The Effect of Breast Feeding on Growth Development, Bone Mineral Density of Carpus, and Nutrient Intakes in Preschool Children (모유 경험이 미취학 아동의 신체발달, 손목 골밀도 및 영양섭취에 미치는 영향)

  • The purpose of this study was to examine the long term effects of breast feeding on growth, bone development and nutrient intakes in preschool children. Subjects were 62 preschool children. Anthropometric characteristics and bone mineral density of carpus were measured using DEXA. The questionnaire was composed of health status, life style, dietary behaviors, and dietary intakes and was completed by the children's mothers. The average age of the boys was 62.4 months and that of the girls was 62.1 months. Average birth height and weight of the subjects was 50.9 cm and 3.4 kg for boys and 50.3 cm and 3.3 kg for girls, respectively. The average height, weight, $\%$ body fat, and obesity index were 111.7 cm, 19.6 kg, $15.0\%$, $-2.1\%$ in boys and 109.4 em, 18.7 kg, $17.5\%$, $0.2\%$ in girls, respectively. Forty children were fed colostrum, 21 were fed breast milk, 29 were fed formula, and 12 were fed mixed milk. There was no significant difference in growth status between children who were fed colostrum and those who were not. Children who had mixed feedings were significantly taller than those who did not (p <0.001), and children who had colostrum had significantly higher bone mineral density than those who did not ($0.25 \pm 0.04 g/ cm^2$ vs. $0.23 \pm 0.04 g/cm^2$, p < 0.05). However, the different feeding methods did not show any difference in bone mineral density. Except vitamin $B_6$, folic acid and vitamin E, vitamin, mineral and calorie intakes did not meet the Korean RDAs. Calcium intake was especially lower than recommendations by as much as $62\%$ and $70\%$ in boys and girls, respectively. There was no significant difference in nutrient intakes between children who had colostrum and those who did not. However, children who were formula consumed significantly more animal fat than those who did not (p < 0.05). Neonatal feeding and breast or formula feeding seems to associate with height, bone mineral density and animal fat intakes in preschool children, based on the results of this study. A longitudinal study is needed to clarify this relationship.

Variations of Immunoglobulins in Colostrum and Immune Milk as Affected by Antigen Releasing Devices

  • This work was conducted to examine the variation of immunoglobulins (Igs) in serum, immune milk, normal milk and colostrum upon implantation of a new Antigen Releasing Device (ARD). The core of each ARD housed an immunostimulating complex (ISCOM) that was made of adjuvant Quil A and type XIII lipase from a Pseudomonas sp. Each ARD was coated with polylactic acid, known as polylactide, that controls antigen release. Twenty lactating Chinese Holstein cows were divided into 2 groups (n = 10): test group and control group. All cows in the test group were implanted with a single injection in the right iliac lymph node with 3 types of ARDs, which were designed to release the antigens at d 0, 14 and 28 post-implantation. Blood and milk samples were collected from both groups, and colostrum samples were also collected from other post-partum cows in the same farm. Concentrations of $IgG_1$, IgA and IgM in whey and serum were measured by sandwich ELISA. The results showed that the $IgG_1$, IgA and IgM concentrations in serum and whey from the test group were higher than from the control group. Among the three Igs measured, the $IgG_1$ concentration in serum was significantly higher at d 40 after ARD implantation, and the $IgG_1$ concentration in whey peaked at d 9, 17 and 30, which corresponded with release of the antigen. Based on Pearson's correlation between Ig concentration and production parameters, IgA concentration in normal milk was positively correlated with lactation period, which reflected IgA changes during the lactation period in immune milk. In colostrum, $IgG_1$, IgA and IgM decreased abruptly from d 0 to 3, and then decreased slightly. In conclusion, serum $IgG_1$ concentration can be affected by controlled release of the ARD, while whey IgA levels are primarily affected by lactation period. These results may be useful in future studies designed to regulate concentrations of Igs in immune milk.

