Effects of Dietary Taurine on the Lipid Metabolism in Laying Hens (사료내 타우린 첨가가 산란계의 지방대사에 미치는 영향)

  • Two experiments were conducted to investigate the effect of taurine supplementation on lipid metabolism in laying hens. In experiment 1, 19-wk-old laying hens were given one of four taurine supplemented diets (0 (control), 0.4, 0.8, and 1.2% taurine) fur 10 weeks. Abdominal fat weight was lower in the 1.2% diet by 29.2% compared to the control. Serum concentrations of triacylglycerol and HDL-cholesterol were not different among the treatments. However, seam concentration of total cholesterol was higher by 22.4% in the 1.2% diet compared to the control. Concentration of triacylglycerol or total cholesterol in the liver were decreased by 26.1% or 26.4% and 28.2% or 26.4%, respectively in the 0.8% and 1.2% diets compared to the control. The concentration of HDL-cholesterol in liver was also lower by 33.9% in the 1.2% diet compared to the control. In experiment 2, 81-wk-old laying hens were allocated to one of three taurine supplemented diets (0 (control), 1 and 2% taurine) fur 6 weeks. Abdominal fat weight was lower by 25% in 1% taurine supplementation compared to the control. Serum concentrations of triacylglycerol, total cholesterol and HDL-cholesterol of hens fed with 1% diet were not different from those of control group. However, sew concentrations of triacylglycerol and total cholesterol were lower by 44.0% and 19.8%, respectively in the 2% diet compared to the control. Furthermore, serum concentration of HDL -cholesterol in the 2% diet was higher by 75% compared to the control. Concentrations of triacylglycerol and total cholesterol in the liver in the 2% diet were decreased in the 1% diet by 36.8 and 23%, respectively, but increased by 78.4% and 70%, respectively, compared to the control. The concentration of HDL-cholesterol in the liver was not different between the 1% diet and the control, but higher by 62.8% in the 2% diet compared to the control. These results indicated that taurine supplementation decreased the fat storage in abdominal cavity, which was accompanied by the changes in triacylglycerol and cholesterol metabolisms of laying hens.

LIPID PEROXIDATION AND ANTIOXIDANTS IN WORMERS EXPOSED TO LEAD (연 폭로 근로자들의 지질과산화 및 항산화상태에 관한 연구)

  • This study was started to find out if plasma malondialdehyde(MDA), $\alpha-tocopherol$ and erythrocyte superoxide dismutase(SOD) activity could be markers of biological activity resulting from exposed to lead in workers. Blood samples were randomly obtained from lead-exposed workers(n=29), $CO_2$, welders(n=60) and office workers(n=60). We used whole blood to analyse blood lead with atomic absorption spectrophotometer. Superoxide dismutase activity in erythrocyte was measured with spetrophotometer. MDA and $\alpha-tocopherol$ in plasma were measured with high performance liquid chromatography. Lead-exposed workers was significantly high in blood lead concentration$(29.37{\mu}g/d\ell)$ compared with welders$(6.42{\mu}g/d\ell)$ and office workers$(5.01{\mu}g/d\ell)$. The level of plasma MDA was significantly higher in the lead-exposed workers($1.87{\mu}mol/g$ cholesterol) than the welders($1.41{\mu}mol/g$ cholesterol) and office workers($1.41{\mu}mol/g$ cholesterol). Erythrocyte SOD activity in lead-exposed workers(56.80 U/g Hb) was significantly increased than those of welders(37.63 U/g Hb) and office workers(20.47 U/g Hb). The plasma $\alpha-tocopherol$ level of lead-exposed workers($4.93{\mu}g/g$ cholesterol) was statistically different from welders($4.25{\mu}g/g$ cholesterol) and office workers$4.28{\mu}g/g$ cholesterol). Neither age nor smoking was related to SOD or MDA level. Blood lead was significantly correlated with erythrocyte SOD activity(r=0.405), plasma MDA(r=0.296) and $\alpha-tocopherol$ (r=0.207). Plasma MDA was also significantly correlated with SOD (r=0.217). In multiple regression analysis, the change of MDA and SOD activity level related to the blood lead concentration. These results suggested that the increase of plasma MDA and erythrocyte SOD activity in lead-exposed workers had a close relationship with the oxidative stress induced by lead.

