Reproduction Strategies of Clonal Plants of Potentilla conferta in Uzbekistan and Mongol

  • Clonal plants combine sexual and clonal reproduction, which contribute differently to plant fitness. Reproductive analyses have highlighted the importance of clonal growth in shaping the spatial genetic structure in Potentilla conferta Bunge, a herbaceous rhizomatic clonal distributed in hot sand dunes. We investigated the reproduction system of P. conferta at two populations in Mongol and three natural populations in Uzbekistan. The measurements of 19 quantitative or qualitative morphological characters were taken on each of total individuals directly from their natural habitats. Some morphological characteristics between Mongolia and Uzbekistan populations showed a slight heterogeneity of variance. Especially, the length of internodes (LFI and LSI) and characteristics of root (LLR and NOR) were shown a significant difference between two countries (P<0.05). P. conferta of Uzbekistan has most ramets at short distance intervals 30~100 cm. In light conditions, P. conferta of Uzbekistan was significantly less resilience than P. conferta of Mogol. In drought conditions, although there was not shown significant difference, P. conferta in Uzbekistan was less resilience than that in Mogol. The core analysis indicates that P. conferta in Uzbekistan is the more resistant than that of Mongol and seems to do by sexual reproduction strategy during several strong environmental disadvantages such as drought events.

Clonal Hematopoiesis of Indeterminate Potential Is Associated with Current Smoking Status and History of Exacerbation in Patients with Chronic Obstructive Pulmonary Disease

  • Background: There is limited data regarding the clinical outcomes of clonal hematopoiesis of indeterminate potential (CHIP) in patients with chronic obstructive pulmonary disease (COPD). This study aimed to evaluate the clinical significance of CHIP as a COPD biomarker. Methods: This retrospective study was conducted on patients with COPD who were enrolled prospectively in the Seoul National University Hospital Airway Registry from January 2013 to December 2019 and underwent pulmonary function and blood tests. We evaluated the CHIP score according to smoking status and severity of airflow obstruction. Results: We analyzed next-generation sequencing data to detect CHIP in 125 patients with COPD. Current smokers had a higher prevalence of CHIP in combination of DNMT3A, TET2, and PPM1D (DTP), DNA methyltransferase 3 alpha (DNMT3A), and protein phosphatase, Mg2+/Mn2+ dependent 1D (PPM1D) genes than in never- or ex-smokers. CHIP of DTP and DNMT3A genes was significantly associated with current smokers (adjusted odds ratio [aOR], 2.80; 95% confidence interval [CI], 1.01 to 7.79) (aOR, 4.03; 95% CI, 1.09 to 14.0). Patients with moderate-to-severe airflow obstruction had a higher prevalence of CHIP in most of the explored genes than those with mild obstruction, although the difference was not statistically significant. CHIP in ASXL transcriptional regulator 1 (ASXL1) genes was significantly associated with history of mild, severe, and total acute exacerbation. Conclusion: Given that CHIP in specific genes was significantly associated with current smoking status and acute exacerbation, CHIP can be considered as a candidate biomarker for COPD patients.

삼나무의 내한성(耐寒性) - 품종별(品種別) 채취시기별(採取時期別) 차이(差異) - (Freezing Resistance of Cryptomeria japonica - Its clonal and Seasonal Differences -)

