Effect of Various Biodegradable Chelating Agents on Root Growth of Plants under Mercury Stress (생분해되는 다양한 킬레이트들이 수은에 노출된 식물의 뿌리성장에 미치는 영향)

  • Phytoextraction is a technique that uses plants to remediate metal-contaminated soils. However, this technique cannot be applied in highly metal-contaminated areas, as plants cannot normally grow under such conditions. Therefore, this study investigated the introduction of various biodegradable chelating agents to increase the bioavailability of metals in highly metal-contaminated areas. Mercury (Hg) was selected as the target metal, while cysteine (Cys), histidine (His), malate, succinate, oxalate, citrate, and ethylenediamine (EDA) were used as biodegradable chelating agents. Plants were grown on agar media containing various chelating agents and Hg to analyze the effect on plant root growth. Cys and EDA were both found to diminish the inhibitory effect of Hg on plant root growth, whereas His, citrate, and ethylenediamine tetraacetic acid (EDTA) did not show any significant effects, and malate, succinate, and oxalate even promoted the inhibitory effect of Hg on plant root growth. Thus, Cys and EDA would seem to be promising biodegradable chelating agents for highly Hg-contaminated areas.

  • Deliveries of tar, nicotine and carbon monoxide in mainstream smoke decreased with increment of exogenous potassium content of tobacco. Potassium malate was more effective than potassium nitrate, potassium carbonate, potassium citrate and potassium oxalate for the reduction of carbon monoxide, but potassium nitrate was more effective than potassium palate, potassium carbonat, potassium citrate and potassium oxalate for the reduction of carbon in the case of nicotine. Puff count and burning rate were slightly changed with added potassium salts.

  • Using the synchronous culture method and the manometric technique, changes in respiratory activities, utilization of some organic acids (succinate, malate, lactate and acetate etc.) and its effect on glucose metabolism in Chlorella cells at different growing stages were measured. 1) Endogenous respiration of the cells was not active at growing stage and was almost constant throughout the early ripening, maturing and division stages. 2) Lactate was utilized as respiratory substrate better than other organic acids tested. Exogenous respiration of glucose was most active at growing and maturing stages and was decreased strikingly at division stage. 3) Succinate and citrate inhibited endogenous and glucose respiration of the cells throughout the all life cycle. 4) Malate and acetate were utilized in the cells at early growing and division stages better, and malate enhanced the glucose respiration while in case of acetate it was depressed. 5) Calcium ion inhibited not only permeability of respiratory substrate but endogenous respiration itself.

  • The utilization of acetate by Dunaliella tertiolecta was examined, and the detections and assays of the enzymes of the tricarboxylic acid cycle and the glyoxylate pathway were described. Acetate could not be utilized as a sole carbon source for the growth. The carboxyl carbon of acetate was incorporated more rapidly into CO2 than the methyl carbon. It was identified that malate, succinate, citrate and etc., were accumulated whne [U-14C] acetate was supplied to the cell free homogenate. The following enzyme activities were measured; acetothiokinase, isocitrate dehydrogenase, fumarase, malate dehydrogenase and aconitase. Though isocitratase, malate synthetase, succinate dehydrogenase and oxoglutarate dehydrogenase could not be detected, 14C from succinate was easily contributed to CO2 and cell component. The evidence suggested that the glyoxylate pathway was not operative and showed that the TCA cycle was the all important pathway in the oxidation of acetate to CO2 in Dunaliella.

Lead (Pb) as a target metal and cysteine (Cys), histidine (His), citrate, malate, oxalate, succinate, and ethylenediamine (EDA) as biodegradable chelating agents were selected. Ethylenediamine tetraacetic acid (EDTA) was used as a comparative standard. Plants were grown on agar media containing various chelating agents with Pb to analyze the effect on root growth. Cys strongly increased the inhibitory effect of Pb on root growth of plants, while, His did not affect on it significantly. The inhibitory effect of oxalate is weak, and malate, citrate, and succinate did not show significant effects. Both EDTA and EDA diminished the inhibitory effect of Pb on root growth. The effect of EDA is correlated with decreased Pb uptake into the plants. In conclusion, as biodegradable chelating agents, EDA is a good candidate for highly Pb-contaminated area.

Ethylenediamine as a Promising and Biodegradable Chelating Agent in Growth of Plant Under Zinc Stress (아연 스트레스를 받는 식물의 성장을 위한 생분해되는 킬레이트로서 에틸렌디아민)

  • Zinc (Zn) is an essential element required for growth and development of plants. However, Zn can be toxic to plants when it presents excessive amount. Phytoextraction is an economic and environment-friendly technique using plants to clean-up metal-contaminated soils. However, the technique cannot be applied in highly metal-contaminated areas because plants will not normally grow in such conditions. Therefore, this research focuses on identifying chelating agents which are biodegradable and applicable to highly metalcontaminated areas.   • A 150-fold purified preparation of NADPH-specific glutamate dehydrogenase of Corynebacterium glutamicum (1) was used for the determination of kinetic parameters of the substrates, NADPH, NH$_4$Cl, and $\alpha$-ketoglutarate in the direction of glutamate synthesis. The kinetic constants determined from this study suggest a biosynthetic role for the enzyme, Based on the analysis of the result derived from initial velocity, the reaction mechanism was postulated to be ordered addition with NADPH as a first substrate to bind in the forward direction. Of the several metabolites tested for a possible function in the regulation of glutamate dehydrogenase activity, only malate and citrate were appeared to have an appreciable influence on the enzyme, Potassium chloride showed to be the most effective for the enzyme activity.

