진피의 수치 조건에 따른 성분 및 항산화 활성 변화 (Changes in the Constituents and Antioxidant Activity in Accordance with the Processing Conditions of Citrus unshiu Markovich)

  • Processing of medicinal plants is one of the processing methods for reducing of toxicity or improving of effect on medicinal plants. In this study, we studied the changes in the constituents and antioxidant activity in accordance with the processing conditions by the salt water of Citrus unshiu Markovich. Changes in antioxidant activity was measured by the DPPH method and changes in the components were analyzed by high performance liquid chromatography. As a result, the content of the main constituents (narirutin, hesperidin) and total polyphenol were increased by increasing the concentration of salt water. Moreover, antioxidant activity was increased gradually in proportion to the increase of the total polyphenol content.

  • 감귤 부위별 및 감귤 주스 가공 후 폐기되는 착즙박을 배지로 하여 재배된 간, 표고, 새송이, 산호침, 참부채 및 영지 버섯) 균사체의 GC/MS에 의한 휘발성 물질을 분석하였다. 감귤 분말에서 휘발성 물질은 29종류이었는데, ${\delta}-elemene$, ${\beta}-elemene$, ${\beta}-caryophyllene$, caryophyllene, ${\gamma}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 감귤류의 essential oil 중의 성분들이었으나 8-hydroxy-linalool, ${\beta}-selinene$, tetradecanoic acid 및 pentadecanal은 essential oil 중의 성분들이 변화한 것이었다. 이외에도 tangeretin, nobiletin, 및 stigmasterol이 검출되었다. 버섯 균사체 분말에서 검출된 휘발성 성분들은 18종이었으며, ${\beta}-elemene$, germacrene-D 및 ${\delta}-cadinene$은 버섯이나 균사체에서 보고되지 않은 것으로 보아 균사체 배지로 사용한 착즙박에서 유래한 것으로 생각된다. caryophyllene, hexadecanoic acid, decanoic acid 및 tetradecanoic acid는 기 보고된 버섯이나 균사체의 성분들이었으며, Limonene-1,2-epoxide, ethyllinoleate, 1,2-benzenedicarboxylic acid, dicyclohexyl ester 및 ($3{\beta}$,24S)-stigmast-5-en-3-ol (${\gamma}-sitosterol$)은 감귤이나 버섯 균사체에서 보고되지 않은 성분들이 검출되었다.

Contents of Polyphenols and Limonoids in Citron (Citrus junos Sieb. ex Tanaka) Seed Extracts and Their Antioxidant Properties

  • Contents of phytochemicals in citron (Citrus junos Sieb. ex Tanaka) seeds and their effects on antioxidant activities were investigated. Methanol extract of defatted citron seeds contained the highest amounts of total polyphenols followed by 70% ethanol and water extracts. Neohesperidin was the most predominant citrus flavanones in these extracts. The highest amounts of limonoids were found in methanol extract, and this extract was the most efficient in scavenging both DPPH and ABTS radicals. All 3 extracts also exhibited good antioxidant activities against attack of linoleate free radicals on $\beta$-carotene. When methanol extract was sequentially fractionated into ether, ethyl acetate, n-butanol, and water fractions, butanol fraction contained the highest amounts of polyphenols otherwise most of limonoids were concentrated in ethyl acetate fraction. A positive relationship between radical scavenging activities and total polyphenol contents in fractions was observed while antioxidant activity on $\beta$-carotene seemed more related with contents of limonoids and other hydrophobic polyphenols.

감귤류 종자 추출물의 향균활성 (Antimicrobial Activity of Extracts from Citrus Seeds)

