Post-Harvest Strategies to Improve Tenderness of Underutilized Mature Beef: A Review

  • Beef muscles from mature cows and bulls, especially those originating from the extremities of the carcass, are considered as underutilized due to unsatisfactory palatability. However, beef from culled animals comprises a substantial proportion of the total slaughter in the US and globally. Modern consumers typically favor cuts suitable for fast, dry-heat cookery, thereby creating challenges for the industry to market inherently tough muscles. In general, cull cow beef would be categorized as having a lower extent of postmortem proteolysis compared to youthful carcasses, coupled with a high amount of background toughness. The extent of cross-linking and resulting insolubility of intramuscular connective tissues typically serves as the limiting factor for tenderness development of mature beef. Thus, numerous post-harvest strategies have been developed to improve the quality and palatability attributes, often aimed at overcoming deficiencies in tenderness through enhancing the degradation of myofibrillar and stromal proteins or physically disrupting the tissue structure. The aim of this review is to highlight existing and recent innovations in the field that have been demonstrated as effective to enhance the tenderness and palatability traits of mature beef during the chilling and postmortem aging processes, as well as the use of physical interventions and enhancement.

하고초 추출물의 항혈전 효능 및 혈소판 응집 억제작용 (Antithrombotic and Antiplatelet Activity of Extract from Prunella vulgaris)

  • 한방생약제의 항혈전 및 혈소판 응집 억제 효능을 탐색하기 위하여 하고초의 물 추출물로 혈전 용해능 활성과 혈액 응고시간 지연효과 즉 PT (prothrombin time), APTT (activated partial thromboplastin time)와 혈소판 응집억제 활성 등에 대해 항혈전 효능을 평가하였다. 혈전용해도를 측정하는 fibrin plate가 용해되어 형성된 투명환의 넓이를 측정하는 실험을 진행한 결과 혈전용해도가 농도의존적으로 효능을 나타내었다. 혈액 응고 cascade에 미치는 영향을 알아보기 위해 혈액 응고 시간 지연 및 단축 효과를 확인하고자 APTT와 PT에 미치는 영향을 조사한 결과 PT의 경우 10 mg/ml, 5 mg/ml의 경우에는 대조군보다 우수한 지연효과를 보였다. APTT의 경우에는 10 mg/ml, 5 mg/ml는 대조군과 비교하여 매우 탁월한 지연효과를 보이고, 2.5 mg/ml, 1.25 mg/ml에서도 높은 지연효과를 나타냈다. 혈소판의 응집에 따라 형성되는 두 전극 사이에 형성된 전기적 저항의 변화로 나타나는 실험을 시행한 결과 하고초의 ADP와 collagen에서 뛰어난 응집억제 활성을 보였다. 따라서 위의 항혈전 효능평가 실험결과를 볼 때 하고초를 향후에 혈전 질환의 치료제 개발에 효과적으로 이용될 수 있을 것으로 사료된다.

LB30057 Inhibits Platelet Aggregation and Vascular Relaxation Induced by Thrombin

  • Previous study showed that an amidrazonophenylalanine derivative, LB30057, which has high water solubility, inhibited the catalytic activity of thrombin potently by interaction with the active site of thrombin. In the current investigation, we examined whether LB30057 inhibited platelet aggregation and vascular relaxation induced by thrombin. Treatment with LB30057 to plateletrich plasma (PRP) isolated from human blood resulted in a concentration-dependent inhibition of thrombin-induced aggregation. Values for $IC_{50}$ and $IC_{100}$ were $54{\pm}4$ nM and $96{\pm}3$ nM, respectively. This inhibition was agonist (thrombin) specific, since $IC_{50}$ values for collagen and ADP were \much greater than those for thrombin. In addition, concentration-dependent inhibitory effects were observed on the serotonin secretion induced by thrombin in PRP. Consistent with these findings, thrombin-induced increase in cytosolic calcium levels was inhibited in a concentration-dependent manner. When LB30057 was treated with aortic rings isolated from rats, LB30057 resulted in a concentration-dependent inhibition of thrombin-induced vascular relaxation. All these results suggest that LB30057 is a potent inhibitor of platelet aggregation and blood vessel relaxation induced by thrombin.

