Effect of Coix on Plasma Cholesterol and Lipid Metabolism in Rats (율무쌀이 쥐의 혈장콜레스테롤 및 지질대사에 미치는 영향)

  • This study was done to determine the effects of coix on the content of cholesterol. lipids, phosphoilpids, total bile acids in liver, plasma, and feces of rats fed containing 1% cholesterol. Four groups of experimental diets, such as soybean oil-starch diet, soybean oil-coix diet, lard-starch diet, lard-coix diet were fed ad libitum to the 6 weeks old white male rat of Spargue-Dawley strain for 27 days. The results obtained are summarized as follows: 1) Plasma cholesterol level of the lard-starch group was significantly higher than the other groups (p<0.05), and that of the lard-coix group was the lowest. The content of plasma triglyceride, total bile acids was not different among the lard-starch group was significantly higher than the other groups(p<0.05). 2) The contents of the liver lipids, cholesterol, and triglycerides of the lard-coix group were significantly lower than the other groups(p<0.05). The contents of phospholipid of the liver of the lard-starch group was significantly lower than the other groups(p<0.05), and those of the soybean oil-coix group and the lard-coix group were high. 3) The contents of intestinal lipids, triglyceride, and phospholipid of the lard-coix group were significantly lower than the other groups(p<0.05). The contents of the intestinal tissue cholesterol of the lard-coix group was significantly lower than the other groups(p<0.05). 4) Cholesterol content of the feces was not different among the experimental groups. Triglyceride content of the feces of the soybean oil-starch group was significantly higher than the other groups, but total bile acid content of the feces was not different among the experimental groups. 5) Dry matter digestibility was not different among the experimental groups, but cholesterol retention was rather low when soybean oil-diet was fed. The results of this study reveales that coix has an effect to lower the plasma cholesterol level. Coix were decreased the content of liver cholesterol, increased the contend of liver phospholipid, and induced triglyceride excretion of the feces in the lard diet which lead to hypercholesterolemia. It could be suggested that the coix has influesced liped metabolsim in body.

Effects of Citrus Flavonoid, Hesperidin and Naringin on Lipid Metabolism in HepG2 Cells (간배양 HepG2 세포의 지질대사에 미치는 Hesperidin 및 Naringin의 영향)

  • The effects of citrus flavonoids, hesperidin and naringin, on the lipid metabolism were investigated in cultured human hepatocyte HePG2 cells. HepG2 cells were cultured for 6 h and 24 h to the control medium or the media containing hespridin and narigin, which concentrations were 0.5 and 5.0 mg/$m\ell$. There were no significant effects on cell proliferation and cellular protein content, except for increased in these parameters by adding both citrus flavonoids (0.5 mg/$m\ell$). The cellular content of triacylglycerol after 6 h incubation with 0.5 mg/$m\ell$ hesperidin and naringin was markedly increased, and after 24 h incubation that was decreased in both citrus flavonoids supplementation. The supplementation of 5.0 mg/$m\ell$ hesperidin caused a marked decrease in the cellular cholesterol content following 6 h incubation, and that was also reduced markdly, in a dose-dependent manner, during incubation for 24 h. However, there was no significant difference in the cellular cholesterol content in medium supplemented with naringin. The effect of hesperidin and naringin on acyl-CoA: cholesterol acyltransferase (ACAT) activity was studied in vivo and in vitro. The data confirmed that hesperidin inhibit ACAT activity in vivo and in vitro, whereas naringin had no such effect on ACAT activity in vivo but not in vitro. The present study suggests that hesperidin reduces the cellular triacyglycerol and cholesterol contents in human hepatocyte HepG2 cells.

Influence of Feeding β-Cyclodextrin to Laying Hens on the Egg Production and Cholesterol Content of Egg Yolk

  • The main objective of the present study was to determine the effect of dietary $\beta$-cyclodextrin ($\beta$-CD) on egg laying performance and cholesterol content of egg yolk. Feed intake, egg production and egg weight varied slightly, but not significantly, between hens fed either a control diet or a diet containing one of an increasing series of $\beta$-CD concentrations (2%, 4% or 6%). However, feed intake, egg production and egg weight were all lower in the hens fed on 8% $\beta$-CD (p<0.05). No difference in haugh unit values, egg yolk color and egg shell thickness were found between the different treatments. The cholesterol content of egg yolks (mg/g yolk) was significantly decreased by 0.71, 2.98, 4.00 and 4.24 mg in eggs from hens maintained on 2%, 4%, 6% and 8% $\beta$-CD, respectively (p<0.05). These observations indicate that appropriate supplementation of diets with $\beta$-CD can reduce the cholesterol content of eggyolks, thus raising the prospect of the production of a healthier functional food.