Studies on the Mineral Content of Edible Mushrooms (식용 버섯류의 무기물 함량)

  • To investigate on the trace element content of twelve edible mushrooms and Aloe arborescens, i. e., Lentinus edodes, Ganoderma lucidum (culturing in wood and soil), Tricholoma matsutake, Agaricus bisporus, Cyrophora esculenta, Auricularia auricula-Jude (produced in Korea and China), Sarcodon asparatus, Pleurotus ostreatus, Coriolus versicolor, Smilax rotundifolia and Aloe arborescerts were analyzed by Atomic absorption spectrometer. The obtained results were summerized as follows: 1. Potassium, sodium, magnesium and iron content for the most part samples were in large quantities, especially phosphorus content of those was highest ammount for the all samples. 2. Sodium content was much ammount in the Lentinus edodes (39mg) and Ganoderma lucidurn (20 mg), Culturing in wood and soil, while potassium was very high ammount in the Aloe arborescens and other samples. Mush ammount of magnesium as compared with others was Lentinus edodes (144mg), Ganoderma lucidurn (128mg), Aloe arborescerts (50mg) and pleurotus ostreatus (60mg). Phosphorus content of Ganoderma lucidurn, Lentinus edodes, Gyrophora esculenta, Auricularia polytricha and Agaricus bisporus was much ammount while iron content of all samples equality higher ammount. Sodium content of Aloe arborescens was not analyzed out for almost all, its potassium (82mg), magnesium (50mg) and iron (18rng) content comparatively higher quentity than others minerals and phosphorus volume (4.9mg) as compared with others, was conspicuously lower detect. 4. Cadimium and lead content of harmful metal element were detected on trace quentity for the most part samples 5. Organic acids of samples i.e., Legtinus edodes, Agaricus bisporus, Pleurotus ostreatus and Ganoderma lucidum were Citrate, Malate, Fumalate, Succinate, Oxalate, Acetate, Lactate, and Tartarate and Citrate, Malate and Fumarate contents were higher amount remarkbly than other organic acids. Tartarate content was trace amount.

The Mitochondrial Tricarboxylate Carrier of Silver Eel: Chemical Modification by Sulfhydryl Reagents

  • The tricarboxylate (or citrate) carrier was purified from eel liver mitochondria and functionally reconstituted into liposomes. Incubation of the proteoliposomes with various sulfhydryl reagents led to inhibition of the reconstituted citrate transport activity. Preincubation of the proteoliposomes with reversible SH reagents, such as mercurials and methanethiosulfonates, protected the eel liver tricarboxylate carrier against inactivation by the irreversible reagent N-(1-pyrenyl)maleimide (PM). Citrate and L-malate, two substrates of the tricarboxylate carrier, protected the protein against inactivation by sulfhydryl reagents and decreased the fluorescent PM bound to the purified protein. These results suggest that the eel liver tricarboxylate carrier requires a single population of free cysteine(s) in order to manifest catalytic activity. The reactive cysteine(s) is most probably located at or near the substrate binding site of the carrier protein.

Preparation Conditions of Agar Oligosaccharides with Organic Acids (한천 올리고당 제조를 위한 유기산 처리 조건)

  • The optimum organic acid and temperature conditions were investigated for the preparation of oligosaccharides from agar. The tested organic acids were acetate, citrate, lactate, malate, and succinate and the conditions for oligosaccharides preparation were $0.3\%,\;0.5%;and\;0.7\%$ organic acid concentrations at $80\~120^{\circ}C.$ The low concentration of organic acid below $0.3\%$ decreased the degrading ratio and the high concentration up $0.5\%$ could not changed the degrading ratio. Conditions below $100^{\circ}C$ was not good for degrading agar. But $100^{\circ}C\;or\;120^{\circ}C$ was optimal temperature conditions for agarooligosaccharides according to the organic acid type and concentration. The organic acid concentration was $0.5\%$ and organic acid was the citrate or malate. The treatment time considered optimum was 120$\~$180 min. The maximal degrading ratio giving optimum conditions such as $100^{\circ}C\;and\;120^{\circ}C\;was\;35.5\%\;and\;38.7\%,$ respectively. The agarooligosaccharides prepared by autoclaving at $100^{\circ}C\;and\;120^{\circ}C$ were 2$\~$7 species oligomer.
