Relationship between Clearance Rates and Metal Uptake Rates of Corbicula fluminea, Potamocorbula amurensis and Macoma balthica: Influence of Water Temperature and Body Size (세 종의 이매패류 Corbicula fluminea, Potamocorbula amurensis, Macoma balthica의 여수율과 금속 흡수율과의 관계에 대한 연구)

  • A series of radiotracer studies were conducted to evaluate the influence of water temperature and/or body size on the clearance rates and uptake rates of Cd, Se and Zn in the Asiatic clam, Corbicula fluminea, Asian clam Potamocorbula amurensis and Balthic clam, Macoma balthica. Uptake rates of Cd, Se and Zn were estimated simultaneously with clearance rate of clams under 3 different water temperature conditions (5, 13 and $21^{\circ}C$). The weight specific clearance and metal uptake rates of P. amurensiswere increased with temperature, however, no consistent temperature effect was observed for the other clams. The variation of uptake rates of Cd, Se and Zn along with temperature or body size in each clam species was well associated with clearance rates. The inter-species as well as the intra-species difference of metal uptake rates could be well explained by the variation of clearance rates of clam individuals.

Screening and Purification of an Antimicrobial Peptide from the Gill of the Manila Clam Ruditapes philippinarum (바지락(Ruditapes philippinarum) 아가미로부터 항균 펩타이드의 탐색 및 정제)

  • This study screened the biological activity of an acidified gill extract of the Manila clam Ruditapes philippinarum including antimicrobial, hemolytic, membrane permeabilization, and DNA-binding activity, and purified the antimicrobial material. The acidified gill extract showed potent antimicrobial activity against Bacillus subtilis and Escherichia coli without significant hemolytic activity, but showed no membrane permeabilization or DNA-binding ability. An antimicrobial material was purified from the acidified gill extract using C18 reversed-phase and cation-exchange high-performance liquid chromatography (HPLC). Treatment of the purified material with trypsin completely abolished all of the antibacterial activity against Bacillus subtilis, suggesting that the purified material is a proteinaceous antibiotic. The molecular weight of the purified material was 2571.9 Da, but no primary structural information was obtained due to N-terminal blocking. A future study should confirm the primary structure. Our results suggest that the Manila clam gill contains proteinaceous antibiotics that have a role in first-line defense. This information could be used to better understand the Manila clam innate immune system.

Studies on the Myofibrillar Proteins of Mollusca (연체류의 근원섬유단백질에 관한 연구)

  • In order to compare and examine the general characteristics of myofibrillar proteins which is an important protein source as a food resource and relates directly with muscle contraction, we have extracted the myofibrillar proteins from squid and clam. The ionic strength of myofibrillar proteins connected with Ca-ATPase activity, Mg-ATPase activity and EDTA-ATPase activity showed distinct differences between squid and clam. In the activity-pH curve, actomyosin of the clam had a weak biphasic response. In the low concentration of dioxane, myofibrillar proteins of the squid showed a sudden decrease in activity but myofibrillar proteins of the clam showed in increase in activity. Ethanol and metanol in low concentration caused myosin and HMM from the squid and clam to increase their activities. If we cause modification by NEM, under 10-6M concentration, the activity was increased but above 10-5M concentration, there was a sudden decrease in activity.

  • The liquid lead-lithium (Pb-17Li) blanket has many applications in fusion reactors due to its good tritium breeding performance, high heat transfer efficiency and safety. The compatibility of liquid Pb-17Li alloy with the structural material of blanket under magnetic field is one of the concerns. In this study, corrosion experiments China low activation martensitic (CLAM) steel and 316L steel were carried out in a forced convection Pb-17Li loop under 1.0 T magnetic field at 480 ℃ for 1000 h. The corrosion results on 316L steel showed the characteristic with a superficial porous layer resulted from selective leaching of high-soluble alloy elements and subsequent phase transformation from austenitic matrix to ferritic phase. Then the porous layers were eroded by high-velocity jet fluid. The main corrosion mechanism of CLAM steel was selective dissolution-base corrosion attack on the microstructure boundary regions and exclusively on high residual stress areas. CLAM steel performed a better corrosion resistance than that of 316L steel. The high Ni dissolution rate and the erosion of corroded layers are the main causes for the severe corrosion of 316L steel.