Effects of Dietary Ginger and Scutellaria Dry Powder on Antioxidant Capacity and IgG Concentration in Colostrum and Plasma of Sows (모돈에 대한 생강 및 황금 분말 급여가 초유와 혈청의 항산화력과 IgG 농도에 미치는 영향)

  • The objective of this study was to investigate the effects of dietary ginger and Scutellaria on antioxidant capacity and IgG concentration in plasma and colostrum of sows. Twenty-four sows (Landrace $\times$ Yorkshire) in parity 2 were assigned to 3 treatment groups (8 sows per treatment) from 30 day before farrowing up to weaning on day 21 after farrowing. The treatment diets included 1) CON (Basal diet), 2) GDP (0.3% ginger dry powder was used as the replacement of wheat in basal diet) and 3) SDP (0.3% Scutellaria dry powder was used as the replacement of wheat in basal diet). No differences were found in antioxidant capacity and IgG concentration in plasma of sows among all the three treatments. Antioxidant capacity in colostrum was not affected by replacement of ginger and Scutellaria dry powder. IgG and protein concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. However, fat and lactose concentration were not affected by treatments. The total amino acid concentration in colostrum was significantly higher (P<0.05) in GDP than that in CON and SDP. Plasma antioxidant capacity was not affected by treatments in piglet on 0 day and 21 day, but tended to increase by supplementation of ginger. On 0 day, plasma IgG in piglet was not detected by replacement of ginger and Scutellaria dry powder. However, plasma IgG concentration in piglets on 21 day was significantly higher (P<0.05) in GDP than that in CON and SDP. These results indicate that colostrum IgG concentration in sows and plasma IgG concentration in piglets were improved by feeding ginger dry powder 30 days before farrowing in comparison to basal diet.

A Study n Infant Feeding Practices in Seogypo Area and South Chejukun Area (서귀포시 지역과 남제주군 지역 여성의 영아영양법에 관한 실태조사)

  • This study was conducted to survey the feeding methods in Seogypo city and south Chejukun area. A random sample for this study a total of 355 mothers having babies under the age of 3 were surveyed. were boys. 45.5% were girls. infants' colostrum intakes in Seogypo sith and south Chejukun area were 69.9% and 63.1% respectability. Nother's education level affected colostrum intakes significantly. Baby's sex birth weight and mother's job affected the infants' feeding methods. The case of girl normal birth weight and mother's employment showed high percentage of bottle feeding. Bottle feeding was higher in Seogypo small city than in south Chejugun the country. Most of reasons for bottle feeding or mixed feeding was the lack of breast milk. The highest percentage of recommenders for breast feeding were baby's mother in Seogypo city area and were baby's grandmother in south Chejugun area. In Seogypo city area mother's education level did not affect the infant's feeding methods but in south Chjukun area mother's education level affected significantly.

Iron, Copper and Zinc Levels in Human Milk and Estimated Intake of the Minerals by Breast-Fed Infants during the Early Lactation (수유 초기 모유 중 철, 구리, 아연의 함량과 영아의 섭취량 추정)

  • The objectives of the present study were to measure the content of iron, copper and zinc in human milk and to estimate the intake of iron, copper and zinc of breast-fed infants during the early period of lactation. Twenty-five lactating women who delivered in a hospital in Seoul volunteered for the study. Milk samples were collected at day 2, 3, 4, 5, 6, 7, 15 and 30 postpartum. The contents of iron, copper and zinc were determined using the atomic absorption spectrophotometer after wet digestion. The intakes of iron, copper and zinc of infants were estimated by multiplization with the infant milk intake reported in our laboratory. The content of iron was 0.58 $\mu\textrm{g}$/g in colostrum, 0.48 $\mu\textrm{g}$/g in transitional milk and 0.39 $\mu\textrm{g}$/g in mature milk while the estimated iron intake of infants was 271, 255 and 259 $\mu\textrm{g}$/day, respectively. The content of copper in the milk was 0.45 $\mu\textrm{g}$/g in colostrum, 0.43 $\mu\textrm{g}$/g in transitional milk and 0.33 $\mu\textrm{g}$/g in mature milk while the estimated copper intake of infants was 210, 229 and 220 $\mu\textrm{g}$/day, respectively. The content of zinc in the milk was 5.24 $\mu\textrm{g}$/g in colostrum, 3.70 $\mu\textrm{g}$/g in transitional milk, 2.93 $\mu\textrm{g}$/g in mature milk while the estimated zinc intake of infants was 2452, 1968, and 1949 $\mu\textrm{g}$/day, respectively. These results suggest that copper and zinc intake of infants are met to RDA but iron is not.

Changes in Compositions of Holstein Colostrum during Lactation Period (Holstein 젖소 초유의 성분 변화에 관한 연구)

  • To investigate the compositions of Holstein colostrums, samples were collected at 12 hour-interval after 12hrs postpartum. Milk protein, milk fat, SNF and total solid content of the colostum rapidly decreased from 12 hours to 48 hours after calving whereas lactose was the lowest at 12 hours after calving. Ash content was not shown to changes during lactation periods. Immunoglobulin G(IgG) concentration was also significantly(p<0.05) high in both primiparous and multiparous colostrum collected at 12 and 24 hours after calving. IgG concentration of primiparous and multiparous colostrums at 12 hours after calving was 44mg/ml and 44.27mg/ml, respectively. There was no apparent difference in IgG level between primipara and multipara. Fatty acid composition of colostral lipid was not shown to changes during lactation period. However, lauric acid, myristic acid and total saturated, fatty acid were slightly decreased in multiparous colostral lipid. Capric acid, myristic acid and palmitoleic acid composition in primiparous colostral lipid were slightly higher than those of multiparous colostral lipid throughout all lactation periods.