Effect of Soy Protein Hydrolyzate on Lipid Metabolism and Antioxidant Activity in the Rat (대두단백가수분해물이 흰쥐의 지질대사와 항산화에 미치는 영향)

  • This study was conducted to investigate the effect of soy protein hydrolyzate on lipid metabolism and antioxidant activity in the rat. Thirty-eight male rats of Sprague-Dawley strain were divided into five groups: casein, isolated soy protein (ISP), seoritae protein hydrolyzate (SH), soluble soy protein hydrolyzate (SS), and insoluble soy protein hydrolyzate (IS). The control diet (casein group) contained 20% casein protein and experimental diet contained 10% casein and 10% isolated soy-protein or soy-protein hydrolyzate. Fecal lipid content was increased and lipid apparent absorption rate was decreased significantly by the ISP group at the first week of experimental period. Blood triglyceride, total cholesterol, LDL-cholesterol and atherogenic index (AI) were decreased by soy protein hydrolyzate groups than casein group. Liver total lipid, triglyceride and cholesterol were not different among groups, but showed decreasing tendencies in soyprotein hydrolyzate groups. The lipid lowering effect was prominent in the IS group among soy protein hydrolyzate groups. Total antioxidant activity showed increasing tendency in the seoritae hydrolyzate group. Liver superoxide dismutase (SOD), glutathione peroxidase (GPx) and catalase activities also showed higher tendencies in the seoritae hydrolyzate group than other groups. In conclusion, insoluble soyprotein hydrolyzate was more effective in lowering body lipids and seoritae hydrolyzate had higher antioxidant capacity among soy protein hydrolyzates.

Lipid Lowering Effect of Anthocyanin-Pigmented Rice Bran in Streptozotocin-Induced Diabetic Male Rats

  • Oryza sativa cv. Heugjinjubyeo, an anthocyanin-pigmented rice variety, is well known to contain high levels of bioactive phytochemicals, anthocyanin, quinolone alkaloids and phenolic acids. Here, we studied the inhibitory effect of Oryza sativa cv. Heugjinjubyeo bran on the absorption of dietary fat in streptozotocin-induced diabetic Sprague-Dawley male rats. For these experiments, experimental animals were divided into four groups: normal, diabetic-control and two experimental groups that were fed 1.0 g or 2.0 g/kg body weight/day of Oryza sativa cv. Heugjinjubyeo bran supplement for 14 days. As a result, liver glycogen levels increased significantly by 65% and 32% in groups receiving 1.0 g and 2.0 g/kg body weight/day, respectively, compared to diabetic-control. Liver cholesterol levels were significantly lower by 8.3% and 14.5% in the groups fed 1.0 g and 2.0 g of anthocyanin- pigmented rice extracts, respectively.

Effect of Silk Fibroin on the Protection of Alcoholic Hepatotoxicity in the Liver of Alcohol Preference Mouse

  • Silk fibroin (SF) derided from the domestic silk worm, bombyx mori, is the natural protein and widely used as bio-functional materials as well as apparels. We studied the livers protective effect of SF from alcohol-induced hepatotoxicity in the alcohol preference mouse. To increase more absorption of SF in experimental animals, molecular weight of SF was lowered by 2N of HCI aqueous solution at 10$0^{\circ}C$ for 48 hrs. SF was added to liquid diet with alcohol and fed to the alcohol preference mice for 4 weeks. To assess the liver function, the concentration of alanine aminotransferase (AlT), aspartate aminotransferase (AST) and cholesterol present in either blood or liver tissue were measured. As compared with non-SF treated groups the SF-treated showed significantly low concentrations of ALT, AST, cholesterol and triacylglycerol values, respectively. Histopathological examination revealed that the extent of hepatocyte injury in the SF-treated group was reduced when it was compared with non SF-treated group. These results suggest that SF may have liver protective effects against alcohol-induced hepatotoxicity.

