This study is manufacturing method and analysis of feasibility about collagen peptide from fish scale. This is processed by enzyme hydrolysis, isolating and refining etc. The results of analysis of nutritional composition showed protein content of collagen peptide. In the analysis of constitutive amino acids, the ratio of contents of hydroxyproline and glycine, the characteristics of collagen peptides appeared similar and the contents of glutamic acid and aspartic acid which are involved in protein metabolism. As a result of measurement of total polyphenol content and total flavonoid, it showed that collagen peptide had more contents generally, and the effect of bioactivity of pig-skin collagen peptide appeared higher although different kinds of scale collagen peptide showed a little DPPH radical scavenging ability, total antioxidant capacity by ABTS, ACE inhibitory.
본 연구에서는 단백질 가수분해효소 0.1%와 1% Protamex를 사용하여 저분자 콜라겐을 제조하였다. 토종 닭발의 조단백질과 콜라겐의 함량은 일반 육계에 비해 높은 함량이 나타났다. 단백질 가수분해 효소농도와 반응시간이 증가할수록 낮은 분자량의 콜라겐을 얻을 수 있는 것으로 나타났다. 특히 1% Protamex로 7시간 처리한 시료가 1,000-5,000 Da의 저분자 콜라겐 함량이 55.6%로 나타났으며, 평균 분자량은 5,390 Da로 가장 낮은 분자량이 나타났다. 이는 단백질 가수분해효소 Protamex가 고분자 펩타이드 결합을 저분자 펩타이드로 분해했기 때문이다. 효소처리 콜라겐의 조직감은 고분자 펩타이드의 콜라겐이 저분자 펩타이드로 분해되어 gel을 형성하지 못하고 sol의 형태를 유지하였다. 효소농도와 효소반응시간이 증가할수록 콜라겐의 평균분자량은 작아지나 효소반응 5시간부터 평균분자량의 감소가 미미해지는 경향이 나타났다. 따라서 저분자 콜라겐 효소반응시간은 경제적으로 볼 때 5시간에서 7시간 사이가 적합하다고 할 수 있다. 이 연구결과는 향후 산업적 효소를 이용한 저분자 콜라겐 제조 및 식품 소재 활용의 기초자료로 이용할 수 있을 것이다.
생체에서 가장 많은 비율로 분포하고 있는 콜라겐 펩타이드는 동물의 뼈와 해양 생물의 비늘에 많이 함유되어 있다. 콜라겐은 동물 생체의 여러 결합조직에서 구조 단백질로 흔하게 발견된다. 또한, 이들은 생물 의료 자재, 제약, 화장품, 식품 및 가죽 산업에 널리 이용된다. 각종 어류 비늘에서 추출된 펩타이드는 UVB 조사에 의해 유도된 피부의 손상 및 광 노화에 대한 보호 효과가 있었다. 그럼에도 불구하고, UVB 조사에 대한 옥돔 비늘 유래의 펩타이드 특성은 명확히 알려져 있지 않다. 이 연구에서는 옥돔 비늘 추출물에서 분리된 1 kDa 이상(HMP)과 1 kDa 이하(LMP)의 펩타이드를 이용하여 UVB 조사에 의해 유도된 피부 손상과 광 노화에 대한 효과를 연구하였다. 이들 펩타이드는 농도 의존적으로 DPPH 라디칼 소거능을 보였으며 LMP는 HaCaT 인간 피부세포에서 UVB 조사에 의해 유도된 세포 지질 과산화 산물인 8-isoprostane 생성을 억제하였다. 그리고 LMP와 HMP는 B16F10 마우스 흑색종 세포에서 tyrosinase 활성 및 melanin 함량을 감소시켰으며 또한 HaCaT 세포에서 UVB로 유도된 elastase 활성을 감소시켰고 matrix metalloproteinase-1의 활성을 감소시켰다. 이러한 결과는 옥돔 비늘에서 유래된 펩타이드가 미백효과, 항산화제 및 광 노화 억제제로서 유용한 물질이 될 것으로 기대된다.
The effects of low molecular-weight collagen peptides derived from porcine skin were investigated on the physicochemical and sensorial properties of chocolate ice cream. Collagen peptides less than 1 kDa in weight were obtained by sub-critical water hydrolysis at a temperature of $300^{\circ}C$ and a pressure of 80 bar. Ice cream was then prepared with gelatin powder and porcine skin hydrolysate (PSH) stabilizers mixed at seven different ratios (for a total of 0.5 wt%). There was no significant difference in color between the resulting ice cream mixtures. The increase in apparent viscosity and shear thinning of the ice cream was more moderate with PSH added than with gelatin. Moreover, the samples containing more than 0.2 wt% PSH had enhanced melting resistance, while the mixture with 0.2 wt% PSH had the lowest storage modulus at $-20^{\circ}C$ and the second highest loss modulus at 10℃, indicating that this combination of hydrocolloids leads to relatively softer and creamier chocolate ice cream. Among the seven types of ice creams tested, the mixture with 0.2 wt% PSH and 0.3 wt% gelatin had the best physicochemical properties. However, in sensory evaluations, the samples containing PSH had lower chocolate flavor scores and higher off-flavor scores than the sample prepared with just 0.5 wt% gelatin due to the strong off-flavor of PSH.

