COLONIZATION OF ALKALI-TREATED FIBROUS ROUGHAGES BY ANAEROBIC RUMEN FUNGI

  • This study reports light and electron microscope examination of rumen fungal colonization of alkali-treated roughage feeds incubated in decron bags in the rumen of cannulated sheep for varying time intervals. Six roughages, pre-treated with ammonium hydroxide or sodium hydroxide at 4% (w/w) level were examined together with untreated control samples. Alkali pre-treatment was associated with an earlier and more pronounced fungal colonization than all control roughages. Sodium hydroxide pre-treatment was significantly more effective than ammonium hydroxide in improving the susceptibility of roughages to rumen fungal colonization and studies by SEM showed that the pre-treatment permitted greater penetration of feeds by fungi. Sodium hydroxide pre-treatment also significantly increased dry matter disappearance from feed held in Dacron bags in the rumen with all feeds except Lucerne stem. It is not known to what extent fungal activity contributed to increased breakdown of the feeds.

Colonization and Extinction Patterns of a Metapopulation of Gold-spotted Pond Frogs, Rana plancyi chosenica

  • We investigated colonization and extinction patterns in a meta population of the gold-spotted pond frog (Rana planeyi ehoseniea) near the Korea National University of Education, Chungbuk, Korea, by surveying the frogs in the nine occupied habitat patches in the study area four times per breeding season for three years (2006$\sim$2008) and recording whether the patches were occupied by frogs as well as how many frogs were calling in the patches. We then developed five a priori year-specific models using the Akaike Information Criterion (AIC). The models predicted that: 1) probabilities of colonization and local extinction of the frogs were better explained by year-dependent models than by constant models, 2) there are high local extinction and low colonization probabilities, 3) approximately 31% number of patches will be occupied at equilibrium, and 4) that considerable variation in occupation rate should occur over a 30-year period, due to demographic stochasticity (in our model, the occupation rate ranged from 0.222 to 0.889). Our results suggest that colonization is important in this metapopulation system, which is governed by mainly stochastic components, and that more constructive conservation effects are needed to increase local colonization rates.

Endotracheal Colonization and Ventilator-associated Pneumonia in Mechanically Ventilated Patients according to Type of Endotracheal Suction System (기관 흡인술 유형에 따른 인공호흡기 적용 환자의 기관 내 균집락화와 폐렴 발생률)

  • Purpose: This study was conducted to identify endotracheal colonization and the incidence of ventilator-associated pneumonia related to the type of endotracheal suction system. Methods: The participants in this study were ICU patients hospitalized between October 2009 to March 2010 who used ventilators for over 48 hr with closed (CSS, n=30) or open (OSS, n=32) suction systems. To standardize the pre-intervention suction system, a suctioning protocol was taught to the ICU nurses. Collected data were analyzed using ${\chi}^2$-test, Fisher's exact test, Wilcoxon rank sums test, Wilcoxon test, Log-rank test and Poisson regression. Results: Endotracheal colonization was higher in OSS than CSS from day 1 to day 8 while using a ventilator and there was a significant difference between the two groups. The CSS reached 50% of endotracheal colonization by the 4th day, whereas for the OSS, it was the 2nd day (p=.04). The incidence of ventilator-associated pneumonia showed no significant difference. Conclusion: For patients with a high risk of pneumonia, CSS must be used to lower endotracheal colonization.

Seasonal Dynamics of Arbuscular Mycorrhizal Fungi (AMF) in Forest Trees of Chittagong University Campus in Bangladesh

  • Status of Arbuscular Mycorrhizal (AM) colonization in seven tree species (Albizia saman, Acacia auriculiformis A. Cunn. ex Benth., Albizia lebbeck, Chickrassia tabularis A. Juss., Eucalyptus camaldulensis Dehnn., Gmelina arborea (Roxb) DC, Swietenia macrophylla King.) collected from the hilly areas of Chittagong University (CU) was investigated. Roots and rhizosphere soil samples were collected in different seasons (pre-monsoon, monsoon and post monsoon). Percentage of AM colonization in root and number of spores/100 gm dry soil were assessed. The result of the investigation reveals that the intensity and percentage of AM colonization varied in different forest tree species in different seasons. In this study, maximum AM colonization and spore population were found in pre-monsoon and minimum were in monsoon season. The intensity of colonization was maximum in C. tabularis (74.43%) in pre-monsoon, A. lebbeck (69.45%) in monsoon and S. macrophylla (67.8%) in post monsoon seasons and minimum in A. auriculiformis (53.75%) during pre-monsoon, A. saman (24.4%) in monsoon and A. saman (19.36%) in post monsoon. The number of spores found per 100 g dry soil ranged between 164-376 during pre-monsoon, 27-310 during monsoon and 194-299 in post monsoon season. Out of six recognized genera of AM fungi, Glomus, Sclerocystis, Entrophospora, Scutellospora, Acaulospora and other unidentified spores were observed.

