Growth, Photosynthesis and Chlorophyll Fluorescence of Chinese Cabbage in Response to High Temperature (고온 스트레스에 대한 배추의 생장과 광합성 및 엽록소형광 반응)

  • In order to gain insight into the physiological responses of plants to high temperature stress, the effects of temperature on Chinese cabbage (Brassica campestris subsp. napus var. pekinensis cv. Detong) were investigated through analyses of photosynthesis and chlorophyll fluorescence under 3 different temperatures in the temperature gradient tunnel. Growth (leaf length and number of leaves) during the rosette stage was greater at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures than at ambient temperature. Photosynthetic $CO_2$ fixation rates of Chinese cabbage grown under the different temperatures did not differ significantly. However, dark respiration rate was significantly higher in the cabbage that developed under ambient temperature relative to elevated temperature. Furthermore, elevated growth temperature increased transpiration rate and stomatal conductance resulting in an overall decrease of water use efficiency. The chlorophyll a fluorescence transient was also considerably affected by high temperature stress; the fluorescence yield $F_J$, $F_I$, and $F_P$ decreased considerably at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, with induction of $F_K$ and decrease of $F_V/F_O$. The values of RC/CS, ABS/CS, TRo/CS, and ETo/CS decreased considerably, while DIo/CS increased with increased growth temperature. The symptoms of soft-rot disease were observed in the inner part of the cabbage heads after 7, 9, and/or 10 weeks of cultivation at ambient $+4^{\circ}C$ and ambient $+7^{\circ}C$ temperatures, but not in the cabbage heads growing at ambient temperature. These results show that Chinese cabbage could be negatively affected by high temperature under a future climate change scenario. Therefore, to maintain the high productivity and quality of Chinese cabbage, it may be necessary to develop new high temperature tolerant cultivars or to markedly improve cropping systems. In addition, it would be possible to use the non-invasive fluorescence parameters $F_O$, $F_V/F_M$, and $F_V/F_O$, as well as $F_K$, $M_O$, $S_M$, RC/CS, ETo/CS, $PI_{abs}$, and $SFI_{abs}$ (which were selected in this study), to quantitatively determine the physiological status of plants in response to high temperature stresses.

CHLOROPHYLL FLUORESCENCE IN CUCUMBER (Cucumis safivus L.) AND PEA (Pisum sativum L.) LEAVES UNDER CHILLING STRESS IN THE LIGHT AND DURING THE SUBSEQUENT RECOVERY PERIOD

  • To investigate the chilling sensitivity related injuries in the photosynthetic apparatus of cucumber leaves, the light-chilling induced alterations of chlorophyll fluorescence transients in cucumber leaves were compared with those in pea leaves. As an early effect of light-chilling, an increase in Fp/Fm$^*$ was observed in both pea and cucumber leaves, which was saturated by about 6 h chilling. However, the saturated value of Fp/Fm was almost 1.0 in cucumber, in contrast to about 0.8 in pea. During the recovery period after 24 h chilling, the light-chilling induced changes in pea seemed to be reversed, but those in cucumber leaves were thought to be irreversible, because Fo was increased significantly. Light-chilling caused significant decreases in qQ and qE in cucumber leaves, but qR was increased until 6 h, and decreased thereafter. In both pea and cucumber leaves, Fm was increased by 2 h dark treatment. The Fm from the predarkened pea leaf discs was higher than the value from the preilluminated ones during the whole period of light-chilling (500 $\mu$mol m$^{-2}$s$^{-1}$ PAR). However, the predarkened cucumber leaf discs showed a reduction in Fm and an increase in Fo during the 2 h chilling in the light. These results indicate that the causes of chilling sensitivities in photosynthetic apparatus of cucumber leaves are possibly related with the damage in PSI reaction center and the ability of acidification of lumen by PSII.