  • To develope natural food preservatives antimicrobial effect of the natural products against food-related bacteria and yeast The purpose of this study was evaluate antimicrobial effect of the citrus seeds. antimicrobial activities of methanol extracts from the citrus seeds investigate against Escherichia coli O26, Staphylococcus aureus 6358, Saccharomyces cerevisiae IBM 4274, Bacillus licheuiformis 9945a and Alcoligenes faecalis. Citrus seeds is containing to moisture 4∼6.5%, curd protein 11∼15%, curd lipid 32∼46%, curd carbohydrate 22∼45 % and ash 2∼4 %, that is containing to flavornoid 12∼48mg% and phenolic compound 22∼53mg%. Solidity content of the methanol extract from the citrus seeds was 0.8∼1.2%. Almost all of the methanol extracts from citrus seeds exhibited growth inhibiting activities for most of microorganisms tested. The methanol extracts from Citrus grandis, C. sunki, C. sulcata showed the growth inhibitory effects against Escherichia coli O26. The methanol extracts from C. obovoidea, C. sulcata, C. aurantium showed the growth inhibitory effects against Staphylococcus aureus 6358. The methanol extracts from C. obovoidea, C. sulcata, C. tangerina showed the growth inhibitory effects against Saccharomyces cerevisiae IBM 4274. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. tangerinan, C. natsudaidai, C. iyo, C. aurantium showed the growth inhibitory effects against Bacillus licheuiformis 9945a. The methanol extracts from C. obovoidea, C sunki, C. sulcata, C. aurantium showed the growth inhibitory effects against Alcoligenes faecalis. Among this especially, Showed growth inhibiting activity of the methanol extracts from Citrus sulcata that about microorganisms investigated. If apply searching suitable application method about such the citrus seeds antimicrobial activity, role as good antimicorbial material in storage or cooking of food, processing is expected.

가공용 감귤 신품종 하밀감 과피 추출물의 항산화 활성 (Antioxidative Activities of New Citrus Hybrid 'Hamilgam' Peel Extracts)

  • The major objective of this study was to investigate the usability as cultivar for processing of new Citrus hybrid 'Hamilgam'. We investigated various antioxidant activities, such as DPPH radical, ABTS radical, Hydroxyl radical and Superoxide anion radical effect, along with total polyphenol content and total flavonoid content of Citrus hybrid 'Hamilgam', C. unshiu, C. natsudaidai. The total polyphenol content and total flavonoid content were the highest in the Hamilgam. The major flavonoids of Hamilgam and Natsudaidai were naringin and neohesperidin, and those of Unshu were narirutin and hesperidin, as determined by HPLC. Especially, the neohesperidin content of Hamilgam showed much higher (100 times over) than that of Unshu. However, the aglycone form was not found. DPPH and ABTS radical scavenging activities were the highest in Hamilgam peels. Hydroxyl radical scavenging activity was high in the order of Unshu, Hamilgam and Natsudaidai. Superoxide anion radical scavenging of 3 cultivars peels displayed low activities compared to DPPH and ABTS radical scavenging. Based on these results, Hamilgam peel extracts possess antioxidant activities and may thus serve as potential sources of functional food, cosmetic products, etc.

칼라 영상처리에 의한 과일분류시스템 (Classification system of fruits by color image processing)

  • 일반적으로 농산물의 품질은 무게나 크기 등을 직접 측정하여 판정하거나 간접 또는 비파괴방법을 이용하여 판정하게 된다. 본 논문에서는 비파괴를 이용한 판정방법인 칼라 영상처리를 이용하여 농산물의 품질과 등급을 결정하는 알고리즘을 개발하여, 이를 실시간 귤 분류시스템에 적용하였다. 개발된 시스템은 귤의 크기와 색상을 칼라 영상처리 하여 초당 최대 6개를 실시간 분류 할 수 있는 성능을 가지고 있다. 또한 다른 농산물의 품질판정을 위한 제어기를 설계할 경우에도 본 논문에서 개발한 알고리즘이 적용될 수 있을 것으로 기대된다.

감마선 이용 미생물학적 및 관능적 품질이 우수한 아이스크림의 개발 (Development of Ice Cream with Improved Microbiological Safety and Acceptable Organoleptic Quality Using Irradiation)

  • 감마선을 이용하여 미생물학적으로 안전하면서 관능적 품질이 우수한 아이스크림을 개발하기 위해 감마선에 안정적인 spearmint, mint 및 citrus향을 선발하고 이를 이용하여 감마선 조사에 의한 특유의 냄새변화가 없는 아이스크림을 제조한 후 품질을 비교하였다. 일반적인 조성은 향 무첨가 대조구와 처리구 사이에 차이가 없었으며, 총균수는 향 무첨가 대조구, spearmint, mint 및 citrus향 첨가구가 각각 2.38, 1.23, 1.38 및 1.15 log CFU/g 수준에서 검출되었다. 그러나 대조구는 3 kGy, 그 외 처리구는 1 kGy 선량에서 균이 검출되지 않았다. 관능검사 결과 mint향을 첨가하고 1 kGy 감마선 처리한 아이스크림과 citrus향을 첨가하고 3 kGy에서 감마선 처리한 아이스크림이 유의적으로 높은 기호도를 보였다. 따라서 3 kGy 이하의 감마선 조사와 mint 또는 citrus향의 선택적 첨가는 관능적으로 문제가 없으면서 유아, 노약자, 환자 등 민감한 소비자들에 대한 미생물학적 안전성을 확보하는 기술로 사용될 수 있을 것으로 판단된다.

