed in a series of previous researches. Included in the previous studies are design and construction methods for the precast segmental coping, the column-coping connection, the column-segment connection, column-foundation connection. In this paper, seismic performance of the fabricated ICH CFT columns was extensively investigated experimentally. Two test specimens were prepared depending on the connection methods of segments; one by mortar-grouting method and the other by reinforcement method using stiffeners.

  • Stone column is one of the ground improvement systems which is being used for accelerating consolidation and increasing bearing capacity for settlement sensitive structures like load embankments, bridge abutments, oil storage tanks etc. The effects of this method are enhancement of ground bearing capacity, reduction of settlement, prevention of liquefaction and prevention of lateral ground movement. Recently, geosynthetic reinforced (encased) stone column approach has been developed to improve its load carrying capacity through increasing confinement effect. Although such a concept has successfully been applied in practice, fundamentals of the method have not been fully explored. This paper presents the results of an investigation on the bearing capacity and failure mechanism of geogrid-encased stone column by model tests. The results of the analyses indicated improved bearing capacity of the geogrid reinforced stone column method over the conventional strone column method with no encasing.

Size Verification of Small and Large Bubbles in a Bubble Column (기포탑에서 작은기포와 큰기포의 크기 구별)

  • Size verification of small and large bubbles in a bubble column was investigated by employing the dynamic gas disengagement (DGD) method and dual electrical resistivity probe (DRP) method, simultancously. The holdups of large and small bubbles in the bubble column in a given operating condition were obtained by means of the DGD method by measuring the pressure drop variation in the column with a variation of time after stopping the gas input into the column. The size and frequency of bubbles were measured by the DRP method in the same operating condition, from which the bubble holdup of each range of size was obtained. The verification of size in determining the large or small bubbles was decided by comparing the holdups of large or small bubbles measured by the DGD method with that measured by the DRP method. Filtered compressed air and tap water were used as a gas and a continuous liquid medium. The diameter and height of the bubble column were 0.102 m and 1.5 m, respectively. The demarcation size between the large and the small bubbles in the bubble column was 4.0~5.0 mm; the demarcation size was about 5.0 mm when the gas velocity was in the relatively low range, but about 4.0 mm when the gas velocity was in the relatively high range, within this experimental conditions.
