배추김치즙의 Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus 및 Enterobacter cloacae에 대한 항균작용 (The Antibacterial Action of Chinese Cabbage Kimchi Juice on Staphylococcus aureus, Salmonella enteritidis, Vibrio parahaemolyticus and Enterobacter cloacae)

  • 배추김치의 항균작용을 실험한 본 연구에서 1개월간 발효된 배추김치즙 시료(염분 2.8%, 산도 1.34%, PH 4.1)를 2% 함유한 보통 한천 배지에서 실험군의 집락수 억제율은Salmonella enteritidis ATCC 13076이 가장 낮은 63%이었고 Enterobacter cloacae ATCC 13047과 Staphylococcus aureus ATCC 6538p은 각각 72%, 76%이었고 Vibrio parahaemolyticus ATCC 17802은 가장 높은 90%를 보였다. Vibrio parahaemolyticus ATCC 17802는 배추김치즙 함량이 6%에서 100%발육 억제효과를 보여 실험균 중 배추김치즙에 대한 저항력이 가장 약했으나 동일한 배추김치즙 시료농도(6%)에서 Salmonella enteritidis ATCC 13076, Enterobacter cloacae ATCC 13047, Staphylococcus aureus ATCC 6538p는 각각 89%, 90%, 91% 의 집락수 억제 효과를 보였다. 배추김치즙 함량 10%에서 모든 실험군의 집락가100% 억제되는 효과를 보였다. 실험결과 1개월간 충분히 발효된 배추김치에는 어떤 식중독균도 생존할 수 없을 것으로 밝혀졌다.

레이저와 불소가 인공우식병소의 항균성 및 내산성에 미치는 영향에 관한 연구 (EFFECTS OF LASER AND FLUORIDE TREATMENT ON THE RESISTANCE TO MICROORGANISM AND ACID IN ARTIFICIAL CARIOUS LESION)

  • The main purpose of this study was to evaluate the acid resistance and antimicrobial effect of fluoride-laser combined application. Recently extracted third molars were used. $5{\times}3mm$ of the buccal and lingual specimens were exposed and incipient artificial carious lesions were formed by keeping them in the artificial cariogenic solution for 5 days. They were divided into five groups and treated with fluoride and laser according to the predetermined regimen. The acid resistance was compared between groups by chemical quantitative analysis of the calcium and phosphorous released into the test solution after single or combined application of fluoride and laser. The antimicrobial effect of each group was analyzed by counting the number of colony forming units after microbial incubation. The results from the present study can be summarized as follows; 1. Experimental groups showed lower values in calcium and phosphorous contents as well as in $CFU/m{\ell}$(colony forming units) than control group. Combined application groups showed lower values than single application groups. 2. Acid resistance and antimicrobial effect by fluoride and laser treatment were confirmed in this study. 3. Based upon the above-mentioned results of this study, it can be assumed that the use of laser-fluoride combined application may provide the child and adolescent patient population with antimicrobial effect as well as acid resistance. Further studies using various materials and experimental conditions are being encouraged.

치마버섯(Schizophyllum commune)으로부터 $A{\alpha}$ mating locus의 분리 및 특성 (Isolation and Characterization of $A{\alpha}$ mating locus from Schizophyllum commune)

  • 본 연구는 고등균류중 담자균류에 속하는 치마버섯에 있어 자실체 형성을 직접적으로 조절하는 mating locus의 분리 및 특성을 규명하고자 하였다. 북미 자생의 치마버섯 UVM 1-34 균주로부터 $A{\alpha}3$ allele 분리를 위하여 만든 genomic library의 전체 숫자는 약 $2{\times}10^4$ cells로서 이중 약 90%가 약 35kb의 inserted DNA를 가진 것으로 나타났으며, colony 및 southern hybridization을 통해 얻은 6개의 clone 모두가 mating activity를 나타내었다. 이 중에서 1개 clone을 선정하여 남미자생의 UVM 1-71 $A{\alpha}3$ allele의 Z, Y region을 포함하는 5.7kb의 단편을 pBluescript ll KS(+)에 subcloning 시켜 trp1 gene 함유의 pTC20 cosmid와 함께 cotransformation 시킨 결과 약 50%의 clamp cell 형성을 보임으로서 이 clone이 mating activity를 가지는 것으로 나타났다.