Aspergillus niger로 발효한 감귤과피 추출물의 항염효과 (Anti-inflammatory Effect of Citrus unshiu Peels Fermented with Aspergillus niger)

  • 본 연구에서는 미생물을 이용하여 감귤과피 발효를 실시하고, 발효 추출물의 생리활성 변화를 확인하였다. 분말화시킨 온주밀감 과피에 Aspergillus niger를 첨가하고 5일간 진탕 배양하고 감귤발효물을 ethyl acetate로 추출하였다. 대조구는 발효 전 감귤과피 추출물을 사용하였으며, 각 추출물의 성분변화는 HPLC를 이용하여 분석하였다. 그 결과, neohesperidin 함량이 현저히 증가하면서, 발효에 의해 compound 1, 2가 생합성되었다. 감귤발효 추출물의 항염 활성은 LPS로 염증을 유발시킨 RAW 264.7 macrophage cell에서 확인하였다. 감귤발효 추출물이 대조군에 비해 NO (Nitric oxide) 생성이 유의적으로 감소하였으며, 염증반응과 관련된 단백질 iNOS와 COX-2 의 발현도 감귤발효 추출물에서 유의적으로 감소하였다. 감귤발효 추출물은 pro-inflammatory cytokine인 TNF-${\alpha}$와 IL-6 생성을 억제하는 효과도 나타내었다. 본 연구 결과를 통하여 감귤 과피의 flavonoid 배당체 화합물들을 aglycone 형태로 전환함과 동시에 유효 생리활성 성분의 효능을 증대시켜 부산물로서 발생되는 감귤의 새로운 활용 방안으로 기대된다.

Simultaneous Determination of Flavanone Glycosides in the Fruit of Citrus paradisi and C. grandis by HPLC-PDA

  • An HPLC (high-performance liquid chromatography)-PDA (photodiode array) detection method was established for the determination of naringin, hesperidin and neohesperidin in the fruit of Citrus paradisi and C. grandis. The flavonoids were separated in less than 20 min using an YMC RP 18 column with isocratic elution using acetonitrile and water (23 : 77, v/v) at a flow rate of 1 ml/min, and a PDA detector. The levels of naringin, hesperidin and neohesperidin were 1345.92, 950.62, and 2078.82 ${\mu}g/g$, respectively, in the peel, and 102.43, 59.13, and 86.68 ${\mu}g/g$, respectively, in the flesh of C. paradisi. In C. grandis, the levels of naringin, hesperidin and neohesperidin were 3530.56, 80.00, and 5.26 ${\mu}g/g$, respectively, in the peel, and 59.59, 7.43, and 38.41 ${\mu}g/g$, respectively, in the flesh. The total content was highest in the peel, reaching 0.44% and 0.36% in C. paradisi and C. grandis, respectively, while the flesh contained only 0.025% and 0.011%, respectively. Therefore, the peels of C. paradisi and C. grandis are necessary for the processing and utilization of flavonoids.

제주산 온주밀감의 품질평가에 미치는 요인 (Factors Affecting on Quality Evaluation of Citrus unshiu Produced in Cheju)

  • Soluble solids($^{\circ}$Brix), acid content and edible part ratio of Citrus unshiu collected at sorting Places in south Cheju area wee more lowered, and peel thickness was more thickened with increasing fruit size. Compared to early variety of Citurs unshiu(C. unshiu Miyakawa Mar), peel thickness of medium variety of Citurs unshiu(C. unshiu Hayashi Mar) was more thickened, and edible part ratio was more lowered with increasing fruit size. Soluble solids of medium variety of Citurs unshiu were lowered, and acid contents were increased compared to early variety. Processing properties for pressing concentrated juice was good for early variety of Citurs unshiu, especially on fruit diameter of 50-65mm. Soluble solids, acid contents and juice ratio were decreased with increasing peel thickness. The quality properties for fresh fruit with peel thickness had a good correlation on early variety of Citurs unshiu, and these data are supposed to be applied to the quality evaluation of Citurs unshiu produced in Cheju.
