INFECTIOUS STUNTING SYNDROME OF BROILER CHICKS I. CLINICAL SIGNS AND PATHOLOGICAL LESIONS

  • This study represented an endeavor to observe clinical signs and pathological lesions in broiler chicks suffering from experimental Infectious stunting syndrome(ISS). One hundred and twenty day old broiler chicks were divide randomly into two equal groups i.e. control (A) and inoculated (B). At day one of age each chick of the groups (A and B) was dosed with one ml of either tryptose phosphate broth or prepared inoculum respectively. Chicks of both the groups were housed separately under similar standard management. Inoculation induced characteristic clinical changes in birds of treatment group like of brownish diarrhea, lameness, feather developing problems and paleness of combs, wattles and shanks. By day-29 of the experiment all the stunted birds from group-B and an equal number of birds from group-A were slaughtered. These birds were examined thoroughly to record the gross changes in various structures and then the severely affected organs were processed for histopathological examination. The skeletons of affected birds were brittle, keel bones showed quite prominence while the muscles and subcutaneous tissues were almost devoid of fat. Grossly it was observed that pancreas, spleen and bursa of Fabricius were severely atrophied while the intestines were ballooned with undigested feed and gases. Histopathological examination of pancreas and spleen revealed a classical picture necessary for understanding the pathogenesis of the syndrome. The acivar cells of pancreas were atrophied and underwent vacuolation, degeneration and vecrosis. The zymogen granules were almost absent from the acinar cells. A characteristic change was an inflammatory reaction in one or more pancreatic ducts where the epithelium and fibrous tissues occluded the lumen of the ducts and led to the obstruction in pancreatic drainage.

Heterophil Phagocytic Activity Stimulated by Lactobacillus salivarius L61 and L55 Supplementation in Broilers with Salmonella Infection

  • Newborn chicks are susceptible to Salmonella enterica serovar Enteritidis (SE). The objective of this study was to evaluate the effect of Lactobacillus probiotic isolated from chicken feces on heterophil phagocytosis in broiler chicks. A total of 150 newborn broiler chicks were divided into 5 groups (30 chicks per group) as follows: group 1 (normal control), given feed and water only, group 2 (positive control) given feed, water and SE infection, group 3 (L61 treated) given feed, water, SE infection followed by Lactobacillus salivarius L61 treatment, group 4 (L55 treated) given feed, water, SE infection followed by L. salivarius L55 treatment, and group 5 given feed, water, SE infection followed by L. salivarius L61 + L55 combination treatment. After SE infection, L. salivarius treatment lasted for 7 days. The results showed that L. salivarius L61 and L. salivarius L55 treatment, either alone or combination of both, increased the survival rate after SE infection, and upregulated heterophil phagocytosis and phagocytic index (PI). Conversely, chick groups treated with Lactobacillus showed lower SE recovery rate from cecal tonsils than that of the positive control group. The PI values of the chicken group with SE infection, followed by the combination of L. salivarius L61 and L. salivarius L55 were the highest as compared to either positive control or normal control group. Two Lactobacillus strains supplementation group showed significantly (p<0.05) higher PI value at 48 h than 24 h after treatment.

  • Over 40 per cent Indian population needs to be attended for nutritional improvement. Traditional nutritious food resources though abundantly available but presently less used, if incorporated in the diets, can yield low-cost nutritionally balanced diets. The linear programming (LP)model was used for optimization of food resources to satisfy protein -energy requirements of hard working rural poor. Soybean-a highly nutritious food legume-available at reasonably low price was included along with chick -pea and pigeon -pea for comparison .The three cereals predominantly used in Indian diets -rice, wheat and sorghum -and three typical leafy vegetables namely, spinach , drumstick leaves and rajagira (Amaranths paniculantus) leaves were used for optimization . The contribution of food legume was restricted to supply only 50 per cent of daily protein requirement for maximum protein value. The quantities of other food stuffs namely , milk , sugar , tubers etc. were restricted to nati nal per capita availability. the nutrition satisfaction levels in per cent RDA by the model are 149-250 for protein , 51-106 for fat, 134-362 for iron an d143-1158 for Vitamin-A. Quantities of food legumes, cereals and leafy vegetables ranged from 289-601, 3000-3700 and 360-1200 g/day respectively for a reference family of five hard working rural poor. Daily food resources cost ranged from US$ 0.6 to 1.0 for the family. Amongst food legumes, soybean based diet combinations provide maximum nutritional benefits at minimum cost indicating its suitability for wider adoption by hard working poor.

