Comparison on phosphate solubilization ability of Pantoea rodasil and Burkholderia stabilis isolated from button mushroom media (양송이배지로부터 분리한 Pantoea rodasil 와 Burkholderia stabilis의 인산가용화능 비교)

  • This study investigated the synergistic effect of single inoculation and co-inoculation of phosphate-solubilizing bacteria (PSB) Pantoea rodasil LH-1and Burkholderia stabilis HS-7. Phosphate-solubilizing abilities of these strains were assessed by measuring phosphorus content in culture media that were singly inoculated or co-inoculated with these strains for 7 days. The co-inoculation of P. rodasil LH-1and B. stabilis HS-7 was found to release the highest content of soluble phosphorus ($783.41{\mu}g\;mL^{-1}$) into the medium, followed by single inoculation of B. stabilis HS-7B ($743.90{\mu}g\;mL^{-1}$) and P. rodasil LH-1 ($736.59{\mu}g\;mL^{-1}$). The highest pH reduction, organic acid production, and glucose consumption were also observed in the medium inoculated with both the strains, compared with that in the medium inoculated with the strain alone. Results of a plant growth promotion bioassay showed 7.7% and 15.5% higher leaf and root growth, respectively, in romaine lettuce co-inoculated with P. rodasil LH-1and B. stabilis HS-7 than those inoculated with the strain alone. However, no significant difference was observed between single inoculation and co-inoculation of these strains with respect to phosphorus release and plant growth. Although the results of the present study did remarkdly not show the synergistic effect of phosphate solubilization by co-inoculation of the PSB strains examined, these results indicate that treatment with PSB exerts a beneficial effect on crop growth.

  • The study was aimed to investigate the mixing ratio of kiwi and persimmon juices for the production of good quality wine by Saccharomyces cerevisiae Y28. Firstly, the optimum condition of rapidase treatment for the kiwi and persimmon juices was established, thereafter various mixing ratio (10:0, 9:1, 8:2, 7:3, 6:4, 5:5) of kiwi and persimmon was investigated regarding physiochemical properties and flavor compounds of wine. As the result, the optimum conditions were obtained as 0.3% rapidase for 1 h in kiwi and 0.3% rapidase for 3 h in persimmon. According to higher ration of persimmon, the pH of wines increased from 3.69 to 3.77, while the acidity of wines decreased from 2.07% to 1.51% at 14 days fermentation. The ranges of brix and reducing sugar in wines were decreased which ranges around 9.6 to 8.8 and 6.07 to 6.90 g/L, respectively, after fermentation. Major organic acid in wines were identified as tartaric acid, malic acid, and citric acid. A small amount of free sugar such as sucrose and glucose were detected in wines, but fructose was completely absent. The soluble phenolic contents were decreased that ranges around 1.00 to 1.25 g/L, in contrast, browning degree were increased ranges around 0.212 to 0.412 after fermentation. The major flavor components were identified as ethyl acetate and hydrazine, and 1,1-dimethyl. Importantly, phenylethyl alcohol was detected from the all wines that have a typical rose like flavor. But sensory test results and preference of kiwi-persimmon (7:3) mixing wine was better than the other wines.

Flavor Improvement of Chungkookjang by Addition of Yucca (Yucca shidigera) Extract (유카(Yucca shidigera) 추출물 첨가에 의한 청국장의 풍미 개선에 관한 연구)

  • This study was carried out to investigate the effect of yucca extract on the flavor development in Chungkookjang fermented by Bacrillus sp. b01. The changes in the contents of amino-type N, ammonia type N, volatile compounds and organic acids, and those in the activities of ${\alpha}-amylase$ and protease were also determined with aging period. The amount of amino-type N increased gradually with time and was slightly higher in Chungkookjang containing yucca extract than in control. The content of ammonia-type N in Chungkookjang decreased by the addition of yucca extract. During aging, a little higher amylase activity was detected in Chungkookjang containing yucca extract. However, the amylase activity was the lowest in Chungkookjang containing 1 mg/g of yucca extract. The protease activity was slightly higher in Chungkookjang containing yucca extract. The organic acids, such as citrate, acetate, malate, fumarate, and succinate, were detected. The yucca extract reduced the production of tetramethylpyrazine which was responsible for the unpleasant odor, but enhanced the production of 2,5-dimethylpyrazine and 2,6-dimethylpyrazine which contributed to the taste. Sensory evaluation showed that the addition of yucca extract of 0.5 mg/g significantly improved the flavor and taste of products.

