  • 키틴의 글루코사민 단위에 클로로프로판, 산화프로필렌 그리고 클로로프로판 디올을 반응시켜 프로필 키틴(PPC), 히드록시프로필 키틴(HPC), 디히드록시프로필 키틴(DHPC)등의 에테르 형태의 키틴 유도체를 합성하였고, 이들을 99% 포름산에 용해시켜 고분자 농도 30 wt%이상에서 콜레스테릭한 유방성 액정을 형성하였다. 액정이 형성된 용액으로부터 필름을 제조하여 라이소자임이 포함된 유사 체액, pH 1.2, pH 6.7 그리고 pH 8.2 용액에서 in vitro 분해정도를 관찰한 결과 처음 1주일에서 급격하게 분해되었으며 중성 영역에서 큰 무게 감소와 기계적 강도가 감소하였다. 세가지 시료들 중 DHPC가 가장 좋은 생분해 정도를 나타냈다.

게껍질로부터 CM-Chitin의 합성 (Synthesis of CM-Chitin from Crab Shells)

  • Chitin was isolated from crab shells by modified Hackman's method, which was treated again with aqueous sodium hydroxide solution to give alkali chitin(sodium alkoxide of chitin). And after, the alkali chitin is allowed to react with monochloroacetic acid to synthesize 6-O-(carboxymethyl) chitin [CM-Chitin] under diminished pressure. In order to synthesize high performance CM-Chitin as hygroscopic agent, it was measured that the yield and degree of subsititution of each CM-Chitin according to molar ratio(monochloroacetic acid equivalent mol/N-acetyl-D-glucosamine residue) and their moisture-absorption and moisture-release properties were measured and compared with those of hyaluronic acid. The moisture-absorption and moisture-release properties of CM-Chitin, especially 0.8 in degree of substitution, were found quite similar to those of hyaluronic acid. The preliminary results show that CM-Chitin might be used as hygroscopic agent instead of hyaluronic acid in field of cosmetics.

Interacting Domain Between Yeast Chitin Synthase 3 and Chitin Synthase 4 is Involved in Biogenesis of Chitin Ring, but not for Cell Wall Chitin

  • Recently, we identified a domain, termed MIRC3-4, for the protein-protein interaction between yeast chitin synthase 3 (CHS3) and chitin synthase 4 (CHS4). In this study, the functional roles of MIRC3-4 were examined at the G1 phase and cytokinesis of the cell cycle by Calcofluor staining and FISH. Some mutations in MIRC3-4 resulted in disappearance of the chitin ring in the early G1 phase, but did not affect chitin synthesis in the cell wall at cytokinesis. The chitin distribution in chs4 mutant cells indicated that CHS4 was involved in the synthesis of chitinring in the G1 phase and in the synthesis of cell wall chitin after cytokinesis, suggesting that Chs4p regulates chitin synthase 3 activity differently in G1 and cytokinesis. Absence of the chitin ring could be caused either by delocalization of Chs3p to the bud-neck or by improper interaction with Chs4p. When mutant cells were immunostained with a Chs3p-specific antibody to discriminate between these two alternatives, the mutated Ch3p was found to localize to the neck in all MIRC3-4 mutants. These results strongly irdicate that Chs4p regulates Chs3p as an activator but not a recruiter.

  • DEAE-chitin은 alkali-chitin 용액에 DEAE HCl을 이용하여 제조하였다. 제조된 DEAE-chitin은 물과 유기용매에 대한 향상된 용해성을 나타내었다. DEAE-chitin은 chitin의 탈아세틸화반응보다 온화한 조건인 sodium borohydride를 함유하고 있는 10% 수산화나트륨 수용액으로 $80^{\circ}C$에서 9시간동안 탈아세틸화반응을 행하여 DEAE-chitosan을 제조하였고, DEAE-chitin의 양이 온성을 증가시키기 위해 ethyl halide를 이용하여 4차화시킴으로써 4차 암모늄기를 가진 TEAE-chitin유도체를 각각 합성하였다. 이들 합성유도체들의 반응에 따른 구조변화는 FTIR, $^1H$ NMR 등으로 확인하였고, 카올린 현탁액을 이용한 응집실험결과 pH가 약 알칼리일 때, 그리고 수지농도가 8ppm일 때 최적의 응집성능을 나타냄을 알 수 있었다.

  • When rats are exposed to cadmium in their diet, the cadmium accumulates in the liver. This study was carried out to investigate the effect of chitin, chitosan and dithiocarbamate chitosan on the cadmium accumulation in liver of rats. For this experiment, 10 male Sprague-Dawley were used. The experimental groups were divided into four independent groups which were one control group and three experimental groups by cadmium alone treatment or chitin, chitosan and dithiocarbamate chitosan with cadmium. In order to investigate the eliminative effect of chitin, chitosan and dithiocarbamate chitosan on the cadmium accumulation in liver of rats, the cadmium concentration, the metallothionein level and the enzyme activitys were measured. The results obtained revealed the eliminative effect of cadmium in liver of rats by chitin, chitosan and dithiocarbamate chitosan. The effect of chitin on the cadmium elimination was less than that of chitosan and dithiocarbamate chitosan. Also it shows that the eliminative effect of cadmium by dithiocarbamate chitosan was the highest.

