Flavonoid contents of common buckwheat (cv. Kitawase) and tartary buckwheat (cv. Hokkai T 8, Hokkai T 9 and Hokkai T 10) were determined by high-performance liquid chromatography (HPLC). Moreover, they were measured at different plant developments such as 10, 18, 20, 22 and 30 days after sowing (DAS) and with plant parts including leaf, stem and flower harvested at 30 DAS. Total flavonoids including chlorogenic acid, four kinds of C-glycosylflavones (orientin, isoorientin, vitexin, isovitexin) and rutin of tartary buckwheats (range of 44.2-54.7, mean 44.2) were found 35% higher than those of common buckwheat (28.9 mg/g dry wt.). Among them, rutin was measured above 80% of total flavonoid contents. The other flavonoids (chlorogenic acid and four kinds of C-glycosylflavones) presented the highest level at 10 DAS and decreased according to plant developments. On the other hand, rutin content of Kitawase presented the highest level (33.6 mg/g dry wt.) at 22 DAS and decreased up to 30 DAS. Rutin content in tartary buckwheat temporarily decreased from 10 to 18 DAS and then reversely increased up to 30 DAS presented the highest level as 'U' curve. In Hokkai T 10, rutin content was found the highest level (53.8 mg/g dry wt.) at 30 DAS. In different plant parts harvested at 30 DAS, rutin content of leaf (range of 42.8-68.0, mean 57.0) was 5.3-fold higher than that of stem (range of 8.0-15.9, 10.8 mg/g dry wt.), regardless of cultivar. Significantly, rutin content (78.7) in the flower of Kitawase was 1.8 times higher than in the leaf and 9.8 times in the stem. Especially, chlorogenic acid content (14.6 mg/g dry wt.) in the flower of Kitawase was 63-fold higher than in the leaf, and 20-fold in the stem.

We investigated the protective effects of chlorogenic acid (CGA), a polyphenol compound, on oxidative damage induced by UVB exposure on human HaCaT cells. In a cell-free system, CGA scavenged 1,1-diphenyl-2-picrylhydrazyl radicals, superoxide anions, hydroxyl radicals, and intracellular reactive oxygen species (ROS) generated by hydrogen peroxide and ultraviolet B (UVB). Furthermore, CGA absorbed electromagnetic radiation in the UVB range (280-320 nm). UVB exposure resulted in damage to cellular DNA, as demonstrated in a comet assay; pre-treatment of cells with CGA prior to UVB irradiation prevented DNA damage and increased cell viability. Furthermore, CGA pre-treatment prevented or ameliorated apoptosis-related changes in UVB-exposed cells, including the formation of apoptotic bodies, disruption of mitochondrial membrane potential, and alterations in the levels of the apoptosis-related proteins Bcl-2, Bax, and caspase-3. Our findings suggest that CGA protects cells from oxidative stress induced by UVB radiation.
In this study, we investigated maximum yield of radical scavenging activity (RSA) using each of various extraction solvents and procedure from noni friut (Morinda citrifolia). Noni fruits were either sun-dried (FS) or hot-air dried (FO) at $60^{\circ}C$ after steam blanching. For optimum aqueous extraction, noni fruits should be sun dried and autoclave extraction time should not be over 30 min to produce extraction with high RSA with low cost. In case of 50% ethanol extraction, reflux extracts of FS and FO resulted in $IC_{50}$ of 1.92 mg/mL and 3.06 mg/mL at 8 hr. When $IC_{50}$ values were lower than 5 mg/mL, coefficient of correlation was 0.71 indicating that 71% of the phenolic antioxidants in noni fruits were accounted for the activity by scavenging free DPPH. However, coefficient of correlation significantly decreased to 0.63 over $IC_{50}$ values of 5 mg/mL. Autoclave extraction contained chlorogenic acid of $14.69 \;{\mu}g/mL$ and scopoletin of $3.86 \;{\mu}g/mL$. Reflux extraction showed all three compounds, chlorogenic acid ($26.19 \;{\mu}g/mL$), quercetin ($19.59 \;{\mu}g/mL$), and scopoletin ($17.4 \;{\mu}g/mL$). Therefore, the result of this study indicated that the potential antioxidant activities and functional values were obtained significantly with reflux extraction from noni fruit.