  • Hypercholesterolemia is a main risk factor to develop cardiovascular disease, a major cause of death in Korea currently. Dietary factors which shows hypocholesterolemic effect have been reported, which includes plant proteins, unsaturated fatty acids, milk, calcium, flavonoids. Numerous animal and human studies confirmed the pronounced hypocholesterolemic effects of soluble dietary fiber, such as psyllium husk, pectin, sodium alginate, guar gum. Several hypotheses have been proposed to explain the hypocholesterolemic effect of dietary fiber, including binding of bile acids by fiber, interference of lipid absorption and reduced hepatic cholesterol synthesis by propionate, a product of soluble dietary fiber fermentation. Several other hypotheses have been proposed, and these hypotheses are not mutually exclusive.

The Effect of Aerobic Dancing on Lipid and Ca Metabolism in College Women (에어로빅 운동이 여대생의 체내지질 및 Ca 대사에 미치는 영향)

  • This study was designed to find out the effects of aerobic exercise on lipid and Ca metabolism in seven healthy college women, aged 20 to 22 years. Metabolic studies were conducted before and after a 10-weeks exercise period, during which subjects participated in the planned aerobic dance program every day except Sunday. The initial mean time engaged in the exercise was 28 minutes and it was gradually increased up to 45 minutes around the middle point of the exercise period. During both of metabolic study periods, the subjects ate experimental diets which supplied about 120g of protein and 600mg of Ca daily and during the rest of the experimental period they ate their usual diets. The use of alcoholic beverages and drugs were prohibited. The results were summarized as follows. 1) The effect of exercise on lipid metabolism. (1) Mean body weight decreased significantly after exercise(p<0.01) and it did not show a significant decline in skinfold thickness and total body fat contents. (2) Serum total cholesterol level decreased significantly after exercise(p<0.05) and TG level also tended to be lower than that of pre-exercise period. Exercise did not exert any influence on the level of serum HDL-cholesterol in this study. (3) Exercise did not alter total lipid content in feces and apparent lipid absorption rate. 2) The effect of exercise on Ca and P metabolism. (1) After exercise, focal Ca excretion was slightly reduced, however, urinary Ca excretion was not significantly changed. In the results, a slight increase was shown in body Ca retention after exercise. (2) Exercise tended to increase urinary P excretion, but neither P balance nor fecal excretion was significantly changed after exercise. (3) Bone mineral content was not affected by exercise. In summary, aerobic exercise decreased total cholesterol and TG level in serum and tended to increase body Ca retention. With the results, it can be concluded that the additional physical activities beyond the normal daily life in college women might prevent some degenerative diseases-suchas atherosclerosis and osteoporosis.

Effects of Soyprotein and Casein Mixtures Combined with Various Levels of Pectin on Lipid Metabolism in Rats (Casein과 대두단백의 혼합비율 및 Pectin 첨가 수준이 흰쥐의 체내 지방대사에 미치는 영향)

  • This experiment was designed to investigate the effects of the mixing ratio of soyprotein and casein, and the level of pectin combined with the mixture on lipid metabolism in rats. Forty-eight male weanling rats of Wistar strain weighing 58.8\;{\pm}\;1.9g$ were divided into six groups by completely randomized block design and fed 10% protein diet for four weeks. Two types of protein mixtures (casein to soyprotein mixing ratio of 1 : 3 and 2: 1)combined with 0.5% , 3%, and 5% of pectin were employed for experimental diets. The results obtained in the study are summarized as follows ; 1) Feed efficiency ratio and protein efficiency ratio were not significantly different among six groups for the whole experimental period, but those for high casein-low pectin group were significantly higher than the ones for high soy-high pectin group at 4th week of the experimental period. 2) Gross fecal dried weight and fecal lipid excretion were higher in high pectin groups of both protein combinations. Therefore, the apparent fat digestibility and absorption appeared to be significantly low in high pectin groups. 3) Pectin was effective in lowering serum lipid and cholesterol levels in high casein groups, but no effect of pectin was noted in high soyprotein groups. 4) Lipid and cholesterol contents of the liver were higher in high soy-low pectin group than the others. And no marked differences in lipid and cholesterol contents in the kidney and carcass were observed.

