  • Hong, You-Son($1452{\sim}1529$) overcame his suffering, caused by his low social standing, through Taoism. Hong, You-Son could be infatuated with Taoism because Taoism was still spread all over the country in early Chosun dynasty and in more permissive atmosphere than the atmosphere after the Japanese invasion of Korea in 1592. Therefore, Taoist health prevention, not religion, reflecting theory of Taoist hermit and wishing for perennial youth and long life was prevalent among the intellectual class in that period. In addition, it was very fascinating idea for Chosun intellectual class to prevent health with practice because medicine was not developed in that period. The importance of his health prevention was vitality ; he insisted that people can get health and long life with it and need mind practice to discard greed of this life. His advocacy was agreed with Kim, Si-Seup, and Chung, Ryum, the members of Danhak School, and his health prevention became a base for medicine developments preventive medicine. Hong, You-Son seemed to use Taoism not to avoid temporally, but to overcome his suffering, and furthermore, find out self identity. He became one of major members of Danhak School because the ideas of Taoism such as accomplishing health and long life with self practice and achieving self satisfaction with discarding utilitarianism was fitted for conflict mind of Hong, You-Son. Taoist ideas were reflected in his poetry and his everyday life ; he identified a Taoist hermit with Himself in his poetry and acted like a hermit in his everyday life.

  • The gene for esterase (rEst1) was isolated from a new species of genus Rheinheimera by functional screening of E. coli cells transformed with the pSMART/HaeIII genomic library. E. coli cells harboring the esterase gene insert could grow and produce clear halo zones on tributyrin agar. The rEst1 ORF consisted of 1,029 bp, corresponding to 342 amino acid residues with a molecular mass of 37 kDa. The signal P program 3.0 revealed the presence of a signal peptide of 25 amino acids. Esterase activity, however, was associated with a homotrimeric form of molecular mass 95 kDa and not with the monomeric form. The deduced amino acid sequence showed only 54% sequence identity with the closest lipase from Cellvibrio japonicus strain Ueda 107. Conserved domain search and multiple sequence alignment revealed the presence of an esterase/ lipase conserved domain consisting of a GXSXG motif, HGGG motif (oxyanion hole) and HGF motif, typical of the class IV hormone sensitive lipase family. On the basis of the sequence comparison with known esterases/ lipases, REst1 represents a new esterase belonging to the class IV family. The purified enzyme worked optimally at $50^{\circ}C$ and pH 8, utilized pNP esters of short chain lengths, and showed best catalytic activity with p-nitrophenyl butyrate ($C_4$), indicating that it was an esterase. The enzyme was completely inhibited by PMSF and DEPC and showed moderate organotolerance.

  • Lipid transfer proteins (LTPs) are widely distributed in the plant kingdom, but their functions remain elusive. The proteins AlLTP2-4 were isolated from three related Allium plants: garlic (A. sativum L.), Welsh onion (A. fistulosum L.), and Nanking shallot (A. ascalonicum L.). These novel proteins comprise a new class of LTPs associated with the Ace-AMP1 from onion (A. cepa L.). The AlLTP genes encode proteins harboring 132 common amino acids and also share a high level of sequence identity. Protein characteristics and phylogenetic analysis suggest that LTPs could be classified into five distinct groups. The AlLTPs were clustered into the most distantly related plant LTP subfamily and appeared to be restricted to the Allium species. In particular, the number of amino acids existing between the fourth and fifth Cys residue was suggested as a conserved motif facilitating the categorization of all the LTP-related proteins in the family. Unlike other LTPs, AlLTPs harboring both the putative C-terminal propeptide and N-terminal signal peptide were predicted to be localized to cytoplasmic vacuoles. When a chimeric GFP protein fused with both N-terminal and C-terminal AlLTP2 signal peptides was expressed in rice cells, the fluorescence signal was detected in the endomembrane compartments, thereby confirming that AlLTPs are an unprecedented intracellular type of LTP. Collectively, our present data demonstrate that AlLTPs are a novel type of LTP associated with the Allium species.