  • 본(本) 연구(硏究)는 일본(日本) 및 한국(韓國)에서 내한성(耐寒性)이 강(强)하다고 생각되는 삼나무 클론 및 개체간(個體間)에 내한성도(耐寒性度)의 차이(差異)를 알기 위하여 진주(晋州)와 전주(全州)의 두 지역(地域)에서 생장(生長)하고 있는 삼나무 묘목(苗木) 및 성목(成木)으로 부터 1977~1978년도(年度) 겨울동안 3차(次)에 걸쳐 채취(採取)된 시료(試料)에 대(對)하여 주(主)로 엽(葉), 형성층(形成層), 재부유조직(材部柔組織)을 중심(中心)으로 내한성(耐寒性)을 조사(調査)하였다. 실험결과(實驗結果)와 고찰(考察)을 요약(要約)하면 다음과 같다. 1. 클론 및 개체간(個體間)의 내한성도(耐寒性度) 차이(差異)는 채취지역(採取地域)에 따라 최고(最高) $9^{\circ}C{\sim}15^{\circ}C$정도(程度)였다. 이러한 내한성도(耐寒性度)의 차이(差異)는 선발(選拔)된 모수(母樹)가 자라고 있는 지역(地域)의 기후조건(氣候條件)과 상관관계(相關關係)가 없었다. 대부분(大部分)의 삼나무 삼림(森林)이 인공조림(人工造林)된 것이므로 내한성(耐寒性) 유전자(遺傳子)와 환경(環境)과의 사이에 자연도태(自然淘汰)에 의(依)한 평형(平衡)이 아직도 이루어지지 않는 것으로 해석(解釋)되었다. 2. 한겨울에 채취(採取)된 몇 품종(品種)이 엽(葉)의 내한성도(耐寒性度)가 형성층(形成層)이나 재부유조직(材部柔組織)보다 높은 결과(結果)를 보였던 것 외에는 엽(葉), 형성층(形成層), 재부유조직(材部柔組織) 간(間)에 내한성도(耐寒性度)의 차이(差異)는 거의 없었다. 가장 내한성(耐寒性)이 낮은 부위(部位)는 정아(頂芽)이지만 정아(頂芽)가 동해(凍害)을 받을 경우는 잠아(潛芽)나 부정아(不定芽)에 의(依)하여 생장(生長) 계속되는 것이 관찰(觀察)되었다. 3. 엽(葉), 형성층(形成層), 재부유조직(材部柔組織)의 내한성도(耐寒性度)로 미루어 한국(韓國) 중부지역(中部地域)에서도 내한성(耐寒性)이 강(强)한 삼나무들은 생육(生育)될 수 있을 것이 예상(豫想)된다. 그러나 삼나무의 가장 내한성(耐寒性)이 약(弱)한 부위(部位)는 지면(地面)에 가까운 줄기라고 보고(報告)되었기 때문에 앞으로 더 연구(硏究)가 필요(必要)하다고 생각되며 지제부(地際部)의 동해(凍害)를 방지(防止)할 수 있는 조림학적(造林學的) 방법(方法)들이 타문헌(他文獻)을 통(通)하여 고찰(考察)되었다.

Potentilla속 내 한국의 솜양지꽃(Potentilla discolor)과 몽골의 P. conferta 생식계의 비교 (Comparison of Reproduction Systems of Genus Potentilla, Potentilla discolor in Korea and P. conferta in Mongol)

  • 한국의 솜양지꽃(Potentilla discolor) 9개 집단과 몽골의 양지꽃속 식물 종 P. conferta 두 집단을 이용하여 두 종의 생식계를 비교하였다. 분포지에서 19개 형태 형질을 조사하여 정량분석에 사용하였다. 형태 형질에서 두 종간 많은 형질에 대해 유의한 차이를 나타내었다. 주성분분석에서 마디간 길이와 뿌리의 길이와 개수 등에서 특히 유의한 차이를 나타내어 이들 두 종을 분류하는데 유의한 성분으로 판단된다. P. conferta에서 라메트의 수는 거리의 증가에 따라 현저하게 감소하나 P. discolor는 정규분포 모양의 곡선을 나타내어 $60{\sim}80$ cm에서 가장 많았다. 빛에 대한 감수성은 P. discolor가 P. conferta보다 내성을 가지나 가뭄에 대해서는 P. conferta가 P. discolor보다 더 높은 내성을 나타내었다 . 이는 사막에 근접한 건조한 지역에 적응한 형태로 판단되며 온대 지방인 우리나라에 분포하는 솜양지꽃에 비해 단위면적당 밀도가 높아 이웃간 거리가 짧아 짧은 라메트를 많이 가지며 라메트 수가 많아 영양번식이 더 많이 이루어지고 있음을 나타낸다.