Effect of Red Pepper Varieties on the Microflora, Enzyme Activities and Taste Components of Traditional Kochujang during Fermentation (고추품종을 달리한 전통식 고추장의 숙성중 미생물, 효소활성 및 맛성분의 변화)

  • Microbial counts, enzyme activities and taste components of traditional kochujangs prepared with the powders from 4 different varieties of red pepper, were investigated during 90 days fermentation for the industrial production of traditional kochujang. The viable cell counts of anaerobic bacteria in kochujangs did not change remarkably during fermentation, however, aerobic bacterial counts showed a rapid increase up to 90 days of aging. The yeasts in all kochujang samples increased until 60 days of aging and than decreased. After 90 days of aging, the count of aerobic bacteria in Kumtop kochujang was higher than those of others. The activities of liquefying amylase decreased during the aging, but those of saccharogenic amylase increased at 60 days of aging. The activities of neutral protease were higher than those of acidic protease, and increased during the middle and last period of aging. The major free sugars of kochujang were maltose and glucose, and their contents were higher in Hongkwang kochujang. The major organic acids of kochujang were succinic, formic and citric acid, followed by lactic acid. Succinic acid content in kochujang decreased during fermentation, whereas formic and citirc acids were increased. The major free amino acids were serine, proline, glutamic acid, aspartic acid and alanine. Kumtop kochujang contained the highest amount of total free amino acids. Among the nucleotides and related components in kochujang, cytidine-5-monophosphate was the most abundant component at the begining of aging period, while hypoxanthine increased remarkably during fermentation. Hanwang kochujang was higher in the content of nucleotides than others. Capsaicin contents of kochujang decreased during aging and those of Jangter kochujang was higher than that of others. Sensory evaluation showed that Jangter kochujang was significantly better than Kumtop kochujang in overall acceptability, but there were no appreciable differences in color and flavor.

On the Mushroom Cultivation of Oak (Quercus) Chip and Used Culture Medium of Lentinus into Feedstuff (참나무류(類) 칩을 이용(利用)한 표고버섯재배(栽培)와 폐잔사(廢殘渣) 사료화(飼料化))

  • To increase both the production of mushrooms and the usages of the residues (used media) for roughage, the pinchips(PC) and sawdusts(SD) of three Korean oak species were tested as cultivating media for Lentinus edodes with varing the composition of the media. The results are as follows. 1. Organic acids(tannic acid and citric acid), which were added in the PC medium to enrich the quality of mushrooms, did not increase the growths of the mycelia and the quality. 2. The mushrooms were able to be harvested from six months after beginning the cultivation. No significant differences were found among the qualities of the mushrooms produced by various treatments. 3. The contents of crude proteins(1.82-4.55%) in the used media were higher than those in woods (c.a. 0.7%), and the total digestible nutrients (44.0-46.0%) in the used medium were not much different from those in rice straws (c.a.48.0%). 4. The contents of some essential amino acids (methionine in the used medium of the tannic acid added PC or S.D. and isoleucine, phenylalanine and lysine in the used medium of the untreated PC or SD) were higher than those in the control (raw media). The contents of a few other essential amino acids (threonine, valine and arginine) were lower in the used media. Most nonessential amino acids in the used media, particularly in the used bed-log, appeared to decrease than those in the control.