Partition of Amino Acid Requirements of Broilers between Maintenance and Growth. V. Isoleucine and Valine

  • Two experiments were conducted to subdivide isoleucine (exp. 1) and valine (exp. 2) requirements for maintenance from the requirements for growth of broilers aged 1 to 3 weeks. Purified diets were used, containing five graded levels of isoleucine and valine. Based on weight gain response, the isoleucine requirement for growth was 7.50 mg/g weight gain and the daily isoleucine need for maintenance (mg) was 0.044 per unit metabolic body size ($(Wg^{0.75})$). Based on the N gain response, the isoleucine requirement for growth was 0.317 mg/mg N gain and the daily isoleucine need for maintenance (mg) was 0.040 per unit metabolic body size $(Wg^{0.75})$. Based on weight gain and N gain response, the total isoleucine requirement was calculated 244 mg/day or 0.59% of the diet, 274 mg/day or 0.66% of the diet, respectively. From the relationship of weight gain and N gain, 5.07% of the retained protein was comprised of isoleucine; the reported isoleucine content of chick muscle was 4.42%. The valine requirement for growth was 9.84 mg/g weight gain and 0.36 mg/mg N gain whereas the maintenance requirement was 0.046 or 0.052 mg per unit of metabolic body size (Wgo.11. According to the model developed to estimate valine requirement, the total requirement was 319 mg/day or 0.77% of the diet, 315 mg/day or 0.76% of the diet, respectively. Previous reported valine requirements for growing chicks of 7~24 days old were in close agreement with these estimates. As a percentage of retained protein, valine was calculated to be 5.81% ; the reported valine concentration of crude protein of chicks' body including feathers was 6.72%.

  • Two experiments were conducted to subdivide threonine (exp. 1) and glycine (exp. 2) requirements of broilers into maintenance and growth requirements. Purified diets containing five graded levels of threonine (exp. 1) and glycine (exp. 2) were fed to growing chicks to estimate threonine (exp. 1) and glycine (exp. 2) requirements for growth and maintenance. A model developed to divide threonine requirement for maintenance from that for growth yielded a requirement for growth of 8.946 mg/g weight gain and 0.341 mg/mg N gain; the maintenance requirement was 0.033 or 0.030 mg per unit of metabolic body size $(Wg^{0.75})$. The plateau of plasma threonine concentration occurred at 279.4 mg threonine intake/day. The total threonine requirement was 289.1 mg/day or 0.69% of the diet, 294.1 mg/day or 0.71% of the diet based on weight gain and nitrogen gain responses, respectively. These estimates were in close agreement with previous estimates of threonine requirements. From the relationship of weight gain to N gain, 5.46% of the retained protein consisted of threonine; the reported threonine content of chick muscle was 4.02%. The glycine requirement for maintenance could not be determined due to failure to obtain data allowing extrapolation to zero response. However, ADG increased slightly up to 0.56% glycine.

  • The control of pathogenic microorganisms is important issue in human environments, especially in surface area. However, surface disinfection has not been fully researched. In this study, the surface disinfection under UV irradiation was performed to investigate the quantitative kinetics for Bacillus subtilis spore inactivation in several experimental conditions, such as light intensity, temperature and surface roughness. This study reports that UV light would apparently inactivates the microorganisms and the required IT value for 2 log (99%) Bacillus subtilis spore inactivation was found to be 14.5 $mJ/cm^2$ in plain surface, as predicted by the Delayed Chick-Watson model. When UV was irradiated, there were no significant temperature effects. However, the experimental result shows that the more increased IT values are required at larger surface roughness.

  • To determine the site at which -1 ribosomal frameshifting occurs within the gag-pro overlap of HTL V-I. DNA fragment corresponding to a portion of the gene overlap was cloned into a SP6 vector. The resultant plasmid harbors the hybrid gene consisting of a synthetic gene encoding 5 amino acids derived from chick prelysozyme including the initiator methionine plus 141 nucleotides of gag-pro overlapping region followed by Staphylococcus aurcus protein A gene fragment. In vitro transcription by SP6 RNA polymerase with this DNA template made an abundant amount of single species mRNA. Cell-free translation programmed with the RNA transcribed in vitro yielded a polypeptide of 21 kDal in size. which could be purified into homogeneity by IgG-Sepharose affinity chromatography. In vitro system described in this study must be useful for rapid purification and sequencing of the Gag-Pro transframe protein. allowing to determine the exact frameshift site on mRNA and to identify the tRNA involved in frameshifting event for the expression of pro gene.

A Study on Heating of Hatching Eggs to Improve Hatchability : A Field Study (부화율 향상을 위한 종란의 가열방법에 관한 연구)

  • This study was conducted to evaluate the effects of heating hatching eggs on the number of day-old chicks, egg temperature and egg weight during extended storage, and to provide basic information for improving hatchability to livestock producers. Eggs (Hy-line) were subjected to the following treatments: "control": eggs were maintained in an incubator after storage for 8 days; "T1": eggs were preheated for 8 hours at $23.9^{\circ}C$ after storage for 8 days in a hatchery; "T2": eggs were initially heated for 8 hours at $37.8^{\circ}C$ in an incubator and then preheated for 8 hours at $23.9^{\circ}C$ in a hatchery after storage for 8 days. The results were as follows: First, at the end of the experiment, the total number of day-old chicks was higher in T1, followed by T2 and then the control. This indicated that chick hatchability may be improved when eggs are preheated. Second, compared with the control, the number of day-old female chicks was expected to be higher in treatments with pre-heating; however, the results indicated the opposite effect. Third, as storage time lengthened, the factor that influenced preheating (the main effect and interactions) was not egg weight but egg temperature measured in the upper, middle and bottom parts of incubator. The temperatures recorded in all treatments ranged from 37.97 to $38.40^{\circ}C$ in the upper parts of incubator, 37.80 to $38.26^{\circ}C$ in the middle parts of incubator, and 37.94 to $38.59^{\circ}C$ in the bottom parts of incubator over storage. In conclusion, preheating was very effective in improving hatchability, and egg temperature was the main factor affecting preheating and hatchability.