  • Colonization studies previously performed with a green-fluorescent-protein, GFP, labeled derivative of Bacillus amyloliquefaciens FZB42 revealed that the bacterium behaved different in colonizing surfaces of plant roots of different species (Fan et al., 2012). In order to extend these studies and to elucidate which genes are crucial for root colonization, we applied targeted mutant strains to Arabidopsis seedlings. The fates of root colonization in mutant strains impaired in synthesis of alternative sigma factors, non-ribosomal synthesis of lipopeptides and polyketides, biofilm formation, swarming motility, and plant growth promoting activity were analyzed by confocal laser scanning microscopy. Whilst the wild-type strain heavily colonized surfaces of root tips and lateral roots, the mutant strains were impaired in their ability to colonize root tips and most of them were unable to colonize lateral roots. Ability to colonize plant roots is not only dependent on the ability to form biofilms or swarming motility. Six mutants, deficient in abrB-, sigH-, sigD-, nrfA-, yusV and RBAM017410, but not affected in biofilm formation, displayed significantly reduced root colonization. The nrfA- and yusV-mutant strains colonized border cells and, partly, root surfaces but did not colonize root tips or lateral roots.

  • Degradation of rice straw was observed to be higher (p<0.01) in the buffaloes than in cattle. At 48 h, the dry matter (DM) loss of straw for buffaloes was 53.6 0.8% and that for cattle was 48.7 2.6%. Palm press fibre (PPF) was poorly degraded in the rumen of both animal species. A loss of about 21% DM was observed in both cattle and buffaloes after 48 h of incubation in the rumen. The pattern of bacterial and fungal colonization of straw and PPF seemed to be similar in both cattle and buffaloes. Microbial colonization was restricted by plant structures like the silica crystals in both straw and PPF. The predominant bacteria colonizing both straw and PPF fragments were the rods. Eroded zones and digestion pits were pronounced in straw fragments after 1 h of incubation. The PPF fragments appeared undegraded even after 6 h of incubation. Fungal colonization of straw was rapid and extensive in both cattle and buffaloes. The sporangia observed in straw were mainly spherical or oval in shape, but fusiform sporangia with acuminate tip were predominantly seen in PPF fragments.

A Case Report on the Clearance of Vancomycin-Resistant Enterococcus Colonization in a Patient with Traumatic Subarachnoid Hemorrhage and Subdural Hematoma Treated with Hwanglyeonhaedok-tang (황련해독탕 투여 후 Vancomycin-resistant Enterococcus 집락이 해제된 외상성 지주막하출혈 및 경막하혈종 환자 증례보고 1례)

  • This study aimed to investigate the effect of Hwanglyeonhaedok-tang on the clearance of vancomycin-resistant Enterococcus (VRE) colonization in a patient with traumatic subarachnoid hemorrhage and subdural hematoma. A 73-year-old man who was diagnosed with traumatic subarachnoid hemorrhage and subdural hematoma with isolation of VRE colonization was administered Hwanglyeonhaedok-tang and followed up with a rectal swab VRE culture every week. After treatment with Hwanglyeonhaedok-tang, VRE colonization was no longer detected. This case report proved that Hwanglyeonhaedok-tang could be an effective treatment for clearance of VRE colonization in a patient with traumatic subarachnoid hemorrhage and subdural hematoma.

The Effects of Bojungiki-tang by Withdraw of Isolation of Vancomycin-resistant enterococci Colonization - Case Series (Vancomycin 내성 장구균 집락 해제에 대한 보중익기탕의 효과)

  • Bojungiki-tang is a traditional oriental medicine to boost the immune system. In this study, we investigated the effects of Bojungiki-tang by withdrawal of isolation of VRE colonization. Four cases of post-stroke patients with VRE colonization took Bojungiki-tang and continuously were followed up with stool cultures. After three times negative stool VRE, we withdrew isolation of VRE colonization. One case patient was diagnosed with VRE colonization in another hospital and had no withdrawal during that period. He was admitted to Kyung-hee University Oriental Medicine where he took Bojungiki-tang. After three times negative stool VRE, we withdrew isolation of the patient. These results demonstrate that Bojungiki-tang is effective on boosting immunity level. Further studies are needed to better characterize this protective effect of Bojungiki-tang.