Effect of Feeding Strategies on Milk Production of Holstein Dairy Cows Managed by Small-Farmers Alpine Grassland in Korea

  • The main objective of this experiment was to investigate effects of two different feeding systems on body weight, milk yield, milk composition, and mineral and fatty acids content of Holstein dairy cows' milk. Sixteen of 25 months-old Holstein dairy cows were assigned to two groups (n=8) to study effects of the feeding system for 150 days. Two feeding systems were compared for five months; Group 1 was housed indoors and mainly fed a concentrate diet, Group 2 was maintained outdoors for five-seven hours/day on various kinds grass in a pasture. The experiment was conducted June-October 2017. Results revealed the indoor-fed cows had higher body weight, that was significant compared with the outdoor-based feeding system of Holstein dairy cows (p<0.05). Indoor-raised milking cows had higher milk yield (32.45 kg) as compared with pasture-raised milk yield (26.44 kg). Cows fed indoors significantly increased milk yield, total protein content, lactose, citric acid level, and lowered level of total solid and free fatty acids relative to the pasture-fed milking cows (p>0.05). There were higher levels of mineral content and fatty acid content in the milk of indoor-fed dairy cows than the pasture-raised dairy cows (p>0.05). Our study results demonstrated the potential benefits of the indoor feeding system for increased body weight, milk yield, mineral and fatty acids content summer through autumn when low pasture growth rates and quality may otherwise limit production.

Effect of 1-methylcyclopropene and ethylene-absorbent treatments on quality changes of Prunus mume fruit during storage (1-MCP 처리 및 에틸렌 흡착제가 '백가하' 매실 저장에 미치는 영향)

  • This study was conducted to investigate the effect of 1-methylcyclopropene (1-MCP) and ethylene-absorbent on the qualities of Prunus mume fruit. Prunus mume fruits were stored without film packaging (Cont), packed in LDPE film (Cont-P), and packed with ethylene-absorbent (Cont-PE). Fruits were treated with 1-MCP (1 ppm) for 24 hr at $1^{\circ}C$. After treatment, fruits were packed in LDPE film (MCP-P) and with ethylene-absorbent (MCP-PE) and then stored at $1^{\circ}C$ for 8 weeks. Total soluble solids increased during storage but decreased after 6 weeks while total acidity decreased during storage. Cont was almost completely decayed after 8 weeks of storage while Cont-P, Cont-PE, MCP-PE, and MCP-P were 46, 69, 83, and 5% decayed, respectively. L value decreased but a value increased during storage in all samples. Firmness of peel and flesh of samples decreased gradually for 8 weeks. Respiration rate did not show any significant difference among samples. Ethylene production of Cont showed $0.05{\mu}L/kg/h$ but immediately after 1-MCP treatment, it showed $0.02{\mu}L/kg/h$. Oxalic and malic acids decreased while citric acid increased during storage; fructose and glucose substantially decreased after 8 weeks whereas sorbitol and sucrose increased upto 4 weeks and then decreased thereafter. Based on these results, packing the fruits treated with 1-MCP could extend the freshness of Prunus mume fruit.

Effect of Cholesterol-loaded-cyclodextrin in Presence and Absence of Egg Yolk during Freezing Step on Quality of Markhoz Buck's Spermatozoa

  • Cryopreservation protocols induce partially irreversible damage to mammalian sperm plasma membranes. Previous studies have indicated that adding cholesterol to the plasma membrane, as cholesterol-loaded-cyclodextrins, improves cryosurvival of sperm. Therefore, the purpose of this study was to determine if treating sperm of Markhoz bucks with cholesterol-loaded-cyclodextrins (CLC) (0, 0.75, 1.5, 2.25 and 3 mg/ml diluted $240{\times}10^6$ sperm/ml) in Tris-citric acid-glucose diluents with and without egg yolk (containing 5% glycerol) would improve the post-thaw sperm quality. The motion characteristics were evaluated with a Computer Assisted System Analyzer (CASA); acrosome integrity and vitality were measured with the triple-stain technique. Samples were recovered before and after freezing by means of putting straws into $37^{\circ}C$ water for 30 sec and then parameters were assessed. The results showed that the treatments significantly affected motility, progressive motility, recovery rate, curvilinear velocity, beat cross frequency, live sperm with reacted acrosome, live sperm with unreacted acrosome, dead sperm with reacted acrosorne, and dead sperm with unreacted acrosome during freezing (p<0.05). However; no significant differences were found for average path velocity, straight line velocity, amplitude of lateral head displacement, straightness and linearity (p>0.05). The best results were observed for extender containing 2.25 mg/ml ($240{\times}10^6$ sperm/ml) CLC supplemented with 2.6% egg yolk. In conclusion, the findings of this study indicate improved Markhoz sperm viability and motility following treatment in the presence of egg yolk.