Is There Additive Therapeutic Effect When GCSF Combined with Adipose-Derived Stem Cell in a Rat Model of Acute Spinal Cord Injury?

  • Objective : Functional and neural tissue recovery has been reported in many animal studies conducted with stem cells. However, the combined effect of cytokines and stem cells has not yet been adequately researched. Here, we analyzed the additive effects of granulocyte colony-stimulating factor (GCSF) on adipose-derived stem cells (ADSCs) infusion in the treatment of acute spinal cord injury (SCI) in rats. Methods : Four days after intrathecal infusion tubes implantation in Sprague-Dawley rats, SCI was induced with an infinite horizon impactor. In the Sham group (n=5), phosphate-buffered saline was injected 3, 7, and 14 days after SCI. GCSF, ADSCs, and ADSCs with GCSF were injected at the same time in the GCSF (n=8), ADSC (n=8), and ADSC+GCSF groups (n=7), respectively. Results : The ADSC and ADSC+GCSF groups, but not the GCSF group, showed significantly higher Basso-Beattie-Bresnahan scores than the Sham group during 8 weeks (p<0.01), but no significant difference between the ADSC and ADSC+GCSF groups. In the ladder rung test, all four groups were significantly different from each other, with the ADSC+GCSF group showing the best improvement (p<0.01). On immunofluorescent staining (GAP43, MAP2), western blotting (GAP43), and reverse transcription polymerase chain reaction (GAP43, nerve growth factor), the ADSC and ADSC+GCSF groups showed higher levels than the Sham and GCSF groups. Conclusion : Our analyses suggest that the combination of GCSF and ADSCs infusions in acute SCI in the rat does not have a significant additive effect. Hence, when combination agents for SCI stem cell therapy are considered, molecules other than GCSF, or modifications to the methodology, should be investigated.

  • Open space planners and designers should consider scientific and quantified functions of trees when they have to locate where to plant the tree. However, until now, most planners and designers could not consider them because of lack of tool for considering scientific and quantitative tree functions. This study introduces a tree location supporting tool which focuses on the multi-objective including scientific function using ACO (Ant colony optimization). We choose shading effect (scientific function), shielding, and openness as objectives for test application. The results show that when the user give a high weight to a particular objective, they can obtain the optimal results with high value of that objective. When we allocate higher weight for the shading effect, the tree plans provide larger shadow value. Even when compared with current tree plan, the study result has a larger shading effect plan. This result will reduce incident radiation to the ground and make thermal friendly open space in the summer. If planners and designers utilize this tool and control the objectives, they would get diverse optimal tree plans and it will allow them to make use of the many environmental benefits from trees.

Antibacterial activity of enrofloxacin loaded gelatin-sodium alginate composite nanogels against intracellular Staphylococcus aureus small colony variants

  • Background: The poor intracellular concentration of enrofloxacin might lead to treatment failure of cow mastitis caused by Staphylococcus aureus small colony variants (SASCVs). Objectives: In this study, enrofloxacin composite nanogels were developed to increase the intracellular therapeutic drug concentrations and enhance the efficacy of enrofloxacin against cow mastitis caused by intracellular SASCVs. Methods: Enrofloxacin composite nanogels were formulated by an electrostatic interaction between gelatin (positive charge) and sodium alginate (SA; negative charge) with the help of CaCl2 (ionic crosslinkers) and optimized by a single factor test using the particle diameter, zeta potential (ZP), polydispersity index (PDI), loading capacity (LC), and encapsulation efficiency (EE) as indexes. The formation mechanism, structural characteristics, bioadhesion ability, cellular uptake, and the antibacterial activity of the enrofloxacin composite nanogels against intracellular SASCVs strain were studied systematically. Results: The optimized formulation was comprised of 10 mg/mL (gelatin), 5 mg/mL (SA), and 0.25 mg/mL (CaCl2). The size, LC, EE, PDI, and ZP of the optimized enrofloxacin composite nanogels were 323.2 ± 4.3 nm, 15.4% ± 0.2%, 69.6% ± 1.3%, 0.11 ± 0.02, and -34.4 ± 0.8 mV, respectively. Transmission electron microscopy showed that the enrofloxacin composite nanogels were spherical with a smooth surface and good particle size distributions. In addition, the enrofloxacin composite nanogels could enhance the bioadhesion capacity of enrofloxacin for the SASCVs strain by adhesive studies. The minimum inhibitory concentration, minimum bactericidal concentration, minimum biofilm inhibitory concentration, and minimum biofilm eradication concentration were 2, 4, 4, and 8 ㎍/mL, respectively. The killing rate curve had a concentration-dependent bactericidal effect as increasing drug concentrations induced swifter and more radical killing effects. Conclusions: This study provides a good tendency for developing enrofloxacin composite nanogels for treating cow mastitis caused by intracellular SASCVs and other intracellular bacterial infections.

