Methanol Extract of Cinnamomum cassia Represses Cellular Proliferation and Gli-mediated Transcription in PANC-1 Human Pancreatic Cancer Cells

  • Twenty five methanolic plant extracts were investigated to determine the anticancer activity against sonic hedgehog (shh)/Gli signaling pathway dependent cancer, PANC-1 human pancreatic cancer cells, through three screening programs. All extracts were inspected their inhibitory properties on sonic hedgehog-conditioned medium (shh-CM) induced alkaline phosphatase (ALP) activity in C3H10T1/2 mouse mesenchymal stem cells to examine whether the plant extracts affect the shh/Gli signaling pathway. Next, plant extracts were screened the ability to suppress the cell proliferation of PANC-1 human pancreatic cancer cells. Finally, active plant extracts from the two screening systems were evaluated for the suppressive effect on Gli-mediated transcriptional activity in PANC-1 cells. Among active plants, Cinnamomum cassia suppressed Gli-mediated transcriptional activity leading to the down-regulated expression of Gli-target genes such as Gli-1 and Patched-1 (Ptch-1). This study provides the consideration for the important role of natural products in drug discovery process as well as the basis for the further analysis of active plant and potential identification of novel bioactive compounds as inhibitors of Gli and therapeutic candidates against shh/Gli signaling pathway dependent cancers.

  • Objectives: The prevalence of obesity and metabolic syndrome is increasing worldwide. Regulation of cellular energy metabolis has the potential to be manipulated therapeutically to serve as a target for obesity and insulin resistance. Skeletal muscle is regarded as a target for regulation of energy metabolism and insulin resistance. In this study, the authors investigated the regulatory effect of (Cinnamomum cassia Blume, CCB), Aconitum carmichaeli Debx (ACD), and Benth (Pueraria lobata Benth, PLB) on energy and glucose metabolism in C2C12 myotubes. Methods: The water extracts of CCB, ACD, and PLB (0.5 mg/ml) were treated in differentiated C2C12 myotubes. The expressions of adenosine monophosphate-activated protein kinase (AMPK) and phosphorylation AMPK were detected with western blotting. Glucose metabolism was investigated with glucose uptake assay and glucose consumption assay, total adenosine triphosphate (ATP) content was also analyzed. Results: CCB, ACD, and PLB activated the phophorylation of AMPK, they also increased the glucose metabolism and total ATP contents in C2C12 myotubes. Conclusions: This study suggests that CCB, ACD, and PLB have the potential to increase energy and glucose metabolism in skeletal muscle.

계피 에탄올 추출물의 유효성분 분석 및 항산화 효능 평가 (Antioxidant Potential of Cinnamomum cassia Ethanolic Extract: Identification Of Compounds)

  • Objectives : Natural products containing bioactive compounds with high antioxidant activity are potentially important sources that can contribute to the improvement of various diseases. Therefore, the aim of this study was to investigate phenolic compounds of Cinnamomum cassia (C. cassia) ethanolic extract (CCEE). And then we evaluated the antioxidant effect. Methods : We used liquid chromatography with tandem mass spectrometry (LC-MS/MS) to identify the compounds in CCEE. LC-MS/MS was performed in positive ion mode using Shimadzu, Nexera HPLC system and IDA TOF mass system. Solvent A was distilled water and solvent B was acetonitrile as mobile phase. The analysis was performed at a flow rate of 0.5 ml/min, column temperature of 35 ℃ and wavelength of 284 nm. The antioxidant effect of CCEE was analyzed using DPPH (2,2-diphenyl-2-picrylhydrazyl free radical) and ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)). In addition, total phenolics and total flavonoids contents were measured to determine antioxidant effects. Results : Analysis using LC-MS/MS identified four compounds: Coumarin, Trans-cinnamaldehyde, Trans-cinnamic acid, and 2-Methoxycinnamaldehyde. Free radicals decreased in a concentration-dependent manner starting from 10 ㎍/ml of CCEE, and decreased to a level similar to Ascorbic acid (AA) from a concentration of 60 ㎍/ml onwards. Conclusions : Based on the findings, CCEE exhibits strong antioxidant activity as evidenced by the presence of Coumarin, Trans-cinnamaldehyde, Trans-cinnamic acid, and 2-Methoxycinnamaldehyde. Consequently, this study suggests that CCEE can serve as an important source of natural antioxidants and can be efficiently used in the management of oxidative stress diseases.

