Caenorhabditis elegans exhibits sophisticated chemotaxis behavior through two parallel strategies, klinokinesis and klinotaxis, executed entirely by a small nervous circuit. It is therefore suitable for inspiring fast and energy-efficient solutions for autonomous navigation. As a random search strategy, the Lévy walk is optimal for diverse animals when foraging without external chemical cues. In this study, by combining these biological strategies for the first time, we propose a spiking neural network model for search and contour tracking of specific concentrations of environmental variables. Specifically, we first design a klinotaxis module using spiking neurons. This module works in conjunction with a klinokinesis module, allowing rapid searches for the concentration setpoint and subsequent contour tracking with small deviations. Second, we build a random exploration module. It generates a Lévy walk in the absence of concentration gradients, increasing the chance of encountering gradients. Third, considering local extrema traps, we develop a termination module combined with an escape module to initiate or terminate the escape in a timely manner. Experimental results demonstrate that the proposed model integrating these modules can switch strategies autonomously according to the information from a single sensor and control steering through output spikes, enabling the model worm to efficiently navigate across various scenarios.
This paper demonstrates a microfluidic chip incorporating patterned hollow silica beads that can be effectively used for chemotaxis assay. The hollow silica bead has been exploited to develop a carrier for chemoattractant to induce cell migration. The microfluidic chip contains a patterned array of microfabricated docks which can hold only one bead per docking site. The hollow bead placed inside microfluidic chip releases chemotactic reagent (PDGF-BB) around its periphery in a controlled fashion which generates a signal for chemotatic migration of fibroblast cells. The number of cells migrated close to each bead has been assessed. On-chip cell migration assay showed a remarkable result proving the high efficiency and reliable accuracy in quantitative analysis. Therefore, the device could be extensively used in cell migration assay and other various studies related to cellular movements.
House dust mite (HDM) is important in the pathogenesis of allergic diseases including asthma and atopic dermatitis. Dermatophagoides pteronissinus (Dp) is one of major HDM allergens. In this study, we investigated that Dp extract (DpE) affects on the chemotactic activity of monocytes isolated from the peripheral blood. DpE inhibited the migration of human monocytes in response to CC chemokines such as MIP-$1{\alpha}$, RANTES, HCC-4, MCP-1, and TARC. DpE did not alter the expression of CC chemokine receptors (CCRs) such as CCR1, CCR2, CCR3, CCR4, and CCR5. These results indicate that DpE blocks the chemotaxis of human monocytes and its mechanism is not involved in alteration of CCR expression. Better understanding of the effect of DpE on monocytes will enable elucidation of the role of Dp in the development of allergic diseases.
Induction of antibody production in C3H/He mice by bacterial infection is regulated through the processing exerted by antigen presenting cells. From the studies with Psudomonas aeruginosa, Salmonella typhimurium, and Micrococcus luteu, lipopolysaccharides (LPS) in Gram negative bacteria, which are known to be T-cell independent B cell mitogen, seem to be the major factor stimulating immune responses via activation of macrophages. Activation of macrophage, however, does not seem to correlate with antibody production. M. luteus was easily eliminatd by activated macrophages, while the processed antigens were immediately releasedd into culture medium before presentation. Nevertheless, antigens from Gram positive bacteria, Staphylococcus aureus and Bacillus subtilis, were very very active in chemotaxis and activation of periotoneal macrophages as well as in antien presnetation, while the very nature of the antigens is not yet clearly understood.
고양이 말초혈액 다형핵백혈구(PMNC)의 유주성에 있어서 인삼 saponins(ginseng total ssponin(GTS), ginseng PT saponin(GfT) 및 ginseng PD saponin(GPD))의 면역증강 효과를 검토하였다. PMNC에 대한 유주성을 hoyden chamber 변법으로 측정한 결과, GTS, GPT, GPD는 PMNC에 대해 직접적인 유주활성을 나타내지 않았다. 또한 인삼 saponins을 첨가하여 배양한 PMNC 배양상충액의 경우게는 미약한 유주활성을 나타내었다. 그러나 GPf및 GPD를 첨가하여 배양한 말초혈액 단핵구세포(MNC)배양상충액에서는 PMNC의 유주활성이 현저하게 증가하였다. PMNC에 대해 유주활성이 인정된 GPI)로 배양한 MNC 배양상충액을 이용하여 checkerboard assay를 실시한 결과 배양상충액의 농도차에 의존하는 진의 유주활성임을 알 수 있었다. 이상의 결과로부터 인삼 saponine 중 GPT 및 GPD가 고양 이 말초혈액 다형핵백혈구의 유주성을 중강시키는 효과가 있으며, 이것은 이들 saponins에 의해 활성화된 단핵구세포에서 분비되는 유주성인자에 의해 다형핵백혈구의 유주활성이 증 강되는 것으로 사료되었다.