Severe congenital neutropenia mimicking chronic idiopathic neutropenia: a case report

  • Severe chronic neutropenia is classified as severe congenital, cyclic, autoimmune, or idiopathic. However, there is a lot of uncertainty regarding the diagnosis of severe congenital neutropenia (SCN) and chronic idiopathic neutropenia, and this uncertainty affects further evaluations and treatments. A 20-year-old man presented with fever and knee abrasions after a bicycle accident. On admission, his initial absolute neutrophil count (ANC) was 30/µL. He had no medical history of persistent severe neutropenia with periodic oscillation of ANC. Although his fever resolved after appropriate antibiotic therapy, ANC remained at 80/µL. Bone marrow (BM) aspiration and biopsy were performed, and a BM smear showed myeloid maturation arrest. Moreover, genetic mutation test results showed a heterozygous missense variant in exon 4 of the neutrophil elastase ELANE: c597+1G>C (pV190-F199del). The patient was diagnosed with SCN. After discharge, we routinely checked his ANC level and monitored any signs of infection with minimum use of granulocyte colony-stimulating factor (G-CSF), considering its potential risk of leukemic transformation. Considering that SCN can be fatal, timely diagnosis and appropriate management with G-CSF are essential. We report the case of a patient with SCN caused by ELANE mutation who had atypical clinical manifestations. For a more accurate diagnosis and treatment of severe chronic neutropenia, further studies are needed to elucidate the various clinical features of ELANE.

목조문화재의 원형보존을 위한 충해 방제방안 (Pest control managements for preservation of wooden cultural properties)

  • The cultural properties are damaged by various causes according to the characteristics of material, the condition of preservation, and the period of time. Especially, biodeterioration makes lots of damages in organic properties than inorganic ones. The damages of wooden cultural properties by insects usually are caused by the three orders; Isoptera, Coleoptera, and Hymenoptera. As the result of investigation on the state of 141 buildings of wooden cultural properties in 1999, some of them were damaged by many kinds off actors; wasp, powder post beetle, cigarette beetle, termite, decay, and physical cracking. And it was found that the patterns of damages were related to species-specific habits of insects. There are several methods of pest control for the prevention of wooden cultural properties from damages caused by insects. Those are as follows; physical control, chemical control, biological control, and integrated pest management. When insects and fungi were detected at the wooden buildings, the fumigation is best treatment to stop biodeterioration. And then, wood materials also need to be treated with insecticidal and antiseptic chemicals to avoid a reinfestation, because the fumigant is volatile. The six commercial chemicals which are applied to the insecticidal and antiseptic treatment of wooden cultural properties were purchased to test their abilities. According to the comparative results of efficacy of them in laboratory, chemical D showed excellent efficacy in all items, including antiseptic and termiticidal items. The goal of these pest controls is to protect wooden buildings from insects and microorganisms. The most effective method used currently is chemical control(fumigation, insecticidal and anticeptic chemical treatment), but it has to be treated periodically to control pest effectively. Recently environmentally-friendly control methods such as bait system or biological treatments are replacing traditional barrier treatments using large amounts of chemicals. Especially, termite is a social insect which makes a colony. Although a building with fumigation treatment is safe for a while, once attacked building has a risk of damage by reinfestation of termite. Therefore, to control termites from damaged building, the entire colony including reproductives(queen and king) and larvae around buildings must beeliminated. Bait system can be used as a preventive measure in early detection of them through termites colony monitoring and baiting. It would be the most effective for termite control if bait system would be used together with the chemical controls.

