This experiment was performed to study the effects of radioactive phosphorus($^{32}p$) on the growth of chick embryo and young chick. Radioactive phosphorus($^{32}p$) was administered into the yolk sac of chick embryo in doses of 2 uci/gm and 1 uci/gm and was administered intraperitoneally to the young chick in doses of 1 uci/gm and 0.5 uci/gm. The chick embryos were sacrificed on 6th, 7th, 8th, 9th and 10th day and the chicks were sacrificed on 1st, 3rd, 6th and 10th day after the administration and the liver, kidney, spleen, brain, eye ball and femur were weighed to observe the effects of growth inhibition on them. The results obtained were as follows. 1. A marked growth inhibitory effect was found on 8th, 9th and 10th day after the administration of $^{32}p$ in chick embryos and the same effect was found on 6 th and 10 th day after the administration in chicks. 2. The growth of the liver, kidney, spleen and femur was inhibited markedly but the brain and eye ball were not affected in chick embryos and chicks.
Pathological and virological investigations were conducted on suspected outbreaks of infectious bursal disease (IBD) in a broiler farm and five pullet-raising poultry farms of Mymensingh and Tangail districts of Bangladesh. About 80 to 100 percent chicks were affected at the age of 26 to 45 days and mortality varied from 20 to 30 percent in broilers and 40 to 80 percent in layer chicks. Signs, symptoms, gross and microscopic lesions were typical of acute IBD. Several isolates of virus could be obtained by embryo inoculation and the virus was diagnosed as infectious bursal disease virus (IBDV) by agar gel immunodiffusion test (AGID). The virus isolate belonged to the very virulent pathotype of IBDV causing 100 percent mortality in three weeks old chicks on experimental infection.

이 연구의 목적은 주입구의 위치에 따른 병아리 배아의 생존율을 서로 비교하고 가장 적합한 주입구 위치를 찾기 위하여 실시되었다. 멸균처리된 핀셋을 사용하여 난각의 첨단부와 둔단부 그리고 옆부분에 주입구를 각각 만들었다. 연구 결과, 둔단부에 주입구 (BE1)를 만든 수정란의 발생율이 가장 높았으나 내부난각막이 불투명하여 혈관내 미세주입이 어렵다. 따라서 본 연구에서는 첨단부에 주입구를 만든 다음 이 주입구를 통하여 약 2 $\mu$L의 DMEM 용액을 2.5일령된 배자의 혈관에 주입하였고, 주입부위의 출혈을 막기 위해 DMEM 용액을 주입한 후 공기방울을 넣은 결과 생존율이 약 17.0% 이었다. 따라서 이러한 주입구에 의한 방법은 생식세포가 조작된 germline chimera 또는 형질전환닭을 생산하는데 매우 유용한 시스템으로 이용될 수 있을 것이다.
닭의 성장과정에서 어린 병아리의 발달에 가장 결정적인 시간으로 인식되는 시점은 부화 전 수일부터 부화 후 수일간이다. 부화 전후의 충분한 영양소 공급은 배아의 최종 발달과 병아리의 초기성장에 결정적 기여를 한다고 볼 수 있다. In ovo feeding에 의한 탄수화물 또는 아미노산의 공급은 부화 전 후의 소화장기 발달에 긍정적으로 작용한다. Prebiotics과 synbiotics 등에 대한 in ovo 주입은 건강한 미생물 균총 형성 등을 제공하여 영양적, 면역적 측면에서 좋은 결과들을 볼 수 있지만, 연구 결과의 수적 제한과 in ovo 주입의 양과 주입 부위 및 주입시기 등에 대한 추가적인 연구가 요구된다. 탄수화물을 이용한 in ovo feeding은 부화시 체중 증가를 보이지만 육계의 부화 후 능력에는 크게 영향을 보이지 않으며, 탄수화물 주입에 따른 부화율 감소는 기술적 요인(최적 농도, 주입 장소, 부피 등)이 크게 작용하는 것으로 보인다. In ovo feeding에 의한 아미노산 공급은 병아리의 세포성 면역과 체액성 면역을 높여 초기 면역 향상에 도움이 되는 것으로 판단된다. In ovo feeding은 부화 전후의 최적의 영양소 공급과 면역력 증진 및 절식에 따른 병아리의 스트레스를 완화시켜주는 실용적인 기술이 될 수 있다.