Blood monocytes are the precursors for the lipid-laden foam cells of early atherosclerotic lesions. Monocyte chemoattractant protein-1 (MCP-1), a CC chemokine. and chemokine receptor 2 (CCR2) playa crucial role in the recruitment of monocytes to the vascular lesion. Using the human monocyte THP-1 cell line. we investigated the inhibitory effects of methanol extracts of 127 medicinal herbs on MCP-1-induced chemotaxis. (omitted)
Objectives: The present study was designated to evaluate the direct effects of Fructus Liquidambaris on suppression of eosinophil activity and on suppression of chemokines such as eotaxin, IL-8, ICAM-1, and VCAM-1, in vitro. Methods: A549 cells were stimulated with TNF-$\alpha$ (100 ng/ml), IL-4 (100 ng/ml) or IL-$1{\beta}$(10 ng/ml) to induce chemokines and adhesion molecules involved in eosinophil chemotaxis. Then after treatment of Fructus Liquidambaris, inhibition effect assay such as ELISA, real-time RT-PCR, and chemotaxis assay was performed. Results: Eotaxin level was suppressed in both protein secretion and mRNA expression. Eosinophil recruitment to lung epithelial cells was also reduced by Fructus Liquidambaris, implying the role of eotaxin in eosinophil recruitment. In addition, expression of IL-8 was also suppressed by Fructus Liquidambaris (p<0.05). However, expression of adhesion molecules (ICAM-1, VCAM-1) related to eosinophil was not affected. The eosinophil migration was inhibited at high concentration of Fructus Liquidambaris (p<0.05). Conclusion: These results suggest that Fructus Liquidambaris may regulate a common signaling pathway of eotaxin and IL-8. FS might be of therapeutic value in diseases such as asthma.

Acne vulgaris, the most common skin disease. can be formed as only a few comedons or severe inflammatory lesions. The pathogenesis of acne involves various factors; excessive androgen, excessive sebum production, abnormal alteration of follicular epithelium, proliferation of Propionibacterium acnes, and inflammation. We investigated acne therapy using oriental herbs described in the Korean traditional medical book(Dong-ui-bo-gam). Oriental herbs(Angelica daurica, Arctium lappa, Coptidis rhizoma, and Glycyrrhiza glabra) were chosen based on their respective property of sebum control, anti-inflammatory activity, and anti-bacterial activity. We examined the effect of acne treatment, in terms of chemotactic inhibition, lipogenesis inhibition, and anti-bacterial activity for P. acnes. 1. Neutrophil chemotaxis assay; P. acnes secrete chemotactic factors and other pro-inflammatory extracellular products. Neutrophil chemotactic activity of P. acnes was measured by 48-well chemotaxis method. Angelica daurica clearly suppressed chemotactic activity of P. acnes. 2. Using sebaceous gland of hamster ear lipogenesis assay; Sebaceous lipogenesis was measured using ear biopsies by incubation or $C^{14}$-acetate in culture media. The $C^{14}$-labeled lipids were extracted and determined by liquid scintilation counting. Coptidis rhizoma markedly inhibited sebum production. 3. Anti-bacterial assay for P. acnes(MIC test); Glycyrrhiza glabra showed anti-bacterial activity. P. acnes did not develop resistance against Glycyrrhiza glabra. Retinoids are effectively to inhibit sebum production and regulate follicular keratinization process, with little anti-inflammatory activity. Angelica daurica suppressed neutrophil chemotaxis, Coptidis rhizoma inhibited sebum production, and Glycyrrhiza glabra showed anti-bacterial activity against P. acnes. A combined formulation of Angelica daurica, Coptidis rhizoma. and Glycyrrhiza glabra is expected to provide effective acne treatment.