  • Oat gum, a soluble dietary fiber, was extracted from rolled oat and the content of $\beta$-glucan and the cholesterol adsorption activity of oat gum were examined. The content of oat gum extracted from rolled oat was 5.15% and the amount of $\beta$-glucan in the oat gum was 29.48%. Most of the cholesterol adsorption of oat gum occurred within 10 hours in the dialysis cell and the cholesterol adsorption activity of oat gum was between 12 to 25%.

Effects of Dietary Xylooligosaccharide on Lipid Levels of Serum in Rats Fed High Cholesterol Diet (고콜레스테롤 식이를 급여한 흰쥐에서 Xylooligo당의 혈중 지질개선 효과)

  • The purpose of this study was to investigate the effects of dietary xylooligosaccharide on lipid levels of serum in rats fed high cholesterol diet. Male Sprage-Dawley rats weiging 100$\pm$10g were randomly assigned to groups of two normal(N, N+10X) and four high cholesterol diets which contained 1%(w/w) cholesterol. High cholesterol diet groups were classified to xylooligosaccharide free diet (C group), 5% xylooligosaccharide diet(C+5X group), 10% xylooligosaccharide diet(C+10X group) and 15% xylooligosaccharide diet(C+15X group) according to the level of dietary xylooligosaccharide supplementation. These experimental diets were fed ad libidum for 4 weeks. The weight gain of high cholesterol diet group were significantly increased more than that of normal group, but those of 10% and 15% dietary xylooligosaccharide groups were significantly decreased more than that of high cholesterol diet(C)group. The higher content of xylooligosaccharide, the more food intake was increased. The food efficiencies of 10%, 15% cholesterol diet groups were lower than that of high cholesterol diet(C)group. The levels of serum triglyceride(TG) and total cholestoral were significantly high in cholesterol diet groups compared with normal diet group but were decreased in groups fed 5% and 10% dietary xylooligosaccharide. Especially, the lowest level showed in group fed high cholesterol diet containing 10% xylooligosacchride. High cholesterol diet group containing 10% xylooligosaccharide increased HDL-cholesterol level and then decreased LDL-cholesterol level and atherogenic index compared with other groups. The level of TBARS(thiobarbituric acid reactive substances) in serum was decreased in rat group fed xylooligosaccharide in high cholesterol diet. The higher content of xylooligosacchride, the more gastrointestinal transit time was decreased. The results indicate that dietary xylooligosaccharide can improve status of TG and total cholesterol and repress lipid peroxidation in serum lipid at hypercholesterolemia induced by high cholesterol diet.

Antihyperglycemic and lipid lowering effect of Tectona grandis in alloxan induced diabetic rats

  • In India, Tectona grandis is traditionally used in the treatment of diabetes mellitus and lipid disorder. In the present study, the antihyperglycemic and lipid lowering effect of ethanolic extract of bark of Tectona grandis (TG) was evaluated using alloxan induced diabetes in rats. Alloxan was given at the dose of 140 mg/kg, i.p. After induction of diabetes, TG was administered for 42 days p. o. and simultaneously different biochemical parameters like plasma glucose, liver glycogen content, serum triglyceride, cholesterol, LDL-cholesterol and HDL-cholesterol were estimated. Diabetic control showed significant increase (P < 0.01) in plasma glucose, serum triglyceride, cholesterol, LDL-cholesterol and significant decrease (P < 0.01) in serum HDL-cholesterol and liver glycogen content. Treatment with TG showed significant reduction (P < 0.01) in plasma glucose when compared with diabetic control. The elevated levels of serum triglyceride and cholesterol levels were significantly reduced (P < 0.01) by TG. TG treatment for 42 days showed significant decrease in serum LDL-cholesterol (P < 0.01) and significant increase in serum HDL-cholesterol level (P < 0.01). Moreover, diabetic control there was significant decrease in liver glycogen content which was significantly increased (P < 0.05) by treatment with TG. Hence, from the result obtained in the present study it can be concluded that Tectona grandis has the potential to treat diabetes condition and associated lipid disorder.