Evaluation of Inflammation Parameters in Philadelphia Negative Chronic Myeloproliferative Neoplasia Patients

  • Background: Chronic myeloproliferative diseases are clonal stem cell diseases which occur as a result of uncontrollable growth and reproduction of hematopoietic stem cells, which are the myeloid series source in bone marrow. Recent studies have suggested that chronic inflammation can be a triggering factor in the clonal change in chronic myeloproliferative neoplasia (CMPN). In our study, we evaluated the existence of a chronic inflammation process in our Philadelphia negative (Ph-)CMPN patients using inflammation parameters in combination with demographic, laboratory and clinical characteristics of the patients. Materials and Methods: Demographic characteristics, clinical and laboratorial data, and thrombosis histories of 99 Ph-CMPN patients, who were diagnosed at our outpatient clinic of hematology in accordance with WHO 2008 criteria, were analyzed retrospectively,with 80 healthy individuals of matching gender and age included as controls. Complete blood counts, sedimentation, C reactive protein (CRP), JAK V617F gene mutations, abdomen ultrasound images and previous thrombosis histories of these patients were retrospectively analyzed. Results: Ph-CMPN and healthy control groups included 99 and 80 cases, respectively. PV, ET and MF diagnoses of patients were 43 (%43.4), 44 (44.4%) and 12 (12.1%), respectively. JAK V617F gene mutation was found to be positive in 64 (71.1%) of all cases and in 27(65.8%), 32 (82%), 5 (50%) of the cases in PV, ET and PMF groups, respectively. Thrombosis was determined as 12 (12%) in the entire group, 12.5% in the JAK V617F negative and 15.3% in the positive patients, with no statistical significance (p=0.758). No significant difference was observed between patients with and without previous thrombosis history in respect to hemogram parameters, sedimentation and CRP (p>0.05), neutrophil to lymphocyte ratio (NLR), erythrocyte distribution width (RDW), mean platelet volume (MPV) and sedimentation levels of the patient.

사시나무 (Populus davidiana Dode)의 액아(腋芽)를 이용한 기내대량증식(器內大量增殖) (In Vitro Plant Multiplication from Axillary Buds of Populus davidiana Dode)

  • Populus davidiana는 종자의 활력이 짧고(약 3주), 삼목시 발근율이 저조하여 전통적인 유성 무성번식법의 어려움이 있어서 효과적인 기내증식법을 연구하였다. 10가지의 기본 배지를 이용하여, 1-2개의 액아를 포함한 잎이 붙어 있는 줄기절편체를 0.2mg/l의 BAP가 첨가된 배지에서 배양한 결과 MS 배지와 LP배지에서 가장 많은 신초를 생산하였다. 두 배지에서 5주간 배양한 결과 절편체당 9개 이상씩 신초를 생산하였다. 신초생산과 신초의 생장에 있어 클론간의 상당한 차이를 보였다. 몇 클론은 생장조절물질을 첨가하지 않아도 신초를 생산하였다. 기내배양된 신초 중 60% 이상이 0.2mg/l의 IBA가 첨가된 1/2 GD배지에서 발근이 가능하였다.

Phosphotyrosine Protein Phosphatase Activity Is Inversely Related to Metastatic Ability in Rat Prostatic Tumor Cell Subclonal Lines

  • In clonal sublines with different metastatic ability derived from Dunning rat prostate tumor, phosphoamino acid levels of cellular proteins were determined. Cell lines with high metastatic ability exhibited 5-fold higher phosphotyrosine level than did cell lines with low metastatic ability, while the contents of phosphoserine and phosphothreonine were similar among cell lines examined, All cell lines showed similar activities of protein tyrosine kinases as well as overall protein kinases. Phosphotyrosine protein phosphatase (PTPP) activities of the cells with high metastatic ability were very low, compared to those of the cells with low metastatic ability, suggesting that the different phosphotyrosine levels among the cell lines were due to the difference in PTPP activities rather than protein tyrosine kinase activities. Cellular activities of prostatic acid phosphatase (PAcP), which has been reported to possess phosphotyrosine protein phosphatase activity, were shown to be inversely related to the phosphotyrosine levels and metastatic abilities of the prostate tumor cells, These results suggest that cellular PAcP activity, regulating phosphotyrosine levels of cellular proteins, is closely connected with the metastatic process in prostate tumor cells and can be utilized as a good biochemical marker for the diagnosis of metastasis of prostate tumor.