마이크로에멀젼을 이용한 우르솔릭산 피부 적용제제의 설계 및 평가 (Formulation Design and Evaluation of Ursolic Acid Microemulsion Delivery System for Topical Formulation)

  • Ursolic acid (UA), a bioactive triterpene acid, has been known to increase collagen content in human skin in addition to other actions such as anti-inflammatory, skin-tumor prevention and anti-invasion. However, it is poorly soluble in water. Therefore, we firstly prepared microemulsion system with benzyl alcohol, ethanol and Cremophor EL, RH 40 and Brij 35 as surfactant in order to increase solubility of UA and then prepared microemulsion was dispersed in o/w cream base for the topical delivery of UA in an effort to improve anti-wrinkle effect. The pseudo-ternary phase diagrams were developed and various microemulsion formulations were prepared using benzyl alcohol as an oil, Cremophor EL, RH 40 and Brij 35 as a surfactant. The droplet size of microemulsions was characterized by dynamic light scattering. The accumulation of VA in the skin from topical cream was evaluated in vitro using hairless mouse skins. The mean droplet size was $26.8{\pm}6.6$ nm for microemulsions II with Cremophor EL. All UA creams showed pseudoplastic flow and hysterisis loop in their rheogram, depending on the type of materials added in topical creams. The in vitro accumulation data demonstrated the UA topical cream prepared with the combination of Poloxamer 407 and Xanthan gum as a copolymer showed higher accumulation percentage than those prepared with either Poloxamer 407 or Xanthan gum. These results suggest that UA topical cream using microemulsion systems may be promising for the topical delivery of UA.

Comparison of Single and Double Combination of Temperature-time in Sous Vide Treated Semitendinosus Muscle from Cattle and Goat

  • This study observed the effects of the double combination of temperatures-times (2 temperatures: 2 times combination, 2T2T) in sous vide cooking method on the physicochemical properties and collagen solubility (CS) of semitendinosus muscle from cattle and goat as a comparison to common sous vide treatment (1 temperature: 1 time combination, 1T1T). The new invention of sous vide cooking method (2T2T) cooked at the first temperature at $45^{\circ}C$ for 3 h, and the second temperature at $60^{\circ}C$, $65^{\circ}C$, and $70^{\circ}C$ for 3 h, and labeled as N45-60, N45-65, and N45-70, respectively. While, common sous vide treatment (1T1T) were cooked directly for 6 h at $60^{\circ}C$ (T60), $65^{\circ}C$ (T65), and $70^{\circ}C$ (T70). Results revealed that cooking with 2T2T treatment improved the water-holding capacity and reduced the cooking loss of both beef and goat meat. The $L^*$ values have no apparent changes between treatment in beef and goat meat, while $a^*$ values of N45-60 treated goat presented markedly higher values than other treatments but an only slight increase in beef at the same treatment (p>0.05). Again, 2T2T treatment tended to decrease mean shear force (SF) values for beef and goat meat with the lowest SF values recorded at N45-60, and the CS no or less influenced this value. Therefore, the application of innovative sous vides cooking method (2T2T) presented comparable values in the treated beef and goat meat as compared to common sous vide method (1T1T).

Nutritional Quality and Variation of Meat and Bone Meal

  • Meat and bone meal is a valuable protein and mineral source in diets of production animals and contributes to the protein, energy and mineral component of diets. The aim of the present study was to more accurately characterise the apparent ileal amino acid digestibility of meat and bone meals produced in New Zealand and evaluate routine in vitro assays used in practise to measure meat and bone meal quality. A total of 94 commercial meat and bone meals from 25 New Zealand rendering plants over a two and a half year period were analysed for proximates, gross energy, gross amino acid content (incl. hydroxyproline, hydroxylysine and lanthionine), apparent ileal amino acid digestibility, pepsin nitrogen digestibility, protein solubility and bone content. The mean crude protein content of the 94 meat and bone meal samples was 56.8% with a range of >35% units and a coefficient of variation of 9.8%. The mean crude fat and ash content were 10.0 and 28.4% respectively. These latter components showed a large range (16 and 43%, respectively) with coefficients of variation above 22%. Amino acid digestibility between samples was highly variable with lysine and sulphur amino acids digestibility ranging between 45.8-89.0 and 38.2-85.5%, respectively. Pearson correlation coefficients are presented between crude protein content and individual gross amino acids, crude protein content and individual digestible amino acid content, and pepsin N digestibility and individual digestible amino acid content. There was a significant relationship between the digestible amino acid nitrogen content and the crude protein content while pepsin nitrogen digestibility was not correlated to ileal amino acid nitrogen digestibility (r=-0.06). Meat meals with a high protein content had relatively low hydroxyproline and hydroxylysine levels something that was attributed to the levels of collagen from bone. The data indicated that lanthionine (formed upon heat treatment of cysteine with a hydroprotein) is not a good indicator of the heat treatment employed to meat and bone meals. Step-wise multiple regression equations to predict the apparent digestible content of amino acids from rapid in vitro assays are presented. The most selected variables included ash and crude fat content. In general the equations derived for the essential amino acids had a higher degrees of fit (R2) compared to the non-essential amino acids. The R2 for the essential amino acids ranged from 0.43 for histidine and 0.68 for leucine. These equations provide a means of more rapidly estimating the apparent ileal digestible amino acid content (protein quality) of meat and bone meal using standard analyses.

