Monitoring of Pesticides in the Yeongsan and Seomjin River Basin (영산강 및 섬진강 수계 중 농약 분포 조사)

  • BACKGROUND: A lasting release of low levels of persistence chemicals including pesticides and pharmaceuticals into river has a bad influence on aquatic ecosystems and humans. The present study monitored pesticide residues in the Yeongsan and Seomjin river basins and their tributaries as a fundamental study for water quality standard of pesticides.METHODS AND RESULTS: Nine pesticides(aldicarb, carbaryl, carbofuran, chlorpyrifos, 2,4-D, MCPA, methomyl, metolachlor, and molinate) were determined from water samples using SPE-Oasis HLB(pH 2) and LC/MS/MS. Validation of the method was conducted through matrix-matched internal calibration curve, method detection limit(MDL), limit of quantification(LOQ), accuracy, precision, and recovery. MDLs of all pesticides satisfied the GV/10 values. Linearity(r2) was 0.9965- 0.9999, and a percentage of accuracy, precision, and recovery was 89.4-113.6%, 3.1-14.0%, and 90.8-106.2%, respectively. All pesticides exclusive of aldicarb were determined in the river samples, and there was a connection between the positive monitoring results and agricultural use of the pesticides.CONCLUSION: Monitoring outcomes of the present study implied that pesticides were a possible non-point pollutant source in the Yeongsan and Seomjin river basins and tributaries. Therefore, it is required to produce and accumulate more monitoring results on pesticides in river waters to set water quality standards, finally to preserve aquatic ecosystems.

Comparing Farming Methods in Pollutant runoff loads from Paddy Fields using the CREAMS-PADDY Model (영농방법에 따른 논에서의 배출부하량 모의)

  • BACKGROUND: For Non-Point Source(NPS) loads reduction, pollutant loads need to be quantified for major farming methods. The objective of this study was to evaluate impacts of farming methods on NPS pollutant loads from a paddy rice field during the growing season. METHODS AND RESULTS: The height of drainage outlet, amount of fertilizer, irrigation water quality were considered as farming factors for scenarios development. The control was derived from conventional farming methods and four different scenarios were developed based combination of farming factors. A field scale model, CREAMS-PADDY(Chemicals, Runoff, and Erosion from Agricultural Management Systems for PADDY), was used to calculate pollutant nutrient loads. The data collected from an experimental plot located downstream of the Idong reservoir were used for model calibration and validation. The simulation results agreed well with observed values during the calibration and validation periods. The calibrated model was used to evaluate farming scenarios in terms of NPS loads. Pollutant loads for T-N, T-P were reduced by 5~62%, 8~37% with increasing the height of drainage outlet from 100 mm of 100 mm, respectively. When amount of fertilizer was changed from standard to conventional, T-N, T-P pollutant loads were reduced by 0~22%, 0~24%. Irrigation water quality below water criteria IV of reservoir increased T-N of 9~65%, T-P of 9~47% in comparison with conventional. CONCLUSION(S): The results indicated that applying increased the height of drainage after midsummer drainage, standard fertilization level during non-rainy seasons, irrigation water quality below water criteria IV of reservoir were effective farming methods to reduce NPS pollutant loads from paddy in Korea.

Characteristics of Pesticide Runoff and Persistence on Agricultural Watersheds in Korea (영농지역에서 작물재배 형태에 따른 농약의 잔류성과 유출특성)

  • To evaluate the exposure of non-point source pesticide pollution in agricultural watershed and to investigate pesticide distribution and runoff from agricultural land, paddy field, upland and orchard, this experiment was carry out during crop growing seasons. The pesticide were detected twenty pesticides fungicide 4, insecticide 10, herbicide 6) in water of Neungchon agricultural watershed and detection concentrations were range 0.008${\sim}$7.59 ppb. Most of the detection pesticides were using pesticides to rice paddy fields to control fungi, insects, weeds. During the crop cultivation, the pesticide were detected total thirty pesticides by pepper field soil 6, orchard soil 4, sesame field soil 3 and rice paddy field soil 5, and pesticide concentrations were range 0.001${\sim}$0.109 ppm. Especially the herbicides were detected mainly in May and June in the stream water. The pesticide were detected thirty pesticides by fungicide 2, insecticide 6, herbicide 5 in water of Jungam Koseong agricultural watershed and detection concentrations were range 0.01${\sim}$7.21 ppb. In regard to the detected pesticides, the concentration of individual pesticides measured in surface water of the study areas never exceeded guidelines for agriculture chemicals concerning water quality-effluent from paddy fields in Japan (Katayama, 2003). Runoff rate of pesticides was range 0.07${\sim}$3.06 % from Kongju agricultural land to watershed after applied pesticides.