Monochromatic green light-emitting diodes (LED) light stimuli influences the posthatch growth performance of chicks. This study was undertaken with the following objectives: i) to examine whether the green LED light stimuli induces an overheating effect by determining weight loss rate of fertile eggs during incubation period; ii) to look for the development of eyes and other primary organs at different ages of embryos and newly hatched chicks. Arbor Acres fertile broiler eggs (n = 480) were randomly assigned to 3 incubation groups and exposed to continuous white light, green light, or a dark environment (control) from the first day to 19 d of incubation. The light sourced from LED lamps with the intensity of 30 lx at eggshell level. The results showed that either green or white light stimuli during incubation did not significantly affect the weight loss rate of fertile eggs, hatching time, hatchability, chick embryo, or body weight (BW), the weight percentage of heart, liver, and eyes, as well as obvious systematic abnormalities in eye weight, side-to-side, back-to-front, or corneal diameter from 15 d of embryogenesis to 6 d of posthatch (p>0.05). Compared with the dark condition, green light stimuli during incubation tended to increase feed intake (p = 0.080), improved the BW gain of chicks during 0 to 6 day posthatch (p<0.05), and increased the percentage of pectoral muscle to the BW on 3- and 6-day-old chicks. In addition, embryos or chicks in green light had lower weight percentage of yolk retention on 19 d of embryogenesis and 1 d of posthatch in comparison to those in dark or white group (p<0.05). These results suggest that providing 30 lx green LED light stimuli during incubation has no detrimental effect on the development of eyes, heart and liver of embryos and hatchlings, but does have potential benefits in terms of enhancement of the chick growth during the early posthatch stages. In addition, the fertile broiler eggs stimulated with 30 lx green LED light during incubation does not cause an overheating effect.
The objective of these experiments was to develop an attenuated thermostable Newcastle disease virus(NDV), CBP-1 strain isolated from infected pheasants. Safety, pathogenicity and protective effects against velogenic NDV were also investigated to evaluate if the attenuated NDV, CBP-1 strain could be a candidate for a new NDV vaccine strain. CBP-1 strain was passaged up to the 173 times by nine days old embryonated eggs and chicken embryo fibroblast(CEF) cell cultures. Its sensitivitly to lipid solvents and low pH, thermostability, mean death time(MDT), intracerebral pathogenicity index(ICPI) of one day old chicks and intravenous pathogenicity index(IVPI) of four weeks old chicks were examined. Safety, boosting and protective effects were tested by chicks mortality. CBP-1 NDV strain had significant thermostability at 56$\^{C}$ for 30 minutes. by hemagglutinin activity and egg infectivity test, but was not resistant to lipid solvent. It showed possibility to use as a feed or water vaccine because of the resistance to low pH. MDT, ICPI and IVPI of CBP-1 were attenuated from 51.5, 1.96, 2.60 to 112.4, 1.12, 1.45. These results implied that the 173rd passages in embryonated egg and CEF cell cultures induced a substantial attenuation of the pathogenicity of the parent virus, changing the virulence from velogenic to intermediate between mesogenic and lentogenic. After vaccination with CBP-1 at one day old by drinking water mortality was 17.5%. However, spray vaccination with B1 at one day old, CBP-1 at two weeks ild and challenge with velogenic Kyojeongwon strain at four weeks old showed 93.5% survival rate. Mortality of chicks, vaccination with 173rd passaged CBP-1 strain at one day old, two weeks old and challenge with Kyokeongwon strain at four weeks old, was 20.0%. The results of these studies indicated that partial attenuated CBP-1 strain tended to be a low safety for ND of broiler chicks and would need to be more successive attenuation.