Flora Colonization and Oral Glucose Levels During the Early Postnatal Period in High-Risk Newborns (고위험신생아의 생후 초기 구강 내 균집락 형성과 당농도 및 영향요인)

  • Purpose: A longitudinal study was conducted to explore flora colonization and oral glucose high-risk newborns during the first 7 days after birth. Methods: Oral secretions of hospitalized newborns were obtained for microbial cultures and glucose test at days 1-7 after birth. Results: Among the total 112 newborns, 40% were girls and 73% were premature. Mean gestational age was $34.4{\pm}3.2$ weeks and weight was $2,266{\pm}697.5$ grams. The most common flora included Streptococcus (28.2%), Methicillin-resistant Staphylococcus aureus (MRSA, 10.9%), Staphylococcus (6.0%) and Coagulase-Negative Staphylococcus (CNS, 4.0%). The average oral glucose level was $29.2{\pm}23.0mg/dL{\sim}58.2{\pm}39.5mg/dL$. Newborns with higher oral glucose than serum (crude odds ratio [ORc] =1.75; 95% confidence interval [CI] =1.03-2.97), phototherapy (ORc=3.30; 95% CI=2.29-4.76) and prone position (ORc= 2.04; 95% CI=1.13-3.69) were more likely to be colonized. Having oral tubes (ORc=0.42; 95% CI=0.29-0.59), parental nutrition (ORc=0.21; 95% CI=0.13-0.32) and antibiotics (ORc=0.51; 95% CI=0.36-0.73) had protective effects. For oral glucose statistical significances existed on time effect among newborns with Streptococcus (F=9.78, p=.024), MRSA (F=7.60, p=.037) or CNS (F=11.15, p=.019) and interaction between time and colonization among newborns with all of four flora (F=2.73, p=.029) or colonization with only Staphylococcus (F=2.91, p=.034). Conclusion: High-risk newborns develop flora colonization at an early period of life. Their clinical features were associated with types and time of oral flora colonization. They need close monitoring and multifaceted intervention to improve oral environment and infection control.

The Incidences of Catheter Colonization and Central Line-Associated Bloodstream Infection According to Tegaderm vs. Chlorhexidine Gluconate (CHG)-Tegaderm Dressing (중심정맥관 번들이행에 따른 중심정맥관 균집락과 중심정맥관 관련 혈류감염: CHG테가덤과 일반테가덤 드레싱비교연구)

  • Purpose: In spite of the recent application of a general infection control method, central line-associated infections is still relatively high in Korea. Central line bundle with Chlorhexidine gluconate (CHG) tegaderm dressing was reported to be effective in reducing catheter colonization and central line-associated bloodstream infections (CLABSI). Therefore, this study aimed to examine the incidences of catheter colonization occurrence and CLABSI while using Tegaderm vs. CHG Tegaderm dressings. Methods: We used a descriptive design. 400 patients who had central venous catheters were selected from four hospitals in the Korean National Healthcare-associated Infections Surveillance System. Of all subjects, 200 used Tegaderm™ (Tegaderm group), and the remaining 200 used CHG Tegaderm (CHG Tegaderm group) dressing at the catheter insertion site. Data were analyzed using the χ2 test or Fisher's exact test, t-test, and logistic regression analysis using SPSS WIN 21.0. Results: In the Tegaderm and CHG Tegaderm groups, CLABSI incidences were 5.89 and 1.79 per 1,000 catheter-days, catheter colonization incidences were 3.93 and 1.43 per 1,000 catheter-days, and central line bundle compliance rates were 26.0% and 49.0%, respectively. Catheter colonization risk factors were 'reinsertion after failure' and 'Tegaderm dressing' at the central line insertion site. CLABSI risk factors were 'incomplete performance of 7 central line bundle items' and 'Tegaderm dressing' at the central line insertion site. Conclusion: A further prospective study is needed to examine the effects of central line bundle with CHG Tegaderm dressing, avoiding central line reinsertion after failure, and improving the bundle compliance in reducing catheter colonization and CLABSI.
