Isolation and characterization of cultured chicken oviduct epithelial cells and in vitro validation of constructed ovalbumin promoter in these cells

  • Objective: Transgenic hens hold a great promise to produce various valuable proteins. Through virus transduction into stage X embryo, the transgene expression under the control of constructed chicken ovalbumin promoters has been successfully achieved. However, a validation system that can evaluate differently developed ovalbumin promoters in in vitro, remains to be developed. Methods: In the present study, chicken oviduct epithelial cells (cOECs) were isolated from oviduct tissue and shortly cultured with keratinocyte complete medium supplemented with chicken serum. The isolated cells were characterized with immunofluorescence, western blot, and flow cytometry using oviduct-specific marker. Chicken mutated ovalbumin promoter (Mut-4.4-kb-pOV) was validated in these cells using luciferase reporter analysis. Results: The isolated cOECs revealed that the oviduct-specific marker, ovalbumin protein, was clearly detected by immunofluorescence, western blot, and flow cytometry analysis revealed that approximately 79.40% of the cells contained this protein. Also, luciferase reporter analysis showed that the constructed Mut-4.4-kb-pOV exhibited 7.1-fold (p<0.001) higher activity in the cOECs. Conclusion: Collectively, these results demonstrate the efficient isolation and characterization of cOECs and validate the activity of the constructed ovalbumin promoter in the cultured cOECs. The in vitro validation of the recombinant promoter activity in cOECs can facilitate the production of efficient transgenic chickens for potential use as bioreactors.

Effects of Chicken Treated with Hwangki-Beni Koji Sauces on Body Weight, Serum and Hepatic Lipid Profiles of Rats Fed High Fat and High Cholesterol Diets (황기홍국소스를 처리한 계육이 고지방 및 고콜레스테롤 식이 흰쥐의 체중과 혈청 및 간 조직의 지질 함량에 미치는 영향)

  • Dietary effects of chicken treated with Hwangki-Beni Koji sauces (HBS) on serum and hepatic lipid profiles in rats fed high a fat and high cholesterol diet were investigated. Experimental plots (5 rats per each group) were divided into normal control diet group (NC), high fat and high cholesterol diet group (HFC), HFC plus 15% base sauce treated chicken supplemented diet group (HFC-BS), HFC plus 15% HBS-treated chicken supplemented diet group (HFC-HBS), and fed for 5 weeks. Feed intakes in HFC-HBS group were higher than those of HFC and HFC-BS groups, whereas body weight gain and feed efficiency ratio of the HFC-HBS group were lower than those of the HFC and HFC-BS groups. Serum alanine aminotransferase and aspartate aminotransferase activities of HFC-HBS group were significantly lower than those of HFC and HFC-BS groups. Level of serum triglyceride, total cholesterol and LDL-cholesterol in HFC-HBS group were 34.06%, 6.28% and 31.00% lower than those of HFC-BS group, respectively, whereas HDL-cholesterol level and atherogenic index in HFC-HBS group were 1.72-fold higher and 2.52-fold lower, respectively, than those of HFC-BS group. Level of hepatic total lipid, triglyceride and total cholesterol in HFC-HBS group were 9.53%, 5.25% and 22.20% lower than those of HFC-BS group, respectively. In the morphological results of liver, the hepatocytes of central vein lesion and intact hepatic cell plate are preserved well from steatosis in HFC-HBS group, whereas the hepatocytes in HFC and HFC-BS groups are distended by accumulation of multiple fat droplets, and sinusoids are occluded. In the above results, we expected that chicken treated with Hwangki-Beni Koji sauce may have functionalities of anti-obesity, anti-cholesterol and also, lowering anti-atherogenic index in high fat and high cholesterol diets.