Senescence as A Consequence of Ginsenoside Rg1 Response on K562 Human Leukemia Cell Line

  • Aims and Background: Traditional chemotherapy strategies for human leukemia commonly use drugs based on cytotoxicity to eradicate cancer cells. One predicament is that substantial damage to normal tissues is likely to occur in the course of standard treatments. Obviously, it is urgent to explore therapies that can effectively eliminate malignant cells without affecting normal cells. Our previous studies indicated that ginsenoside $Rg_1$ ($Rg_1$), a major active pharmacological ingredient of ginseng, could delay normal hematopoietic stem cell senescence. However, whether $Rg_1$ can induce cancer cell senescence is still unclear. Methods: In the current study, human leukemia K562 cells were subjected to $Rg_1$ exposure. The optimal drug concentration and duration with K562 cells was obtained by MTT colorimetric test. Effects of $Rg_1$ on cell cycle were analyzed using flow cytometry and by SA-${\beta}$-Gal staining. Colony-forming ability was measured by colony-assay. Telomere lengths were assessed by Southern blotting and expression of senescence-associated proteins P21, P16 and RB by Western blotting. Ultrastructural morphology changes were observed by transmission electron microscopy. Results: K562 cells demonstrated a maximum proliferation inhibition rate with an $Rg_1$ concentration of $20{\mu}\;mol{\cdot}L^{-1}$ for 48h, the cells exhibiting dramatic morphological alterations including an enlarged and flat cellular morphology, larger mitochondria and increased number of lysosomes. Senescence associated-${\beta}$-galactosidase (SA-${\beta}$-Gal) activity was increased. K562 cells also had decreased ability for colony formation, and shortened telomere length as well as reduction of proliferating potential and arrestin $G_2$/M phase after $Rg_1$ interaction. The senescence associated proteins P21, P16 and RB were significantly up-regulated. Conclusion: Ginsenoside $Rg_1$ can induce a state of senescence in human leukemia K562 cells, which is associated with p21-Rb and p16-Rb pathways.

Isolation of Probiotic Piliated Lactobacillus rhamnosus Strains from Human Fecal Microbiota Using SpaA Antiserum-Based Colony Immunoblotting

  • Piliated Lactobacillus rhamnosus (pLR) strains possess higher adherent capacity than non-piliated strains. The objective of this study was to isolate and characterize probiotic pLR strains in human fecal samples. To this end, mouse polyclonal antiserum (anti-SpaA) against the recombinant pilus protein (SpaA) of L. rhamnosus strain GG (LGG) was prepared and tested for its reactivity and specificity. With the anti-SpaA, a method combining the de Man, Rogosa, and Sharpe (MRS) agar plating separation and colony immunoblotting (CIB) was developed to isolate pLR from 124 human fecal samples. The genetic and phenotypic characteristics of the resultant pLR isolates were compared by randomly amplified polymorphic DNA (RAPD) fingerprinting, and examination of adhesion to Caco-2 cells, hydrophobicity, autoaggregation, and in vitro gastrointestinal tolerance. Anti-SpaA specifically reacted with three pLR strains of 25 test strains, as assessed by western blotting, immunofluorescence flow cytometry, and immunoelectron microscopy (IEM) assays. The optimized MRS agar separation plus anti-SpaA-based CIB procedure could quantitatively detect $2.5{\times}10^3CFU/ml$ of pLR colonies spiked in $10^6CFU/ml$ of background bacteria. Eight pLR strains were identified in 124 human fecal samples, and were confirmed by 16S RNA gene sequencing and IEM identification. RAPD fingerprinting of the pLR strains revealed seven different patterns, of which only two isolates from infants showed the same RAPD profiles with LGG. Strain PLR06 was obtained with high adhesion and autoaggregation activities, hydrophobicity, and gastrointestinal tolerance. Anti-SpaA-based CIB is a rapid and inexpensive method for the preliminary screening of novel adherent L. rhamnosus strains for commercial purposes.