Comparison of Movement Distance and Home Range Size of Gold-spotted Pond Frog (Pelophylax chosenicus) between Rice Paddy and Ecological park - Focus on the Planning Alternative Habitat - (논과 생태공원에서 금개구리 이동 거리 및 서식영역 크기 비교 - 대체서식지 조성 중심으로 -)

  • The movement distance and home range size of Pelophylax chosenicus were identified in the rice paddy and ecological park as alternative habitats from July to November 2017. A total of 39 frogs were tracked by radio tracking method. As a result, the average move distance in the population of rice paddy was 11.7 ± 1.9 m (n = 64) and the population of ecological park was 24.7 ± 4.3 m (n = 39). The move distance between the two populations was significantly different. The mean MCP of the population of rice paddy was 181.2 ± 110.8 m2 (n = 11) and the population of ecological park was 416.1 ± 276.2 m2 (n = 10), but there was no significant difference. The population area of rice paddy was 4,160 m2 (Kernel density 95%) and the core area was 1,080 m2 (Kernel density 50%). The population area (Kernel density 95%) of ecological park was 5,391 m2 and the core area (Kernel density 50%) was 736 m2. This study shows that it is appropriate to construct the area of alternative habitat for P. chosenicus at least 1.33 ha, and it is more advantageous for the ecological park to be constructed than the paddy field with high development pressure and human interference. If the rice paddies were to be abandoned for several years, or to be used traditional farming methods, such as refraining from using agricultural machinery and chemicals, they could be used as alternative habitat for P. chosenicus.

Evaluation of Compost Qualities with or without Microbial Inoculation for Food Waste Composting (미생물제 첨가유무에 따른 음식물 쓰레기 퇴비 부숙도 평가)

  • This studies were conducted to evaluate efficiency of microbial inoculator for active composting of food wastes. The Microbial inoculators used in this studies were purchased from different comparise to evaluate their effectiveness for composting of food waste in Korea. The number of bacteria growing at $30^{\circ}C$ in commercial inoculator collected were below $91.0{\times}10^8\;CFU/g$ which were counted from well cured compost made by animal manure. The number of bacteria in commercial microbial inoculator, such as FL, VP, B9, CM and GE were higher than that of composted at $50^{\circ}C$ or $60^{\circ}C$ of incubation temperature. Fungi were counted in GR, VP and B9 as over $10^3CFU/g$ at $30^{\circ}C$ of incubation temperature, while fungi of all the commercial inoculator collected could not grown at $50^{\circ}C$ and $60^{\circ}C$. Actinomycetes in most of the these had higher number($10^5CFU/g$) than that of compost : however, it was not detected at $60^{\circ}C$ incubation temperature from all the samples collected. The amount of carbon dioxid production was order to VP>HU>B9>GE>CM>Control>Compost in the lab scale composting test with or without inoculation of commercial inoculators, however, but the difference in carbon dioxide production was similar among each treatments. The effect of inoculation on composting parmeter such as pH changes, temperature increasing and change of chemicals properties were a little among each treatments, with or without inoculation of commercial inoculator in active composting of food waste. Using commercial inoculator did not show any statistical difference in food waste composting process under various condition such as pH changes, temperature changes, etc.

  • The growth of Lactobacillus bulgaricus treated with vanillin, ortho-vanillin and guaiaco1 was studied on synthetic medium in mechanically agitated chemostat culture, The exponential-phase growth rate exhibited a maximum at the cells treated with 50 ppm vanillin. That stimulation, however, appears to be an effect on growth rate rather than total cell growth. And the others were inhibited by the chemicals. Much greater inhibition in growth of the cells treated with 100 ppm of each chemical than oars treated with 50 ppm was observed after 25 hour fomentation. For aerobic microbes, the alcohol dehydrogenase reaction is enhanced for the reproduction of NAD, which consequently cause to stimulate fermentation. For micro-aerophilic microbes , however, the same effect was not observed at the present study at least in the case of cell concentration. However except f or one treated with 50 ppm vanillin the same effect was observed in the case of growth is to. From the result using the glucose as a substrate, it was found that the cell concentrations measured in terms of ultimate optical density (UOB/ml), were 0.96 and 0.92, when treated with 50 and 100 ppm vanillin; 0.40 and 0.45 when treated with ortho-vanillin 50 and 100 ppm: 0.49 and 0.47, when treated with guaiacol 50 and 100 ppm. The specific growth rates were 0.44, 0.15, 0.25, 0.29, 0.37, and 0.34; the specific production rates wire 0.33, 0.15, 0.16, 0.22, 0.28, and 0.26 and the glucose concentrations (g/1) after 25 hour fermentation were 23.5, 32.8, 31.5, 29.5, 28.0 and 28.8, these all in the same sequences as the first.