The Effects of the High Pressure Boiled Extracts (HPBE) of the Ogol Chicken with Herbs on the Hormones, Cytokine, Specific Antibody of Serum in the Rat (오골계 증탕액 급여가 흰쥐의 혈중 호르몬, Cytokine 및 특이항체에 미치는 영향)

  • This study was conducted to investigate feeding effects of the high pressure boiled extracts (HPBE) of the Ogol chicken with herbs on glucose, hormones and immunological response (cytokine, specific antibody) of serum in the rat which fed either with normal feed (T$_1$), normal feed + herb HPBE (T$_2$), normal feed + Ogol chicken HPBE (T$_3$), normal feed + mixture of cross-bred Ogol chicken HPBE (T$_4$) hydrolyzed with Flavourzyme 0.1% for 35 days. During experimental period, there was a weak trend to have a higher glucose content for the T$_4$ group with 102.27${\pm}$5.95 mg/dL, but it was not significantly higher than other treatments. For insulin level, T$_1$ group showed numerically a slightly higher level with 6.79${\pm}$4.64 ${\mu}$IU/mL, but the difference was not significant in statistic term due likely to a large variation in comparison with other treatments. The treatments did not significantly alter testosterone level in rat plasma with 1.09, 1.46, 0.98, 1.13 ng/mL in T$_1$, T$_2$, T$_3$ and T$_4$, respectively. T$_4$ treatment increased the aldosterone level to a significantly (p<0.05) higher level (273.33 ng/dL) than other treatments. The extract treated rat showed a tendency in the cortisol level of lower levels than the control group, particularly, it was significantly (p<0.05) lower in T$_3$ group than other groups. T$_3$ and T$_4$ groups showed higher levels for interlukin-4 (IL-4) and anti-BSA IgG in immune cells and plasma. T$_2$, T$_3$ and T$_4$ treatments showed a slightly higher levels in v-interferon (INF-r) than the control, with a greater effect for T4 treatments. These results suggested that HPBE of the cross-bred Ogol chicken hydrolyzed with Flavourzyme increased immunological activity and decreased the concentration of cortisol and aldosterone hormones.

Effect of Supplementing Betaine on Performance, Carcass Traits and Immune Responses in Broiler Chicken Fed Diets Containing Different Concentrations of Methionine

  • An experiment was conducted with broiler (Cobb 400) male chicks (n = 480) to determine the effect of betaine (Bet) supplementation (0 and 800 mg/kg) to diets containing five concentrations (15, 18, 20, 22 and 24 g/kg crude protein, CP) of methionine (Met) in a $2{\times}5$ factorial study for performance, carcass traits, immune responses, and serum parameters. Each diet was fed ad libitum from 1 to 42 d of age to 8 replicates of 6 chicks. Birds were housed in battery brooders placed in an open-sided poultry shed. Body weight gain, feed intake, feed conversion efficiency and slaughter variables were recorded at 21 and 42 d of age. Serum biochemical profile, antibody production against Newcastle disease (ND) and lymphocyte proliferation ratio (LPR) were analysed at 42 d of age. Supplementing Bet to diets containing sub-optimal concentrations of Met (15 g/kg) improved weight gain and breast yield at 21 d of age (p<0.01), and feed conversion efficiency at 42 d of age (p<0.05). Feed efficiency at 21 d of age, body weight gain at 42 d of age, slaughter variables except breast yield at 21 d of age and ND antibody titres were not affected (p>0.05) by the interaction. LPR increased (p<0.05) with Bet supplementation at 20 g Met/kg CP equal to those broilers fed 24 g Met without Bet. Bet supplementation enhanced the concentrations of protein, globulin and cholesterol in serum of broilers fed sub-optimal concentrations of Met. Results suggested that Bet supplementation (800 mg/kg diet) enhanced growth (21 d), feed conversion efficiency (42 d), breast yield and lymphocyte proliferation in broilers fed a diet containing 15 g Met/kg CP.