Seasonal and Regional Effects on Milk Composition of Dairy Cows in South Korea

  • For a period of over 6 years, more than 160,000 milk samples were collected and analyzed to determine the influence of different seasonal temperatures and geographic regional location on milk composition in South Korea. Fat, protein, lactose, non fat milk solids (NFMS) and total solids (TS) contents were significantly higher among dairy cows milked in winter season than other seasons (p<0.05). In contrast, freezing point (FP), milk urea nitrogen (MUN) and somatic cell count (SCC) were significantly higher in summer season than other seasons (p<0.05). The average SCC in the autumn season was $358{\times}10^3$/ml, which was lower than any other seasons (p<0.05). These results may be due to the changes in temperature during different seasons. Meanwhile, milk produced by dairy cows in central region had higher fat, protein, lactose, NFMS, TS and MUN and had lower SCC compared to other regions (p<0.05). Fat, TS, FP, MUN and citric acid in northeast region were lower than other regions (p<0.05). The SCC was significantly higher in southeast region than those of other regions (p<0.05). As a result, it might be possible that the differences in feeding management in each different region may affect the milk composition. In conclusion, present results indicated that milk composition is clearly influenced by both season and regional location. Therefore, based on these results, development of different feeding systems, according to season and region is needed to produce high quality and satiable milk production.

Glycolysis Mediated Sarcoplasmic Reticulum Ca2+ Signal Regulates Mitochondria Ca2+ during Skeletal Muscle Contraction (근수축시 해당작용에 의한 근형질 세망의 Ca2+ 변화가 미토콘드리아 Ca2+ 증가에 미치는 영향)

  • PURPOSE: This study was to investigate the Glycolysis mediated sarcoplasmic reticulum (SR) $Ca^{2+}$ signal regulates mitochondria $Ca^{2+}$ during skeletal muscle contraction by using glycolysis inhibitor. METHODS: To examine the effect of Glycolysis inhibitor on SR and mitochondria $Ca^{2+}$ content, we used skeletal muscle fiber from gastrocnemius muscle. 2-deoxy glucose and 3-bromo pyruvate used as glycolysis inhibitor, it applied to electrically stimulated muscle contraction experiment. Intracellular $Ca^{2+}$ content, SR, mitochondria $Ca^{2+}$ level and mitochondria membrane potential (MMP) was detected by confocal microscope. Mitochondrial energy metabolism related enzyme, citric acid synthase activity also examined for mitochondrial function during the muscle contraction. RESULTS: Treatment of 2-DG and 3BP decreased the muscle contraction induced SR $Ca^{2+}$ increase however the mitochondria $Ca^{2+}$ level was increased by treatment of inhibitors and showed and overloading as compared with the control group. Glycolysis inhibitor and thapsigargin treatment showed a significant decrease in MPP of skeletal muscle cells compared to the control group. CS activity significantly decreased after pretreatment of glycolysis inhibitor during skeletal muscle contraction. These results suggest that regulation of mitochondrial $Ca^{2+}$ levels by glycolysis is an important factor in mitochondrial energy production during skeletal muscle contraction CONCLUSIONS: These results suggest that mitochondria $Ca^{2+}$ level can be regulated by SR $Ca^{2+}$ level and glycolytic regulation of intraocular $Ca^{2+}$ signal play pivotal role in regulation of mitochondria energy metabolism during the muscle contraction.
