Effect of Photoperiod, Temperature and True-leaf Stage in Bolting Rate of Chicory (Cichorium intybus L. var. sativus)

  • Root chicory (Cichorium intybus L. var. sativus) is potential alternative medicinal and sugar crop which accumulates a high amount of linear polyfructan, inulin in its roots. A problem in root production is that over-wintered stock plants often flower. Once the plant becomes reproductive, stem elongation and root growth slows and floral buds arise from every node, rendering the plants useless for propagation. The objectives of this research was to examine the effectiveness of manipulating environmental factors containing photoperiod, temperature and number of leaf states. The experiment was performed in growth chamber to create two photoperiods (8 h, and 16 h) with three temperature regimes (5$^{\circ}C$/3$^{\circ}C$, 1$0^{\circ}C$/8$^{\circ}C$ and 15$^{\circ}C$/13$^{\circ}C$ day/night temperature) for a total of six treatments on three type of true-leaf stage of plant. Data of bolting rate, shoot and root length, shoot and fresh weight was invetigated in each treatments. This is the first report on changes in bolting rate and shoots and roots production during a whole growing season and differences in the effect of cold and photoperiod treatment depending on the true-leaf stage of plant.

Production of Inulo-oligosaccharides from Chicory(Cichorium intybus, L.) with Endo-inulinase from Arthrobacter sp.537 (새로운 endo-inulinase를 이용한 치커리 추출물로부터 Inulo올리고당의 생산)

  • For the effective production of functional oligosaccharides(DP 3-5) from inulin in chicory extracts, the acid hydrolysis and enzymatic endo-inulinase reaction were compared. Acid hydrolysis was unfavorable ; the content of oligosacharides in total sugar increased to 26.0% for 12 min at $55^{\circ}C$ and 24.6% at 6 min at $65^{\circ}C$ and showed little change for 30 min. The content of high DP(DP 6) decreased from 83.5 to 49.5% and 23.0% for 30 min, repectively. Glucose, fructose and sucrose increased to 24.6% and 50.3%, respectively. Hydrolysis of chicory extracts with purified endo-inulinase from Arthrobacter sp. S37 was carried out at $40^{\circ}C$ and pH 7.5 for 44 hrs. The content of high DP($DP{\geq}6$) in total sugar decreased from 83.5 to 23.0% and that of inulobiose(F2) and DP 3-5 increased to 66.1%. Glucose, fructose and sucrose were not produced. The hydrolysis of chicory extracts without DP 1 and DP 2 with crude or with purified enzyme were also carried out. In contrast to the hydrolysate of crude enzyme, that of purified endo-inulinase did not contain glucose, fructose, sucrose, F2 and 1-kestose(GF2). The content of oligosaccharides in the hydrolysate of the purified endo-inulinase were 79.2%, composed mainly of inulotriose(F3), inulotetraose(F4) and inulopentaose(F5), which shows that the enzymatic hydrolysis using purified endo-inulinase from Arthrobacter sp. S37 is the best method for oligosaccharides production from inulin in chicory extracts.

  • The cycloinulooligosaccharide fructanotransferase (CFTase) gene (cft) from Paenibacillus macerans was subcloned into an E. coli-yeast shuttle vector, pYES2.0, resulting in pYGECFTN. The plasmid pYGECFTN (8.6 kb) was introduced into Saccharomyces cerevisiae SEY2102 cells and then the transformants were selected on the synthetic defined media lacking uracil. The cft gene expression in yeast transformant was demonstrated by the analyses cyclofructan (CF) spots on thin-layer chromatogram. The recombinant CFTase was not secreted into the medium and localized in the periplasmic space. The production of CF was observed after 5 min of the enzymatic reaction with inulin. The optimun pH and temperature for CF production were found to be at pH 8.0 and $45^{\circ}C$, respectively. Enzyme activity was stably maintained up to $55^{\circ}C$. The CF was produced from all inulin sources and was most efficiently produced from dahlia tubers and Jerusalem artichokes.

Optimization of the Viability of Probiotics in a Fermented Milk Drink by the Response Surface Method

  • Growth promoters were added to skim milk to retain the viability of Lactobacillus acidophilus and Bifidobacterium longum to help the product meet the "therapeutic minimum" at the time of consumption. The experiments were divided into two parts. The first part of the study used chicory inulin, isomalto-oligosaccharides and sucrose to investigate the effects of sugars on the activity of L. acidophilus and B. longum. The results indicated that the addition of isomalto-oligosaccharides stimulated growth of L. acidophilus and B. longum, resulting in a higher level of the probiotics after one month storage and yielded better $\beta$-galactosidase activity during fermentation. The second part studied the effects of three growth promoters on the viability of the probiotic cultures and the response surface method was employed to find the optimal ratio for addition of the growth promoters. The optimal ratio for added calcium gluconate, sodium gluconate and N-acetylglucosamine in fermented milk drinks were established. The response surface method proved to be a very effective way of optimizing the activity of probiotic cultures when developing a new fermented milk drink.

Studies in Quantitative Analysis of Inulin-type and Levan-type Fructan in some Korean Foods (국산 식품에서 이눌린타입과 레반타입 플럭탄 정량분석 연구)

  • Fructan, a fructose homopolymer, is found in various foods, including onion, garlic, chicory, Jerusalem artichoke, banana, and Cheonggukjang. This study aimed to quantitatively analyze both levan-type and insulin-type fructan using acid analysis and enzyme treatment. In order to analyze fructan contents, we applied optimized conditions to various fructan-rich foods using products from 2017. In the case of oxalic acid hydrolysis, fructose concentrations increased as oxalic acid concentration increased. Inulinase treatment was better than invertase treatment in terms of fructose liberation from fructan. We applied three different methods to fructan-rich foods, including onion, garlic, banana, and Cheonggukjang and found that fructose released from fructan-rich foods was the highest in oxalic acid hydrolysis among three different methods. Except for Cheonggukjang, inulinase treatment produced better results in terms of fructose contents than invertase treatment. From our study, estimated daily fructan intakes by Koreans were 1,172~3,402 mg from onion and garlic. In conclusion, we believe that information on fructan-rich foods may be helpful to understand their roles in the human digestive system.