Evaluation of light-emitting diode colors and intensities on slaughter performance, meat quality and serum antioxidant capacity in caged broilers

  • Objective: This study was to evaluate the interaction of three different light-emitting diode (LED) light colors (white, green, and blue) and three intensities (5, 10, and 15 lx) on slaughter performance, meat quality and serum antioxidant capacity of broilers raised in three-layer cages. Methods: A total of 648 (8-days-old) male broiler chicks (Cobb-500) were randomly assigned in 3×3 factorially arranged treatments: three light colors (specifically, white, blue, and green) and three light intensities (namely, 5, 10, and 15 lx) for 35 days. Each treatment consisted of 6 replicates of 12 chicks. The test lasted for 35 days. Results: The semi-eviscerated weight percentage (SEWP) in 5 lx white was higher than that in 15 lx (p<0.01). The eviscerated weight percentage (EWP) (p<0.05) and water-loss percentage (WLP) (p<0.01) decreased in 10 lx white light than those in green light. Under blue light, the content of hypoxanthine (Hx) in muscle was lower than that under white and green light (p<0.01). The content of malondialdehyde (MDA) in 15 lx blue light was higher than that in 10 lx green light (p<0.05). Light color had an extremely significant effect on thigh muscle percentage, WLP, Hx, and crude protein content (p<0.01). Light intensity had a significant effect on SEWP (p<0.05), EWP (p<0.05), lightness (L*) value (p<0.05), WLP (p<0.01), and the contents of superoxide dismutase (p<0.05), MDA (p<0.01), glutathione peroxidase (p<0.01). Conclusion: Using white LED light with 10 lx light intensity can significantly improve the chicken quality of caged Cobb broilers, improve the content of inosine acid in chicken breast and enhance the antioxidant capacity of the body. We suggest that the broiler farm can use 10 lx white LED light source for lighting in 8 to 42 days.

Comparisons of Anthropometric and Environmental Factors, and Food Behaviors of Normocholesterolemia and Hypercholesterolemia in the Postmenopausal Women (정상콜레스테롤혈증과 고콜레스테롤혈증을 가진 폐경 후 여성의 신체적, 환경적 요인과 식행동 비교)

  • Women have a greater incidence of coronary heart disease(CHD) after menopause. It relates to hormone imbalance-induced changes in known CHD risk factors, especially hyperlipidemia. Accordingly, once women are diagnosed, the rate of morbidity and mortality is greater than for men. Thus, preventive strategies are critical to improve the quality oflife in the later years for women. The purpose of ths study was to explore the difference of anthropometric, environmental factors and food behaviors between normocholesterolemia and hypercholesterolemia within postmenopausal women. The subjects were classified as normocholesterolemia and hypercholesterolemia based on the Guideline for Korean Hyperlipidemia. The results obtained are summarized as follows. The distributions of general characteristics such as age, family income, occupation status, age at menarche, and menopausal period were not significantly different between the normocholesterolemia group and hypercholesterolemia group. The anthropometric factors such as body mass index(BMI) and waist hip ratio(WHR) were not significantly different between the normocholesteroemia group and the hypercholesterolemia group but odds ratio of hypercholesterolemia group was higher than that of normocholesterolemia but not significant. Aerobic activity more than 1hours/day was significantly related to low serum cholesterol concentration. Eating behaviors related to regular meal time, regular breakfast habit, and slow meal speed were related to low serum cholesterol concentration. Eating the chicken without skin was significantly related to low serum cholesterol concentration. Our data indicate that desirable eating habit, aerobic activity more than 1hr/day are important factors of serum cholesterol concentrations in postmenopausal women. The result of this study provide information that is important in designing appropriate dietary guidelines for hypercholesterolemia in postmenopausal women.