Preliminary Study on the Toxicity and Transfer of Heavy Metals and Tributyltin to Seagrass Zostera marina (잘피의 광합성에 대한 중금속 및 TBT의 독성 영향과 중금속 흡수에 대한 연구)

  • Uptake kinetics of Cd and Zn by leaves and rhizome of the seagrass Zostera marina were examined in controlled laboratory radiotracer experiments. Subsequently, acute toxicity of Cd, Cu and TBT on photosynthetic quantum yield (ΔF/Fm’ of Z. marina were determined, and the differential sensitivities of rapid light curve (RLC) to those harmful substances were also compared. All measurements on photosynthetic activity were determined by chlorophyll a fluorescence method using pulse amplitude modulation (PAM). Metal uptake by Z. marina was saturated with increasing exposure time in leaves and rhizomes. Uptake of Zn by Z. marina was faster than that of Cd. Metal uptake rates in Z. marina decreased with the increase of dissolved metal concentrations and also with the increase of biomass. The adverse effect of TBT on effective quantum yield was stronger than other pollutants. Average acute toxicity on the RLC of the seagrass exposed to TBT and two heavy metals (Cd and Cu) was going to decrease as follows: TBT > Cd > Cu. Our preliminary results in this study suggested that Z. marina potentially can be used as a biomonitor of harmful substances contamination in coastal waters.

The Effect of Substrate on Ecophysiological Characteristics of Green Macroalga Ulva pertusa Kjellman (Chlorophyta) (구멍갈파래(Ulva pertusa Kjellman)의 생태생리에 대한 생육기질의 효과)

  • Seashore joining with land and sea, which is typical habitat for marine macroalgae, is classified two types of shore as soft- and hard-bottom shore according to topographical (geological) and ecological features. We compared two of Ulva pertusa Kjellman from two contrasting habitats, sandy (soft-bottom, Haenam) and rocky shore (hard-bottom, Hadong) in terms of chlorophyll-a fluorescence and its parameters, and various photosynthetic pigment and nutrient content in the tissue of those. Both of habitats were different in the light environment such as light attenuation coefficient and even in nutrient concentration of ambient seawater. Electron transport rate (ETR) of Ulva from sandy shore was higher than from rocky shore. The range of photosynthetic pigment content in the tissue of U. pertusa was significantly much more in from sandy shore, and also nitrogen and phosphorus content were significantly higher except for carbon content. However, there were no significant differences in the ratio of among photosynthetic pigments, and N:P ratio was similar between each other, even though significantly different. Our result implied on the reason of why most of green tides in the worldwide concentrated and frequently occurred at sites with sandy, muddy and silty bottoms, being classified as soft-bottom shore.

LIGHT-DEPENDENT CHANGES OF CHLOROPHYLL FLUORESCENCE AND XANTHOPHYLL CYCLE PIGMENTS IN MAIZE LEAVES DURING DESICCATION

  • Changes of chlorophyll fluorescence and xanthophyll cycle pigment contents in maize leaves were investigated dunng desiccation in darkness or in the light. In darkness, a drastic dehydration of detached maize leaves down to 50% relative water content (RWC) affected photochemical efficiency of photosystem II (Fv/Fm) and pht)tochemical quenching (qP) only slightly. In contrast, desiccation in the light with a moderate intensity led to a pronounced reduction in Fv/Fm with a Fo quenching when RWC was greater than 70%. This reduction in Fv/Fm could be recovered in darkness under hutrod condition. In leaves with RWC below 70%, significant reduction in Fv/Fm was accompanied by an increase of Fo, which could not be reversed within 5 h in darkness under humid condition. The nonphotochemical quenching increased during desiccation in the light with a concomitant rise in zeaxanthin at the expense of violaxanthin. Pretreatment with dithiothreitol (DTT), an inhibitor of zeaxanthin synthesis, inhibited the development of nonphotochemical quenching and prevented the xanthophyll interconversion during desiccation in the light. These results suggest that even light with a moderate intensity becomes excessive under dehydration and zeaxanthin-associated photoprotection of photosynthetic apparatus against photodamage is involved, but the protection is not complete against severe desiccation.