대기오염에 의한 폐장조직 손상 -연구방향의 설정을 위한 논의- (Human Lung Insults due Air Pollutant -A Review for Priority Setting in the Research-)

  • Much progress has been made in understanding the subcellular events of the human lung injuries after acute exposure to environmental air pollutants. Host of those events represent oxidative damages mediated by reactive oxygen species such as superoxide, hydrogen peroxide, and the hydroxy, free radical. Recently, nitric oxide (NO) was found to be endogenously produced by endothelial cells and cells of the reticulo-endothelial system as endothelialderived relaxation factor (EDRF) which is a vasoactive and neurotransmitter substance. Together with superoxide, NO can form another strong oxidant, peroxonitrite. The relative importance of exogenous sources of $N0/N0_2$ and endogenous production of NO by the EDRF producing enzymes in the oxidative stresses to the heman lung has to be elucidated. The exact events leading to chronic irreversible damage are still yet to be known. From chronic exposure to oxidant gases, progressive epithelial and interstitial damages develop. Type I epithelial cells become thicker and cover a smaller average alveolar surface area while thee II cells proliferate instead. Under acute damages, the extent of loss of the alveolar epithelial cell lining, especially type II cells appears to be a good predictor of the ensuing irreversible damage to alveolar compartment. Interstitial matrix undergo remodeling during chronic exposure with increased collagen fibers and interstitial fibroblasts. However, Inany of these changes can be reversed after cessation of exposure. Among chronic lung injuries, genetic damages and repair responses received particular attention in view of the known increased lung cancer risks from exposure to several air pollutants. Heavy metals from foundry emission, automobile traffics, and total suspended particulate, especially polycystic aromatic hydrocarbons have been positively linked with the development of lung cancer. Asbestos in another air pollutant with known risk of lung cancer and mesothelioma, but asbestos fibers are nonauthentic in most bioassays. Studies using the electron spin resonance spin trapping method show that the presence of iron in asbestos accelerates the production of the hydroxy, radical in vitro. Interactions of these reactive oxygen species with particular cellular components and disruption of cell defense mechanisms still await further studies to elucidate the carcinogenic potential of asbestos fibers of different size and chemical composition. The distribution of inhaled pollutants and the magnitude of their eventual effects on the respiratory tract are determined by pollutant-independent physical factors such as anatomy of the respiratory tract and level and pattern of breathing, as well as by pollutant-specific phyco-chemical factors such as the reactivity, solubility, and diffusivity of the foreign gas in mucus, blood and tissue. Many of these individual factors determining dose can be quantified in vitro. However, mathematical models based on these factors should be validated for its integrity by using data from intact human lungs.

피혁분의 효율적인 탈 크롬 방법에 관한 연구 (Effective Chrome Removal Process from Shaving Dust)

  • 본 연구에서는 피혁분에서 크롬을 제거하여 여러 면으로 활용 가능한 단백질자원을 얻고자 하였다. 피혁분은 크롬이 콜라겐사이에 가교결합을 하여 크롬-콜라겐염(Cr-collagenate)을 형성하므로 1단계 공정에서 $Ca(OH)_2$용액에 침지(steeping)하여 팽윤(swelling)-팽창(plumping)시켜, $H_2SO_4$용액으로 크롬을 용출시켰다. 2단계 공정에서 NaOH용액으로 팽윤-팽창시키고 $H_2O_2$용액으로 용해도가 큰 Cr(VI)으로 산화하여 $H_2SO_4$용액으로 용출시켜 탈 크롬을 마무리하였다. 1단계 공정에서 $3%-Ca(OH)_2$용액, $0.8%-H_2SO_4$용액으로 2단계 공정에서 0.1%-NaOH용액, $3%-H_2O_2$용액, $1%-H_2SO_4$용액으로 순차적으로 침지하여 효율적으로 완전 탈 크롬을 할 수 있었다. 완전 탈 크롬된 피혁분에는 수분 10.68%일 경우, 조 단백질이 79.81% 함유하였으며, 각 공정의 침지용액들은 3회까지 반복 사용하여도 완전히 크롬을 제거할 수 있었다.