Accumulation of Heavy Metals in the Antarctic Clam Laternula elliptica

  • Immunohistochemical and ultrastructural experiments were conducted to find out heavy metal accumulation in some selected organs such as the kidney, the hepatopancreas, and the gills of the Antarctic Clam Laternula elliptica According to the immunohistochemical study the subject organs of the clam showed reactions indicating the presence of MT(metallothionein), a metal-binding protein involved in metal detoxifying process. Examination under the transmission electron microscope also revealed that other ligands(e.g. metal-rich granules in the kidney) may play a role in metal accumulating and detoxifying process in L. elliptica. In the artificial exposure of the clam to Cd, it showed immediate subcellular responses, suggesting that this species can be used as rapid and efficient bioindicators for Cd exposure in natural environment.

Genetic Similarity and Difference of Marsh Clam (Corbicula leana) Obtained by RAPD-PCR

  • Genomic DNA from the muscle of marsh clam (Corbicula leana) from Gochang was extracted in order to identify genetic differences and similarity by randomly amplified polymorphic DNAs-polymerase chain reaction. 3.28 of the 23.0 polymorphic bands per lane were found to be polymorphic in marsh clam. Also, about 4.34% of total polymorphic bands were either specific to marsh clam. The major common bands of 0.28 kb generated by primer OPB-15 (GGAGGGTGTT) were present in every individuals, respectively, which were polymorphic. This common bands which present in every individuals should be diagnostic of specific strains, species and/or their relatedness. Primer OPB-19 (ACCCCCGAAG) produced the highest number of specific bands, which was 12. The specific minor band of 0.07 kb was present in lane 22, which were polymorphic. Especially, only a specific band (1.35 kb) identifying individuals was observed in lane 22.

  • Chemical composition, free amino acid and fatty acid composition of wild and cultured cockle clam were studied. 1. The content of crude protein was higher in Anadara suberenata thanthat ini Anadara granosa. The contents of crude protein and ash increased after boiling. 2 Free amino acid composition of Anadara granosa was similar to that of Anadara suberenata. Both glutamic acid and aspartic acid among free amino acid were the most abundant in cockle clam. Next, leucine, arginine and lysine were abundant in order. They all composed of approximately 50%^ of total amino acid content. Whereas the contents of cystine and methionine were poor. 3. In cockle clam the major fatty acids consisted of C16 :0, C18: 0, C18 ;1 and C18:3 in order. The level of C16 :0 was the highest in lipids. The levels of C18 : 0 and C14 :0 were higher in Anadara granosa than those in Anadara suberenata, while the levels of C16 :0, C18 :3 and C16 : 1 were higher in Amadara suberenata than those in Anadara granosa.

Unique substrate preference of Ostrea denselamellosa Lischke, 1869 (Mollusca: Ostreidae) at Haechang Bay, on the south coast of Korea

  • In the present study, we observed a unique association of the flat oyster, Ostrea denselamellosa obtained from a muddy substrate at Haechang Bay, the south coast of Korea in the spring of 2013. Fossilized or semi-fossilized veneriid clam shells, possibly Ruditapes philippinarum, were found adhering to the umbonal area of the flat oyster valves. This unique association of the flat oyster shells with the fossilized clam shells suggested that the flat oyster larvae utilized the clam shells as substrate during settlement. Since availability of clam shells in the muddy subtidal environment is limited, this unique substrata for the flat oyster larvae may limit recruitment of the flat oysters in the bay.

ESR Spectroscopy for Detecting Gamma-Irradiated Shellfishes (방사선 조사된 조개류의 확인을 위한 ESR Spectroscopy의 이용)

  • Electron spin resonance (ESR) spectroscopy was used to investigate the effect of irradiation dose on the ESR signal intensity and to identify the stability of radicals after 9 weeks of storage in order to detect irradiated shellfishes. The irradiated shellfishes (short-necked clam, purplish washington clam, freshwater clam, jackknifed clam, scallop and hard-shell mussel) presented an asymmetric absorption in shape at $g_{1}$=2.002~2.003 and $g_{2}$=1.998. The strength of the ESR signal increased linearly with the applied doses (1~7 kGy). A highly positive correlation coefficients ($R^{2}$=0.9136~0.9896) were obtained between the irradiation dose and corresponding ESR signal intensity. The intensity of the signals after irradiation was stable even after 9 weeks of storage at 5$\pm$1$^{\circ}C$.