Recently, consume for functional cosmetics containing natural products has been greatly increased. In order to develop it as a natural cosmetic material, we selected Ixeridium dentatum (Thunb.) Tzvelev, Plantago asiatica L. and Rumex crispus L. that have antioxidant and anti-inflammatory effects. In this study, simultaneous quantitative analysis of the isolated compounds and natural product complexes (Mix.) were validated using high performance liquid chromatography (HPLC). The isolated six compounds were shown in a large linearity with a correlation coefficient (R2) of 0.99. The limit of detection (LOD) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 0.36 ㎍/mL, 0.36 ㎍/mL, 0.37 ㎍/mL, 0.30 ㎍/mL, 0.22 ㎍/mL and 0.12 ㎍/mL, respectively. The limit of quantification (LOQ) of chlorogenic acid, plantamajoside, acteoside, emodin chrysophanol and physcion were 1.10 ㎍/mL, 1.08 ㎍/mL, 1.12 ㎍/mL, 0.99 ㎍/mL, 0.66 ㎍/mL and 0.35 ㎍/mL, respectively. Content analysis showed chlorogenic acid (0.19 ± 0.02%), plantamajoside (0.48 ± 0.01%), acteoside (0.65 ± 0.01%), emodin (1.15 ± 0.11%), chrysophanol (0.73 ± 0.01%) and physcion (0.69 ± 0.09%). Therefore, the results of this study may provide for basic data of standardization research natural cosmetic material development on the I. dentatum, P. asiatica and R. cripus.
본 연구의 목적은 클로로겐산이 함유된 피부 패치를 피부에 부착하여 체지방과 체중에 미치는 변화를 규명하여, 유용성을 평가하는 것이다. 시험군 8명, 대조군 8명으로 총 16명의 대상자가 연구에 참여하였고, 연구 기간은 2020년 08월 03일부터 2020년 08월 31일까지 4주간 실시하였다. 군-간 효과검정 결과 체지방의 변화는 전체적으로 유의한 차이가 없었으나(F=4.38, p<.055), 1주 후와 4주 후 검사에서는 의미있는 변화(p<.001)가 있었다. 또한, 체중은 유의미한 차이로 시험군이 대조군보다 체중 감소가 큰 것으로 나타났다(F=9.43, p<.008). 이번 연구의 결과는 비만 관련 연구에 패치를 적용한 예비연구로써 의미가 있다고 할 수 있으며, 연구의 결과가 향후 연구될 피부 패치 연구의 기초 자료로 활용될 수 있을 것으로 판단된다.

Background: Short-term hydroponic-cultured ginseng (sHCG), which is 1-year-old ginseng seedlings cultivated for 4 weeks in a hydroponic system, is a functional food item with several biological effects. However, the optimal extraction conditions for sHCG, and the bioactivity of its extracts, have not been evaluated. Methods: Chlorogenic acid (CGA) and ginsenoside contents were evaluated in sHCG, white ginseng (WG), and red ginseng (RG) using high-performance liquid chromatography. Response surface methodology (RSM) was used to optimize the extraction conditions (temperature and ethanol concentration) to maximize the yield of dry matter, CGA, and four ginsenosides (Re, Rg1, Rb1, and Rd) from sHCG. The optimal extraction conditions were applied to pilot-scale production of sHCG extracts. The expression levels of tumor necrosis factor (TNF)-α/interferon (IFN)-γ-induced thymic and activation-regulated chemokines (TARC/CCL17) were measured after treatment with sHCG, WG, and RG extracts, and the effects of their bioactive compounds (CGA and four ginsenosides) on human skin keratinocytes (HaCaTs) were evaluated. Results: CGA and four ginsenosides, which are bioactive compounds of sHCG, significantly inhibited TNF-α/IFN-γ-induced TARC/CCL17 expression. The optimal sHCG extraction conditions predicted by the RSM models were 80 ℃ and 60% ethanol (v/v). The sHCG extracts produced at the pilot scale under optimal conditions greatly alleviated TNF-α/IFN-γ-induced TARC/CCL17 production compared with WG and RG extracts. Conclusions: Pesticide-free sHCG extracts, which contain high levels of CGA and the ginsenosides Re, Rg1, Rb1, and Rd as bioactive compounds, may have therapeutic potential for atopic diseases.