Anti-oxidation and Fat Accumulation Inhibitory Effects of Silbi-san (실비산의 항산화 및 지방축적 억제 효과)

  • Objectives The purpose of this study was to investigate the effects of Silbi-san on the antioxidant and fat accumulation inhibition and to analyze the anti-obesity effect by analyzing the changes in serum lipid composition in obese mice. Methods We compared contents of phytochemicals like total polyphenols and total flavonoid and antioxidant activities such as 2,2-dipheny-1-picrylhydrazyl and 2.2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity. After Silbi-san in 3T3-L1 cells in vitro and mouse adipose tissue ex vivo, we quantified intracellular triglyceride accumulation and lipolysis. Moreover, the anti-obesity activity though inhibiting pancreatic lipase were analyzed. In 3T3-L1 cells, morphological changes showed that control cells had many lipid while cells treated with Silbi-san had less lipid accumulation. 30% EtOH Silbi-san treatment also suppressed the fat absorption by inhibiting the activity of pancreatic lipase and led to high lipolysis through promoting glycerol release. The experimental group was divided into four groups: Normal group fed normal feed, Control group fed 60% high fat diet (HFD) and distilled water, drug group fed 60% high fat diet and 200 mg/kg of Silbi-san water extract, drug group fed 60% HFD and 200 mg/kg of Silbi-san 30% ethanol extract. Results Serum total cholesterol content and serum low density lipoprotein-cholesterol content were significantly decreased in the Silbi-san extract group compared to the control group, serum high density lipoprotein-cholesterol content was significantly increased in Silbi-san extract group. Conclusions In this study, the antioxidant and fat accumulation inhibitory effects of Silbi-san were confirmed.

A Study on the Effects and Mechanisms of the Combination Extract of Ephedrae Herba and Coicis Semen on Lipid Accumulation and Glucose Absorption in Non-Alcoholic Fatty Liver Disease (마황과 의이인 혼합추출물이 비알콜성 지방간 모델에서 지질 축적 및 포도당 흡수에 미치는 효과 및 기전 연구)

  • Objectives: Ephedrae herba (EH) and Coicis semen (CS) has been frequently prescribed for the treatment of obesity. However, effects of combinational extracts of these two herbs on non-alcoholic fatty liver disease are unknown. The aim of the present study was to investigate the effects of EH and CS on lipid accumulation and glucose absorption in free fatty acids (FFAs) or palmitic acid (PA)-treated HepG2 cells. Methods: Five samples of EH and CS were extracted by combination ratios (S1=0:100, S2=25:75, S3=50:50, S4=75:25, S5=100:0). Oil Red O staining was used to measure lipid accumulation in FFAs-induced steatosis cells. Intracellular triglycerides and total cholesterol levels were measured in FFAs-induced steatotic HepG2 cells. In PA-treated cells, intracellular 2-NBDG was detected using a fluorescence microplate reader and flow cytometry. Phosphorylation of key metabolism-related factors of AMP-activated protein kinase and acetyl-CoA carboxylase, expression of key lipid synthesis-related factors carnitine palmitoyltransferase 1 alpha (CPT1α), sterol regulatory element-binding protein 1 (SREBP1), peroxisome proliferator-activated receptor gamma (PPARγ) and CCAAT enhancer binding protein alpha (C/EBPα) were confirmed by western blot. Results: Treatment of EH-CS combination in the FFAs-induced steatotic HepG2 cells significantly reduced lipid accumulation. As the relative ratio of Ephedrae herba increased, the lipid-lowering effects of the combination were increased. However, S1 and S5 of Ephedrae herba and Coicis semen did not significantly reduce triglycerides and total cholesterol induced by FFAs. However, the combination of Ephedrae herba and Coicis semen restored glucose absorption in PA-induced HepG2 cells. Major makers of SREBP1, PPARγ, C/EBPα, and CPT1α expression tended to decrease with EH ratio. Conclusions: The EH-CS combination has advantages over sole EH and CS extracts in improving lipid and glucose metabolism in liver steatosis models.