  • The purpose of this paper is to make a critique of racial aspects of Caribbean literature more ethical through a constant concern with history and political philosophy. The first step I take for this purpose is a comparative reading of C. L. R. James's view of Toussaint L'Ouverture's position and Frantz Fanon's view of race and class in the historical context of the Caribbean power-relations. In so doing, I examine how Toussaint's and Fanon's wills to negotiation were thwarted in the New World history. To elaborate upon this ethico-political approach, I have recourse to the so-called later Derrida, focusing on his books, such as The Politics of Friendship, Of Hospitality, On Cosmopolitanism and Forgiveness, etc. Taking an up-close look at Derrida's thought, I argue that his political contemplation of ethics is as effective as his deconstruction of "otherness" in dealing with the nature of ethnic clashes in both the real world and minority literature. In the second half of my paper, I reexamine the issues of race, gender, and class in the three novels of Jamaica Kincaid - Annie John, Lucy, and The Autobiography of My Mother. It is conceivable that from the feminist perspective Kincaid's fiction has been read as a postcolonial Bildungsroman. In my supplementary attempts to this criticism, I reveal that the teenage narrator's precocious awareness is still under the colonial influence in the Annie John section. My analysis of Lucy contends that the reasons why the white woman fails to make friends with the young black woman should be sought in the long history of the U.S. racial politics. In the section of The Autobiography of My Mother, I discuss how difficult it is for a minority woman to liberate from the spell of history insofar as she is engaged in the issue of identity. In closing, I pose a need of consolation that literature may grant us by becoming able to produce a different interpretation on all the bleaker reality.

  • Hanok is Korea's traditional housing. From the 1970s, the vast majority of citizens in urban areas have not lived in Hanok housing but an apartment. Consequently, many people no longer familiar with the experience of living in traditional Hanok. People who live in the environment of residential apartments eargle like new residential and pursue eco-friendly homes, health homes, espacially among the hanok was reassure potential. There have been many debate how Hanok housing can become more sustainable, yet keep characteristics of its identity. The purpose of this study is to examine the process by which traditional Hanoks and modern architecture can influence each other, how the popularization of modern homes was formed and this relationship with the new less popular Hanoks. Finally, We will exmine how old Hanok can be renovated into new, modern Hanoks. The subject areas are Seoul, Jeonju, and Chongju in early twentieth century; by discussing with upper-class Hanok to improve the general standard of most traditional Hanok.

  • Glycolysis is responsible for the conversion of glucose into pyruvate and for supplying reducing power and several metabolites. Fructose-1,6-bisphosphate aldolase (AtFBA1), a central enzyme in the glycolysis pathway, was isolated by functional complementation of the salt-sensitive phenotype of a calcineurin (CaN)-deficient yeast mutant. Under high salinity conditions, aldolase activity and the concentration of NADH were compromised. However, expression of AtFBA1 maintained aldolase activity and the NADH level in yeast cells. AtFBA1 shares a high degree of sequence identity with known class I type aldolases, and its expression was negatively regulated by stress conditions including NaCl. The fusion protein GFP-AtFBA1 was localized in the cytosol of Arabidopsis protoplasts. The seed germination and root elongation of AtFBA1 knock-out plants exhibited sensitivity to ABA and salt stress. These results indicate that AtFBA1 expression and aldolase activity is important for stress tolerance in yeast and plants.

  • Increased consensus among gerontologists exist on the need to pay greater attention to the reciprocal relationship between the social and spatial in order to understand the construction of aged identity and everyday lives of old people. With urbanization, spaces are increasingly age-graded and as a consequence, urban elders are socially isolated from the community. In this study, we examine the social interaction patterns in various places specifically designated for the elderly, such as the Senior Center, Senior Welfare Center, and Hall for the Aged in Seoul. Main purpose of this study is to understand everyday life experiences of space the elderly people residing in the city go through. Data were gathered through qualitative, case study method. Results show that such spaces were important sites for social interaction for urban elderly who lacked social spaces after retirement and active parenting role. Also, it was shown that each place presents different contexts for social interactions and certain components of social class differences existed. Heterogeneity within the participants of each place was also observed. Quite strong stereotypes about certain places were also observed among the urban elders. Based on these results, importance of developing a 'community perspective' in the study of old age is discussed.

  • The ectomycorrhizal fungus Tricholoma matsutake grows symbiotically with Pinus densiflora. Phenylalanine ammonia-lyase (E.C. 4.3.1.24) catalyzes the conversion of L-phenylalanine to trans-cinnamic acid. The role of fungal phenylalanine ammonia-lyase, however, has not been clear until now. In this study, the gene encoding phenylalanine ammonia-lyase (PAL), which was isolated from T. matsutake, was cloned and characterized. The PAL gene (tmpal) consists of 2,160 nucleotides, coding for a polypeptide containing 719 amino acid residues. The deduced amino acid sequence of tmpal from T. matsutake shows high identity (70%) with that from Laccaria bicolor. Comparative analysis of the PAL genes among T. matsutake and other species of the class Agaricomycetes showed that both active sites and binding sites were significantly conserved among these genes. The transcriptional analysis of the PAL gene revealed a differential gene expression pattern depending on the developmental stages (mycelium, primordium, stipe, pileus, and gills) of T. matsutake. These results suggest that the PAL gene in T. matsutake plays an important role in multiple physiological functions.
