화학공장에서의 정량적 위험성 평가를 이용한 비상 대응 계획 시스템 개발 (Development of Emergency Response Plan System Using Quantitative Risk Assessment in Chemical Plants)

  • In this study, we analyzed the accident scenarios of chemical plants through the analysis of lots of chemical accidents and using experts knowledge, and looked into the method of prevention and response. Moreover, we developed a systemic and actual Emergency Response Plan Software(ERPS) that could prevent, prepare and respond totally for the chemical industry facilities using the data from the accident effect estimation from the local society and the geographic information of a chemical plant. The ERPS consists of the information for the plant and process, the consequence analysis and the ERPTA(Emergency Response Plan Tree Analysis). In conclusion, the program developed in this study could help effectively all the chemical industry facilities to prevent and respond to possible accidents.

Effect of Stage of Growth and Cultivar on Chemical Composition of Whole Maize Plant and Its Morphological Fractions

  • Samples of whole plant, leaf and stem of Akbar, Neelum, UM-81 and lZ-31 cultivars of maize fodder harvested up to 14 weeks at different growth stages were drawn and analysed for dry matter contents and various cell wall constituents such as NDF, ADF, hemicellulose, cellulose, lignin, cutin and silica. The dry matter contents of whole maize plant, leaf and stem increased significantly (p<0.01) with advancing plant age. Maximum dry matter was found in the leaf fraction of the plant. The cell wall components continued to increase significantly (p<0.001) in whole maize plant and its morphological fractions as the age advanced. Maximum values for NDF, ADF, cellulose and lignin were observed in stem followed by whole plant and leaf, whereas hemicellulose, cutin and silica contents were higher in leaf fraction of the plant. The cultivars were observed to have some effects on chemical composition of all plant fraction. The results indicated that maturity had a much greater effect on the concentration of all the structural components than did the cultivars. It was concluded that maize fodder should be cut preferably between 8th to 9th week of age (flowering stage) to obtain more nutritious and digestible feed for livestock. Among the maize cultivars, Neelum proved to be the best, due to its higher dry matter contents and lower lignin concentration.

Biological Control of Oomycete Soilborne Diseases Caused by Phytophthora capsici, Phytophthora infestans, and Phytophthora nicotianae in Solanaceous Crops

  • Oomycete pathogens that belong to the genus Phytophthora cause devastating diseases in solanaceous crops such as pepper, potato, and tobacco, resulting in crop production losses worldwide. Although the application of fungicides efficiently controls these diseases, it has been shown to trigger negative side effects such as environmental pollution, phytotoxicity, and fungicide resistance in plant pathogens. Therefore, biological control of Phytophthora-induced diseases was proposed as an environmentally sound alternative to conventional chemical control. In this review, progress on biological control of the soilborne oomycete plant pathogens, Phytophthora capsici, Phytophthora infestans, and Phytophthora nicotianae, infecting pepper, potato, and tobacco is described. Bacterial (e.g., Acinetobacter, Bacillus, Chryseobacterium, Paenibacillus, Pseudomonas, and Streptomyces) and fungal (e.g., Trichoderma and arbuscular mycorrhizal fungi) agents, and yeasts (e.g., Aureobasidium, Curvibasidium, and Metschnikowia) have been reported as successful biocontrol agents of Phytophthora pathogens. These microorganisms antagonize Phytophthora spp. via antimicrobial compounds with inhibitory activities against mycelial growth, sporulation, and zoospore germination. They also trigger plant immunity-inducing systemic resistance via several pathways, resulting in enhanced defense responses in their hosts. Along with plant protection, some of the microorganisms promote plant growth, thereby enhancing their beneficial relations with host plants. Although the beneficial effects of the biocontrol microorganisms are acceptable, single applications of antagonistic microorganisms tend to lack consistent efficacy compared with chemical analogues. Therefore, strategies to improve the biocontrol performance of these prominent antagonists are also discussed in this review.

Nematicidal and Plant Growth-Promoting Activity of Enterobacter asburiae HK169: Genome Analysis Provides Insight into Its Biological Activities

  • In the course of screening for microbes with nematicidal activity, we found that Enterobacter asburiae HK169 displayed promising nematicidal activity against the root-knot nematode Meloidogyne incognita, along with plant growth-promoting properties. Soil drenching of a culture of HK169 reduced gall formation by 66% while also increasing root and shoot weights by 251% and 160%, respectively, compared with an untreated control. The cell-free culture filtrate of the HK169 culture killed all juveniles of M. incognita within 48 h. In addition, the nematicidal activity of the culture filtrate was dramatically reduced by a protease inhibitor, suggesting that proteolytic enzymes contribute to the nematicidal activity of HK169. In order to obtain genomic information about the HK169 isolate related to its nematicidal and plant growth-promoting activities, we sequenced and analyzed the whole genome of the HK169 isolate, and the resulting information provided evidence that the HK169 isolate has nematicidal and plant growth-promoting activities. Taken together, these observations enable the future application of E. asburiae HK169 as a biocontrol agent for nematode control and promote our understanding of the beneficial interactions between E. asburiae HK169 and plants.