Blue-green algae as a Potential agent Causing Turf Leaf Disease (잔디 엽병을 유발하는 잠재인자로서의 남조류(Blue-Green algae)에 대한 관찰보고)

  • Recently irregular dark-colored patches were found on the Kentucky teeing ground in a golf course in Gyunggi providence. Interestingly, blue-green algae from the leaf tissue sample containing black spot-stained symptoms were largely observed through microscopic study. In general, algae present on the upper soil surface or in the upper layer of root zone form dark brown layers of scum or crust, which invoked harmful effects to turf growth such as poor drainage, inhibition of new root development. In this observation, unlike the algae were sometime found in senescing leaves on contacted soil in July and August, the blue-green algae were detected within black spot-stained Kentucky bluegrass leaf tissues including leaf blade, ligule, auriclea as well as leaf sheath. The blue-green algae were also detected on the leaf and stem tissue adjacent to the symptomatic leaf tissues. Two species of blue-green algae, Phomidium and Oscillatoria, were greatly observed. Oscillatoria species was more commonly notified in all samples. In addition, the two species were found on a putting green showing yellow spot disease at another golf course in Gyunggi providence. The data from chemical control assay revealed that chemicals such as propiconazole, iprodione, and azoxystrobin decreased blue-green algae population and leaf spots, which finally resulted in enhanced leaf quality. All taken together, we strongly suggested that the disease-like phenomenon by blue-green algae might be very closely mediated with infection/translocation process in relation with turfgrass. It indicates that blue-green algae in turf management may play an adverse role as a secondary barrier as well as a pathogenic agent. This report may be helpful for superintendents to recognize and understand the fact that algae control should be provided more cautiously and seriously than we did previously in upcoming golf course management.

Effects of Bisphenol A on the Hatching of fertilized Eggs and Spawning of Adult Fish in Songsari, Oryzias latipes (Bisphenol A가 송사리, Oryzias latipes의 수정란 부화와 어미의 산란에 미치는 영향)

  • The effects of bisphenol A (BPA), known as one of the endocrine disrupting chemicals (EDCs), on the hatching of fertilized eggs (FE) and the spawning of adult fish in songsari, Oryzias latipes were investigated. In condition of the experimental groups, incubated water temperature was $25.0{\pm}0.5^{\circ}C$ and photoperiod was 16 h (light) : 8 h (dark). fE were exposed to aqueous solutions of BPA at nominal concentration of 50, 100 and $200 {\mu}g/l$. The time required in hatching of FE was long in the BPA $50 and 100 {\mu}g/l$ treatment groups when compared to the controls, and in the BPA $200 {\mu}g/l$ treatment group, FE failed to hatch. And also hatching rate was decreased in the BPA treatment groups in comparison to the controls, Adult fishes were reared to oral administration via a diet of 50, 100 and $200 {\mu}g/g$ body weight BPA concentration for 3 weeks, Number of total eggs spawned in the adult fish were fewer in the BPA 100 and 200 treatment groups than in the controls and BPA 50 treatment group. Frequency of abnormal eggs in the total eggs spawned was 11.8 and $16.2{\%}$ in the control and acetone carrier control lower than 36.8, 46,8 and $74.1{\%}$ in the BPA 50, 100 and 200 treatment groups, respectively. And hatching rate of FE decreased in the BPA treatment groups in comparison to the controls. In these results, inhibition of the hatching of FE and the quantity and quality of spawned eggs in adult fish were observed from BPA treatment groups when compared to the controls.