Asterias pectinifera, a species of starfish and cause of concern in the aquaculture industry, was recently identified as a source of non-toxic and highly water-soluble collagen peptides. In this study, we investigated the antioxidant and anti-photoaging functions of compounds formulated using collagen peptides from extracts of Asterias pectinifera and Halocynthia roretzi (AH). Our results showed that AH compounds have various skin protective functions, including antioxidant effects, determined by measuring the scavenging activity of 2,2-diphenyl-1-picrylhydrazyl radicals, as well as anti-melanogenic effects, determined by measuring tyrosinase inhibition activity. To determine whether ethosome-encapsulated AH compounds (E(AH)) exert ultraviolet (UV)-protective effects, human dermal fibroblasts or keratinocytes were incubated with E(AH) before and after exposure to UVA or UVB. E(AH) treatment led to inhibition of photoaging-induced secretion of matrix metalloproteinase-1 and interleukin-6 and -8, which are associated with inflammatory responses during UV irradiation. Finally, the antibacterial effects of AH and E(AH) were confirmed against both gram-negative and gram-positive bacteria. Our results indicate that E(AH) has the potential for use in the development of cosmetics with a range of skin protective functions.

Skin aging appears to be the result of overlapping intrinsic (including genetic and hormonal factors) and extrinsic (external environment including chronic light exposure, chemicals, and toxins) processes. These factors cause decreases in the synthesis of collagen type I and elastin in fibroblasts and increases in the melanin in melanocytes. Collagen Type I is the most abundant type of collagen and is a major structural protein in human body tissues. In previous studies, many products containing collagen derived from land and marine animals as well as other sources have been used for a wide range of purposes in cosmetics and food. However, to our knowledge, the effects of human collagen-derived peptides on improvements in skin condition have not been investigated. Here we isolate and identify the domain of a human COL1A2-derived protein which promotes fibroblast cell proliferation and collagen type I synthesis. This human COL 1A2-derived peptide enhances wound healing and elastin production. Finally, the human collagen alpha-2 type I-derived peptide (SMM) ameliorates collagen type I synthesis, cell proliferation, cell migration, and elastin synthesis, supporting a significant anti-wrinkle effect. Collectively, these results demonstrate that human collagen alpha-2 type I-derived peptides is practically accessible in both cosmetics and food, with the goal of improving skin condition.
The prevalence rate of osteoporosis in older men and women has recently increased, and thus bone health is a major concern in Korea. This concern, along with increasing concern regarding youth height, has led to growth of the functional food industry for children, reaching approximately 200 billion won. It is necessary to develop safe and effective functional materials for bone growth and health. Foods are excellent sources of functional material as they are safe to use. It is well known that the phosphopeptide casein, which is derived from milk, is effective against osteoporosis by aiding in the absorption of calcium. In our study, collagen peptides derived from porcine skin were evaluated as a functional material for bone growth, as several peptides have been shown to aid in osteoblast formation.
In this study, novel peptides (NIPP-1, NIPP-2) derived from Navicula incerta (microalgae) protein hydrolysate were explored for their inhibitory effects on collagen release in hepatic fibrosis with the investigation of its underlying mechanism of action. TGF-${\beta}1$ activated fibrosis in LX-2 cells was examined in the presence or absence of purified peptides NIPP-1 and NIPP-2. Besides the mechanisms of liver cell injury, protective effects of NIPP-1 and NIPP-2 were studied to show the protective mechanism against TGF-${\beta}1$ stimulated fibrogenesis. Our results showed that the core protein of NIPP-1 peptide prevented fibril formation of type I collagen, elevated the MMP level and inhibited TIMP production in a dose-dependent manner. The treatment of NIPP-1 and NIPP-2 on TGF-${\beta}1$ induced LX-2 cells alleviated hepatic fibrosis. Moreover, ${\alpha}$-SMA, TIMPs, collagen and PDGF in the NIPP-1 treated groups were significantly decreased. Therefore, it could be suggested that NIPP-1 has potential to be used in anti-fibrosis treatment.
숭어(Mugil cephalus) 비늘에서 추출한 가용성 펩타이드 추출물이 streptozotocin (STZ 55 mg/kg BW, IP injection)으로 유도된 당뇨성 Sprague Dawley계 수컷 흰쥐의 있어서, 혈당 및 혈청 지질개선효과와 당질대사 이상 등에 관여하는 효소의 활성 변동을 생리생화학적 측면에서 검토하기 위하여 본 실험을 수행하였다. 대조군인 CG군을 비롯한 STZ 당뇨 유발군(SW군), 당뇨 유발에 콜라겐 펩타이드를 섭취시킨 군(SFW군)을 5주간 실험 사육한 결과, 혈당 농도는 당뇨 유발군에 콜라겐 펩타이드를 급여함으로서 유의적인 감소 효과를 보였다. 또한 혈청 총 콜레스테롤, 동맥경화지수, LDL, LDL-콜레스테롤, 중성지방 및 인지질 농도, 유리 콜레스테롤, 콜레스테롤 에스테르 비 등은 콜라겐 펩타이드를 섭취시킨 군(SFW군)에서 농도가 저하되는 것으로 나타났다. 한편, HDL-콜레스테롤 및 총 콜레스테롤에 대한 HDL-콜레스테롤 비 등은 콜라겐펩타이드 섭취(SFW군)에 의해서 증가되는 것으로 나타났다. 혈청 중 alkaline phosphatase (ALP) 및 aminotransferase(AST, ALT)의 활성은 STZ으로 당뇨를 유도 시킨 후 콜라겐펩타이드를 섭취시킴으로서 저하되는 것으로 나타났다. 따라서 본 실험 결과 등에서 콜라겐 펩타이드 추출물이 STZ으로 유발된 당뇨성 흰쥐의 혈당조절 기능 및 지질대사 이상 등에서 오는 당뇨성 질환의 예방 및 개선에 효과가 있을 것으로 사료된다.