Piglets' Surface Temperature Change at Different Weights at Birth

  • The study was carried out in order to verify the effects of piglets' weight at birth on their surface temperature change (ST) after birth, and its relationship with ingestion time of colostrum. Piglets from four different sows were weighed at birth and divided into a totally randomized design with three treatments according to birth weight (PBW): T1 - less than 1.00 kg, T2 - 1.00 to 1.39 kg, and T3 - higher than or equal to 1.40 kg. The time spent for the first colostrum ingestion was recorded (TFS). Images of piglets' surface by thermal imaging camera were recorded at birth (STB) and 15, 30, 45, 60, and 120 min after birth. The air temperature and relative humidity were recorded every 30 min and the indexes of temperature and humidity (THI) were calculated. A ST drop after 15 min from birth was observed, increasing again after sixty minutes. Positive correlations were found between the PBW and the ST at 30 and 45 min after birth. The PBW was negatively correlated with the TFS. The THI showed high negative correlations (-0.824 and -0.815) with STB and after 15 min from birth. The piglet's surface temperature at birth was positively correlated with temperature thereof to 15 min, influencing therefore the temperatures in the interval of 45 to 120 min. The birth weight contributes significantly to postnatal hypothermia and consequently to the time it takes for piglets ingest colostrum, requiring special attention to those of low birth weight.

  • The fatty acid composition of human milk was determined in 20 milk samples (ten colostrum and 10 mature) obtained at different stages of lactation. Human colostrum contained a lower percentage of total lipids than mature milk. In comparison with mature milk, human colostrum was characterized by a lower percentage of saturated fatty acids, a higher percentage of monounsaturated fatty acids and a higher percentage of extra-long-chain polyunsaturated fatty acid metabolites of both linoleic acid ($\omega$ 6 series) and linolenic acid ($\omega$ 3 series). The linoleic acid levels reported here are considerably higher than those reported previously in Korea. This shift has paralleled an increase in the use of vegetable oils in Korean diet. The human mature milk differed from marketing cow's milk in fatty acid composition, as it had a lower content of short-, medium-and long-chain saturated fatty acids and a higher content of long-chain monounsaturated and polyunsaturated fatty acids.

Biochemical Properties and Antibacterial Activity of Lactoferrin from Korean Native Cow (한우 Lactoferrin의 생화학적 특성 및 항균 활성)

  • The purpose of this study was to demonstrate biochemical properties and antibacterial activity of lactoferrin(Lf) obtained from the colostrum of Korean native cow. Lactoferrin was isolated from the colostrum of Korean native cow by purification steps using batch extraction, ion exchange chromatography, gel filtration chromatography, affinity chromatography. Other whey protein components that is similar molecular weight and affinity to lactoferrin were gradually removed from crude Korean Native cow's lactoferrin during the purification steps. The molecular weight of the purified Korean native cow's Lf(K-Lf) was 81 kDa, the isoelectric point was 9, and the content of iron was 0.56mg/g, which is indicated that iron saturation of the K-Lf was 40.6%. Amino acid composition and a-helix content were different K-Lf from bovine Lf(B-Lf). Antibacterial activity of E. coli O111 by K-Lf was lower than that of B-Lf. A minimal inhibitory concentration(MIC) of K-Lf and B-Lf was 2.75mg/ml and 1.5mg/ml respectively.

  • The purpose of this study was to demonstrate biochemical properties of lactoferrin (Lf) obtained from the colostrum of Korea native cow. The molecular weight of the purified Korean native cow's Lf (K-Lf) was 81kDa, the isoelectric point was 9, and the content of iron was 0.56 mg/g, which is indicated that iron saturation of the lactoferrin was 40.6%. Amino acid composition and a-helix content were different K-Lf from bovine Lf (B-Lf). Immunological cross reactivity was observed between K-Lf and B-Lf but not between K-Lf and human Lf (H-Lf) by immunodiffusion test and Western blot analysis. Out results indicate that structure of K-Lf is different from that of B-Lf although K-Lf and B-Lf were immunologically cross-reactive.