Chitin.Chitosan이 흰쥐의 콜레스테롤 대사에 미치는 영향 (Effects of Chitin and Chitosan on Lipid Metabolism in Rats)

  • The effects of chitin and chitosan on the contents of plasma and liver lipids and the fecal excretion of neutral and acidic steroids were investigated in rats fed with atherogenic diet. Dietary chitin did not affect plasma total cholesterol, LDL cholesterol and HDL cholesterol, while chitosan diet de creased total cholesterol and LDL cholesterol. Plasma content of triglyceride was reduced signif icantly by chitin diet but not by chitosan. Increases in fecal weight were observed in rats fed with either chitin or chitosan, and the increase was greater with chitosan than with chitin. Dietary chitosan resulted in an increase of fecal excretion of neutral steroids without any changes in the excretion of acidic sterols. Putting the results together, dietary chitosan reduces plasma cholesterol probably due to the reduction of cholesterol absorption. The decrease in plasma triglyceride level by dietary chitin requires further research to understand the mechinism.

  • Chitin, a linked polysaccharide composed of 2-acetamido-2-deoxy-$\beta$-D-glucopytanose residues, is distributed widely in nature. It has been utilized on various application field due to the development of chitin derivatives such as chitosan, partial deacetylated chitin, carboxylmethyl chitin, sulfated chitin, and so on. Chitin and chitosan have been recently interested in antitumor and antimicrobial activities, because of a powerful tumor inhibitory effect against experimental mouse tumors. Especially, the oligosaccharides obtained by partial degradation of them exhibited a remarkable antitumor effect against sarcoma 180, MM 48 and Meth Asolid tumors and antimetastatic effect against Lewis lung carcinoma in mice. This review describes on antitumor effects of chitin, chitosan and their oligosaccharides by their mechanism of action involving enhancement of immunological system.

갑각류인 꽃게 껍질을 이용한 키틴/키토산의 제조에 관한 연구 (A Study on the Preparation of Chitin/Chitosan Using Protunus Trituberculatus Shells Such as Crustacea)

  • Chitin/chitosan be known as biodegradable natural polymer. However, commercial use of chitin has been limited due to highly resistance to chemicals and the absense of proper solvents. Therefore, we was studied that chitin was prepared by the application of Hackman's method from Protunus trituberculatus shells. And another viscosity chitosan were prepared from chitin which were deacetylated under various concentration of alkali, reaction time and temperature by the application of Mima's method. And crosslinked chitin/chitosan was preparaed from chitin/chitosan with crosslink agent followed by crosslinkage. The major parameters for chitosan manufacturing methods were found to be concentration of alkali solution, reaction time and temperature etc. The effects of these parameters on chitin, another viscosity(molecular weight) chitosan and crosslinked chitin/chitosan were investigated by various analysis apparatus.

Extraction of high thermally stable and nanofibrous chitin from Cicada (Cicadoidea)

  • Due to the increasing interest in natural biopolymers including chitin, the exploitation of economic and easily accessible chitin sources with good physicochemical properties is nowadays required. In view of this fact, in the current study chitin was extracted and physicochemically characterized from six Cicadas (Hemiptera: Homoptera: Cicadoidea) species collected from Mediterranean region of Turkey (2014-15). Chitin was extracted using a classic extraction method that includes acid and base treatment. TGA results revealed a remarkable increase ($410-412^{\circ}C$) for all the six Cicada species compared to other chitin samples extracted from various sources. For all of the six selected species the chitin contents on the dry basis were determined as 6.7% for Cicadatra atra, 5.51% for C. hyalina, 8.84% for C. platyptera, 4.97% for Cicada lodosi, 6.49% for C. mordoganensis, and 5.88% for Cicadetta tibialis. The surface morphology of chitin isolates from Cicada species was observed to consist of nanofibers and nanopores.

Inhibition of Chitin Sulfate on Human Low Density Lipoprotein(LDL) Oxidation by Macrophages

  • 본 연구는 동맥경화의 원인으로 알려진 사람 oxidized low density lipoprotein (LDL)에 대한 chitin sulfate의 산화억제 효과에 대하여 실험하였다. 사람 LDL을 쥐의 마크로파지 유도 oxidized LDL로 산화시킬 때 chitin sulfate를 첨가하여 TBARS와 전기영동에 의한 이동상 및 $O_2$소거 기능을 확인하였다. 사람 LDL에 macrophage를 이용해서 LDL을 산화시킬 때 chitin sulfate를 50~100 $\mu$M 첨가하였을 때 억제 효과가 높았으며, chitin sulfate를 100 $\mu$M 첨가하였을 때 LDL의 산화가 거의 억제되었고, 전기영동에 의한 이동거리도 100 $\mu$M chitin sulfate에서 완전히 억제되었다. 그리고 phorbol myristate acetate를 처리한 마크로파아지 유도 활성 산소의 소거 효과는 chitin sulfate의 농도가 100$\mu$M일 때 거의 소거하였다. 이상의 결과로 보아 chitin sulfate는 동맥경화의 예방에 효과적이라는 결론을 얻었다.