건강기능성식품의 원료로 이용되고 있는 석류의 주요성분에 대한 효율적인 이용을 위하여 반응표면분석에 의하여 각 성분의 열수 추출조건을 분석하여 최적 추출조건을 확립하였다. 주요성분의 최적 추출조건 예측을 위하여 추출온도$(60{\sim}100^{\circ}C)$, 추출시간$(1{\sim}5hr)$을 독립변수로 하여 중심합성계획에 의해 10구의 추출조건을 설정하였다. 각 추출물의 주요성분 함량 즉, chlorogenic acid, kaempferol, $17-{\alpha}-estradiol$ 및 $17-{\beta}-estradiol$ 함량을 측정하여 SAS program에 의해 회귀분석을 하여 각각의 특성을 분석하였다. 그 결과 chlorogenic acid, kaempferol 및 $17-{\alpha}-estradiol$ 함량은 주로 추출온도에 영향을 받는 것으로 나타났으며, $17-{\beta}-estradiol$ 함량은 추출시간에 영향을 받는 것으로 나타났다. 각 변수에 대한 회귀식을 도출하여 공정인자별 석류 추출물의 chlorogenic acid, kaempferol, $17-{\alpha}-estradiol$ 및 $17-{\beta}-estradiol$ 함량에 대한 최적 추출조건을 superimpose한 결과 추출온도 $98{\sim}100^{\circ}C$, 추출시간 $3{\sim}4hr$로 각각 예측되었다.
Some seeds of the Rutaceae family, Zanthoxylum piperitum, Z. schinifolium officinalis, Poncirus trifoliata, Citrus unshin, were investigated to clarify their antioxidative components. Finely powdered samples were extracted by hexane, followed by dichioromethane and then 70% methanol in a hot bath. Its unsaponifiables containing X-and Y-tocopherol with trace amount of ${\beta}-and$\;{\delta}-tocopherol$. also showed comparatively weak activity, although the hexane fraction itself had no significant antioxidative effect on lard. Levels of total tocopherols in the samples averages 42. 24-154. 11 mg/lOOg total extractives. The dichloromethane-and 70% methanol extractives showed strong antioxidative activity, from which antioxidative substances were purified with benzene-acetone(6:5, V/V) on a silica gel column, and with a solvent mixture of acetonitrile-methanol-$H_2O$(40:40:20, V/V/V) on a Sep-Pak $C_{18}$ hydrolyzed by 5% KOH-ethanol. The recovered unsaponifiables were, then, separated on a column of high performance liquid chromatography. The unsaponifiables produced by hydrolysis of the isolates from dichloromethane extractives has epi-catechin(40.0-57.1%) and (+)-catechin<$l9.1{\sim}24.4%$ to total phenolic substances, on area base) as major component, accompanied by chlorogenic acid, gallic acid(?), trans-p-coumaric acid and tralls-p-ferulic acid including some unknown components, and those derived from 70% methanol extractives also comprise (+)-catechin($31.3{\sim}39.6%$ to total components, on area base), epi-catechin($2O.2{\sim}36.4%$), trans-p-cournaric acid(8.4-15.3%) and trans-p-ferulic acid($7.7{\sim}14.1%$) as predominant component with some minor coponents, but the fraction supposed to be gallic acid(?) is not present. The antioxidative activities of the phenolic components isolated in this work were in order of epi-catechin>catechin>chlorogenic acid>trans-p-ferluic acid>trans-p-coumaric acid.
Objectives : Lonicerae japonicae Flos(LJF) has been reported to exhibit anti-oxidant, anti-inflammatory, anti-viral, anti-rheumatoid properties. However, it is still largely unknown whether LJF inhibits the ultraviolet(UV)B-induced oxidative damage in human HaCaT keratinocytes. Therefore in this paper, we investigated the anti-oxidative capacity and protective effect of LJF against UVB-induced oxidative demage in human HaCaT keratinocytes. Methods : To evaluate the anti-oxidative activity of LJF extracts, we measured total phenolic contents, total flavonoid contents, antioxidant capacity, and superoxide scavenging activity. To give an oxidative stress to HaCaT cells, UVB was irradiated with $200mJ/cm^2$ to HaCaT cells. To detect the protective effect of LJF against UVB, we measured cell viability, DNA fragmentation and reactive oxygen species (ROS) production. In addition, we performed high-performance liquid chromatography (HPLC) analysis to find a major component of LJF. Results : LJF contained phenolic and flavonoid contents, and showed the anti-oxidant and superoxide scavenging activity. The UVB-induced oxidative conditions led to the cell death, DNA fragmentation and reactive oxygen species (ROS) production. However, pretreatment with LJF reduced oxidative conditions, including inhibition of cell death, DNA fragmentation and ROS production. In addition, we found out chlorogenic acid as major component of LJF. Conclusions : These results could suggest that LJF contained anti-oxidative contents and exhibited protective effects against UVB on human HaCaT keratinocytes. And the effective compound of LJF which could show protective activities against UVB is chlorogenic acid. Thus, LJF and chlorogenic acid would be useful for the development of drug or cosmetics treating skin troubles.