Efficacy of Soil Amendment with Medicinal Plant Materials for the Control of Root-knot Nematode (Meloidogyne incognita) in Tomato

  • Soil amendments with oriental herbal medicines such as fruit of Anethum graveolens, flower buds of Syzygium aromaticum, rhizome of Cnidium officinale, rhizome of Coptis chinensis, root bark of Paeonia suffructicosa, stem bark of phellodendron amurense, and stem bark of Cinnamomum cassia at the rate of 0.2% (weight by volume of soil) significantly reduced Meloidogyne incognita infection (root gall formation) of tomato seedlings compared with the control. The most effective treatments were root bark of p. suffructicosa and stem bark of C. cassia as they gave minimum numbers of galls on tomato roots (4.7% and 8.9%, respectively, relative to control) as compared to other treatments. Another study with root bark of p. suffructicosa and C. cassia at different application doses also showed consistent results in reducing gall number. The control efficacy decreased as the application doses were lower-ed, indicating their dose-dependent control activities. These treatments significantly enhanced aboveground plant growths (total masses).

식물정유 7종의 항산화능 분석 및 Candida albicans 생장 억제 정유의 생리활성 성분 구명 (Antioxidant Properties of 7 Domestic Essential Oils and Identification of Physiologically Active Components of Essential Oils against Candida albicans)

  • 본 연구에서는 국내 자생식물 7종의 정유 중 우수한 항산화 효과가 있는 온주밀감, 육계나무 정유 2종을 선별하였고, 피부염을 유발하는 Candida albicans에 대한 항미생물 활성 평가를 통해 식물정유의 항미생물 활성성분을 구명하고자 하였다. 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거활성 측정 결과 SC50값이 온주밀감 정유 0.010 mg/mL, 육계나무 정유 0.09 mg/mL을 나타냈으며, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) 라디칼 소거활성 측정 결과 온주밀감 정유 0.09 mg/mL, 육계나무 정유 0.06 mg/mL로 높은 항산화 활성을 나타냈다. Minimum Inhibitory Concentration는 육계나무 정유 1.25 mg/mL, 온주밀감 정유 5 mg/mL 값으로 육계나무 정유에서 뛰어난 항미생물 활성이 나타났다. Thin layer chromatography (TLC) bioassay를 통해 2종 정유의 성분 분획에 따른 C. albicans에 대한 항미생물 활성을 평가하였고, preparative TLC (prep. TLC)를 통해 활성분획을 획득하고, 동일 Rf값을 가지는 성분들에 대해서 GC/MS 분석을 수행하여 항미생물 활성성분을 구명하였다. 그 결과 항산화 및 항미생물 활성을 나타내는 주성분은 각각 육계나무 정유의 경우 cinnamyl acetate, eucalyptol, linalool, citral 온주밀감 정유의 경우 linalool이었다. 2종 정유에 공통적으로 항산화 및 항미생물 활성을 나타낸 분획부 성분분석 결과 linalool은 항산화, cinnamyl acetate, eucalyptol, citral, geranyl acetate은 항산화와 항미생물 능력이 있다고 사료된다.

한약재 주정 추출물이 종자발아와 유묘생장에 미치는 영향 (Effect of Ethanol Extract of Herbal Medicine on Crop Growth Inhibitor)

  • BACKGROUND: Herbal extracts have been screened for their inhibitory effect of seed germination and root development on weeds, but there is a scarcity of reports for crop growth regulation. The objective of this research was to develop a growth inhibitor on Brassica campestris, and its effective extraction method from herbal medicine extract. METHODS AND RESULTS: Eighty four herbal medicine extracts were tested for their plant growth inhibition activity on B. campestris. The alcohol extracts of Artemisia annua, Cinnamomum cassia, and Mentha arvensis inhibited over 30% of germination and the extract of A. annua, and C. cassia inhibited over 70% of radicle growth at 0.1 % w/w treatment. The partially purified extracts of A. annua, and C. cassia with dichloromethane and hexane showed stronger radicle growth inhibition than the crude extracts on B. campestris. The diethyl ether extract of A. annua showed a similar 50% radicle growth inhibition ($RI_{50}$ = 45 mg/L) to its partially purified extract with dichloromethane or hexane, but the diethyl ether extract of C. cassia showed a worse $RI_{50}$ than the purified extract. CONCLUSION: The alcohol extracts of A. annua, and C. cassia showed potent radicle growth inhibition properties on B. campestris. Diethyl ether proved to be a good solvent for simple extraction from A. annua.