Albumin Fractions from Different Species Stimulate In Vitro Progesterone Production by Granulosa Cells in Buffalo

  • The ovarian follicular fluid was found to contain steroidogenesis stimulatory protein similar to albumin from human and buffalo. Therefore, the albumins from various species, commercial and purified, were studied for their steroidogenic effect on progesterone secretion by granulosa cells from buffalo ovaries, during culture. A dose of $20{\mu}g$ of bovine serum albumin was optimum to exhibit maximum progesterone secretion on day 6 of culture, in medium ($350{\mu}l$) containing $10^5$ cells. Among commercial albumins, chicken albumin showed highest effect on progesterone secretion, which was followed by albumins from goat, bovine, human, sheep and rat, respectively at day 6 of culture. The albumins were also purified from blood serum of buffalo, goat and rat using salt fractionation, ion-exchange chromatography, gel filtration and SDS-PAGE. The highest stimulatory effect on progesterone secretion was shown by albumin purified from buffalo blood serum and lowest by that from rat blood. Comparatively the buffalo and goat albumins were more biologically active than commercial albumins. The presence of some active molecules conjugated with freshly purified albumins may be responsible for better stimulatory effect.

INFLUENCE OF PHENYLALANINE IN THE MEDIUM ON PROTEIN SYNTHESIS OF CHICKEN EMBRYO FIBROBLASTS

  • The influence of phenylalanine (Phe) in the medium on protein synthesis of chicken embryo fibroblasts (CEF) was examined. CEF was derived from 9-d-old embryos by trypsin-EDTA digestion. To examine the deficiency of Phe in the medium, CEF was cultured in Dulbecco's modified Eagle's medium (DMEM) with or without Phe. CEF was also cultured in Dulbecco's phosphate buffered saline (PBS ($Ca^{2+}$, $Mg^{2+}$)) with or without $400{\mu}m$ Phe in order to examine the effect of Phe supplementation. All media were supplemented with 10% (v/v) fetal calf serum. After incubation for 6, 30 and 54 h, protein synthesis was measured by the incorporation of L-[2, $6-^{3}H$] Phe into CEF for further 18 h. Protein synthesis of CEF cultured in DMEM was higher than that in PBS ($Ca^{2+}$, $Mg^{2+}$). High specific radioactivity of Phe due to the low concentration of Phe in the medium resulted in the apparent increase in protein synthesis of CEF. Protein synthesis cultured in PBS ($Ca^{2+}$, $Mg^{2+}$) with Phe did not increase during 72 h of cell culture.

Role of the Promoter Region of a Chicken H3 Histone Gene in Its Cell Cycle Dependent Expression

  • We fused the promoter region of an H3.2 chicken histone gene, whose expression is dependent on the cell cycle, to the 5' coding region of an H3.3 chicken histone gene, which is expressed constitutively at a low level throughout the cell cycle. This fusion gene showed a cell cycle-regulated pattern of expression, but in a different manner. The mRNA level of the fusion gene increase during the S phase of the cell cycle by about 3.7-fold at 6 h and 2.7-fold at 12 h after the serum stimulation. The mRNA level of the intact H3.2 gene, however, increased by an average of 3.6-fold at 6 h and 8.7-fold at 12 h. This different expression pattern might be due to the differences in their 3' end region that is responsible for mRNA stability. The 3' end of the H3.2 mRNA contains a stem-loop structure, instead of a poly(A) tail present in the H3.3 mRNA. We also constructed a similar fusion gene using a H3.3 histone gene whose introns had been eliminated to rule out the possibility of involvement of the introns in cell cycle-regulated expression. The expression of this fusion gene was almost identical to the fusion gene made previously. These results indicate that the promoter region of the H3.2 gene is only partially responsible for its expression during the S phase of the cell cycle.

  • There are considerable reports that the expression of insulin-like growth factor-Ⅰ (IGF-Ⅰ) is related to ovarian regulation and oviductal development in poultry. Korean Native Ogol Chicken (KNOC) have been inbred to keep a pure line so that there has been limitation in the improvement of egg productivity by genetic studies. Therefore, this study was conducted to investigate the early egg productivity of KNOC pre-selected by IGF-Ⅰ expression. (omitted)