LOW DISSIPATION OF EXCITATION ENERGY IN THE PHOTOSYNTHETIC MACHINERY OF CHILLING-SENSITIVE PLANTS DURING LOWTEMPERATURE PHOTOINHIBITION

  • Using a squash plant, a chilling-sensitive species, and a spinach plant, a chilling-resistant one, effects of chilling temperature on the photosynthetic machinery were studied in terms of chlorophyll fluorescence. When thylakoid membranes were isolated and subjected to incubation at different temperatures, spinach showed stable photosystem II activity at the low temperature side, in contrast to squash which showed quite severe inactivation at low temperature. When parameters of chlorophyll fluorescence were examined, chilling in darkness did not affect either Fv/Fm or photochemical and non-photochemical quenching, in both types of plants. However, chilling of squash plants under irradiance of medium intensity caused a specific decrease in Fv/Fm accompanied by a decline in energy-dependent quenching. Contrastingly, photosystem li of spinach plants were not much affected by light-chilling. When the pool size of zeaxanthin was examined after exposure to high light at different temperatures, squash plants was shown to have a much lower content of antheraxanthin + zeaxanthin, as compared to spinach plants, during low-temperature photoinhibition. These results suggest that chilling-sensitive plants have low capacity to dissipate excitation energy nonradiatively, when they are exposed to low-temperature photoinhibition, and, as a consequence, more vulnerable to photoinhibitory, damage to the photosynthetic apparatus.

Salt Tolerance Assessment with NaCl of Stauntonia hexaphylla (Thunb.) Decene. and Raphiolepis indica var. umbellata (Thunb.) Ohashi (NaCl 처리에 따른 멀꿀과 다정큼나무의 내염성 평가)

  • Stauntonia hexaphylla and Raphiolepis indica, cold-tolerant broadleaved evergreens ranging through the southern region of South Korea, were assessed on salt tolerance with NaCl treatment using visual damage, chlorophyll florescence image, and malondialdehyde (MDA) analysis. As NaCl concentrations increased, the soil pH decreased and EC increased, and the soil of S. hexaphylla was affected more strongly by the treatment than that of R. indica. In visual damage, S. hexaphylla withered above 200 mM NaCl at 20 days after the treatment. All individuals of R. indica survived during the experiment though the leaves of R. indica showed visual damages up to 400 mM NaCl. The color changes in chlorophyll fluorescence showed a strong correlation with the degree of visual damage. As NaCl increased, the red color of the leaves of S. hexaphylla was distinctly changed to blue and chlorophyll fluorescence decreased starting from the margin to the middle of a leaf. R. indica showed subtle color changes and remained in red color during the experiment. At five days after the NaCl treatment, the MDA of S. hexaphylla was above $4.56nmol{\cdot}g^{-1}$ when plants showed the highest visual damage and EC. The MDA of R. indica in all treatments showed below $1.5nmol{\cdot}g^{-1}$ except 400 mM NaCl treatment during the experiment.

Behavior of Nutrients along the Salinity Gradients in the Seomjin River Estuary (섬진강 하구역에서 염분경사에 따른 영양염의 거동)

  • Behavior of nutrients along the salinity gradients in the Seomjin River estuary was investigated in March July, September and November, 1999. Sampling sites were set based on the surface salinity during each cruise rather than geographic locations. The results suggest that source of nitrate and silicate was the Seomjin River discharge, while that of nitrite and phosphate was waste disposal from the Gwangynng Bay near the mouth of Seomjin River estuary. Ammonia was supplied inside the estuary at the region about $6\~8$ km far from Nancho Island. Strong removal behavior of some nutrient such as ammonia, phosphate and silicate was observed at $5\~15$ psu salinity area in November, where high concentrations of $chlorophyll\;a\;(>8{\mu}g/L$) occurred. High N : P ratios and entirely removal of phosphate at chlorophyll a peak region suggest that phosphate is the limiting factor for phytoplankton growth. Relatively high ratios of Rb to Ra (Rb: Fluorescence before acidification, Ra: Fluorescence after acidification) at $5\~15$ psu salinity region in November indicate that phytoplankton were in good physiological condition.