  • 216종의 생약을 대상으로 해서 bacterial 알파에 대해 저해 활성을 나타내는 생약을 선별하였으며, 2차 선별시험 결과 Areca catechu L.(빈랑) Cinnamomum cassia Presl. (계피) 및 Ephedra sinica Stapf(마황)가 비교적 강한 저해 활성을 나타내었다.

  • The effects of MeOH extracts from 28 herbal medicines on monoamine oxidase (MAO) activity were investigated. MAO was purified from mouse brain and its activity was determined by fluoro-photometry using kynuramine as a substrate. Three MeOH extracts, Coptis japonica, Cinnamomum cassia and Poncirus trifoliate from the herbal medicines showed a strong inhibitory effect with less than $100\;{\mu}g/ml$ in their inhibitory amounts of $50\%$ ($IC_{50}$ values) on MAO activity. Four MeOH extracts including Evodia officinalis exhibited a mild inhibition of MAO activity with $100-200\;{\mu}g/ml$ in their $IC_{50}$ values.

Scavenging Effect of Plant-Derived Materials on Free Radieals and Active Oxygen Species

  • The in vitro antioxidative activities of the 33 plant-derived essential oils and 37 phytochemicals including 3 Mentha arvensis leaf-, 2 Thymus vulgaris leaf- and 2 Syzygium aromaticum flower-derived isolates were determined by the inhibition of linoleic acid autoxidation, the generation of superoxide anion and scavenging of DPPH radical. They were then compared to those of the widely used plant-derived antioxidants (pyrogallol and quercetin) and synthetic antioxidant BHT. At a concentration of 0.01%, potent antioxidative effect was observed in the essential oils from Cinnamomum cassia roots, Mentha arvensis leaves, Ginkgo biloba fruits, and Syzygium aromaticum flowers. Of the phytochemicals used, eugenol and isoeugenol at 0.01% showed potent antioxidative activity, and their activities were comparable to those of pyrogallol, quercetin, and BHT. The Cinnamomum root-, the Mentha leaf-, the Ginkgo fruit-, and the Syzygium flower-derived materials may be a good source for an alternative to the currently used antioxidants.

Antioxidant activity of Cinnamomum cassia extract and quality of raw chicken patties added with C. cassia powder and Pleurotus sajor-caju powder as functional ingredients during storage

  • Objective: The aim of this study was to investigate antioxidant activities of cinnamon (Cinnamomum cassia) extracts (extracted with different solvents) at various concentrations and to determine product quality of raw chicken patties added with different levels of cinnamon powder (CP) and oyster mushroon powder (OMP) during storage. Methods: After cinnamon was made into oven dried CP and extracted with water and different levels (50%, 80%, and 100%) of ethanol, antioxidant activities of these extracts were determined. CP and OMP were combined at different levels and added to raw chicken patties. Physicochemical properties and microbial counts were measured during refrigerated storage. Results: Cinnamon ethanol (80%) extract showed the highest (p<0.05) by 2,2-diphenyl-1picrylhydrazyl-radical scavenging activity and reducing power. Cinnamon water extract (CWE) had the highest iron chelating ability (p<0.05), while CP 100% ethanol extract had the highest content of total phenolic compound. Then, CP and OMP were applied to chicken patties at different levels (0.1% to 0.2%). After the addition of CPs, pH, L* (lightness), 2-thiobarbituric acid reactive substance, and volatile basic nitrogen values were decreased, whereas a* (redness) and b* (yellowness) values were increased. Microbial counts of total bacteria and Enterobacteriaceace were decreased with the addition of CP 0.2% regardless of the OMP level. Conclusion: The addition of CP in combination with OMP can increase the shelf-life of chicken patties during storage.