  • 본 연구에서는 상아질의 항복인장강도 (UTS)나 탄성계수 (E)와 같은 물리적 특성이 적용된 용매틀의 각각에 대한 Hoy의 수소결합 용해도 매개변수에 반비례한다는 가설을 설정하고 실험한 결과 가설이 입증되었으며 이를 토대로 다음과 같은 결론을 유도하여 보았다. 첫째는 탈회된 상아질의 인장 특성 및 물성이 가해진 극성 용매의 수소결합능에 밀접히 연관되어 있다는 것이며, 둘째는 낮은 수소결합능을 가진 용매는 교원섬유 층 내에서 새로운 펩타이드간 수소결합을 유도함으로써 탈회된 상아질의 인장력 및 탄성계수를 증가시킨다는 결과이다. 셋째로는 이러한 결과들을 토대로 높은 수소결합능을 가진 용매들은 새로운 펩타이드간 수소결합의 형성을 차단하여 탈회된 상아질의 구조적 특성을 유지시킬 수 있다는 결론을 도출하였다.

ME와 CP가 다른 유기사료 급여가 토종닭의 생산능력 계육품질 및 혈액성상에 미치는 영향 (Performance, Meat Quality and Blood Composition of Cross Bred Chicks Fed Various Organic Dietary CP and ME)

  • 본 연구는 토종닭(한협 3호)에 사육 기간별 대사에너지 및 단백질 수준이 다른 유기사료의 급여가 생산성, 계육품질, 혈액성상에 미치는 영향을 구명하기 위하여 수행하였다. 실험설계는 전기, 중기, 후기에 각각 $2{\times}3$(ME 3,000, 3,100kcal/kg; CP 21, 22, 23%), $3{\times}3$(ME 3,100, 3,150, 3,200kcal/kg; CP 18, 19, 20%), $3{\times}3$(ME 3,100, 3,150, 3,200kcal/kg; CP 15, 16, 17%) 요인실험이며 처리구당 총 공시수 및 반복은 전기에 720수, 4반복, 중기에 702수, 6반복, 후기에 468수, 4반복으로 하였다. 사육 전기 4주간에 사료에너지 수준은 생산성에 영향을 미치지 않았지만 CP는 23% 처리구에서 증체량, 사료섭취량이 21, 22% 처리구보다 매우 증가하였다(P<0.05). 사료요구율은 사료내 단백질 수준이 높을수록 개선되었다(P<0.05). 사육 중기에 증체량은 사료내 에너지가 낮을수록, 단백질은 높을수록 증가하는 경향을 나타내었다. 사료내 에너지 및 단백질 수준은 사료섭취량에 영향을 미치지 않았다. 사료에너지 수준에 따른 사료요구율은 CP 19, 20% 처리구에서 18% 처리구보다 개선되었다(P<0.05). 사육 후기 증체량은 사료에너지와 단백질 수준에 따른 차이가 없었다. 사료섭취량은 ME 3,150, 3,200kcal/kg 처리구에서 ME 3,100 kcal/kg보다 감소하였지만 사료요구율은 개선되었다(P<0.05). 혈중 총단백질, 알부민, 글루코오스, 중성지방은 에너지 및 단백질수준에 따른 차이가 없었다. 혈중 총콜레스테롤은 ME 3,100kcal/kg 처리구에서 ME 3,200kcal/kg 처리구보다 감소하였고 CP 처리구간 차이는 없었다(P<0.05). 가열감량은 ME 3,150kcal/kg 처리구에서 ME 3,200kcal/kg 처리구보다 감소하였다(P<0.05). 수분함량, 전단력, pH은 ME, CP 처리구간 차이를 나타내지 않았다. 단백질용해성은 에너지 및 단백질수준이 높을수록 증가하였다. 계육내 콜라겐(collagen) 함량은 에너지 수준이 높을수록 증가하는 경향이 있다. 계육의 백색도는 사료내 에너지수준이 감소할수록, 단백질수준은 증가할수록 개선되는 경향이 있었다. 적색도는 ME 3,150kcal/kg 처리구에서 ME 3,100kcal/kg 보다 증가하였다(P<0.05). 황색도는 ME 3,100, 3,200kcal/kg 처리구에서 낮아졌으며, CP 15, 17% 처리구에서 CP 16% 처리구보다 매우 낮아졌다(P<0.05). 따라서, 사육기간별 적정 유기사료내 에너지 및 단백질수준은 사육전기 4주간에 ME 3,000kcal/kg, CP 23%, 중기 4주간에 ME 3,100kcal/kg, CP 19%, 후기 2주간에 ME 3,150kcal/kg, CP 16% 급여가 생산성 개선에 적합하였다.