EFFECT OF LIPOPOLYSACCHARIDE AND INTERFERON-${\gamma}$ ON THE FORMATION OF OSTEOCLAST-LIKE MULTINUCLEATED CELL FROM CHICKEN BONE MARROW CELLS IN VITRO (세포 배양시 닭 골수세포로부터 파골세포앙 세포형성에 지질다당류와 인터페론 감마가 미치는 영향)

  • 파골세포는 조혈기관 단핵의 세포로부터 생성되어 골 홉수에 중요한 역할올 담당하며, 지질다당류는 그람음성균의 세포벽을 이루는 성분으로서 치주질환시 치조골 홉수에 관여한다고 알려져 왔다. 활성화된 림프구, 대식세포와 단핵세포로부터 생성되는 당단백질인 인터페론 감마는 파골세포에 의한 골홉수를 억제한다고 밝혀졌다. 이 연구 논문의 목적은 지질다당류와 인터페론 감마가 닭 골수의 미분화세포가 파골양세포로 전환되는데 어떠한 영향올 주는지를 알아보기 위함이다. 16${\sim}$18 일째의 닭의 배 (chick embryo) 에서 경골을 분리하고 횡절개하여 혈청없는 M-199 배양액에 보관했다. 이것을 9${\mu}m$ filter로 여과시켜서 이미 분화된 파골세포와 기타 다른 분화 세포를 분리했다. 여기에서 파골세포의 전구세포를 얻어 LPS와 IFN-${\gamma}$를 단독 또는 복합처리 하고나서 4일 후에 tartrate resistant acid phosphatase (TRAP) Stain을 시행하고 TRAP 양성이며 핵이 세개 이상인 다핵의 세포형성을 관찰하여 세포를 계수하여 다음과 같은 결과를 얻었다. 1. 닭에서 분리해낸 미분화세포에 0.1. 0.5. 1.0 ${\mu}/ml$ 의 LPS 농도를 처리하고 1 주일간 배양한 결과. 0.1 ${\mu}/ml$ 의 농도에서는 대조군에 비해 TRAP 양성인 파골양세포가 증가하는 경향을 보였으며, 반면에 LPS는 0.5 와 1.0 ${\mu}/ml$ 의 농도에서 세포독성을 보였다.(P<0.05) 2. IFN-${\gamma}$는 50. 500U/ml 의 농도에서 대조군에 비해 TRAP 양성인 파골양세포의 수가 감소하는 경향올 보였다 .3. INF-${\gamma}$는 LPS 에의해 유도된 TRAP 양성인 파골양세포의 형성을 감소시켰고 특히 . 250.500U/ml 의 농도에서 유의 성 있는 감소를 보였다. 위의 결과로부터 LPS는 닭의 골수세포로부터 파골양세포의 형성을 증가시키며 IFN-${\gamma}$는 LPS에의해 유도된 파골양세포수를 감소시킨다는 결론을 얻었다.

Echinostoma revolutum: Freshwater Snails as the Second Intermediate Hosts in Chiang Mai, Thailand

  • The occurrence of 37-collar spined echinostome metacercariae in freshwater snails was investigated in 6 districts of Chiang Mai Province, Thailand, from October 2011 to April 2012. A total of 2,914 snails that belong to 12 species were examined, and 7 snail species (Clea helena, Eyriesia eyriesi, Bithynia funiculata, Bithynia siamensis siamensis, Filopaludina doliaris, Filopaludina sumatrensis polygramma, and Filopaludina martensi martensi) were found infected with echinostome metacercariae. The prevalence of metacercariae was the highest in Filopaludina spp. (38.5-58.7%) followed by B. funiculata (44.0%), E. eyriesi (12.5%), B. siamensis siamensis (8.2%), and C. helena (5.1%). Metacercariae were experimentally fed to hamsters and domestic chicks, and adult flukes were recovered from both hosts at days 15 and 20 post-infection. The adult flukes were identified based on morphological features, morphometrics, host-parasite relationships, and geographical distribution. They were compatible to Echinostoma revolutum or Echinostoma jurini, with only minor differences. As the adults were recovered from both hamsters and chicks, our specimens were more compatible to E. revolutum rather than E. jurini (reported only from mammals). This is the first report for metacercariae of E. revolutum in the snail host, C. helena, and also confirmed that Filopaludina spp., E. eryresi, and Bithynia spp. act as the second intermediate hosts of E. revolutum under natural conditions, which are indigenously distributed in Chiang Mai province.