Effects of Nuruk or Wheat Bran Supplemented Diet on the Serum Levels of Cholesterol and Activities of Hepatic Oxygen Free Radical Metabolizing Enzymes in Rats (누룩 또는 밀기울 첨가식이로 성장시킨 흰쥐의 혈중 Cholesterol 및 간조직 유해산소 대사효소 활성 변동)

  • The effects of nuruk and wheat bran on cholesterol level in serum and activities of free radical metabolizing enzymes were investigated in rats. The rats were fed a diet containing nuruk or wheat bran for one month. Body weight and food intake were measured. Animals were sacrificed after one month. The increased food efficiency ratio throughout whole growth period was observed in the rats fed with either nuruk containing Aspergillus terreus or wheat bran compared with control group on normal diet. In the rats fed with nuruk, hepatic GSH content, glutathione S transferase activity, hepatic cytochrome P 450 content, and aniline hydroxylase activities were generally increased. In the rats fed with nuruk containing other fungi except Aspergillus terreus, xanthine oxidase activity was decreased. The decreased cholesterol level in serum was observed in rats fed with nuruk prepared from Aspergillus terreus and wheat bran. LDL cholesterol level was decreased in rats fed with nuruk prepared with other fungi such as Penicillium sp. and Rhizopus sp. But HDL cholesterol level was increased in all groups fed with nuruk from any fungi and wheat bran. These results suggested that nuruk or wheat bran supplemented diet might exert their effect by decreasing cholesterol level in serum and amount of oxygen free radical level.

Experimental Study on the Effects of Mangumtang and Gamimangumtang on Hyperlipidemia (만금탕(萬金湯) 및 가미만금탕(加味萬金湯)이 고지혈증(高脂血症)에 미치는 영향(影響))

  • In order to investigate the effects of Mangumtang(MT) and Gamimangumtang(GMT) experimental studies were performed in Ex.1(treated with cholesterol and administration of sample at a time) and Ex.2(administration of sample after the treatment of cholesterol). The results were summerized as follows : 1. The content of Total cholesterol in the serum was not significantly decreased in MT group, whereas in GMT group significantly decreased compared with control group in Ex.1 and all significantly decreased in MT group and GMT group in Ex.2. 2. The content of Triglyseride in the serum was all significantly decreased in MT group and GMT group in all case. 3. The content of Phospholipid in the serum was not significantly decreased in MT group, whereas in GMT group was significantly decreased compared with control group in Ex.1 or Ex.2. 4. The content of Free Fatty Acid in the serum was all significantly decreased in MT group and GMT group in all case. 5. The content of HDL-Cholesterol in the serum was tended to decreased compared with control group but did not show a significance in Ex.1 or Ex.2. 6. The content of LDL-Cholesterol in the serum was not significantly decreased in MT group and GMT group in Ex.1, whereas in Ex.2 not significantly decreased in MT group but significantly decreased in GMT group. According to the above results, it is assumed that Mangumtang and Gamimangumtang have a valid effect on Hyperlipidemia, especially Gamimangumtang have a more valid effect on prevention and cure of Hyperlipidemia.

  • The oral administration of deer horn extract was proved to cause a considerable amelioration to the impairment of hepatic oxidative phosphorylation and to the decrease of ATP content in liver tissue induced by cholesterol ingestion. Deer horn extract was shown to accelerate considerably incorporation of acetate-1-C$^{14}$ into cholesterol in liver tissue and it tends to restore a decrease of GOT activity of liver tissue, caused by cholesterol administration. Histological examination showed that deer horn extract had a remarkable preventive effect against fatty infiltration of organs such as liver, heart, spleen, and adrenal gland of cholesterol given rabbits.

Effects of Various Cooking and Re-heating Methods on Cholesterol Oxidation Products of Beef Loin

  • The objective of this study was to define the effects of various cooking and re-heating methods (pan roasting, steaming, oven grilling and micro-waving) on the cholesterol and formation of cholesterol oxidation products in beef loin during storage at $4^{\circ}C$. Raw samples showed lower total cholesterol content than cooked products sampled during storage for 6 d. The following cholesterol oxidation products (COPs) were separated by gas chromatography: $7{\beta}$-hydroxy cholesterol, $20{\alpha}$-hydroxy cholesterol, 25-hydroxy cholesterol, cholestane-$3{\beta}$, $5{\alpha}$, $6{\beta}$ triol (triol), ${\alpha}$-epoxide and 7-ketocholesterol. Total amounts of COPs/cholesterol at 0 d were 0.74, 0.63, 0.76, 1.23 and 0.83% for the raw sample, pan roasting, steaming, oven grilling and micro waving methods, respectively. After 6 d storage almost of the samples had higher content of total COPs than at 0 and 3 d; the lowest (0.55%) COPs was found in the steaming cooking and re-heating method. The highest (5.96%) of COPs was found in the pan roasting cooking and re-heating method after 6 d storage. In conclusion, the concentration of total cholesterol and cholesterol oxidation of beef loin were increased as a consequence of cooking and re-heating methods. Steaming and micro-waving methods showed the lowest of cholesterol oxidation products under refrigerated storage for 6 d. However, each cooking and re-heating method had its own distinctive cooking effects.