It has been recognized that the hen, like its mammalian counterparts, provides young chicks with antibodies as protection against hostile invaders. This system facilitates the transfer of specific antibodies from serum to egg yolk, and provides a supply of antibodies called immunoglobulin Y(IgY) to the developing embryo and the hatched chick. The protection against pathogens that the relatively immune-incompetent newly hatched chick has, is through transmission of antibodies from the mother via the egg. Egg yolk, therefore, can be loaded with a large amount of IgY against pathogens which can immobilize the existing or invading pathogens during the embryo development or in day-old chicks. Thus, the immunization of laying hens to various pathogens results in production of different antigen-specific IgY in eggs. Egg yolk contains 8∼20 mg of jmmunoglobulins (IgY) per ml or 136∼340 mg per yolk suggesting that more than 30 g of IgY can be obtained from one immunized hen in a year. By immunizing laying hens with antigens and collecting IgY from egg yolk, low cost antibodies at less than $10 per g compared to more than $20,000 per g of mammalian IgG can be obtained. This IgY technology opens new potential market applications in medicine, public health, veterinary medicine and food safety. A broader use of IgY technology could be applied as biological or diagnostic tool, nutraceutical or functional food development, oral-supplementation for prophylaxis, and as pathogen-specific antimicrobial agents for infectious disease control. This paper has emphasized that when IgY-loaded chicken eggs are produced and consumed, the specific antibody binds, immobilizes and consequently reduces or inhibits the growth or colony forming abilities of microbial pathogens. This concept could serve as an alternative agent to replace the use of antibiotics, since today, more and more antibiotics are less effective in the treatment of infections, due to the emergence of drug-resistant bacteria.

Objective: The clustered regularly interspaced short palindromic repeat (CRISPR)/CRISPR-associated protein 9 (Cas9) system, which is the most efficient and reliable tool for precisely targeted modification of the genome of living cells, has generated considerable excitement for industrial applications as well as scientific research. In this study, we developed a gene-editing and detection system for chick embryo sexing during the embryonic stage. Methods: By combining the CRISPR/Cas9 technical platform and germ cell-mediated germline transmission, we not only generated Z chromosome-targeted knockin chickens but also developed a detection system for fluorescence-positive male chicks in the embryonic stage. Results: We targeted a green fluorescent protein (GFP) transgene into a specific locus on the Z chromosome of chicken primordial germ cells (PGCs), resulting in the production of ZGFP-knockin chickens. By mating ZGFP-knockin females (ZGFP/W) with wild males (Z/Z) and using a GFP detection system, we could identify chick sex, as the GFP transgene was expressed on the Z chromosome only in male offspring (ZGFP/Z) even before hatching. Conclusion: Our results demonstrate that the CRISPR/Cas9 technical platform with chicken PGCs facilitates the production of specific genome-edited chickens for basic research as well as practical applications.
Attempts to isolate chicken anemia agent (CAA) were made by inoculating tissue homogenates into MDCC-MSBl or LSCC-1104B1 cell lines and passaging the cells serially. CAA was isolated from the liver and thymus of 11 weeks old layer chickens and from the liver of 10 weeks old broiler breeder chickens. The layer flock experienced approximately 45% mortality during 9 to 14 week of age from gangrenous dermatitis and lymphoid organs of affected chickens were severely atrophied. The broiler breeder flock experienced approximately 7% mortality during 7 to 9 weeks of age and affected birds showed lesions of colibacillosis, staphylococcal arthritis, and coccidiosis together with atrophied lymphoid organs. The isolated viruses were identified as CAA by the indirect fluorescent antibody test and virus neutralization test using CAA immune sera including one to Gifu-1 strain of CAA. The CAA isolate 89-69, when inoculated into susceptible 1 day old SPF chicks, induced anemia 14 to 16 days after inoculation. It did not induce any cytopathic effects in chicken embryo liver and chicken embryo fibroblast cell cultures. Infectivity of the isolate was not affected by the treatment of chloroform or heat ($70^{\circ}C$ for 15 minutes).

Reovirus의 감염성 측정시 계 태아섬유아세포 및 가세포배양과 Vero cell 배양에서 세포분주와 동시에 바이러스를 접종하거나 단층세포가 형성된 후접종하거나 간에 별다른 차이없이 감염 4-5일후에 end point에 도달되었다. 분리된 reovirus를 계태아간세포배양에 5-10대 계대배양한 후 $10^{5.0}$ TCI $D_{50}$의 바이러스를 10일령 발육계란의 장요막상에 접종하였을 때 평균치사시간이 국내분리주는 54-59시간인데 반하여 S-1133주는 73시간으로 약간 길었다.