Diversity and Functions of Endophytic Fungi Associated with Roots and Leaves of Stipa purpurea in an Alpine Steppe at Qinghai-Tibet Plateau

  • Stipa purpurea is a unique and dominant herbaceous plant species in the alpine steppe and meadows on the Qinghai-Tibet Plateau (QTP). In this work, we analyzed the composition and diversity of the culturable endophytic fungi in S. purpurea according to morphological and molecular identification. Then, we investigated the bioactivities of these fungi against plant pathogenic fungi and 1-aminocyclopropane-1-carboxylate deaminase (ACCD) deaminase activities. A total of 323 fungal isolates were first isolated from S. purpurea, and 33 fungal taxa were identified by internal transcribed spacer primers and grouped into Ascomycota. The diversity of endophytic fungi in S. purpurea was significantly higher in roots as compared to leaves. In addition, more than 40% of the endophytic fungi carried the gene encoding for the ACCD gene. The antibiosis assay demonstrated that 29, 35, 28, 37 and 34 isolates (43.9, 53.1, 42.4, 56.1, and 51.5%) were antagonistic to five plant pathogenic fungi, respectively. Our study provided the first assessment of the diversity of culture-depending endophytic fungi of S. purpurea, demonstrated the potential application of ACCD activity and antifungal activities with potential benefits to the host plant, and contributed to high biomass production and adaptation of S. purpurea to an adverse environment.

화학플랜트에서의 화학물질 누출사고에 대한 배상책임 위험도 산정 (Estimation of the Liability Risk for Release of Chemicals at Chemical Plant)

  • 본 연구는 화학 및 석유화학 플랜트 등의 장치산업에서 사업장 외부로 유해화학물질이 누출·확산되어 발생할 수 있는 배상책임 위험도에 대한 산정 방식을 개선하고자 하였다. 이를 위해 환경부 사고대비 물질(14종)에 대해 누출·확산 시뮬레이션, 화학물질 누출사고 사례 분석, 식물피해 영향 자료 분석 등을 통하여 화학물질 누출사고 배상책임 위험도와의 상관관계인자를 도출하였고, 도출된 결과를 바탕으로 화학물질 배상 책임 위험도 산정 방법을 수정·보완하였다. 14종의 화학물질의 Probit 값과 EURAM 배상책임 위험도의 상관계수는 -0.526로 나타났고, 수정된 화학물질 누출사고 배상책임 위험도와의 상관계수는 0.319로 상관성이 있는 것으로 분석되었다. 수정된 산정 방법론으로 97종에 대한 배상책임 위험도와 ERPG-2 값의 상관계수는 -0.494로 분석되었고, 이는 기존 배상책임 위험도와의 상관관계보다 약 19배 높은 상관관계를 보였고, 부식위험도 값과의 상관계수는 0.91로 분석되었다. 위험도의 증가와 감소에 영향을 미친 상관관계 인자의 표준화 회귀계수(β) 값은 Corrosion Index (0.713), ERPG-2 (0.400), NFPA_Health Index (0.068) 크기 순서로 도출되었다. 이러한 연구결과는 기존과 신규 화학물질의 합리적인 배상책임 위험도 산정이 가능하게 하고, 사업장에서 정량적인 배상책임 위험관리 지표로 활용하는데 도움이 되리라 판단된다.

두 가지 제초제에 대하여 저항성을 가지는 항생제 마커-프리 형질전환 감자 육성 (Development of Antibiotics Marker-free Potato Having Resistance Against Two Herbicides)

  • 본 연구에서는 제초제 저항성 bar 유전자 및 CP4-EPSPS 유전자를 포함하는 발현벡터로 형질전환되고 항생제 마커 유전자를 포함하지 않는 제초제 복합 저항성 감자 식물체를 육성하고자 실험하였다. Bar 유전자를 포함하는 pCAMBIA3300에 CaMV35S 프로모터에 의해 조절되는 CP4-EPSPS 유전자를 도입하여 식물체용 발현 운반체를 제작하고, 이를 Agrobacterium tumafaciens EHA105에 도입하였다. 태동밸리 잎 절편체를 Agrobacterium과 공동배양한 다음, phosphinothricin 0.5 mg/L이 첨가된 배지에서 선발하고 호르몬 무처리 MS발근시켜 형질전환체 (E3-6)를 얻었다. PCR, Southern 분석, 효소면역반응 분석 등을 통해 두 가지 유전자가 도입되었으며 이들이 정상적으로 발현됨이 확인되었다. E3-6 식물체는 glufosinate-ammonium의 어린 식물체 잎 도포처리, glyphosate 용액에 치상한 식물체 조직에서의 shikimate 축적 여부 조사를 통하여 조사한 결과, 두 제초제에 대해 저항성을 나타내었다. 또한 형질전환감자의 전식물체에 대해 glyphosate와 glufosinate-ammonium 각각의 용액 또는 이들의 혼합물을 처리한 후 제초활성 반응을 조사한 결과, E3-6 형질전환 감자는 두 제초제를 각각 단독으로 처리할 때나 혼합하여 동시 처리할 때에도 동일한 저항성이 나타남을 확인하였다.

Loss-of-function and Gain-of-function Rice Mutants from Gamma-Ray Mutagenesis

  • Gamma-ray irradiation is known to induce various mutations in plants caused by chromosome alterations. This study investigated disease responses of selected gamma-ray induced rice mutants generated from seven Japonica-type rice cultivars against three plant diseases. Among the tested 22 mutants, three gain-of-function mutants and six loss-of-function mutants against rice blast were obtained, as well as three loss-of-function mutants against bacterial leaf blight (BLB). Two of the loss-of-function mutants were susceptible to both rice blast and BLB. Gain-of-function mutation has not been frequently observed in rice plants, thus, the mutants can be used to identify loci of novel genes for the regulation of disease resistant response.