가로수용 적색 단풍 느티나무(Zelkova serrata Makino) 신품종 선발 (Selection of New Cultivars with Red Fall Leaf Color in Zelkova serrata Makino as Street Trees)

  • This study was carried out to develop new cultivars of Z. serrata showing red fall leaf as street trees. Z. serrata which had red fall leaves were selected and then examined for contents of leaf anthocyanin and chlorophyll. In addition, for the progeny test, selected trees were grafted. Of 21 trees having red autumn leaves, three individuals, 'S-6', 'S-20', and 'I-24' were finally selected. They contained higher level of anthocyanin in the leaves. Once developed, the red color remained till late autumn. Grafted plants of the selected strains showed high grafting efficiency and red foliage color in autumn. The soil pH of the survey sites ranged from 5.9 to 7.24. However, other soil characteristics did not show much difference among the sites with regard to inorganic nutrients including N(%), $P_2O_5$, CEC(mg/meq), $K^+$, $Ca^{3+}$, and $Mg^{2+}$, 'S-6', 'S-20', and 'I-24', clonal lines with red fall leaf were selected as new cultivars and propagates by grafting.

Prognostic Factors and Survival in Acute Myeloid Leukemia Cases: a Report from the Northeast of Iran

  • Background: Acute myeloid leukemia (AML) is a clonal hematopoietic disorder resulting from genetic alterations in normal hematopoietic stem cells. The aim of this study was to evaluate prognostic factors and survival of AML patients in the Northeast of Iran. Materials and Methods: This retrospective study covered 96 patients with AML referred to Emam Reza Hospital, Mashhad city, Iran, from 2009 to 2015. Age, sex, blood group, type of AML, fever, consumption of amphotericin B, cytogenetic forms and survival were analyzed. Also, WBC, hemoglobin and platelet levels were checked. Mean follow-up was 30.5 months (60.4% mortality). Survival was plotted by GraphPad Prism 5 with Log-rank test. Results: The mean age for all AML patients at diagnosis was 40.4 years (range, 17-77 years). Some 42.7% patients were aged <35 years and 40.6% were male. In all patients, 76% had fever and 50% consumed amphotericin. T(15;17)(q22;q21) had the most prevalence (37.7%) compared to other forms. Out of 92 patients, O+(30.4%) was the most common blood group and AML-M5 (28.3%) the most common subtype. There was a significant difference in survival based on WBC and consumption of amphotericin B (P<0.05). Conclusions: WBC level, fever and consumption of amphotericin B proved to be factors for survival of AML patients. The mean age for patients in Iran is lower than other areas in the World and also survival in this study was higher than in other studies.

미류나무 엽육조직에서 식물체 재분화 (Regeneration of Adventitious Shoots from Populus deltoides Bartr.)

  • 미류나무의 잎절편들을 fructose를 첨가한 WPM에서 배양하였으나, TDZ와 BA 그리고 NAA의 여러 농도와의 조합에서 다른 형태의 기관들(뿌리나 줄기)이 관찰되었으며, 체세포 배와 유사한 기관들의 형태도 관찰되었다. 3계통 미류나무들의 잎절편들을 사이토카이닌(BA 또는 TDZ)와 오옥신(NAA)를 첨가한 WPM 배지에서 접종시켜, 재분화를 유도한 결과, BA (05 mg/L)나 TDZ (0.01 mg/L)와 NAA (0.05 mg/L 또는 0.1 mg/L)의 조합에서 재분화 율은 각각 63%에서 100%이고, 재분화 개체 수는 잎절편당 평균 2.5부터 20개, 그리고 부정줄기 형성체군(organogenic site) 들은 평균 1.8에서 4.5개로 높게 나타났다. 또한 세 계통의 미류나무들은 각기 유사한 처리 조합에서 높은 재분화율을 나타내었다.