  • Dietary fiber is defined as soluble and insoluble polysaccharide consisted in the plant cell wall-associated fibers naturally occurring in fruits, vegetables, and cereal products, and of isolated fibers that are added to processed foods which are also artificially modified. There are so many difference types of dietary fibers as arabinoxylan, polydextrose chicory, oligosccharide. inulin, pectin, bran, cellulose, ${\beta}$-glucan, resistant starch and some seaweed polymers as alginate. Most of them provide many biological benefits in the intestine, as lower risk for developing coronary heart disease, stroke, hypertension, diabetes, obesity and some of the gastrointestinal disease like as colon cancer. And also lowering cholesterol levels, improves glycemic and insulin sensitivity to non-diabetic and diabetic persons including immune system. Beside of many benefits, average consumers in developed and under developing countries take far less amounts of dietary fiber that international organization recommended. Adequate intake of dietary fiber is 14g/1,000kcal base using the energy guide line of 2,000kcal/day for women and 26,000 kcal/day for men, dietary intake is 28g/day of adult women and 36g/day for adult men. The mechanisms behind the reported effects of dietary fiber on metabolic health are not fully well established. It is suggested that changes in intestinal viscosity resulting mucus increasing, macro-nutrients absorption, rate of passage of large intestinal, production of short chain fatty acids by fermentation. Production of gut hormones and changes of microbiota in intestine. It is necessary to do more research in this field in the future and combined interdisciplinary works together.

Effect of Prebiotics on Intestinal Microflora and Fermentation Products in Pig In Vitro Model

  • The objective of this study was to evaluate the effect of the different types and levels of prebiotics on intestinal microflora and fermentation products in the in vitro fermentation model. The prebiotcs used in this study were IMO (iso-malto oligosaccharide), CI (partially digested chicory-inulin), RA (raffinose) and CD (cyclodextrin). Experimental diet for growing pigs was predigested by digestive enzymes and this hydrolyzed diet was mixed with buffer solution containing 5% fresh swine feces. Then, the mixture was fermented with or without prebiotics at the concentrations of 0.5 and 1.0% for 24 h. Samples were taken at 24 h, and viable count of micoflora, gas, pH, volatile organic compounds and short-chain fatty acids were determined. The viable count of Enterobacteriaceae was significantly decreased (p<0.001) in all treatments added with prebiotics in comparison to control without prebiotics. However, the increase of lactic acid bacteria was observed in the prebiotics treatment. Gas production increased as the level of prebiotics increased. The pH values in the fermentation fluid decreased in a dose-dependent manner with increasing the concentration of prebiotics. The fermentation with prebiotics resulted in the reduction of malodorous compounds such as ammonia, hydrogen sulfide, indole and skatole. The increase in short-chain fatty acid (SCFA) production was observed in the treatments with prebiotics. In conclusion, the results of this study demonstrated that the fermentation with prebiotics was effective in reducing the formation of malodorous compounds and increasing lactic acid bacteria and SCFA. These effects depended on the concentration of prebiotics. Moreover, further study is needed to determine whether the in vitro efficacy on the reduction of malodorous compounds and increase of SCFA would also be observed in animals.

Effects of Prebiotics and Probiotics on Swine Intestinal Microflora and Fermentation Products In Vitro Fermentation (In vitro 발효에서 Prebiotics와 Probiotics가 돼지 장내미생물과 발효산물에 미치는 영향)

  • In the present study, the effects of prebiotics and prebiotics+probiotics on intestinal microflora and fermentation products were evaluated in a pig in vitro fermentation model. The substrates used in this study were iso-malto oligosaccharide (IMO), partially digested chicory-inulin (CI), raffinose (RA), and cyclodextrin (CD) as prebiotics and Lactobacillus reiteri as probiotics. For a pig in vitro fermentation, the experimental diet for growing pigs was predigested using digestive enzymes secreted by small intestine and this hydrolyzed diet was mixed with a buffer solution containing 5% fresh swine feces. The mixture was then incubated with either prebiotics or prebiotics+probiotics for 24 h. Samples were taken at 24 h, and viable counts of microflora, gas, pH, volatile organic compounds (VOCs) and short-chain fatty acid (SCFA) were analyzed. The viable count of Enterobacteriaceae was significantly decreased (p<0.001) in all treatments containing prebiotics and prebiotics+probiotics when compared to the control. However, the number of lactic acid bacteria increased in the prebiotics and prebiotics+probiotics treatment. The pH values in the fermentation fluid decreased in all treatments when compared to the control, and their effects were greater in the prebiotics+probiotics group than prebiotics group. Fermentation with prebiotics resulted in a reduction in malodorous compounds such as ammonia, hydrogen sulfide and skatole when compared to the prebiotics+probiotics group. Short-chain fatty acid production was also higher for treatment with prebiotics+probiotics than treatment with prebiotics. In conclusion, the results of this study demonstrated that fermentation with prebiotics was effective in reducing the formation of malodorous compounds and prebiotics+probiotics was effective in increasing lactic acid bacteria and SCFA and reducing the pH. Moreover, further studies will be needed to determine whether the results observed in the in vitro model would occur in pigs that ingest these prebiotics or probiotics.