Disinfection of Fusarium-infected Rice Seeds by Prochloraz and Gaseous Chlorine Dioxide

  • Three species of Fusarium, F. fujikuroi, F. verticillioides and F. proliferatum, are known to be associated with bakanae disease of rice [1, 2]. F. fujikuroi infects rice flowers and survive in endosperm and embryo of the seeds. Infected seed is an important source of primary inoculum of pathogens [3]. Seeds of rice (Oryza sativa cv. Boramchan) collected from bakanae-infected field were found to be 96% infected with Fusarium sp., 52% with F. fujikuroi, 42% with F. verticillioides, and 12% with F. proliferatum as determined by incubation method and species-specific PCR assays. F. fujikuroi was detected at lemma/palea, endosperm and embryo whereas F. verticillioides and F. proliferatum were recovered only from lemma/palea by means of component plating test. Seed disinfection methods have been developed to control bakanae disease and prochloraz has been most widely used for rice seeds. Two chemicals formulated with prochloraz (PC 1) and prochloraz + hexaconazole (PC 2) that inhibit biosynthesis of ergosterol strongly reduced the incidence of Fusarium spp. on selective media to 4.7% and 2.0%, respectively. Disease symptoms of rice seedlings in nursery soil were alleviated by chemical treatment; seedlings with elongated leaves or wide angle between leaf and stem were strikingly reduced from 15.6 to 3.2% (PC 1) and 0 (PC 2), stem rots were reduced from 56.9 to 26.2% (PC 1) and 32.1% (PC 2), and normal seedling increased from 0.4 to 13.3% (PC 2). Prochloraz has some disadvantages and risks such as the occurrence of tolerant pathogens [4] and effects on the sterol synthesis in animals and humans [5]. For these reasons, it is necessary to develop new disinfection method that do not induce fungal tolerance and are safe to humans and animals. Chlorine dioxide ($ClO_2$), that is less toxic, produces no harmful byproducts, and has high oxidizing power, has been reported to be effective at disinfection of several phytopathogenic fungi including Colletotrichum spp. and Alternaria spp. [6]. Gaseous $ClO_2$ applied to rice seeds at a concentration of 20 ppm strongly suppressed mycelial growth of Fusarium fujikuroi, F. verticillioides and F. proliferatum. The incidence of Fusarium spp. in dry seed with 8.7% seed moisture content (SMC) tended to decrease as the concentration of $ClO_2$ increased from 20 to 40 ppm. Applying 40 ppm $ClO_2$ at 90% relative humidity, incidence was reduced to 5.3% and resulted in significant reduction of disease symptoms on MS media. In nursery soil, stem rot was reduced from 56.9 to 15.4% and the number of normal seedlings increased from 0.4 to 25.5%. With water-soaked seeds (33.1% SMC) holding moisture in the endosperm and embryo, the effectiveness of disinfection using $ClO_2$ increased, even when treated with only 20 ppm for four hours. This suggests that moisture was a key element for action of $ClO_2$. Removal of the palea and lemma from seeds significantly decreased the incidence of Fusarium spp. to 3.0%. Seed germination appeared to decrease slightly by water-soaking at $30^{\circ}C$ because of increased SMC and by physical damage of embryos from hulling. These results indicate that the use of gaseous $ClO_2$ was effective as a means to disinfect rice seeds infected with Fusarium spp. and that moisture around the pathogens in the seed was an important factor for the action of $ClO_2$. Further investigations should be conducted to ascertain the best conditions for complete disinfection of Fusarium spp. that infect deep site of rice seeds.

Hepatoprotective and Anticancer Activities of Allomyrina dichotoma Larvae (장수풍뎅이 유충의 간보호 효능 및 항암활성)

  • Beetle larvae have been used as a traditional medicine to treat various human liver diseases. To prove the liver protective function of Allomyrina dichotoma larvae (ADL), we induced liver damage by the intraperitoneal injection of a hepatotoxic reagent, diethylnitrosamine (DEN), to C3H/HeN male mice and orally administered freeze-dried ADL powder. ADL powder lessened DEN-induced hepatotoxicity considering the reduced signs of acute and chronic hepatotoxicities, such as the ALP level in the blood serum, TUNEL-positive hepatocytes, ductural reactions, steatotic hepatocytes, and collagen deposition of the Masson’s trichrome staining. In addition to hepatoprotection, the anti-cancer activity of ADL has been examined. The ADL powder was extracted with ethanol and then fractionated with hexane, ethyl acetate, and water by a solvent partition technique. The ethyl acetate fraction showed cytotoxicity to various cancer cells through induction of apoptosis and necrosis, as well as the perturbed metabolism of the cancer cell to trigger autophagy. Collectively, ADL contains bioactive substances that can protect hepatocytes from toxic chemicals and trigger cell death in cancer cells. Thus, further purification and analyses of ADL fractions could lead to the identification of novel bioactive compounds.
